The expression of microRNA (miRNA) is closely related to radio-chemosensitivity in glioma stem cells (GSCs).Moreover,the growth of glioma stem cells could be inhibited comprehensively by increasing radio-chemosensitivity and apoptosis,simultaneously with the regulation of a single miRNA,which has been confirmed by some researches.Thereby microRNA is prospective for the adoption as a specific agent in targeted therapy of glioma,so as to increase the radio-chemosensitivity in glioma stem cells.

3.The Up-Regulation of the Drug Sensitivity of Human Breast Cancer Cell Line MCF-7 by FasL Gene Transfer

Objective: To investigate whether FasL expression by exogenous FasL gene transfer had regulatory effect on drug sensitivity of tumor cells. Methods: Exogenous Fasl, gene transfer was mediated by lipofectAMINE. The FasL expression was detected bv Immunohistochemistry and RT-PCR. The growth inhibition of cytotoxic drugs on tumor cells was measured by MTT assay. Fas mRNA expression was analyzed bv Semi-quantitative RT-PCR. Results: The eukaryotic expression vector, PcDNA3-FasL and PcDNA3 were successfully transteeted into MCF-7 cells. The transfectant cells was named as MCF-7/FL or MCF-7/Vt respectively. FasL was expressed on MCF-7/FL but not on MCF-7/Vt or MCF-7. The cytolysis ot MCF-7/FL induced by adriamycin at concentration of 0.625 ~ 2.5 ?g/ml, cisplatin of 1 .25 ~ 10 ?g/ml, or methotrexate of 3 ~ 6 ?g/ml respectively were all significantly enhanced compared with the cytolysis of MCF-7. Blocking FasL by using F(ab' )2-anti APO-1 antibody markedly reduced this up-regulated cytolysis induced by these drugs The significant up-regulation of Fas mRNA on MCF-7/FL was observed by Semi-quantitative RT-PCR after these drugs treatment. Conclusion: These findings indicated that FasL expression by gene transfer had up-regulation effect on drug sensitivity of tumor cells, this effect was associated with enhanced Fas expression induced by cytotoxic drugs.
4.Characteristics of emotional memory impairment in untreated patients with early Parkinson' s disease

Objective To characterize emotional memory impairment in untreated patients with Parkinson' s disease (PD) in the early stages of the disease. Methods Emotional memory tasks using standardized sets of emotional pictures including positive, neutral and negative valence were tested in 33 untreated patients with PD in the early stages and 31 healthy controls matched with age, sex, and education.Results Compared with the healthy adults (13.4 ± 1.4), PD patients had significant loss of negative valence picture memorizing ( 8. 9 ± 1.0; t = - 14. 87, P < 0. 01 ).There was no significant difference between PD patients and normal controls in positive ( 11.8 ± 1.0 vs 12. 4 ± 2. 2 , t = - 0. 95 ) and neutral (7.9 ± 1.4 vs 8.2 ± 1.3, t = - 0. 89) valence picture memorizing ( both P > 0. 05 ). ConclusionThe emotional memory for negative valence pictures is impaired but the emotional memory for positive and neutral is relatively spared in early PD patients without treatment.
5.Mir-483 enhances the proliferation,invasion and migration of nasopharyngeal carcinoma cells

Objective To investigate the effects of mir?483 on the proliferation,invasion and migration of nasopharyngeal carcinoma cells . Methods RT?qPCR was used to detect mir?483 expression level in different nasopharyngeal carcinoma cells(CNE?1,CNE?2)and the alteration of mir?483 expression level in these cells after radiation. CNE?1,one of the NPC cells,was selected. Cationic liposomes transient transfection was used to construct cells which overexpressed mir?483 or negative control . MTT assay was conducted to test the cell proliferation ability. Wound scratch assay and transwell migration assay were used to detect the effects of mir?483 on NPC invasion and migration ability. Results In terms of radiation resistance ,the expression level of mir?483 was higher in CNE?1. Overexpressing mir?483 can enhance the proliferation ,invasion and migration ability of CNE?1. Conclusions Mir?483 enhances the proliferation,invasion and migration ability of nasopharyngeal carcinoma cells.
6.Effect of Levodopa on verbal and spatial working memory in elderly patients with Parkinson's disease

Objective To observe the effects of levodopa on verbal and spatial working memory in elderly patients with Parkinson's disease (PD).Methods The modified Smith working memory software were applied to study the verbal and spatial working memory in 32 PD patients before and 8 weeks after treatment with levodopa and benserazide hydrochloride tablets.The results of PD patients were compared with those in 32 normal controls matched for age,sex and education.Results The correct rates of phonological verbal and coordinate visuospatial working memory were significantly lower in the PD patients without treatment than in the normal controls (both P<0.01).The correct rates of semantic verbal and categorical visuospatial working memory had no significant differences between the PD patients without treatment and the normal controls (both P>0.05).The correct rates of phonological verbal working memory and coordinate visuospatial working memory were significantly higher in the PD patients after administration of levodopa and benserazide tablets than in PD patients without treatment [(88.6±6.5)% vs.(82.1 ± 6.6)%,(84.2±8.8)% vs.(75.2±8.9)%,respectively,both P<0.01],but there were no significant differences in the correct rates of semantic verbal working memory and categorical visuospatial working memory between the above two groups (both P>0.05).The correct rates of phonological verbal,semantic verbal,coordinate visuospatial and categorical visuospatial working memory were negatively correlated with the course of disease,and had no correlations with age and the degree of education.Conclusions Levodopa can improve the functions of phonological verbal working memory and the coordinate visuospatial working memory,but has no effect on the semantic verbal working memory and the categorical visuospatial working memory.It is postulated that dopamine replacement therapy may have double dissociable effects on the different working memories.
7.Role of large conductance calcium-activated potassium channels in vascular hyporesponsiveness in rats with obstructive jaundice

Objective To evaluate the role of large conductance calcium-activated potassium (BKCa) channels in vascular hyporesponsiveness in rats with obstructive jaundice.Methods Eighteen male Sprague-Dawley rats,weighing 180-200 g,were randomly divided into 3 groups (n =6 each) using a random number table:control group (group C),sham operation group (group S),and bile duct ligation group (group BDL).Obstructive jaundice was produced by common bile duct ligation.At 7 days after surgery,blood samples were collected for determination of the levels of serum total bilirubin (TBL),direct bilirubin (DBL),indirect bilirubin (IBL),alanine aminotransferase (ALT),and aspartate aminotransferase (AST).Thoracic aortic rings were prepared,and the endothelium was removed.The aortic rings were sequentially perfused with different concentrations of norepinephrine (NE) and sodium nitroprusside (SNP),and the maximum amplitude of contraction and dilatation of aortic rings was recorded.The aortic rings were then perfused with BKCa channel blocker Chtx with the final concentration of 10 7 mol/L,followed by perfusion with different concentrations of NE and SNP again,and the maximum amplitude of contraction and dilatation of aortic rings was recorded under each concentration.The percentage of maximum contraction and dilatation (maximum amplitude after Chtx administration÷maximum amplitude before Chtx administration× 100%) was calculated.Results Compared with C and S groups,the levels of TBL,DBL,IBL,ALT and AST in serum were significantly increased,the maximum amplitude of NE-induced contraction of aortic rings was decreased,and the percentage of the maximum NE-induced dilatation of aortic rings was increased,the maximum amplitude of SNP-induced contraction of aortic rings was increased,and the percentage of the maximum SNP-induced dilatation of aortic rings was decreased in group BDL.Conclusion Excessivc opening of BKCa channels may be involved in the mechanism of vascular hyporesponsiveness in rats with obstructive jaundice.
8.Radiation Injury in the Brain after Radiotherapy for Nasopharyngeal Carcinoma

Objective:To study the clinical features and prognostic factors of brain injury after radiothera-py for nasopharyngeal carcinoma(NPC).Methods:From January 1998 to June 2006,49 NPC patients with Dost-radiation brain injury in our hospital were analyzed retrospectively.Results:The incidence of post-radia-tion brain injury after single-pass radiotherapy and re-course radiotherapy was 2.31%and 9.64%.respectively ,(P<0.05).The median latency period was 50.5 months for single-pass radiotherapy and 25.5 months for re-course radiotherapy.Fourty-nine patients suffered from radiation injury in the brain.The lesions were locat-ed in the temporal lobe in 37 patients(75.5%),in the pens in 9 patients(18.4%)and in mixed position in 3 pa-tients(6.1%).The symptoms and signs of the patients could be alleviated by therapy, but the quality of life was not improved.Conclusion:Radiation brain injury in NPC patients after radiotherapy is related to field de-sign.The incidence of radiation brain injury in the temporal lobe is the highest.Compared with single-pass ra-diotherapy, re-course radiotherapy leads to higher incidence of brain injury and shorter latency period.
9.Regularity and predictors of weight loss during concurrent chemoradiotherapy for nasopharyngeal carcino- ma patients

Objective To investigate the regularity and predictors of weight loss during concurrent chemo-radiotherapy for nasopharyngeal carcinoma patients. Methods A retrospective study was carried out on 82 patients with nasopharyngeal carcinoma ,with average age of 45 years ,who received concurrent chemoradiotherapy at the Affiliated Cancer Hospital of Guangzhou Medical University. Paired t-test was used to analyze the weight loss pattern during chemoradiotherapy. The Spearman′s rank correlation analysis was used to investigate the correlation between high weight loss(HWL)and the clinical factors. Results The paired t-test showed that the weight of the patients decreased continuously every week during chemoradiotherapy(P < 0.05),and the value dropped most significantly during the 4th week. Among these 82 patients,32 were determined as HWL(weight loss > 10%). Spearman′s rank correlation analysis showed that HWL was associated with older age,female,higher T and TNM stage,3D-CRT technology,the higher dose of radiotherapy in nasopharynx and in neck lymph node,and decreases of leukopenia and hemoglobin over degree Ⅱ(P<0.05). Conlousions Weight may decrease continuously every week during chemoradiotherapy for nasopharyngeal carcinoma patients. Age,gender,T stage,TNM stage,radio-therapy dose,and leukopenia and hemoglobin decreases may be predictors for weight loss degree during concurrent chemoradiotherapy for nasopharyngeal carcinoma patients.
10.The influence of the single different radiation dose and time on the microscopic structure and ultrastructure of Balb/c mice.

OBJECTIVE:
To observe the influence of the single different radiation dose and time on the microscopic structure and ultrastructure of Balb/c Mice.
METHOD:
Sixteen four-week-old mice were randomly divided into four groups, control group and three experimental groups, then every experimental group was randomly divided into two groups which were respectively sacrificed on the third and seventh day after exposure to different doses of 8, 12, 16 Gy radiation, then paraffin embedding, tissue section, HE staining and scanning electron microscope, observation were performed after a cochlear specimen collected.
RESULT:
Inner hair cells and outer hair cells arranged in order, no lodging, disorder or missing in control group under scanning electron microscope. Slight lodgy or disorder occured in inner cells and outer hair cells were missing sometimes in exposed animals,which meat irrespective of irradiation dose.
CONCLUSION
Slight changes of ultrastructure of Balb/c Mice occured in the early period when given eaqual to or less than 16 Gy radiation.
