Chinese Journal of Biochemical Pharmaceutics 2014;37(4):81-84
Objective To study the different expressions of cancerous inhibitor of protein phosphatase 2A (CIP2A)in non-small cell lung cancer (NSCLC),investigate its clinical significance and potential prognostic value. Methods Immunohistochemical and real-time polymerase chain reaction (PCR)were used to detect the expressions of CIP2A mRNA and protein in two groups of sixty paired NSCLC specimens and adjacent non-cancer tissues. The relationship between its expression and clinical pathological data and prognostic factors were analysed. Results The expression of CIP2A mRNA in NSCLC specimens was higher than those in adjacent non-cancer tissues(P<0.01 );The expression of CIP2A protein in NSCLC tissues was significantly higher than that in the corresponding adjacent non-cancer tissues (P<0.01 ).The expression of CIP2A was closely correlated with the differentiation and lymph node metastasis of NSCLC tissues (P<0.05 ),while incorrelated with sex, age, pathematology type, tumor size and tumor node metastasis (TNM)stage. Conclusion CIP2A highly expresses in NSCLC,and the expression of CIP2A will increase with the differentiation and lymph node metastasis of NSCLC tissues. The expression of CIP2A and lymph node metastasis can be used as an independent risk factor for the prognosis of patients with NSCLC.

Objective To evaluate the effects of fructose-1,6-diphosphate (FDP) on the preservation and high-energy phosphate metabolism of hypothermic SD rat hearts. Methods Sixteen SD rats were randomly divided into 2 groups. Their hearts were harvested and preserved in UW solution at 4℃ for 12 hours. In the study group (n=8), FDP (5mmol/L) was added to the UW solution, whereas in the control group, FDP was not added. Langendorff perfusion was established with the isolated hearts, and cardiac functions were examined, as well as the activities of lactate dehydrogenase (LDH) and creatine kinase (CK) in the perfusion fluid collected from coronary. Then the high-energy phosphate concentration, ATP, ADP, and AMP contents in the myocardium were determined with HPLC. Results The high-energy phosphate concentration was remarkably higher in the study group than that in the control group (P

The patients undergoing gastroscope and enteroscopy examination were respectively divided into gastric intestinal metaplasia group (n = 397) and non gastric intestinal metaplasia group (n = 360),and the differences in size, number, location and dysplasia of colonic polyps between the two groups were compared. In GIM group, the polyps incidence after classification of polyp size and colonic dysplasia was significantly higher than that in non GIM group (P < 0. 05),while the number and location of colonic polyps were no significant difference between two groups. By the logistic regression analysis, more than 50 years old of age, male,HP infection, the diameter of polyps are larger than 2. 0 cm,or dysplasia showed high possibility to appear gastric intestinal metaplasia (OR > 1,P< 0. 05),the diameter of colonic polyps are 1. 0 ~ 2. 0 cm which showed high possibility to non intestinal metaplasia (OR < 1,P < 0. 05),and there was no significant difference between the two groups in the incidence of GIM in colonic polyps less than 0. 5 cm or 0. 5 ~ 1. 0 cm in diameter. Not only age, gender and HP infection were related to the occurrence of IM,but the risk of GIM increased in patients with colonic polyps' diameter more than 2. 0 cm and dysplasia. Those who found colonic polyps > 2. 0 cm in diameter and polyps with dysplasia were suggested to undergo gastroscopy in order to find GIM early and follow up to improve the detection rate of early gastric cancer.

Objective:
To investigate the trends in incidence and mortality of prostate cancer in Huangpu District, Shanghai Municipality from 2002 to 2019, so as to provide insights into the prevention and treatment of prostate cancer.
Methods:
The incidence and mortality of prostate cancer among men in Huangpu District from 2002 to 2019 were collected from the Shanghai Cancer Registry System. The crude incidence, crude mortality, truncated age-standardized incidence (aged 35 to 64 years) and cumulative incidence (aged 0 to 74 years) of prostate cancer were calculated. The Chinese Fifth National Population Census in 2000 and the Segi's world standard population in 1960 were used to calculate Chinese-standardized rate and world-standardized rate. The trends in incidence and mortality of prostate cancer were evaluated using annual percent change (APC) and average annual percent change (AAPC).
Results:
A total of 2 672 cases of prostate cancer were reported in Huangpu District from 2002 to 2019, and the crude incidence was 33.35/105, the Chinese-standardized incidence was 14.93/105 and the world-standardized incidence was 12.37/105 (AAPC=7.675%, 4.886% and 4.983%, all P<0.05). The incidence of prostate cancer among males at ages of 60 to <70 years and 70 to <80 years appeared increasing trends (AAPC=4.554% and 5.045%, both P<0.05). A total of 1 214 deaths of prostate cancer were reported, and the crude mortality was 15.15/105, the Chinese-standardized mortality was 6.01/105 and the world-standardized mortality was 4.61/105 (AAPC=5.500%, 2.057% and 1.784%, all P<0.05). The mortality of prostate cancer among males at ages of 80 years and above appeared an increasing trend (AAPC=4.220%, P<0.05).
Conclusions
The incidence and mortality of prostate cancer appeared increasing trends in Huangpu District from 2002 to 2019, and the incidence among males at ages of 60 years and above also increased. The screening for prostate cancer among males at ages of 60 years and above should be strengthened.

Objective To study the impacts of alcohol dependence on the anticonvulsant effect of diazepam. Meth?ods Kunming mice (n=36) were divided into 3 groups (n=12 in each group), Alcohol Dependence Group(A group), Diaze?pam Group(D group)and Normal Saline Group(N group). A group received an intraperitoneal injection with a 0.2 mL dose of 0.8%alcohol in NS (normal saline) , while both D and N group received an injection with a 0.2 mL dose of NS without alco?hol , twice a day. Mice’s autonomic activities were monitored every day. After 7 days, the electroconvulsive experiment was performed. Both A and D group were given a weight-based dose of 0.05 mL/10 g of 0.05%diazepam via intraperitoneal injec? tion, while N group was given a 0.05 mL/10 g dose of NS. Before administration and after 15, 30, 60 min of administration, the convulsion threshold of each group was measured. Results The count of autonomic activity of mice in A group was less than that of mice in D and N group during the 2nd day to 6th day(P<0.05). On the 1st and 7th day, the difference of the count of autonomic activity of mice between A group and the other two groups was not statistically significant(P>0.05). The convulsion threshold of mice in A group was higher than that of mice in D and N group before administration(P<0.05). Af?ter administration, the convulsion threshold of mice in N group didn’t show statistically significant difference from that of mice before administration(P>0.05). After 15 min of administration, the convulsion threshold of mice in D group was high?er than that of mice in A and N group(P<0.05), while the convulsion threshold of mice in A group was higher than that of mice in N group(P<0.05). After 30 min and 60 min of administration, both the convulsion thresholds of mice in A and D group were higher than that of mice in N group(P<0.05). However, at this point, the difference of the convulsion thresholds of mice between A and D group was not statistically significant(P>0.05). Conclusion Alcohol dependence has anticon?vulsant effect. Alcohol dependence weakens the anticonvulsant effect of diazepam.

OBJECTIVETo review kidney transplantation in the center and analyze the risk factors affecting long-term allograft survival.
METHODSThirty-two relative variables were analyzed with SAS statistical software. Using Log-rank method, we investigated influence of these variables on short-and long-term survival of grafts. Kaplan-Meier analysis was used to estimate the 1-, 3-, 5-, 10-years graft survival rates and half-life. Proportional hazards regression analysis (Cox model) was used to assess and rank the relative risk of potential variables.
RESULTSThe 1-, 3-, 5-, 10-years graft survival rates were 83%, 75%, 66% and 48%. After excluding the patients died with functioning grafts, the 1-, 3-, 5-, 10 years grafts survival rate increased to 89%, 82%, 75% and 69%, respectively. The mean half-life was 8.78 +/- 0.14 and 14.09 +/- 0.20 years, respectively. By Log-rank analysis, factors affecting short- and long-term graft survival were identified as: renal function, duration of graft function became normal, cold-ischemia time, presence of acute rejection, delayed graft function, immunosuppressive regimen, complication, infection, anti-rejection therapy. Cox model multivariate analysis showed that there were 18 factors affecting graft survival.
CONCLUSIONSNew immunosuppressive agents not only significantly increase short-term graft survival, but also have the better long-term outcome tendency. Making assurance to get high quality donor organ and minimizing the death with graft function may be the most feasible way to prolong graft survival at present.
