1.Clinical value of human papillomavirus genotyping testing in the diagnosis of cervical intraepithelial neoplasia

Objective To Investigate the value of HPV genotyping in diagnosis of cervical intraepithelial neoplasia(CIN).Methods From July 2012 to February 2013,200 women from Tianjin Medical University General Hospital and 244 women from Affiliated Hospital of the Chinese People's Armed Forces Logistics College were selected to be HPV genotyping test and thin liqid-based cytology test.Consequently,132 samples were performed colposcopy test and cervical biopsy.Results HPV prevalence was 26.4% (117/444) in this study.The infection of one type HPV was more common.The top 5 of HPV types were HPV16,58,33,18,and 52.The top 5 of the risk for CIN Ⅱ and above followed HPV16,33,39,52 and 18.There was no significant difference between age and HPV positive rate (x2 =0.948,P > 0.05).Multiple infection and cervical lesions rank correlation analysis(r =0.132,P >0.05).For CIN Ⅱ and above disease,cytology positive rate was 90% (44/49),and HPV positive rate was 96% (47/49) cytology combine HPV positive rate was 98% (48/49,x2 =0.063,P > 0.05).Conclusions HPV infection should increasing trends with age.Cytology test and HPV genotyping test had good consistency.The combination of them can improve the sensitivity for high-grade lesions.
2.Application of breathing pattern parameters to extraction of REM sleep information

Objective To extract breathing pattern parameters during sleep and get the varying law of NREM and REM sleep stages. Method A newly designed respiratory inductive plethysmography (RIP) and a polysomnography (PSG) are utilized to record whole-night-sleep data simultaneously. The breathing pattern parameters obtained by RIP are dealt with according to the results of sleep stages and sleep apnea by PSG. Then the rule found out and summarized from the experiment is applied to distinguish REM sleep. Conclusion RC/VT can be used as an effective parameter to differentiate NREM and REM sleep. Using this parameter, the results of RIP totally accord with the results of PSG.
3.Significance and expression of metdherin in gastric adenocarcinoma and its regulating mechanism of invasion and migration

Objective This study aimed at observing expression and clinical significance of metadherin in gastric adenocarcinoma and exploring the potentially regulating mechanism of metadherin in invation and migration of gastric cancer.Methods Expressions of metadherin and E-cadherin in primay lesion of gastric cancer were detected by immunohistochemistry and their correlation to clinicopathologic characteristics and prognosis were analyzed by Chi-square tests.Transwell assay and wound healing assay were applied for the ability of invasion and metastasis in gastric cancer cells.Then,the down-regulatied metadherin expression in MKN45 cells by RNA interference (siRNA) was carried out and furthermore,the regulation role of metadherin in epithelial-mesenchymal transition was analyzed also in invasion and migration of gastric cancer cells.Results The positive expression of metadherin was correlated to invading depth (P =0.029),lymph node metastasis (P =0.001),TNM stage (P =0.014) and inhibiting E-cadherin expression (P =0.001).The patients with positive metabherin shared poorer prognosis.Furthermore,the down-regulated metabherin in MKN45 cells would result in the increasing expression of E-cadherin,as well as decreasing expression of N-eadherin,Slug and Snail.At the same time,the abilities of invasion (P =0.027) and migration (P =0.008) of MKN45 cells was decreased.Conclusion metabherin induces EMT in metastasis of gastric adenocarcinoma via activating either Slug or Snail but not twist,which would result in the poorer prognosis.
4.Porhyromonas endodontalis liopolysaccharide regulates interleukin-23 expression in mouse osteoblasts

Objective:To investigate the effects of Porhyromonas endodontalis(P.e)liopolysaccharide(LPS)on the expression of IL-23mRNA and protein in mouse osteoblasts MC3T3-E1 and the role of phosphatidylinositol 3-Kinases (PI3K)signaling pathway in this process.Methods:MC3T3-E1 cells were treated with different concentrations of P.e LPS for different hours,or pretreated with LY294002,a special PI3K inhibitor.The IL-23 mRNA and protein expression levels were detected by real-time PCR and Western blot respectively.Results:The level of IL-23 mRNA increased in MC3T3-E1 cells with the increase of concentration and the treatment time of P.e LPS(P <0.05).The IL-23 protein expression was increased by P.e LPS in a time dependent manner(P <0.05).The mRNA and protein of IL-23 decreased(P <0.05)after pretreatment with LY294002.Conclusion:P.e LPS can induce the expression of IL-23 mRNA and protein in MC3T3-E1 cells,and the PI3K signaling pathway may play a part in this process.
5.The clinical characteristics of different treatment in children with esophageal foreign bodies

Objective:To summarize and analyze the clinical characteristics of different treatment in children with esophageal foreign bodies.Methods:This study collected 246 children with esophageal foreign bodies in our hospital from January 2016 to January 2020, which was divided into endoscopic group and operative group.The general and clinical data of children treated with different treatment were collected and statistical analyzed.Results:There were 222 children in endoscopic group and 24 children in operative group, respectively.The rate of surgery was 9.75%.There were no significant differences in gender and location of esophageal foreign bodies.However, the average age of operative group was(2.92±2.67) years, which significantly younger than that in endoscopic group(4.12±3.37)years( P=0.049). The residence time in operative group(median 29.10 h)was remarkable longer than that in operative group(median 11.80 h)( P<0.001). The proportion of sharpness(50.00%) and corrosive(45.83%) foreign bodies in operative group were more than those in endoscopic group[16.22% and 8.11%( P<0.001)]. Moreover, the occurrence rate of major complication in operative group was 83.33%, which was dramatically higher than that in endoscopic group(0.90%)( P<0.001). Conclusion:The younger and longer residence time of esophageal foreign bodies in children contribute to the rate of operative treatment.Additionally, the sharpness and corrosive foreign bodies increase the risk of surgery and serious complications.
6.Low Concentration Contrast Agent and Low Tube Voltage in Light and Moderate Weight’s Abdominal Contrast-enhanced CT Scan

PurposeTo investigate the feasibility of using low concentration contrast agent and low tube voltage in the light and moderate weight's abdominal contrast-enhanced CT scan, in order to find an optimal solution to reduce radiation dose and iodine intake.Materials and Methods Forty patients with light weight whose body mass indexes (BMI) were lower than 20 kg/m2 were randomly divided into group A1 (n=20) and group B1 (n=20). Meanwhile, another 40 patients with moderate weight whose BMI ranged from 20 kg/m2 to 25 kg/m2 were randomly divided into group A2 (n=20) and group B2 (n=20). Low concentration contrast agent and low tube voltage (Visipaque 270 mgI/ml, 100 kV) were used in both group A1 and group A2 in abdominal enhanced CT scan. While both group B1 and group B2 used conventional scan solution (Omnipaque 300 mgI/ml, 120 kV) in abdominal enhanced CT scan. Then the contrast noise ratio (CNR), the image quality score and the effective radiation dose (ED) were compared among the four groups.Results The CNR and image quality score at artery phase and portal phase were neither significantly different between group A1 and group B1, nor between group A2 and group B2 (t=-1.539-0.000,P>0.05). The CNR and image quality score of the liver at artery phase in group B1 were signiifcantly higher than those in group A2 and group B2 (P<0.05).Conclusion The solution of using low concentration contrast agent and low tube voltage in contrast enhanced scan can achieve the same high quality abdominal image with reduced iodine intake and radiation, compared with the application of conventional enhanced scan; BMI has rather great impact on image quality score at arterial phase and little impact on that at portal phase. So it is suggested that the protocol of liver contrast-enhanced CT scan may choose reduction of voltage at portal phase so as to reduce radiation.
7.Study on effect of Chuankezhi in assisted treatment of capillary bronchitis

Objective To observe the clinical curative effect of Chuankezhi Injection atomization inhalation in treating capillary bronchitis.Methods 93 ninety-three children cases of capillary bronchitis were in our hospital from January to December 2015were selected and randomly divided into the treatment group(50 cases) and control group(43 cases) according to the random number table method.The two groups were given the same routine treatment.The treatment group was simuhaneously given 1 mL of Chuankezhi Injection adding into 4 mL of 0.9% Sodium Chloride Injection,the mixture was inhaled by oxygen atomization at a speed of 4-5 L/s for 10-12 min,with 7 d as one treatment course.The clinical effect,clinical symptom disappearance situation and hospitalization time in the two groups were observed.Results The total effective rate in the treatment group was 98.0% (49/50),which was higher than 79.1% (34/43) in the control group;the disappearance time of cough,dyspnea and pulmonary wheezes and crackles and rales,and hospitalizatiorn time in the treatment group were (6.15 ± 1.50)d,(4.5 ± 1.90)d,(4.60 ± 1.70)d,(5.52 ±1.31)d and (6.55±2.30) d respectively,which were significantly better than (7.19 ± 1.85) d,(5.7 ± 2.10) d,(5.81 ± 1.82) d,(6.50 ± 1.83)d and (7.48 ± 2.51) d in the control group,the differences of various indexes between the two groups were statistically significant(P<0.05).Conclusion Chuankezhi Injection in the assisted treatment of capillary bronchitis has significantly effect and better clinical application value.
8.Detecting sleep apnea/hypopnea events with a wearable respiratory inductive plethysmograph system.

The wearable respiratory inductive plethysmograph(RIP) system is a non-intrusive respiratory monitoring system. Sleep monitoring was performed on 9 human subjects suspected of having sleep apnea hypersomnolence syndrome (SAHS) and 7 healthy volunteers using both the wearable RIP system and the conventional polysomnography(PSG), and the sensitivity and specificity of the wearable RIP system were analyzed by comparison with the PSG results. According to the characteristic of the wearable RIP system in detecting sleep apnea/hypopnea event, the diagnostic criteria of sleep apnea/hypopnea event were put forward. All subjects with SAHS diagnosed by the wearable RIP were confirmed by PSG,the sensitivity and specificity of the wearable RIP system for detecting sleep apnea/hypopnea events were 99.0% and 94.6% respectively. The wearable RIP system can be used reliably in detecting sleep apnea/hypopnea events. This system can be used at home for detecting the sleep apnea/hypopnea events non-intrusively.

9.Prognostic value analysis of TOP2A gene expression for bladder cancer

Objective To investigate the relationship between DNA topoisomerase Ⅱ α (TOP2A) gene expression and clinicopathological characteristics and its significance of prognostic evaluation for patients with bladder cancer.Methods Bladder cancer gene expression profile GSE13507 (n =165) and GSE31189 (n =52) were obtained.The expression profile and clinical information of patients with bladder cancer were retrospectively analyzed,and the survival analysis was made.Gene set enrichment analysis (GSEA) was conducted to explore the related pathways which were regulated by TOP2A.Results Compared with normal bladder tissues,TOP2A was upregnlated in bladder cancer tissues (5.823 ± 1.079 vs.4.820 ± 1.129),with a statistically significant difference (t =4.336,P < 0.001).The TOP2A gene expression in patients with bladder cancer was correlated with the age of patients (x2 =5.926,P =0.015),sex (x2 =6.046,P =0.014),T staging (x2 =19.484,P < 0.001),N staging (x2 =9.178,P =0.002),M staging (x2 =21.142,P < 0.001),tumor grade (x2 =47.005,P < 0.001),and progression (x2 =11.735,P =0.001),but it was not correlated with recurrence (x2 =0.808,P =0.369).Survival analysis showed that the specific survival rate in the 100 months of TOP2A gene high expression group and low expression group had a statistically significant difference (66.59% vs.87.95%,x2 =15.820,P < 0.001).The median overall survival time of TOP2A gene high expression group and low expression group were 51.77 months and 134.97 months respectively,with a statistically significant difference (x2 =11.280,P =0.008).The results of GSEA indicated that TOP2A could regulate gene sets related with several pathways like MYC-V1 signaling (P =0.035,FDR =0.132),MYC-V2 signaling (P =0.012,FDR =0.058),E2F signaling (P < 0.001,FDR =0.006) and G2M checkpoint (P =0.006,FDR =0.044).Conclusion The TOP2A gene expression is closely related with clinicopathological characteristics of patients with bladder cancer.TOP2A may function as a potential marker of prognosis for patients with bladder cancer.
10.Effects of different anesthesia methods on cardiac function and postoperative complications in elderly patients undergoing thoracotomy

Objective To investigate the effects of ultrasound-guided continuous thoracic paravertebral nerve block combined with general anesthesia on cardiac function and postoperative complications in elderly patients undergoing thoracotomy.Methods A total of 108 elderly patients who underwent thoracotomy in our hospital from January 2017 to December 2018 were randomly divided into the general anesthesia group(receiving general anesthesia)and the combination group (receiving ultrasound-guided continuous thoracic paravertebral nerve block combined with general anesthesia) (n=54,each group).The anesthesia effect was compared between the two groups.Results The excellence rate of anesthesia was higher in the combination group than in the general anesthesia group(90.7% vs.72.2%,x2 =4.267,P =0.039).Stroke volume variability(SVV),stroke volume(SV),heart rate(HR) and mean artery pressure(MAP)had no significant difference between the two groups at T0(pre-anesthesia)(P>0.05).At T1 (anesthesia),T2 (intraoperative tissue traction),and T3(postoperative suture),there were significant differences in SVV,SV,HR and MAP between the two groups(P<0.05).The incidence of postoperative complications was lower in the combination group than in the general anesthesia group(7.4% vs.22.2%,x2 =4.696,P=0.000).Conclusions Ultrasound-guided continuous thoracic paravertebral nerve block combined with general anesthesia has good anesthetic effects,minor influence on cardiac function and a low incidence of postoperative complications in elderly patients undergoing thoracotomy.
