2.Changes and Clinical Significance of Serum GM-CSF, IL-6 and TNF-α Levels in Patients with Rheumatoid Arthritis

Objective:To investigate the changes and clinical significances of serum levels of granulocyte macrophage colony-stimulating factor (GM-CSF),interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) levels in patients with rheumatoid arthritis (RA).Methods:126 cases of patients with RA in our hospital from June 2013 to December 2016 were selected as RA group and divided into two subgroups as active RA group (69 cases) and remission RA group (57 cases) according to the severity of the disease.55 healthy cases who receive physical check in our hospital at same time were selected as the control group.The serum GM-CSF,IL-6 TNF-α levels were detected by enzyme linked immunosorbent assay (ELISA) and compared between different groups,the correlation of serum GM-CSF,IL-6 TNF-α levels with DAS28 were analyzed.Results:The serum GM-CSF,IL-6 TNF-α levels of RA group were significantly higher than those of the remission RA group and control group (P<0.01),and the serum GM-CSF,IL-6 TNF-α levels of remission RA group were also significantly higher than those of the control group (P<0.01).Positive correlation between serum GM-CSF,IL-6 and TNF-α levels were found(P<.0.01).The level of DAS28 was significantly correlated with the levels of serum GM-CSF,IL-6 and TNF-α (r=0.473,0.584,0.675,all P<0.01).Conclusion:The levels of serum GM-CSF,IL-6 and TNF-α were up-regulated in patients with RA and positively correlated with the activity of RA,which might be of important clinical significance in the early diagnosis,disease evaluation and prognosis of RA.
3.Clinical value of thyroid-stimulating antibody measurement using CHO-hTSHR cell line

Objective To evaluate the clinical value of CHO-hTSHR cells in detecting thyroid stimulating antibody (TSAb). Methods The cAMP production and TSAb activity were measured and cal-culated by stimulating CHO-hTSHR cell line with IgGs of normal control group and Graves" disease ( GD )group. TSAb positive standard was set to more than the mean + 2SD of TSAb activities in control subjects.The positive percentage of TSAb activity in GD group was calculated. Results The cAMP production and TSAb activities of GD group were higher than those of normal group[( 353. 65±126. 34 ) pmol/L vs (237.21±77. 15)pmol/L, ( 149. 08±53. 26)% vs ( 100±32. 52)%, P <0. 05] . The value that higher than 165% was set to be positive for TSAb. The positive percentage of TSAb in GD group was 50% ( 14/28). Conclusion CHO-hTSHR cell line constructed by our group is suitable for detecting TSAb activity in the sera of patients with GD.
4.Mice with focal segmental glomerular sclerosis are induced by adriamycin and its mechanism

Objective To establish adriamycin-induced focal segmental glomerular sclerosis(FSGS) mice model,and observe the expressions of and relation between oxidative stress and p38 MAPK signal pathway in renal injury.Methods Eight-week-old male Balb/c mice were randomly divided into FSGS group (n=20) and control group (n=20).In FSGS group mice were intravenously injected with a single dose of adriamycin (0.01 rag/g),and mice in control group were received saline with the same dose.At day 3,7,14,22 and 32,urine protein-to-urine creatinine ratio (P/C) was detected.At day 22 and 32,serum creatinine,blood urea nitrogen,nitric oxide (NO) and reactive oxygen species (ROS) in blood and urine,and ROS in kidney tissues were detected;changes of pathological morphology in renal tissue were analyzed by HE stain;the expressions of NF-κB,CD36,IL-13,BAX and Bcl-2 mRNA were detected by real time quantitative PCR;the expressions of NF-κB,p-p38 and p-ERK1/2 protein were detected by Western blotting.Results Compared with that in control group,P/C was gradually increasing in FSGS group,and peaked at day 22 (P < 0.05).At day 22 and 32,mice had higher creatinine,serum creatinine,urea nitrogen,ROS and NO in FSGS group than those in control group (all P < 0.05).There were mild hyperplasia of mesangial cells and mesangial matrix,segment with moderate exacerbations,podocytes with significant proliferation,and the capillary loops of the stenosed in the glomerular in FSGS group at day 32.Compared with those in control group,the mRNA expression of NF-κB,BAX,IL-13 and CD36,and the protein expressions of NF-κB and p-p38 MAPK were gradually increased in FSGS group,all showed statistical differences at day 32 (all P< 0.05);the expression of p-ERK1/2 was increased at day 22 (P < 0.05) but was reduced at day 32 (P < 0.05).Conclusions Adriamycin has induced FSGS in mice successfully,which may through oxidative stress activating p38,up-regulating NF-κB,increasing the inflammatory cytokines and inducing apoptosis pathways.
5.Fingerprint of water-soluble composition of Fructus Gardeniae from different breed and production places

AIM:To set up the fingerprint chromatograms of chemical principles of Fructus Gardeniae in Jiangxi Province by HPLC. METHODS:The chromatographic seperation was performed on an Eclipse XDB-C_(18) column(4.6 mm?250 mm,5 ?m,Agilent) with a mobile phase consisting of acetonitrile-0.1% phosphoric acid,gradient eluent,at the flow rate of 1.0 mL/min.The UV detection was set at the maximum absorption wavelength at 238 nm for iridoid glycosides and at 440 nm for crocins. RESULTS:Based on the fingerprint chromatograms of water-soluble composition of 12 batches Fructus gardeniae in authentic regions of Jiangxi,we have pinpointed 11 common peaks. CONCLUSION:This method can provide a basis for controlling the quality of Fructus gardeniae and be a means of germplasm resources.
6.Clinical research on feeding effect of semi-demand feeding and abdominal massage in preterm infants

Objective To evaluate the feeding effect of semi-demand feeding and abdominal massage on the feeding process, feeding performances and clinical safety (including the time of returning to birth weight, feeding tolerance, apnea). Methods A total of 120 preterm infants that met inclusion criteria of the study were randomly assigned to three groups, and each group had 40 cases. Control group:received standard feeding care. Semi-demand feeding group: received 5-10 minutes of oral motor intervention every time before feeding(including oral stimulation and non nutritional sucking), then decided feeding methods according to its consciousness behavior state. Semi-demand feeding combined abdominal massage group: received both semi-demand feeding and abdominal massage before feeding. Observed and recorded the three groups′ transition time, whether there were statistically significant differences in feeding performance and clinical safety. Results The feeding transition time was (11.55± 4.69), (10.98±3.85), (8.50±3.87)d in control group, semi-demand feeding group and semi-demand feeding combined abdominal massage group, and there were significant differences (F=6.088, P=0.003). In terms of feeding performance, the feeding efficiency were (7.27 ± 2.65), (9.74 ± 2.63), (9.52 ± 2.13) ml/min, the feeding proficiency were (66.31 ± 14.51)%, (76.64 ± 12.46)%, (82.26 ± 10.65)%, the volume transfer were (63.99 ± 11.02)%, (80.16 ± 14.41)%, (84.97 ± 11.16)% in control group, semi-demand feeding group and semi-demand feeding combined abdominal massage group, and there were significant differences (F=12.189, 16.383, 31.959, all P < 0.01).Among the clinical safety, the time of restoration of birth weight were (10.55 ± 1.99), (9.28 ± 1.77), (10.05 ± 2.46) d in control group, semi-demand feeding group and semi-demand feeding combined abdominal massage group, and there were significant differences(F=3.759, P=0.026). The incidence of feeding intolerance were 50.0%(20/40), 40.0%(16/40), 22.5%(9/40), and there were significant differences(χ2=6.613, P=0.037). The incidence of apnea were 37.5%(15/40), 20.0%(8/40), 15.0%(6/40), and there were significant differences(χ2=6.093, P=0.048). Conclusions The combined intervention methods could accelerate the feeding process, improve feeding performance as well as reduce the incidence of feeding intolerance and apnea. Hence, semi-demand feeding combined abdominal massage is a recommended choice for clinical use.
8.Tuberous sclerosis complex secondary to fetal heart occupying lesions: a case report and literature review

Objective To explore the clinical characteristics of tuberous sclerosis complex (TSC). Methods The clinical data of one child with TSC were collected. The clinical features and gene mutation were analyzed. Results A 36-day-old girl had abnormal nodules found by echocardiography, which was considered multiple cardiac rhabdomyomas. There were multiple hypomelanotic macules distributed over the skin surface of the trunk and legs. Cranial MRI showed cortical nodules, subependymal nodules and cerebral white matter radial migration line. A mutation in the TSC2 gene (c.4541-4544delCAAA) was found by second generation high-throughput sequencing technology and tuberous sclerosis complex was confirmed. Conclusion Gene detection is helpful in the early diagnosis of tuberous sclerosis complex.
9.Clinical research of the effects of miRNA and hypersensitive C-reactive protein on non-targeted vessels in patients with acute coronary syndromes after percutaneous coronary intervention procedure

Objective To investigate the effects of miRNA and hypersensitive C?reactive protein ( hs?CRP ) on non?targeted vessels in patients with acute coronary syndromes ( ACS ) after percutaneous coronary intervention( PCI) procedure. Methods The serum samples were collected from 217 cases ACS patients to detect the level of miRNA?14?5P and hs?CRP during admission and follow?up periods.All patients underwentPCI with stent implantation and coronary angiography,and CAG was performed at the time of 12?month follow?up.According to CAG results,the patients were divided into non target lesion progression group(progressiongroup) with 76 cases and non target lesion no progression group(no progression group) with 141 cases.Results The expression level of hs?CRP was significantly higher in progression group than the no progression group((1.65±0.18) mg/L vs.(1.52±0.37) mg/L,t = 3.478,P<0.001).The expression level of miRNA?142?5Pwas higher in progression group than the no progression group(27.12±2.11 vs.34.73±2.67,t = 23.035,P<0.001).Multi?factor regression analysis indicated that high expression levels of hs?CR and miRNA?142?5Pduring admission were the predictors of advance of non?targeted vessels patients(OR = 3.496,95%CI 2.046-5.981,P =0.001;OR =1.208,95%CI 1.073-1.361,P =0.002).Conclusion The serum level of hs?CRP andmiRNA?142?5P can predict non?targeted vessels in patients with ACS after PCI.
10.EFFECTS OF GRAPE JUICE ON PROLIFERATION AND APOPTOSIS OF HUMAN PROSTATE CARCINOMA PC-3 CELLS

Objectives: To study the inhibitory effects of grape juice on human prostate carcinoma PC-3 cells and its possible mechanism. Method: PC-3 cells were treated with grape juice in different concentration(16, 32, 64 ?l/ml) and resveratrol for 48 h; and the proliferation of PC -3 cells were measured by growth curve and MTT assay. TUNEL was used to observe the morphology of apoptotic cells and flow cytometry to analyze the PC-3 apoptosis. Results: Resveratrol and grape juice could markedly inhibit the proliferation of PC-3 cells, and TUNEL positive cells were detectable. The percentages of apoptotic cells were 6.0%,18.5%,30.0%, 12.7% in experimental groups and resveratrol group respectively. The effect on PC-3 cells was enhanced with increasing amount of grape juice. The effect of low dose group was weaker than that of resveratrol group, but medium and high dose groups were stronger. Conclusion: The anti-prostate cancer effect of grape juice is not only related to resveratrol contained but also to synergism with other components, and its mechanism might be the induction of PC-3 cells apoptosis.
