Stroke is the disease which caused by many risk factors. The relationship between insulin resistance(IR) and young adult stroke is one of the focuses now in the neurological field.IR refered to the series of pathological and physiological changes,it happens when the response of the target tissue organ to insulin is depressing or losing. There are many ways of detection of IR,the insulin sensitivity index(ISI) is one of the most satisfied indexes to reflect the sensitivity to insulin.IR could cause a series of pathological and physiological changes, which caused hyperinsulinemia,hyperglycemia,hyperpiesia,blood-fat derangement, fibrinolytic system anomaly, blood vessel endothelium change, artherosclerosis,et al,maybe in ways of many metabolic risk factors.Atherosclerosis is the main etiopathogenisis factor in young adult stroke,as a conclusion,IR is also existed in young adult stroke. But,does IR exist in all types of young adult stroke? What is the mechanism? Is the level of IR related with the pathogenetic condition and prognosis? These questions are still not clear at present, and the study on the relationship between IR and young adult stroke is important to prevent young adult stroke.

Quantitative calibration strategy as an essential issue in laser ablation ICP-MS plays an important role for the guarantee of analytical accuracy. In this study, uncertainties of reference values in current available glass certified reference materials (NIST, MPI-DING and USGS) as well as the matrix effect among them were systematically evaluated. The results illustrated that NIST610 was better than other reference materials from aspect of reference value uncertainty. The matrix effect among NIST, MPI-DING and USGS reference materials was insignificant under our experimental conditions.A quantification strategy based on two reference materials (NIST610 and StHs6/80-G) and normalization to 100% (w/w) was proposed to avoid the insufficiency of single reference material strategy, which suffered the very low content and large uncertainty for some elements. The comparision of ML3B-G results obtained from three quantification strategies (single reference material NIST610, single reference material StHs6/80-G and two refterence materials) illustrated that the proposed strategy improved the analytical accuracy.Three reference materials (BCR-2G, CGSG-2 and KL-2G) were quantified with the proposed strategy, and almost all data matched well with reference value, meanwhile the data reported here could supplement the reference value database.

ObjectiveTo investigate the effect of immunoglobulin G (IgG) extravasated from blood circulation on the expression of toll-like receptor 4 (TLR4) induced by peripheral lipopolysaccharide (LPS) in rat brain. Methods The rats were divided into four groups in random, 5 rats in each. Group one received LPS 100μg/kg by intraperitoneal administration, normal saline was given by intravenous injection 6 hours later; group two was injected with adrenalin (AD) 15μg/kg intravenously; group three was treated with LPS intraperitoneally, AD was injected 6 hours later; group four was injected normal saline intravenously as control. For all groups, the animals were sacrificed 30 min after the last injection, and the brains were taken for investigation of the TLR4 expressions by immunofluorescence staining and RT-PCR. Result Immunofluorescence staining showed that IgG immunoreactive product was patch-like, distributed in the brain parenchyma in all the animals that received AD. In the LPS+normal saline group, IgG was found merely around the blood vessels. Meanwhile, in LPS+AD animals, TLR4 immunoreactive product coexisted with microglia marker Iba-1 within the IgG extravasated area. The double-labeled cells dispersed in the brain parenchyma and near to the cerebral vessels. In the LPS+saline group, TLR4 positive cells were endothelial-like. RT-PCR results indicated that the expression level of TLR4 in the LPS+AD group were significantly higher than that in the LPS+saline group or AD group or the saline control (P<0.01). Conclusion Extravasated circulating IgG may enhance the TLR4 expression in the rat brain induced by peripheral LPS.

Objective To observe the protective effects of short-term continuous subcutaneous insulin infusion (CSⅡ) therapy on the peripheral blood circulating endothelial progenitor cells (EPCs) in patients with newly diagnosed type 2 diabetes mellitus (T2DM) and its antioxidation mechanism. Methods Forty T2DM patients (T2DM group) and 30 controls (control group) were involved in this study. Density gradient centrifugation was used to collect peripheral blood mononuelear cells. After 7 days' inducement and differentiation, FITC-UEA and DIL-acLDL double staining cells were identified to be differentiated EPCs by laser scanning conform microscope, and they were counted under inverted microscope. Serum levels of reactive oxygen species (ROS), malondialde-hyde (MDA), glutathione (GSH), superoxide dismutase (SOD) were examined. Results In T2DM group, the EPCs in peripheral blood were significantly reduced (46 ± 3 vs 23 ± 4, P< 0.01), BOS and MDA increased[(1125 ± 687) U/ml vs(2088 ± 765) U/ml, (4.58 ±3.03) μ mol/L vs (8.32 ± 4.15) μ mol/L, P < 0.01] , and GSH and SOD decreased [(263 ± 35) mg/L vs (206 ± 41) mg/L, (103 ± 7) U/ml vs (80 ± 15) U/ml, P < 0.05] . EPCs were negatively correlated with BOS and MDA (r = -0.7352,P < 0.01 ;r = -0.8454, P < 0.01) and was positively correlated with GSH and SOD (r =0.5435,P <0.05;r =0.6437,P <0.05). After short-term CSⅡ therapy, EPCs, GSH, and SOD in patients with T2DM significantly increased (P < 0.05),but BOS and MDA significantly decreased (P <0.05). Conclusion Short-term CSⅡ therapy can significantly reduce oxidative stress in patients with T2DM, promote the restoration of EPCs, and then prevent vascular disease.

Chinese Journal of Immunology 2016;32(6):782-786
Objective:To reveal the dynamic characteristics of the intracellular complement mRNA from mouse macrophage ANA-1 treated with LPS or IL-4. Methods:The polarization models of macrophage ANA-1 were established by treating with LPS(1μg/ml) and IL-4(20 ng/ml),respectively. After treating at 3,8,12 and 24 h,the total RNA were abstracted by Trizol lysis methods . The macrophage polarization were estimated by the expression of IL-1β, CCL2 and Arg-1 mRNA detected by Real-time fluorescent quantitative PCR. The intracellular complement C1q, C3, CfB and CRIg mRNA were quantitatively analyzed. Results: The mouse macrophage ANA-1 cells treated with LPS was polarized to M1 since the levels of IL-1β and CCL2 mRNA were up-regulated significantly,in which their 2-△△Ct value were up to 297. 0±31. 0 and 19. 9±3. 3 respectively at 12 h. On the other hand, the ANA-1 cells treated with IL-4 was polarized to M2 because the level of Arg-1 mRNA was obviously higher( the 2-△△Ct value of Arg-1 mRNA was up to 27.3±9.1 at 24 h)(P<0.05).The intracellular complement C1q,C3,CfB and CRIg mRNAs all were up-regulated in different polarized macrophages. The intracellular C1q and C3 mRNA in polarized M2 were significantly higher,in which the peak value of C1q and C3 were to 94. 9±12. 9 and 11. 3±2. 4 at 12 h,respectively(P<0. 05). Reversely, the CfB mRNA in polarized M1 increased obviously,in which its 2-△△Ct was to 61. 4±6. 2 at 12 h. In addition, the CRIg mRNA in both groups was only up-regulated at 24 h,in which the 2-△△Ct value was 6. 5±1. 8 in M1 and 10. 8±3. 2 in M2(P<0. 05). Conclusion: The macrophage ANA-1 cell polarization models were successfully established by treated with LPS or IL-4. The intracellular complement C1q,C3 and CRIg mRNA in polarized M2 were transcripted more than in M1. But the intracellular CfB mRNA in polarized M1 was up-regulated significantly. These results suggested that the dynamic characteristic of complement components in different polarized macrophage would be correlated with its fun-tions.

Chinese Journal of Clinical Oncology 2014;45(11):679-683
Objective:To determine the function of miR-30b in the metastasis of colorectal cancer cells. Methods:RT-qPCR was performed to test miR-30b expression in 20 fresh primary colorectal cancer tissues and their corresponding adjacent tissues. Transwell and wound healing assays were performed to test the invasion and migration of colorectal cancer cells after miR-30b overexpression or inhibition. Bioinformatics assay was performed to predict miR-30b targets. Western Blot and Dual Luciferase reporter assay were per-formed to test the expressions of Snail and downstream target genes in colorectal cancer cells. Results:The results reveal that miR-30b expression decreased in cancer tissues compared with normal tissues. Transwell and wound healing assays reveal that miR-30b overex-pression inhibited cell invasion and migration, whereas miR-30b inhibition promoted the invasion and migration of colorectal cancer cells. Bioinformatics analyses reveal that miR-30b targets the 3'-UTR of Snail. Dual Luciferase reporter assay confirms that miR-30b af-fects the 3'-UTR of Snail. Western Blot analyses show that Snail and Vimentin expressions were significantly downregulated, whereas E-cadherin expression obviously increased after miR-30b overexpression. However, Snail and Vimentin expressions increased, but E-cadherin expression decreased after miR-30b inhibition. Conclusion:The miR-30b gene is downregulated in colorectal cancer tis-sues. The miR-30b protein may be important in the regulation of cell invasion and migration by targeting Snail in colorectal cancer cells.
