1.Clinical Study on Treatment of Obesity of Gastrointestinal Excessive Heat by Puncturing Acupoints of Stomach Meridian

To investigate the function of the acupoints of the stomach meridian in the treatment of obesity of gastrointestinal excessive heat. Methods: Fifty-one cases of the patients with obesity of gastrointestinal excessive heat were randomly allocated into two groups.The acupoints of the stomach meridian were mainly selected for the treatment group (A), and the combination of Back-Shu and Front-Mu points were used for the control group (B). The treatment was given once every other day, with one month as a course, 3 courses in total, to observe the changes in the symptoms and signs of the patients and to determine the obesity index before and after treatments and the experimental indexes such as leptin, blood lipids,blood sugar and ACHE. Results: In comparison of the control group, various obesity indexes could be significantly reduced (P<0.01), hyperleptinemia and fat metabolic disturbance could significantly improved (both P<0.05) and the imbalance status of the parasympathetic hyperfunction could be significantly reversed (P<0.05) in the patients with obesity in the treatment group. Conclusion: The stomach-meridian acupoints can function on many rings in the occurrence and development of obesity and are key and effective acupoints for obesity of gastrointestinal excessive heat.
2.Change in blood-brain barrier permeability after craniocerebral gunshot injury in dog

Objective To study the change in blood-brain barrier (BBB) permeability in dog with craniocerebral gunshot injury and its significance. Methods Twelve dogs were randomly divided into control group, 15min ofter penetrating craniocerebral injury (PCI) group, 1h after PCI group and 6h after PCI group (n=3). The injury was produced by a small-calibre rifle made in Germany (type 5.56mm, weight of bullet 2.57g). Lanthanum nitrate was infused through the heart. The changes in BBB permeability were observed under electronic microscope. Results In 15min PCI group and 1h PCI group, changes in BBB permeability was observed, with manifestation of opening of tight junctions, with lanthanum nitrate particles passing through BBB into the brain tissues. These changes were more marked in 1h PCI group. Comparing with the above two groups, even more lanthanum nitrate particles were found in the brain tissue in the 6h group (P
3.Clinical analysis on 45 cases of urgent tracheotomy.

OBJECTIVE:
To investigate the clinical characteristics of urgent tracheotomy patients.
METHOD:
Forty-five cases of urgent tracheotomy patients were reviewed retrospectively. The clinical characteristics, pre-operative and post-operative clinical manifestation and complication were studied. Most patients for urgent tracheotomy were head and neck tumor, inflammation and head and neck injury.
RESULT:
Two patients died in operating and two patients died in post-operation. Finding respiratory depression in six patients of long obstruction in post-operation. Eleven patients had complication in operating or post-operation. Hemorrhage and pneumoderma and local trauma were the main complications.
CONCLUSION
Emergent tracheotomy and elective tracheotomy were different-in the short operation time and body position. Complication incidence rate were higher than normal tracheotomy. Patients of long obstruction after post-operation shouldn't breathe in pure dephlogisticated air to avoidance respiratory depression.

4.C-fos gene expression in local cerebral tissues of canines following craniocerebral gunshot wounds

Objective To study the gene expression and the expression change rule of c-fos in the canines posterior to craniocerebral gunshot wounds in order to provide the experimental base for further understanding of the mechanism of gunshot craniocerebral injury. Methods Using the canine model of penetrating craniocerebral injury made by the bullets of the small-calibre rifle made in Germany, the c-fos expressions in the control cerebral tissues and the contusion and concussion areas of the trajectory and the brain stem neurons at different time points after trauma were observed by immunohistochemical method. Results Feeble c-fos expression was detected in the control group. After craniocerebral gunshot wounds, the c-fos expression increased in the contusion and concussion areas of the trajectory and in the brain stem at the 30th minutes, reached the peak at the 2nd hour and began to decrease at the 3rd hour. 5.Effect of gardenoside on the damage of dopaminergic neurons induced by inflammation and its mechanism

Objective To explore the protective effect and mechanism of gardenoside on the damage of dopaminergic neurons induced by lipopolysaccharide (LPS). Methods Both neuron-enriched cultures and neuron-astrocyte cultures were pretreated with vehicle or gardenesides ( 10, 20 and 40 mg/L) for 30 min at 37℃. The culture media were subsequently renewed in order to remove gardenesides. LPS was then added into all culture media at a final concentration of 10 mg/L Twenty-four hours later, the culture media was collected to measure TNF-α, NO, IL-6, GDNF and MMP-9; the cells were collected to count the number of cells labeled with an antibody against tyrosine hydroxylase (TH) and to assess the expression of TH mRNA using reverse transcription-polymerase chain reaction. Results Gardeneside didn't promote the survival of dopaminergic neurons in neuron-enriched culture, but significantly increased the survival of dopaminergic neurons in neuron-astrocyte culture, compared with the vehicle group, the survival of dopaminergic neurons increased from 203.0%±17.4% to 256.7%±15.2% ( F = 17.22, P = 0.001 ) in 40 mg/L gardenaside group. The amount of TNF-α, NO and GDNF released from the neuron-astrocyte cultures after 24 h of addition of LPS was not changed significantly, while the expression of IL-6 and MMP-9 was increased significantly. In this study, the gardenoside concentration-dependently attenuated the LPSinduced increase of the expression of IL-6 and MMP-9, compared with the vehicle group, the expression of IL-6 and MMP-9 decreased to 67.2%±6.4% (F= 12.89,P =0.001 ), 77.3%±9.8% (F =8.27,P = 0.001 ) respectively in 40 mg/L gardenoside group. Conclusions Astrocytes play a neuroprotective role on dopaminergic neurons, which is decreased by LPS via inducing the secretion of pro-inflammatory factors. Gardeneside may protect dopaminergic neurons from LPS-induced injury in an astrocyte-dependent manner and it inhibits the production of proinflammatory factors instead of promoting the secretion of GDNF. From the point of view that a very low toxicity of gardenesides has been well documented, this report may reveal a new way of developing therapeutic interventions for inflammation-related diseases such as Parkinson's disease.
6.Correlation of apolipoprotein M with severity of coronary artery stenosis in patients with coronary heart disease

One hundred and thirty seven pafients diagnosed by coronary angiography were recruited in the study and divided into coronary heart disease group(CAD)and control group.Relationship between serum apolipoprotein M levels and the severity of stenosis of coronary arteries was analyzed.In covariate analysis,serum apolipoprotein M levels were significantly lower in CAD group than those in control group adjusted for sex,age,body mass index(BMI)(P<0.05).By stepwise muhiple regression,serum levels of apolipoprotein M were correlated positively with BMI(r=0.65,P<0.01),and correlated negatively with total cholesterol(TC)(r=-0.53,P<0.01)and low density lipoproteins(LDL-C)(r=-0.42,P<0.01).Serum levels of apolipoprotein M were correlated negatively with the score of coronary stenosis(r=-0.62,P<0.01).The results suggest that apolipoprotein M might be related to the development of coronary artery disease.
7.The role of astrocyte in the lipopolysaccharides-induced damage of dopaminergic neurons

Objective To investigate the role of astrocyte in the lipopolysaccharide-induced damage of dopaminergic neurons. Methods After lipop01ysaccharide was applied to the third generation of rat astrocytes for 24 hours,supernatants of astrocytes culture were collected.The primary middle-brain dopaminergic neuron-enriched culture systems were obtained by neurobasal and ara-c,eoeulture system of both astrocytes and neurons was established by transwell inserts.The lipopolysaccharide was administered into neuron-enriched systems and coculture systems and the change of dopaminergic neurons was detected.At the same time,the supernatants of astrocytes were administered into the neuron-enriched systems,and the survival of dopaminergic neurons and the expression of tyrosine hydroxylase mRNA were observed. Results Lipopolysaccharide had a negative effect on the survival of dopaminergic neurons in a concentration-dependent manner.Both astrocytes and supernatants promoted the survival of dopaminergie neurons,and the former was better than the latter. In the preoccupation of existence of astrocytes,low-concentration lipopolysaccharide promoted the survival of dopaminergic neurons,while high-concentration,decreased.The change of the expression of tyrosine hydroxylase mRNA was similar to the survival of dopaminergic neurons.Conclusions Astrocytes play a protective role in the damage of dopaminergic neurons induced by lipopolysaccharide,and suitable activation of astrocytes would increase the protective effect while excessive activation of astroeytes would attenuate the effect.
8.Application of MR fast scanning techniques in fetal congenital diaphragmatic hernia

Objective To investigate the diagnostic value of MR fast scanning techniques in fetal congenital diaphragmatic hernia. Methods 12 cases diagnosed or suspected fetal congenital diaphragmatic hernia by ultrasound with gestational age (>20 weeks) were underwent MR examination.The fast scan sequences were as follows:Half-Fourier acquisition single-shot fast spinecho se-quence(HASTE)、True fast imaging steady-state precession sequence (True FISP)、Turbo FLASH T1 WI sequence.The axial,co-ronal and sagittal scanning were focused on fetal head and body.The results of prenatal MR examination was compared with neonatal surgical findings or autopsy results after inducing labor.Results All of 12 cases,9 cases were left diaphragmatic hernia,2 cases were right diaphragmatic hernia and 1 case was hiatal hernia.2 cases of them were complicated with other congenital malformations. The features of MRI were as follows:Some portions of bowels and stomach crossed into the thorax were visible in the HASTE and TRUE FISP sequence.The herniated stomach and bowels showed high or scattered high signals.The intestinal types were visible as well.The lung tissue intruded at the apex of the lung and the heart deviated to the opposite side were demonstrated.T1 weighted im-ages obtained from ultra-fast FLASH (Turbo FLASH)T1WI sequence showed characteristic high signals in the meconium,which delineated clearly the shape of the colon and small intestine.The prenatal MRI results of all cases were consistent with surgical (10 cases)and autopsy (2 cases)findings.Conclusion MR fast scanning techniques can clearly show fetal congenital diaphragmatic her-nia.Multiple sequences scanning can comprehensively map the organizational structure and anatomical relationship of fetal congenital diaphragmatic hernia with the surrounding tissues.MR fast scanning techniques play an important role in the diagnosis of fetal con-genital diaphragmatic hernia.
9.Significance of neck dissection during salvage laryngectomy for patients of recurrence after radiotherapy.

OBJECTIVE:
To evaluate the rate of occult metastases detected with elective neck dissection during salvage laryngectomy for patients of recurrence after radiotherapy. The relationship between occult metastases and tumor stage, complications and survival rate after operation was analyzed.
METHOD:
Retrospective review of 24 laryngo-carcinoma patients recurrent after radiotherapy treated with salvage surgery and 28 patients directly received laryngectomy in our hospital between 2005 and 2008.
RESULT:
A higher risk of occult metastases was noted in patients of recurrence after radiotherapy than those without radiation therapy. Cartilage invasion and perineural invasion in the larynx were associated with a higher risk of occult metastases. A statistically significant difference of survival advantage was not noted between these two groups.
CONCLUSION
We recommend bilateral neck dissection at the time of laryngectomy for recurrent staged T3/4 tumors and all patients with recurrent glottis and supraglottic cancers because of the higher rate of occult metastases.

10.STIM1 gene silencing suppresses tumor formation of human hypopharyngeal carcinoma cell line FaDu in nude mice.

OBJECTIVETo evaluate the inhibition effect of STIM1 gene silencing on tumor growth of human hypopharyngeal carcinoma cell lines FaDu in nude mice.
METHODSSTIM1 gene in FaDu was silenced by lentiviral infection, and the effect of inhibition was detected by Real-time PCR and Western blot after lentiviral infection. Nude mice were divided into 2 groups, 5 mice in each group. Inhibition group: subcutaneous inject FaDu cells which STIM1 expression was inhibited.
CONTROL GROUPsubcutaneous inject FaDu cells infected with negative control siRNA-expressing lentivirus. Tumor volumes were measured by calipers, and small animal imaging was detected by NightOWL system on the day 10, 14, 18 and 22 after tumor inoculated. Tumor weights were evaluated in the day 22 after tumor inoculated. Statistical analysis was performed using standard student test(P value threshold was 0.05).
RESULTSThe expressions of human STIM1 gene and protein in FaDu cells were suppressed effectively after STIM1-siRNA lentiviral infection. The mean tumor volumes of control group and inhibition group were (51±25) mm3 and (40±35) mm3, respectively, on the day 10, (262±107) and (106±41) mm3 on the day 14, (716±226) and (340±158) mm3 on the day, (1 682±592) mm3 and (917±252)mm3 on the day 22 (P<0.05). On the day 22, the tumor weight was (1.22±0.41) g in control group and (0.66±0.26) g in STIM1-siRNA group (P<0.05). Small animal imaging showed that the tumors had a smaller fluorescence range with lower signal intensity in STIM1-siRNA group than in control group on the day 14, 18 and 22.
CONCLUSIONThe expression of STIM1 in human hypopharyngeal carcinoma cell lines FaDu can be inhibited effectively by lentiviral infection, causing the inhibition of tumor formation and growth.
