2.Variable-frequency electroacupuncture for elderly patients with knee osteoarthritis

Objective To investigate the therapeutic effects of variable-frequency electroacupuncture treatment for elderly patients with knee osteoarthritis. Methods One hundred and twenty patients with knee oste-oarthritis were randomly divided into a treatment group and a control group. The treatment group was treated with variable-frequency electroaeupuncture, while the control group was treated with muscle strength training. Before treatment and after 8 weeks of treatment, the motor function of their knee joints was evaluated according to Lysholm' s knee scoring scale, quadriceps muscle strength was evaluated according to the maximum load capacity of knee ex-tension, and postural stability was evaluated using a static balance function test. Results Lysholm scores in the treatment group improved significantly compared with those before treatment, except for the item assessing support-ing. While in the control group, compared with before treatment, there were significant differences in the items as-sessing limp, inter-locking, instability, pain and stair activity. There were significant differences in the items as-sessing inter-locking, pain, swelling, stair activity and.squatting between the two groups after treatment. The maxi-mum knee extension load improved significantly in both groups. Static balance with the eyes both open and closed also improved significantly in both groups. There were significant difference in the length, covered area, rectangle-area and length/area of the sway traces of the two groups after treatment. Conclusions Variable-frequency elec-troacupuneture can effectively improve knee function, quadriceps muscle strength and postural stability in elderly patients with knee osteoarthritis.

4.A Reusable Chronocoulometric Aptamer Sensor for Detection of Adenosine Triphosphate

A reusable chronocoulometric adenosine triphosphate (ATP)-aptamer sensor was developed in this work.A short chain of DNA marked as cDNA containing complementary sequence was immobilized on gold electrode based on Au-S self-assembly.The ATP aptamer was hybridized with cDNA.The surface-confined DNA could bind with [Ru(NH3)63+ (RuHex) in the electrolyte via electrostatic interaction.Upon target ATP binding, the aptamer confined onto electrode surface was disassociated from the cDNA oligonucleotides into the solution.Such surface density change of DNA lead to the decrease of chronocoulometric signal for the RuHex which confined on the electrode surface.The chronocoulometric signals showed a linear relationship with logrithm of ATP concentration in the range of 1 nmol/L to 100 μmol/L, and the detection limit of this aptamer sensor could reach 0.5 nmol/L (S/N=3).This aptamer sensor could be regenerated 5 times by simple steps.With this aptamer sensor, the basal level of ATP in the brain cortex micorodialysate was determined to be 19.2±3.7 nmol/L (n=3).

Marrow stromal cells ( MSCs) , component of the mar-row stroma, play an important role in the growth of bone and metabolic balance. This paper mainly summarizes and reviews the molecular regulaory effects of vitamin D and its active form 1, 25 dihydroxy vitamin D3[1,25(OH)2D3] on the osteogenet-ic differentiation of MSCs by Wnt signaling pathway, Wnt5a/ROR2 axis, BMP/TGF-β/Samd signaling pathway and ROS/ERK signaling pathway, so as to clarify the molecular signaling pathway through which vitamin D regulates MSCs metabolically.
6.The diagnosis, treatment and clinicopathologic analysis on gastrointestinal stromal tumors

Objective To investigate the clinicopathologic features of gastrointestinal stromal tumors(GISTs). MethodsThe diagnosis and treatment results of GISTs were studied, and clinicopathologic features and immunohistochemical expressions of CD34, smooth muscle actin (SMA) and S-100 protein were detected in 26 cases of GISTs. ResultsThe tumor was found in the stomach(54%), small intestine(38%) and rectum(8%), and malignant tumors accounted for 77% of the cases. The size of tumors was correlated with the malignancy of GISTs significantly(P0.05). ConclusionsTumor size is a predictor of the malignancy of GISTs, and the immunophenotype is not a predictor.

Objective:To study the effect of family nursing on improving the compliance of drug using and blood glucose levels in diabetic patients.Methods: diabetic patients were collected and randomly divided into observation group received family nursing intervention and control group given usual care, general condition of patients were collected and single factor analysis and multi-factor were conducted. Then medication compliance and blood glucose levels were observed. Results: compliance has positive correlation with cultural level, medical expenses, relationship between doctors and patients, the disease and drug knowledge, family income and negative correlated with reduced physical activity; observation group FBG, PBG, and HbA1c level of observation group were significantly lower than control group, cases of good drug compliance was more than control group.Conclusion: nursing home care can effectively improve the diabetes medication compliance and blood glucose level.
8.Expression of cytokine in mouse graft of corneal allograft rejection

Background Cytokines play a crucial role in mediating immune tolerance or immune rejection of corneal transplantation.However,the study on the expression of cytokines in corneal graft is seldom.ObjectiveThe purpose of the study is to investigate the change of cytokine expression in allografts at different time points after corneal transplantation.Methods BALB/c and C57BL/6 mice aged 6 to 8-week old were used to establish autologous and allografts keratoplasty models.BALB/c mouse was use as donor and receipt in autologous group,and the cornea of C57BL/6 mouse was used to graft on the BALB/c mouse in the allografts group.The graft inflammation was clinically scored,and graft inflammatory scores of ≥5 or opaciflcation scores of ≥2 were identified as rejection.BALB/c mice were randomized into normal control group(3 mice)and allografts group(15 mice).Reverse transcriptPCR (RT-PCR) was used to detect the expression of interleukin-4 (IL-4) mRNA,interferon-γ (IFN-γ) mRNA,IL-10mRNA and tumor necrosis factor(TNF-α) mRNA in graft 6 hours and 1 day,3,7,14 days after operation.Results The corneal graft opacification score was <2 and inflammatory score was <5 in the 10 mice with autologous keratoplasty until 60 days with the survival rate 100%.The edema,opacification and new blood vessel were seen in the BALB/c mice received allografts keratoplasty.The inflammatory score was ≥ 5 and the opacification score was ≥2 24 days after surgery with the rejection rate 100%,in the allografts group,and the graft survival time was (17.80±4.66)days.RT-PCR showed that IL-4 and IFN-γ were positively expressed,and IL-10 and TNF-α were absently expressed in normal mouse cornea.In the allografts keratoplasty mice,positive responses for IL-4,IFN-γ and IL-10 were found in 6hours after operation,but TNF-α was absent.From 1 day through 3 days after operation,the expressions of IL-4,IFN-γand TNF-α were enhanced but IL-10 was disappeared in the graft.IL-10,IFN-γ and TNF-α were expressed till the 7th day,but on the 14th day,only IL-10 was detected in graft in the allografts keratoplasty mice.Conclusions TNF-αis a main factor among the variety of cytokines that may influent corneal allograft rejection locally.
9.THE ISOLATION AND IDENTIFICATION OF ACTINOBACTERIA

The lineage-Actinobacteria class nov.comprises organisms with a DNA base composition which generally is above 50% G+C (with a few exceptions).We set up a method that has been used in isolating Actinobacteria and Actinomycetes. We added 25?g/mL Nalidixic acid and 25?g/mL Aztreonam into isolation media to inhibit the other bacteria and 20?g/mL Benlete to inhibit fungi.We used fluorescent in situ hybridization to identi 1fy Actinobacteria. Using the four probes,PA-1,PA-2,PHGC and PNHGC,we made the identification on the 31 strains of the 56 gram-positive bacteria randomly selected and got 22 positive results,6 negative results and 3 ambiguous results.It was showed that the results of G+C content determination and FISH method were identical.Among 31 strain,there were 24 strains of Actinobacteria,the rate was 77.4%.This proved the isolation and FISH identification methods were effective and reliable.
