AIM: To investigate the effect of mfn2 on mitochondrial function in steatosis hepatocytes. METHODS: Plasmid pEGFP-mfn2 was transfected into hepatocyte strain L02 by Lipofectamine 2000 in vitro, then the steatosis model of hepatocytes was establish by oleic acid induction. RT-PCR was used to evaluate mRNA expression and Western blotting was use to detect the protein expression. ATP level was determined by firefly luciferase bioluminescent. ROS production was measured by fluorescence probe DCFH-DA. Chondrosome transmembrane potential of L02 was observed by labeling of JC-1 and FCM. RESULTS: The stable expression of ectogenesis mitofusin2 in L02 cells was confirmed by RT-PCR and Western blotting. In the model of oleic acids induced lipid formation, Mfn2 obviously inhibited the descent of chondrosome transmembrane potential and ATP level, and increased ROS production in L02 cells. CONCLUSION: Up-regulated expression of mfn2 attenuates mitochondria dysfunction caused by oleic acids induced lipid formation.

Objective To investigate the effects ofdocosahexenoic acid (DHA) on large conductance Ca2+-activated K+ (BK) channels in normal retinal artery smooth muscle cells (RASMCs).Methods Cultured human RASMCs (6 th-8 th generations) were used to patch clamp experiment. The open probabihties (NP0) in BK channels with different concentrations (0.0,1.0,3.0,5.0,7.5,10.0 μmol/L) of DHA were recorded by patch clamp technique in single channel configuration.RASMCs were intervened by different concentrations (0.0,1.0,5.0 μmol/L) of DHA as control group, low and high doses of DHA groups, respectively. The protein expressions of β subunit of BK channels in RASMCs from three groups were measured by Western blot. Results The NP0 of BK channels were 0.044 4±0.001 2,0.081 2±0.004 2,0.209 0±0.006 1,0.310 5±0.005 3,0.465 0±0.007 8 and 0.497 7±0.014 5 with perfusate of 0.0,1.0,3.0,5.0,7.5,10.0 μtmol/L DHA.DHA activated BK channels in a dose-dependent manner (F=2.621,P<0.05).There was no significant difference in the protein expression of control group, low and high doses of DHA groups (F=1 1.657,P>0.05).Conclusion DHA can directly activate BK channels,no increasing in subunit expression of BK channels.

0.05).(2)CagA-positive H.pylori strains of children were only 21.4%. CONCLUSIONS(1)The detection of protein chips technology against H.pylori infection is compared with antral biopsy specimens. The result is without significant difference.(2)Protein chips technology can discover different types of H.pylori strains infection and determine present infection. It is a rapid, accurate method and can detect more factors. It′s a potential detecting method and worthy of using widely.

OBJECTIVE: To optimize the extraction process of total flavonoids from flos lonicerae. METHODS: The time and temperature of extracting, the amount of solvent and the extraction times of total flavonoids from flos lonicerae was optimized using the uniform design. With the yield total flavonoids as markers to conduct U11(116) experiment and optimize the technology. RESULSTS: The optimum extraction process was adding 10 times 5% alcohol extracting for 1 h (only one time) at 85 ℃. CONCLUSION: This extraction process is simple, reliable and serves as a theoretic basis for the extraction process of total flavonoids from flos lonicerae.

OBJECTIVE: To study the in vitro antibacterial activity of the crude extract of total flavonoids from Lonicera japonica against the main pathogenic bacteria.METHODS: With DM130 macroporous resin as stationary phase, the different components(component A contained more sugar, and content B less sugar;C,D and E components were eluted by 20%,40%,and 60% ethanol, respectively) were obtained by gradient elution of the aqueous extract from Lonicera japonica using different concentration of ethanol, then the MIC of different components on main pathogenic bacteria were detected.RESULTS:The antibacterial activity of component B against staphylococcus aureus was more ponent with its MIC at about 1 mg?mL-1.CONCLUSION:Component B has ponent antibacterial action on main pathogenic bacteria.

Chinese Journal of Neurology 2015;48(12):1074-1078
Objective To investigate the level and value of serum uric acid (UA) in patients with multiple system atrophy (MSA).Methods One hundred and one MSA patients were selected as the MSA group and 101 age and sex-matched healthy controls as control group. The UA concentrations and other biochemical indexes were measured by an enzymatic method and carefully analyzed.By comparing the UA concentrations for different genders, subtypes, and different levels of certainty of the MSA group with their counterparts in the control group, we obtained the differences between these two groups and also discussed possible reasons and factors. Results The serum UA levels of MSA patients ((283.74 ± 87.76) μmol/L) were significantly lower than the controls ((317.86 ± 76.95) μmol/L;t =-2.94, P < 0.05).In addition, when the male and female groups were investigated separately, the decrease of serum UA was more significant in the male group compared to the female group (t =-3.88, P < 0.01).Furthermore, by comparing different subtypes of the MSA patients with the control group, we found that the serum UA levels in both P-type or C-type MSA patients were significantly lower than in controls (t =2.92, 2.02;all P < 0.05).By comparing different levels of MSA with the control group, we obtained that UA levels in both probable MSA patients and possible MSA patients were significantly lower than in controls (t =3.13, 2.09;all P < 0.05).By adjusting for the confounding factors, such as age, alanine aminotransferase, aspartate aminotransferase, urea nitrogen, creatinine, high density lipoprotein-cholesterol and low density lipoprotein-cholesterol, the multiple Logistic regression analysis showed that the numbers of MSA patients in the lowest level UA group (UA < 263.0 μmol/L) were higher than the high level UA group (UA > 370.5 μmol/L), though the difference did not reach statistical significance (P > 0.05).Nevertheless, the numbers of male MSA patients in the lowest level and the lower level UA groups (UA =263.0-309.0 μmol/L) were 11.5 and 10.8 times that in the high level UA group (P =0.001, P =0.003).No obviously linear relation between the level of UA and disease progression was found. Conclusions The level of serum UA is significantly decreased in MSA patients, especially in the male patients. The UA, as an antioxidant, may be beneficial to the neurodegenerative diseases.

Objective To study the changes of diamine oxidase (DAO)levels in neonates with hypoxic -is-chemic encephalopathy (HIE)treated with selective brain hypothermi, also to further evaluate Scores for Newborn Gastrointestinal Functior (SNGF).Methods 60 newborns with moderate and severe HIE who were in the NICU of our department from June 2013 to December 2014 were collected. The 60 newborns were randomly divided into hypo-thermia treatment group (HG)and conventional treatment group(CG).The serum was collected and ELISA method was used to test the consistency of DAO on admission and 7 days afterwards, respectively.Meanwhile, the SNGF level of the two groups at the two time points was compared. Results Neither the DAO and SNGF level of the two groups had statistical difference on admission(all P >0.05).Seven days later, both the DAO of the two groups and the SNGF decreased[(12.51 ±1.53)u/mL vs (7.88 ±1.87)u/mL,however,the variation of the hypothermia treatment group was apparently more significant than the change of the conventional treatment group(P =0.011).The SNGF scores of the two groups all decreased, while the hypothermia treatment group was significantly lower than the conventional treat-ment group, the difference were statistically significant (P =0.044,0.006,0.013).Besides,there was remarkably positive correlation between serum DAO level and SNGF (r1 =-0.825,r2 =-0.876,all P <0.05).Conclusion Hypothermia treatment could effectively reduce the injury of asphyxia neonatal gastrointestinal tract by inhibiting the level of DAO,thus improve the gastrointestinal function.
