1.Intravenous transplantation of bone marrow mesenchymal stem cells in combination with recombinant human growth hormone repairs myocardium and vascular tissues in rats with congestive cardiomyopathy

BACKGROUND: It is controversial whether bone marrow mesenchymal stem cells can retain in cardiac injured position, or differentiate into cardiomyocytes or not. OBJECTIVE: To study the effects of recombinant human growth hormone and bone marrow mesenchymal stem cells (BMSCs) intravenous transplantation on myocardium and angiogenesis in rats with congestive cardiomyopathy. METHODS: BMSCs were collected from rats by density gradient centrifugation and adhesive-screening method. Models of cardiac failure were established using adriamycin induction in the model, cell transplantation, growth hormone and combination groups. Following model establishment, cell transplantation group received BrdU-labeled BMSCs (8×10~(13)/L) via vein. Growth hormone group underwent subcutaneous injection of human growth hormone 2 U/kg per day, for 14 consecutive days. Combination group received injection of human growth hormone and BMSC transplantation. At week 4, samples were collected. Immunohistochemical staining for BrdU+MHC and BrdU+Actin was used to determine homing of BMSCs to evaluate the differentiation of transplanted cells into cardiomyocytes and vascular endothelia cells. Hematoxylin-eosin staining was utilized to detect vascular density. RESULTS AND CONCLUSION: Compared with the cell transplantation group, positive rate of Brdu immunohistochemistry was increased in the combination group (P < 0.001). The number of cardiomyocytes and vascular endothelia cells was significantly increased following Brdu+MHC and Brdu+Actin staining (P < 0.001). Compared with the model group, total vascular density, microvessel density and capillary density were significantly increased in the growth hormone, cell transplantation and combination groups (P < 0.001). No significant difference was determined among growth hormone, cell transplantation and combination groups (P > 0.05). Intravenous transplantation of BMSCs could repair cardiomyocytes and vascular endothelial cells by homing into the heart. BMSCs could survive in damaged area and differentiate into cardiomyocytes or vascular endothelial cells and increase the vascular density significantly. Growth hormone could improve microenvironment and raise rates of differentiating into cardiomyocytes or vascular endothelial cells.
2.Effects of bone marrow mesenchymal stem cell intravenous transplantation combined with autologous bone marrow stem cell mobilization on cardiac function in rats with cardiomyopathy-induced heart failure

BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) possess the potential in multi-directional differentiation and can differentiate into cardiomyocytes, which had self-discipline jumpiness. There are few reports on intravenous transplantation or autologous mobilization of BMSCs for treating dilated cardiomyopathy-induced heart failure. OBJECTIVE: To study the effect of BMSC intravenous transplantation combined with bone marrow stem cell autologous mobilization on cardiac function in rats with cardiomyopathy-induced heart failure. DESIGN, TIME AND SETTING: This control experiment was conducted at the National Physiological Laboratory, Molecule Laboratory, Animal Experimental Center and Central Laboratory of Shanxi Medical University from January 10th, 2007 to January 10 th, 2008. MATERIALS: Five Wistar rats served as BMSC donor. Fifty rat models of cardiomyopathy-induced heart failure were created by intraperitoneally injected with 15 mg/kg adriamycin for six weeks. Recombinant human granulocyte stimulating factor (GSF) was bought from Qilu Pharmacy Co., Ltd. CD44, CD34 and CD45 were given by Flow Cytometric Analysis Laboratory, Experimental Center, Shanxi Medical University. METHODS: BMSCs were isolated, purified and cultured in vitro. Surface marker CD45, CDD44 and CD34 were detected with f1ow cytometer. Wistar rat models were randomized into 4 groups. BMSCs and recombinant human GSF (50 ?g/kg) were injected into models by tail vein and hypodermoclysis infusion respectively in the combination treatment group (MG group). Recombinant human GSF (50 ?g/kg) was subcutaneously injected into models in the bone marrow stem cell autologous mobilization group (G group). BMSCs were injected into rat models by tail vein in the BMSC group. Rat models in the heart failure group were intact. MAIN OUTCOME MEASURES: ①Peripheral blood leukocyte and mononuclear cell counts and CD34 expression were obtained before treatment and 6 days after treatment. ②At 4 weeks, cardiac function parameters were evaluated by echocardiography and hemodynamic measurement. Objective To explore the relationship between brain development and early behavior development of infant rats. Methods Infant Spraque - Dawley(SD) rats were used respectively for testing their spontaneous behaviors, inhibitory escaping response and behavior development, and also determine their brain weight and content of Zinc in different part of the brain at age of 1 day, 11 days and 21 days. Results There was a positive correlation between brain development and early behavior development of SD rat, and the high content of zinc in hippocampus and cerebellum. Conclusion It is suggested that the high content of Zinc guarantee furnish security for the late behavior development of infant rat.
4.Mononuclear cells isolated from mobilized bone marrow differentiate into vascular endothelial cells and cardiomyocyte-like cells

BACKGROUND:It is controversial whether bone marrow mobilization can retain in cardiac injured position in congestive cardiomyopathy or differentiate into cardiomyocytes and vascular endothelial cells.
OBJECTIVE:To study the effects of granulocyte colony stimulating factor (G-GSF) on myocardium and angiogenesis in rats with congestive cardiomyopathy.
METHODS:Fifty Wistar rats with heart failure caused by adriamycin-induced cardiomyopathy were divided into heart failure group (n=20) treated with normal saline and bone marrow mobilization (n=30) treated with subcutaneous injection of recombinant human G-GSF. Ten rats from the bone marrow mobilization were kil ed at day 6 of mobilization, and myocardial tissue was taken for CD43 immunofluorescent staining. Blood samples were taken from the rat tail in each group before and 5 days after treatment to count total number of white blood cells and percentage of mononuclear cells. Meanwhile, mononuclear cells extracted from the peripheral blood were used for flow cytometry detection. At day 5 after treatment, bromodeoxyuridine (BrdU, 50 mg/kg) was successively given to al rats for 4 weeks before they were sacrificed. Myocardial tissues were taken to determine the homing of mononuclear cells and evaluate differentiation of mononuclear cells into cardiomyocytes and vascular endothelial cells using BrdU staining, BrdU/myosin heavy chain double staining, and BrdU/actin double staining. Hematoxylin-eosin staining was used for determination of blood vessel density.
RESULTS AND CONCLUSION:G-CSF mobilization increased the number of mononuclear cells that was significantly higher than before treatment (P<0.05). Flow cytometry showed that the number of CD34-positive mononuclear cells in the peripheral blood was higher in the bone marrow mobilization than in the heart failure group (P<0.05). Myocardial CD34 immunofluorescence showed that the heart failure group was negative and the bone marrow mobilization group was positive. In the bone marrow mobilization group, the myocardial tissue was positive for BrdU staining, BrdU/myosin heavy chain double staining and BrdU/actin double staining, while vascular endothelial cells in the region of myocardial injury was positive for BrdU;conversely, the heart failure group was negative. The density of blood vessels in the bone marrow mobilization group was significantly higher than that in the heart failure group (P<0.001). These findings indicate that bone marrow mobilization increases the number of mononuclear cells, and these cells are homing to myocardial injury, thereby playing a repair role in the myocardium and vascular tissue of heart failure rats with congestive cardiomyopathy.
5.Establishment of a Shanxi province centered maintenance network for the management and practical activity of peripherally inserted central catheters

Objective To explore how to establish and safeguard the peripherally inserted central catheters(PICC) maintenance network for the management and practical activity of PICC,so as to provide continuous high-quality PICC maintenance for discharged patients with PICC. Methods 40 nurses each year from 10 cities and 100 country hospitals affiliated with Shanxi Quality Control Center to train in the 1st and 2nd affiliated hospital of Xi`an Jiaotong University were choosen as 214 PICC maintainers. These maintainers were taken into the inner-net of 1st affiliated hospital of Xi`an Jiaotong University management system complete the PICC maintenance net of Shanxi province. Results After the establishment of maintenance network, the number of patients with PICC increased (from 610 cases in 2008 to 1 434 cases in 2014);A large number of PICC management had been down by the qualified nurses in primary hospitals; Duration of PICC in patients days was prolonged; The PICC related complications including thrombophlebitis, catheter infection,occlusion or dislodgment and unplanned extubate were significantly decreased (P < 0.05 respectively). Conclusions Establishment of PICC maintenance network can provide patients with standardized and convenient PICC maintenance service,decrease the incidence of catheter which related complications and ensure patient safety. The application of PICC maintenance network should be used more and more.
6.Study of the patterns of cancer nurse specialist training

Objective To summarize theskill practice as the coremethods and training effect in tumor specialist nurse training , and to explore the suitable effective methods and pattern of China′s national conditions of specialist nurse training in China, for providing a reference for conducting cancer specialist nurse training. Methods Reviewed and analyzed tumor specialist nurse training method to make clear the training effect from 2012 to 2013, 100 students was investigated by using questionnaires , which contain 15 aspects, before and after the training respectively. designskill practice as the coreof the training content (160 hours) and theoretical classes (80 hours), established training evaluation criteria and the organization management system. Results All 100 students had grasped maintenance technology, observation of the complications of treatment, infusion port of the maintenance, the use of chemotherapy drugs, health education for patients with PICC catheter, and the use of effective order, the grasp rate was 100%. Complications rate was decreased significantly after using PICC catheter, (the rate of catheter related blood stream infections and the rate of thrombosis was reduced from the top 35%and 12%before training to 4%and 1%after training respectively (P<0.05). Conclusions In order to train more practical clinical nurse specialists, reduce the complications, raise the overall level of nursing profession, and improve the quality of nursing after using PICC catheter. Skill practice as the core methods in tumor specialist nurse training should be advocated.
7.Perioperative change of erythrocyte complement receptor I activity and malonydinldehyde content in patients undergoing radical recection of breast cancer

Objective To understand the change of erythrocyte immunity and matondialdehyde (MDA) in perioperative patients with mammary cancer. Methods30 patients were divided randomly into two groups:group A(general anesthesia) and group B(epidural block). To estimate the activity of erythrocyte complement receptor I (CR- I ) and the lever of MDA, the venous blood were collected at the time points of before anesthesia, after anesthesia, during operation, and on the 1st, the 3rd, the 5th postoperative day. ResrltsCompared with before anesthesia, the RBCC3bRR, RBCCaR and RFER in group A were decreased statistically at the time points of after anesthesia, during operation, and on the 1st, the 3rd postoperative day(P <0.05 or P <0.01). Above mentioned index in group B were reduced significantly only during operation and on the 1st postoperative day. Between two groups, above mentioned index in group A were lower than those in group B(P<0.05).Compared with before anesthesia, RFIR and MDA in group A were higher at the time points of after anesthesia, during operation, and on the 1st, the 3rd postoperative day(P<0.05 or P<0.01).but above mentioned index in group B rose only on the 1st postoperative day.CR- I and MDA in two groups restored to the lever of be fore anesthesia on the 5th postoperative day. ConclusionThe perioperative immunological function of erythrocyte is obviously depressed by general anesthesia and epidural block, especially the former. This depression is relevant to the disorder of erythrocyte immunity regulating function and the lever of plasma MDA.
8.Effect of inadequate reduction of medial wall on the postoperative varus deformity following calcaneal fracture surgery and other influencing factors of varus deformity

Objective To explore the effect of inadequate reduction of the medial wall on the postoperative varus deformity following calcaneal fracture surgery and other influencing factors of varus deformity.Methods A retrospective analysis was conducted of the 60 patients with calcaneal fracture involving the medial wall (60 feet) who had been treated from October 2005 to September 2015.They were 51 men and 9 women,18 to 55 years of age (average,38.4 years).According to the Sanders classification,24 patients belonged to type Ⅱ,30 to type Ⅲ,and 6 to type Ⅳ.We measured the integrity of the pre-and post-operative medial walls,the height and width of the pre-and post-operative calcanei,the pre-and post-operative B(o)hler angles and the pre-and post-operative calcaneal varus angles for statistical analysis.Results The effect of postoperative height reduction of the medial wall on the postoperative correction of varus angle was significantly larger than that of the lateral wall (P < 0.05).The higher the integrity of the postoperative medial wall,the larger the correction of the postoperative varus angle.The normal rate of postoperative calcaneal varus angle significantly decreased with the increased Sanders classification severity (P =0.001).The postoperative reduction of the height and width of the calcaneus and the integrity of the preand post-operative medial walls were significantly positively correlated to the normal rate of postoperative calcaneal varus angle (P < 0.05).Conclusions The medial wall has a large influence on the postoperative calcaneal varus angle.The higher integrity of the preoperative medial wall,and the more adequate reduction and fixation of the postoperative medial wall as well,the higher rate of postoperative normal calcaneal varus angle.The postoperative reduction of the calcaneal height also affects the varus angle.When the heights of the medial and lateral walls are recovered,the normal rate of varus angle is the highest;when reduction of the medial wall is inadequate but reduction of the lateral wall is fine,the normal rate of varus angle is the lowest.

Objective:To study the influence of DiKang capsule on expression of TGF? receptor Ⅱ on hepatic stellite cells(HSC)in rat with liver fibrosis introduced by DMN.Methods:TGF? receptor Ⅱ on HSC in rat with liver fibrosis introduced by DMN were measured by FCM.Results:In group of 10 ?g/kg DMN injected 3 weeks,expression of TGF? recptor Ⅱ on HSC in these rat fed with DiKang capsule 3 weeks were higher than that not feeding capsule.But in group of injecting 5 ?g/kg DMN continuous 6 weeks,expression of TGF? receptor Ⅱ on HSC does not changed with feeding or not feeding DiKang capsule.Conclusion:DiKang capsule may have a certainly role on expression of TGF? receptor Ⅱ on HSC in rat with liver fibrosis introduced by DMN.
10.The Effect to Expression of NF-?B, Caspase-3 and MMP-2 in Chronic Pelvic Inflammatory Rat by Rectal Ministration with the Penyankangshuan

Objective To study the anti-inflammatory mechanism of the Penyankangshuan (PYKS) in treating chronic pelvic inflammation. Methods After model rats with endometritis were established successflly, 6 groups were formed which were the high, the middle and the low dosage groups of the PYKS, the KFXYS (a kind of patent medicine has a similar effect) group, the model group, the normal group. The treated rats were continuously rectal administration 3 weeks, the expression of NF-?B, Caspase-3 and MMP-2 were observed in the high, the middle dosage groups of the PYKS, the KFXYS group, the model group and the normal group with immunohistochemistry. Results The PYKS obviously raised the expression of Caspase-3 and MMP-2, but decreased the expression of NF-?B. Conclusion The anti-inflammatory mechanism of the PYKS possibly raised the expression of Caspase-3 and MMP-2 of mice endometrial cells, decreased the expression of NF-?B, so as to reduce inflammation soakage in excess and formation of inflammatory medium, accelerate apoptosis and reduce local inflammation.
