1.Study on the risk factors for hip fracture in elderly people in Ningbo

Objective To investigate the risk factors for hip fracture in elderly people in Ningbo,in order to provide the basis for prevention and treatment of hip fracture.Methods A case-control study were conducted in study on 450 cases of hip fracture patients aged over 60 years in Ningbo and 450 subjects of control as 1 ∶ 1 matched case control study.A univariate analysis of hip fracture was performed by x2 test.Risk factors for hip fracture were analyzed by logistic regression.Results Poor self-care ability (OR=5.05),light manual labor occupation (OR=4.10),history of cerebrovascular events (OR=2.59),smoking (OR=2.14) were the risk factors for hip fracture in elderly men,while poor self-care ability (OR=11.36),light manual labor occupation (OR=2.41),osteoporosis (OR=3.19),early menopause (OR=6.89),low body mass index (OR=5.05) were the risk factors for hip fracture in elderly women.The protective factors for hip fracture was milk uptake (OR=0.29) Conclusions Early interventions for hip fracture risk factors,and prevention and treatment of osteoporosis can help reduce the occurrence of hip fracture.
2.Relationship between body composition and bone mineral density in postmenopausal women of different ages

Objective To determine the effect of lean mass and fat mass on bone mineral (BMD) in postmenopausal women of different age groups.Methods A total of 287 postmenopausal women (aged 50 years and over) participated in this study.Individuals were divided into 4 age groups by decades:group 1,50-59 yr (82 cases); group 2,60-69 yr (73 cases); group 3,70-79 yr (66 cases); group 4,80 years and over (66 cases).The BMD of lumbar spine (L2-4),left femoral neck and total hip were measured by dual-energy X-ray absorptiometry.Fat and muscle mass were analyzed by its analysis function and the percent of fat mass and muscle mass were calculated.Then data were processed by the SPSS 18.0 programme.Results One linear regression analysis showed that the L2-4 BMD was significantly associated only with body composition of trunk,while femoral neck (FN) and total hip (TH) BMD were respectively associated with body composition of different parts in group 1.L2-4 BMD was significantly associated only with trunk fat mass and muscle mass (r=0.27,0.25,both P< 0.05),but there were no relationships between BMD of FN and TH and body composition of different parts in group 2.The BMD of L2-4,FN and TH were significantly respectively associated with body composition of different parts in group 3 and 4.Conclusions In postmenopausal women,fat mass and muscle mass play an important role in BMD.In postmenopausal women aged 60-69 yr,the body composition has a minimal impact on BMD,and hormone replacement therapy may be the best for osteoporosis prevention and treatment in this period.Different exercise and loosing of weight in postmenopausal women of different ages can prevent osteoporosis and fragility fractures.
3.Analysis of Drug Utilization in 16 Hospitals of Chengdu Area During 2001~2006

OBJECTIVE:To analyze the utilization status and developing trend of drugs in Chengdu area. METHODS:Data regarding the drug variety,consumption quantity and sum of money in 16 hospitals of Chengdu area between 2001 and 2006 were analyzed retrospectively. RESULTS:The drug consumption sum in 16 Hospitals of Chengdu area from 2001 to 2006 increased by 19.56%,27.19%,17.84%,15.08% and 10.40%,respectively,assuming an downtrend. Among the top 14 chief drug categories on the list of consumption sum,the sequence of anti-infective agents,cardiovascular agents,antineoplastic agents and nervous system agents experienced no obvious changes. CONCLUSION:The drug utilization in Chengdu area tends to be rational and standard.
4.Effects of electroacupuncture on circadian rhythm of temperature and melatonin in depression rats model induced by chronic stress.

OBJECTIVETo observe the effects of electroacupuncture (EA) on circadian rhythm of temperature and melatonin (MT) in depression rats model induced by chronic stress, so as to explore the biological mechanism of EA for depression.
METHODSTwenty-four SD rats were randomly divided into a control group, a model group and an EA group, 8 cases in each one. Rats in the control group were treated with normal diet for 21 days without any treatment. In the model and EA group, rat model was established by chronic unpredictable stress combined with solitarily feeding method, and rats in the EA group was treated with EA at "Baihui" (GV 20), "Yintang" (GV 29) 1 h before stress stimulation everyday, 2 Hz in frequency and intensity was favorable with the head of rat slightly shivering. The needles were retained for 20 min, once a day for totally 21 days. After EA treatment, open-field experiment was adopted to observe the behavioral improvement; the rats temperatures were monitored at six time points (2:00, 6:00, 10:00, 14:00, 18:00, 22:00) and orbital blood sampling was collected. The level of serum MT was tested by enzyme linked immunosorbent assay. The circadian rhythm changes of temperature and serum MT in each group were compared.
RESULTSThe numbers of horizontal movement and vertical movement in the model group were obviously lower than those in the control group (both P < 0.05), while those in the EA group were significantly improved compared with those in the model group (both P < 0.01). The circadian rhythm of temperature and MT disappeared in the model group, which was improved into normal level after EA treatment.
CONCLUSIONThe electroacupuncture has regulation effects on circadian rhythm of temperature and melatonin in depression rat model induced by chronic stress.

Harmful substances are produced during the metabolism of alcohol in the human body, which causes liver disease.The mechanism of oxidative stress is the main pathogenesis of alcoholic liver disease, which plays a major role in the occurrence and development of alcoholic liver disease.Alcohol-induced oxidative stress mediated by reactive oxygen species(ROS)/reactive nitrogen species(RNS) brings about liver disease via apoptotic signaling pathway and MAPK signaling pathway, and alleviates liver disease through Nrf2 signaling pathway.In this paper, the studies on the mechanism of oxidative stress in alcoholic liver disease are reviewed.
6.Effects of sinew-regulating bone-setting manipulations on chondrocytes, IL-1β and NO in rabbits with knee osteoarthritis

Objective: To observe the therapeutic effect of sinew-regulating bone-setting manipulations for knee osteoarthritis (KOA) model rabbits and its impacts on the chondrocyte apoptosis rate and the levels of interleukin (IL)-1β and nitric oxide (NO). Methods: According to the random number table method, 30 New Zealand white rabbits were divided into a normal group (n=9) and a modeling group (n=21). Rabbits in the modeling group were used to establish KOA models with the modified Hulth method. At the 8th week, three rabbits were sacrificed to verify the model and the remaining 18 rabbits were randomly divided into a model group (n=9) and an intervention group (n=9). Rabbits in the normal group and model group were bred routinely without any intervention. Rabbits in the intervention group were treated with the sinew-regulating bone-setting manipulations, 10 min/time, once every other day for a total of 20 times. The Lequesne MG knee function rating was used to evaluate the behavioral differences of the rabbits in each group. The Pelletier score was used to evaluate the general changes of the rabbits. The Mankin score was used to evaluate the pathology of knee cartilages. The enzyme-linked immunosorbent assay and nitrate reductase methods were used to determine the levels of IL-1β and NO in serum and synovial fluid of each group, respectively. In situ terminal deoxynucleotidyl transferase-mediated nick and labeling method was used to determine the apoptosis of chondrocytes in each group. Results: Compared with the normal group, the scores of Lequesne MG, Pelletier and Mankin, and the levels of IL-1β and NO in the model group were increased (P<0.05), which indirectly indicated the success of the model. Compared with the model group, the scores of Lequesne MG, Pelletier and Mankin, IL-1β and NO levels, and chondrocyte apoptosis rate of the intervention group were decreased, and the differences were statistically significant (P<0.05). Conclusion: The sinew-regulating bone-setting manipulations can reduce the levels of IL-1β, NO, and chondrocyte apoptosis rate, and delay the articular cartilage degeneration, therefore, having a good therapeutic effect on KOA.
7.Construction of NF-?B/p65-targeting RNAi combinant adenovirus vector and its suppressive effect on the expression of p65 in ECV304 cells

Objective To construct RNAi combinant adenoviral expressive vectors specific to p65 subunit of NF-?B and to observe their gene silencing effect on p65 subunit.Methods Three pairs of complementary. single-strand DNA oligos targeting three various sites of p65 mRNA were designed and synthesized.Annealling was used to generate double-strand oligos(ds-oligos),and then the ds-oligos were cloned into pENTR~TM/u6 to generate the entry clone named pENTR.Recombination reaction in vitro with the pENTR and pAd/BLOCK-iT~TM- DEST was used to creat the adenovirus plasmid which contains the RNAi cassette.Then,the adenovirus plasmids digested with PacI were transfected into HEK293A cells to product adenovirus,and latter infected the HEK293A cells to amplify the adenoviral stock.Plaque forming assay was used to titer the adenoviral stock.The p65 gene silencing effect induced by the RNAi adenovirus was detected by Western blot and immunocytochemistry assay in ECV304 cells.Results The RNAi adenovirus specific to p65 subunit of NF-?B were produced with titer of 3.0 x 10~9pfu/ml to 2.5?10~10pfu/ml.The expression of p65 protein in ECV304 cells could be down-regulated efficiently by the RNAi adenovirus 48-72 h after infection,which would last for more than 6 days after infection.Conclusion RNAi adenovirus is an important tool inhibiting the expression of target gene efficiently.
8.Expression of a testis-specific gene 1700001022RIK in mice and its bioinformatic analysis.

OBJECTIVETo identify the expression characteristics of the 1700001022RIK (RIKEN cDNA 1700001022) gene in mice and explore its function by bioinformatic analysis.
METHODSUsing the expression profile of gene microarray, we detected the expression of a new testis-specific gene, 1700001022RIK, in mice. We analyzed its expression characteristics in the testis tissue and their changes in different developmental stages of the testis by RT-PCR, real-time RT-PCR, Western blot, and immunohistochemistry. We performed bioinformatic analysis using a bioinformatic software.
RESULTSThe 1700001022RIK gene was specifically expressed in the mouse testis in an age-dependent manner, most highly in the adult mice. The 1700001022RIK protein was mainly expressed in the spermatogonia, spermatocytes, and round spermatids of the adult mice. Bioinformatic analysis showed that the 1700001022RIK protein amino acid sequence had a high similarity in human and mice, which indicated that this gene was highly conserved in mammals.
CONCLUSION1700001022RIK is a testis-specific gene mainly expressed in the spermatogonia, spermatocytes, and round spermatids of seminiferous tubules, which might be involved in the regulation of spermatogenesis.

9. Expression of Hsa_circ_0002360 in lung adenocarcinoma and its clinical significance

Objective The relationship between circular RNAs (circRNA) and lung adenocarcinoma (LAC) remains to be further explored. This study aimed to investigate the expression of Hsa_circ_0002360 in LAC and its value in the clinical diagnosis and treatment of the malignancy. Methods This study included 50 cases of LAC treated in our hospital from May 2018 to June 2019 and another 50 healthy subjects as normal controls. We determined the expression of Hsa_circ_0002360 in the LAC cells by qRT⁃PCR. We cultured human LAC cell lines A549, H1299 and H1975 and human bronchial mucosal epithelial cell line BEAS-2B, inoculated the A549 and H1975 LAC cells in the 6-well plate, with 3 wells for si-circRNAs and the other 3 for negative controls (si-NC), followed by cell transfection experiment. Using the CCK8, colony formation and wound healing assays, we detected the effects of Hsa_circ_0002360 on the activity, proliferation, invasiveness and metastasis of the A549 and H1975 cells. 10.Non-headache symptoms in a migraine attack

Objective To characterize non-headache symptoms during the migraine attack phase in migraine patients. Methods We enrolled in the present investigation 71 patients with migraine with and without aura who had sought medical treatment in our department from January 2010 to January 2011. We used a self-made migraine questionnaire to investigate associations of the non-headache symptoms with the sociodemographic factors, subtypes, severities and attacks of the headache in the patients. Results There were no significant differences in the number of non-headache symptoms among patients with different ages, genders, education levels, subtypes, severities, mean attack frequencies per month (P>0.05). The most frequently reported non-headache symptoms were nausea (87.3%),phonophobia (81.7%) and mood change (69.0%).The incidences ofphonophobia,sleep disorder and osmophobia were significantly higher in patients without aura than in patients with aura (P<0.05).Conclusion Migraine attacks are often accompanied by a variety of non-headache symptoms of which the most frequently reported ones are nausea,phonophobia and mood change.Patients without aura tend to have more phonophobia,sleep disorder and osmophobia than those with aura.
