1.Effect of IFN-? on the growth of human cervical cancer transplanted subcutaneously in nude mice

Purpose:To study the effect of interferon-? on the growth of human cervical cancer transplanted subcutaneously in nude mice.Methods:Twenty-eight nude mice transplanted subcutaneously with human cervical cancer were divided into 4 groups: control group,positive control group(DDP group),IFN-? low dose group(1.5?10 7 u/kg) and IFN-? high dose group(3.0?10 7 u/kg). Different drugs were used from day 14 after transplantation and all mice were sacrificed after 49 days. The weight of the mice and the volume of tumor were measured. FCM was used for measuring the proportion of cell cycle S+G 2 -M(%) in tumor cells. Results:The growth inhibiting rates in DDP group,IFN-? low dose group,IFN-? high dose group were 92.8%,73.8% and 93.1% respectively. Except for mice in DDP group,there was obvious increase in the weight of mice in other three groups. As for the proportion of S+G 2 -M(%),there was no difference in control group,IFN-? low dose group and IFN-? high dose group. Conclusions:IFN-? can inhibit markedly the growth of transplanted cervical cancer,which is correlated with the IFN-? dose. The mechanism of IFN-? inhibiting cervical cancer was not due to the suppression of cancer cell proliferation.
2.Inhibitory effect of paclitaxel combined with nedaplatin on HeLa cells transplanted in nude mice

Background and purpose:Paclitaxel (Taxol,TXL) combined with cisplatin (DDP) has become the common chemotherapeutic regimen for cervical cancer. However, DDP has higher toxicities. Nedaplatin (NDP), a new platinum analogue,has been shown to have a higher antitumor efficiency against cervical cancer and lower toxicity. This study was done to investigate the antitumor effect of TXL+NDP on HeLa cells transplanted tumor compared with TXL+DDP. Methods:HeLa cells were transplanted subcutaneously in nude mice. These nude mice were randomly divided into 6 groups by sorting when tumor diameter reached 5-6 mm: Control, TXL, NDP,DDP, TXL+NDP,TXL+DDP. Tumor volume and body weight were measured during experiment. The volume ratio (T/ C) was the volume of single agent treatment group and control group and used to assess the antitumor effect, and the combination ratio (CR) was used for the combined therapy. More than 20% weight loss was defined as reaching toxicity level. Results:TXL+NDP regimen had a synergistic antitumor effect (CR=0.76) on HeLa cell transplanted tumor, with a maximum weight loss 8.3%. The tumor volume of TXL+NDP was 172?60 mm3, which was significantly smaller than that of TXL(304?55 mm3) group or NDP(356?67 mm3) group(P0.05), but had a higher toxicity with 4 mice death. Conclusion: TXL+NDP regimen has a synergistic inhibitory effect on HeLa cells and lower toxicity. The efficacy of TXL+NDP regimen needs to be confirmed in clinical trials.

Using the self-arrangede questionnaire,it is aimed to investigate the aititude of a normal university's 100 non-psychologystudents' attitudes to some of the porpular standard of mental health in undergraduates.Conclusions:(1)most of the non-psychology students are agree with the 30 items standards of the questionnaire.(2)While using "interpersonal relation","mental state","the attitude toward life","moral code","possessiveness"as the standaes of mental health of undergraduate,there is significant difference between non-psychology boys and girls.(3)While using "be good at copping with affairs","accepting dificiency","appropriate expression of emotion",as the standaes of mental health of undergraduate,there is significant difference between non-psychology sophomore and junior.
6.5-Fluorouracil-loaded polylactic acid nanoparticles have a killing effect on gastric cancer cell lines in vitro

BACKGROUND:5-Fluorouracil occupies an important position in the treatment of gastric cancer, but its long-term use can easily induce adverse reactions such as myelosuppression and leukopenia. Polylactic acid and its copolymers have a higher biocompatibility, and their decomposer cannot gather in the body.
OBJECTIVE:To investigate the in vitro cytotoxicity mechanism of 5-fluorouracil-loaded polylactic acid nanoparticles on gastric cancer cel lines.
METHODS:Ten mice were selected in this study. 5-fluorouracil-loaded polylactic acid nanoparticles (1×10-7, 1×10-6, 1×10-5, 1×10-4 mol/L) were prepared using ultrasonic emulsification method. Kiling effect of polylactic acid nanoparticles on gastric cancer cel lines in vitrowere detected. Then, the inhibition rate was calculated at different concentrations.
RESULTS AND CONCLUSIONS: Under the transmission electron microscope, 5-fluorouracil-loaded polylactic acid nanoparticles had good shape and relatively evenly distributed with no adhesions. After drug administration, the drug concentration was 50% at 24 hours and 62.9% at 72 hours. After 48 hours co-culture with single 5-fluorouracil or 5-fluorouracil-loaded polylactic acid nanoparticles, the viability of gastric cancer cels showed a decrease trend with the increase of drug concentrations, and moreover, 5-fluorouracil-loaded polylactic acid nanoparticles had a better cel inhibition ability than the single 5-fluorouracil (P < 0.05). The IC50value of 5-fluorouracil-loaded polylactic acid nanoparticles was significantly lower than that of 5-fluorouracil (P < 0.05). These findings indicate that polylactic acid nanoparticles as good drug carriers have a strong drug loading capacity and increase drug concentration in the body, but cannot reduce the biological activity of 5-fluorouracil, which provide new ideas for the treatment of gastric cancer.
7.The moderating effect of monoamine oxidase A gene polymorphism in relationship between childhood abuse of male adolescents and externalizing behaviors

ObjectiveTo explore the relationship between male adolescents' childhood abuse and externalizing behaviors,and the moderating effect of monoamine oxidase A (MAOA) gene promoter region variable number tandem repeat (VNTR) polymorphism.MethodsThrough random cluster sampling,352 Han male middle school students from Changsha were tested by Achenbach child behavior checklist-youth self reports (CBCL-YSR)and childhood trauma questionnaire-28 item short form (CTQ-SF),their MAOA genotypes were also identified.A hierarchical multiple regression analysis was used to test the moderating effect.Results ( 1 ) Compared participates with high activity MAOA gene and that with low activity MAOA gene,there were no significant differences on age ( ( 15.82 ± 1.52) vs ( 15.94 ± 1.62 ) ),externalizing behaviors ( ( 15.13 ± 10.14 ) vs ( 14.33 ± 9.70 ) ),total abuse ( (52.59 ± 10.46) vs (51.39 ± 7.56 ) ),emotional abuse( ( 7.63 ± 3.31 ) vs ( 7.11 ± 2.68 ) ),physical abuse ( ( 6.40 ± 2.82) vs (6.12 ± 2.05 ) ),sexual abuse ( ( 6.42 ± 3.24 ) vs ( 5.94 ± 1.72 ) ),emotional neglect ((13.44±5.12) vs (13.16 ±4.83) ),physical neglect( (10.27 ±2.64) vs (10.44±2.53))(t=-1.789~0.678,P > 0.05 ).(2)Except emotional neglect and physical neglect,emotional abuse,physical abuse and sexual abuse could predict externalizing behaviors( Sβ =0.141 ~0.347,P < 0.01 ).(3) MAOA gene was not directly related to externalizing behaviors( Sβ =- 0.023,P > 0.05 ).There was a significant interaction between MAOA gene and emotional abuse( Sβ =-0.148,P < 0.01 ).The interaction between MAOA gene and physical abuse or sexual abuse showed no statistical significance( Sβ =- 0.067,- 0.005,P > 0.05 ).ConclusionMAOA gene polymorphism can moderate the relationship between male adolescents'childhood emotional abuse and externalizing behaviors.
10.Change of airway anaphylatoxin C5 a in patients with asthma

Objective:To investigate the role of anaphylatoxin C 5 a in patients with asthma .Methods:A prospective study was performed between September 2006 and February 2007.A total of 33 patients with acute exacerbation of asthma and 13 healthy subjects were recruited into the study .The patients with acute exacerbation of asthma were also studied when they returned to the remission state .Levels of lung function, levels of C5a in induced sputum and cell differential count in induced sputum were determined . Results:The level of C5 a in induced sputum was significantly higher in patients with acute exacerbation of asthma [0.85(0.68-2.13) μg/L] than that in patients with stable asthma [0.45(0.26-0.88)μg/L, Z=-2.193, P=0.013];Sputum C5a levels in stable asthma patients were significantly higher than those in healthy controls [0.14(0.06-0.45) μg/L, Z=-2.141, P=0.015].The level of C5a in patients with severe exacerbation [2.21(1.27 -9.0) μg/L] was significantly higher than those in patients with mild exacerbation [0.34(0.17-0.63) μg/L] and moderate exacerbation [0.85(0.55-1.67) μg/L,χ2 =12.330, P=0.001].The level of C5a in induced sputum was positively correlated with the number of total cells count (r=0.797, P=0.004), neutrophils (r=0.504, P=0.032) and macrophages ( r =0.424, P=0.036 ) in acute exacerbation of asthma .Conclusion: C5a levels in induced sputum could be identified as an important prognostic biomarker , which involved in asthma ’ s pathogenesis .
