[Objective] To investigate the role SPAG5 play in ovarian adenocarcinoma cell mitosis,Taxol sensitivity and ovarian high grade serous carcinoma patients' prognosis.[Methods] Transient knockdown of SPAG5 in SKOV3 cell were performed, and MTT assay and cell cycle flow cytometry assay were carried out.IHC staining of SPAG5 protein in 110 high grade serous carcinoma patients' tumor tissues were performed, and the expression were analyzed with clinical data and prognosis.Finally,SPAG5 were knocked down in OVCAR3 A2780 and SKOV3 cells followed by 0.5μM Taxol treatment,MTT assay were performed to detect cell viability.[Results] SPAG5 knockdown inhibited cell mitosis of ovarian adenocarcinoma cell SKOV3 by G2/M arrest.High grade serous carcinoma patients after neoadjuvant chemotherapy gained the expression of SPAG5.Patients without neoadjuvant chemotherapy with low SPAG5 expression have poor progress free survival, especially in early stage patients.Patients with low SPAG5 expression also have poorer overall survival, but the difference was not statistically significant.Furthermore,SPAG5 knockdown in OVCAR3 A2780 and SKOV3 cells reduced Taxol sensitivity.[Conclusion] SPAG5 regulated cell mitosis and promoted cell proliferation in ovarian adenocarcinoma cell lines.Expression of SPAG5 in patients' tumor tissues predicted patients' prognosis and Taxol sensitivity. As the results, individualized treatment of high grade serous carcinoma patients is necessary.

Objective To explore the hurt of susceptibility organs and critical orgens followed by rotavirus (RV) infection of whole body in newborn mouse. Methods RV strain was derived from the stool samples of patients with RV diarrhea and was proved to be long type by methods of ELISA and polyacrylamide gel electrophoresis (PAGE). RV was inoculated by the pathways of taking orally and injected to abdominal cavities, respectively. The pathological changes of the newborn mouse model infected with human natural RV by light microscope and electron microscope. The gene probe was marked by digoxin. The direct prove of RV infection in these organs was got by the detection of in situ PCR. Results Pathological changes were found in the small intestinal villus, lamina propria of the stomach and the heart cells of the mice taken RV orally. The mice with intraabdominal RV injection showed pathological changes of the cells in the small intestinal villus, liver and kidneys observed by electron microscope.Shortened small intestinal villus, nuclear membrane disorganization, massive vacuolization, mitochondrial swelling and rough endoplasmic reticulum dilation were observed in the cells of small intestinal. In the liver of the mice, marked mitochondrial swelling and agglutination, cell nucleus pyknosis or collapse, presence of numerous lipid droplets and vacuoles were found in the li-ver cells, with lymphocyte and plasmacyte infiltration.Obvious dilatation and shedding of the microvillus were found in cholangioles. The mitochondria of the proximal convoluted renal tubule showed mild swelling, but the cells in the heart and lung did not display obvious changes. Conclusion RV can damage lots of extra intestinal organs of the newborn mice if RV diffuses to the whole body of the mice.

Animal models with kidney disease are generally divided into two types. One belongs to the models which imitate human kidney disease by the artificial operations, such as anti-glomerular basement membrane antibody nephritis, Heymann nephritis, anti-Thyl. 1 antibody nephritis, BSA nephritis and puromycin nephropathy. The other one pertains to the models which make themselves kidney disease, and appear the pathological characteristics naturally as like as human, such as HIGA mice with IgA nephropathy and NZB/WF1 and MRL/1pr mice with lupus nephritis. In addition, the transgenic animal models with kidney disease can also be established by the modern molecular biologic techniques including gene knockout and siRNA transfection. As for the studies related with kidney disease in pharmacodynamics and pharmacology of Chinese herbal medicine (CHM), it is important to understand deeply the features of each animal model with kidney disease, and select accurately the proper models according to the different experimental objectives, and then, build the special models provided with the combination of disease with syndrome in traditional Chinese medicine (TCM). Therefore,it is the developmental direction for the further study to establish animal models with kidney disease, which should possess the characteristics of syndrome in TCM.

A 77-year-old man was admitted to our hospital at July 5th,2010 with an unexplained massive pericardial effusion for 10 years. With dyspnea for one month and normal vital signs without pulsus paradoxus, other physical examination included a small amount of moist rale, normal heart sounds, jugular vein engorgement, positive hepatojugular reflux, hepatosplenomegaly and pitting edema of the extremities. The patient had a complex past history with lymph node tuberculosis, primary artertial hypertension, polycythernia vera, chronic renal insufficiency and hypothyroidism (Hashimoto's thyroiditis),and moreover, received a high dose radiation of 31p in 1967. Family history is negative. The patient had no cardiac tamponade or pericardial constriction during 10 years,he received pericardiocentesis twice, and pericardial effusion was exudative with a high proportion of monocyte. There was no evidences of tuberculosis infection, hypothyroidism, malignant tumor, severe heart failure, uremia, trauma, severe bacterial or fungus infection, chronic myeloid leukemia or bone marrow fibrosis during the admission. The patient refused anti tuberculosis, indwelling catheter drainage or surgical therapy. In this rare case, the aetiology of chronic massive pericardial effusion is most probably chronic idiopathic recurrent pericarditis.
