Objective To establish a human colon cancer multi-drug resistant cell line LS174T/5-fluorouracil(5-Fu) and to explore its biological characteristics. Methods A resistant human colon cancer line LS174T/5-Fu was established by stepwise increasing concentrations of fluorouracil selection from the parental cell line LS174T.Drug sensitivity was detected by MTT assay,and the changes of biological characteristics were determined by light microscopy,cell counting,flow cytometry and RT-PCR. Results LS174T/5-Fu cell line was developed after 6 months with stable resistance to 5-Fu and a resistance index of 40.24.The cells exhibited cross-resistance to cisplatin,etoposide,doxifluridine and hydeoxycamptothecin.LS174T/5-Fu cells grew more slowly than LS174T cells,which was longer than LS174T cell line.Cell cycle distribution of LS174T/5-Fu cells was changed compared with parental cells.The percentage of cells in G_(1) and S phase was increased,while in G_(2) phase was decreased.The level of MRP mRNA expression was increased according to the concentration of 5-Fu in resistant cell lines. Conclusion LS174T/5-Fu cells is a reliable multi-drug resistant cell subline of human colon cancer.The successful establishment of this cell subline has laid a solid foundation for further study of the multi-drug resistant mechanism of human colon cancer induced by 5-fluorouracil.

Objective To study the expression level of myostatin in the gastrocnemius muscle of cancer cachexia mice and to observe the improvement of the status and the influence on myostatin expression by use of meloxicam.(Methods The) tumor-bearing cachexia mice model was established by Lovo cell line subcutaneous inoculation.Twenty-four BALB/c mice were randomly divided into 3 groups(n=8 for each group):group A,control;group B,tumor-bearing mice plus saline;group C,tumor-bearing mice plus meloxicam(5 mg/kg).The food intake and body composition were documented.The expression of myostatin in the gastrocnemius muscle was investigated by RT-PCR and immunohistochemistry in all the animals,and the correlation analysis between serum TNF? and myostatin was conducted. Objective To survey the current working status of general practitioners (GPs) who received residency training in Shanghai.Methods The current status of general practitioners,who completed residency training in Shanghai from 2013 to 2014,was surveyed with self-designed questionnaire by e-mail.The survey items included current employers,job specification,clinical practice,teaching,scientific research and so on.Results Three hundred and thirteen out of 377 GPs,who completed residency training from 2007 to 2012,participated in the survey (83.0％),including 85 males and 228 females with a mean age of (31.2 ± 2.6) years.Among all participants,298 (95.2％) worked in community health service centers,12.8％ (40/313) of them were involved in general practice teaching,9.9％ (31/313) undertook management,9.3％ (29/313) did academic research in community,and 7.7％ (24/313) had chance for overseas study.The survey also showed that 134 person/times received various commendations and awards.Conclusion Most GPs who completed residency training are undertaking clinical practice in community health service centers,and they are also well involved in management,teaching,and research.

Objective To observe the clinical efficacy of acupuncture plus medication in treating diabetic abducens nerve palsy.Method Twenty-three cases of diabetic abducens nerve palsy were randomized into a treatment group of 12 cases and a control group of 11 cases. The two groups both were intervened by Compound anisodine hydrobromide via para-temporal-superficial-artery injection, based on which, the treatment group received acupuncture at extraocular muscles and body acupoints, while the control group received subconjunctival injection of Dexamethasone beside the endpoint of rectus lateralis. The fasting blood glucose, eye movement and diplopia were observed before and after the intervention for the two groups to evaluate the therapeutic efficacy.Result After the intervention, the two groups didn't show significant intra-group or inter-group differences in fasting blood glucose (P>0.05). The total effective rate was 91.7％ in the treatment group versus 54.5％ in the control group, and the between-group difference was statistically significant (P<0.05).Conclusion Acupuncture plus medication is an effective method in treating diabetic abducens nerve palsy.

Objective To explore the Oneomehnia distribution on electronic map based on gridding orientation pattern of marshlands in Changsha along Xiangjiang River,in order to manage snail information in a visual and digital way.Methods The data of geographic information and snail information were collected by global positioning system(GPS)and database was established using systematic sampling,and then an information management system of electronic map about the distribution of Oncomelania was designed.The degree of Oncomelania was expressed in different colors,the Oncomelania information in the database on the digital map wag reflected accurately using the geographic information system(GIS)technology.Results The electronic map of the distribution information of 20 marshlands in Changsha along Xiangjiang River were drawn.At the same time,the information could be grasped compreheusively.Digit map contains geographic information and database of snail information,such as the row and rnum,the density of alive and infectious snails,the area and geographic information et al.A method of managing and analyzing snail information with higher efficiency was offerred in this article.Conclusion Conventional methods of managing and analyzing Oncomelania information are reformed into digitalization and visualization.

Objective To explore the rule of central region lymph nodes metastasis in differentiated thyroid carcinoma (DTC) and the significance of central region lymph nodes neck dissection in surgical operation for DTC.Methods The clinical and pathological data of 122 lymph nodes in 109 patients (31 female and 78 male) with DTC undergoing neck lymph nodes ( central region lymph nodes included) dissection from Jan.2003 to Jun.2007 in our hospital were analyzed retrospectively.According to preoperative physical examination and imaging analysis,patients were divided into clinical cervical lymph nodes metastasis ( cN + ) group and clinical no cervical lymph nodes metastasis (cNO) group and compared respectively with their pathological data after surgery.Resuits Of the 122 lymph nodes,lymph node metastasis rate was significantly higher in region Ⅵ than in region Ⅱ,Ⅲ and Ⅳ.The difference had statistical significance (P ＜0.01 ).65.6％ (80/122) metastasis was in the central region.81.2％ (56/69) patients in cN + group and 45.3％ (24/53) patients in cN0 group had central region lymph nodes metastasis.Conclusions Central region lymph nodes metastasis is common for patients with DTC.Routine neck dissection in central region should be done in DTC operation.

Objective To investigate the exposure and protection of recurrent laryngeal nerve (RLN) in the reoperation for thyroid diseases.Methods Clinical data of 214 cases undergoing thyroid reoperation were retrospectively analyzed.The patients with a short interval between the 2 thyroid operations or with external-infiltrated thyroid cancer were approached at the lateral strap muscles and the leading edge of the sternocleidomastoid.RLNs were exposed in the lateral region of superior mediastinum tracheoesophageal groove or at the point where RLN enters to throat.RLNs of patients with lymph node metastasis were exposed beside the enlarged lymph nodes.The patients with a long interval between the 2 thyroid operations and with benign tumor or tumor without external infiltration were exposed their thyroids at the anterior midline and then RLNs were exposed at the posterior lateral of the middle thyroid veins or at the inferior thyroid artery.Results Among the 214 cases,344 RLNs were anatomically exposed including 188 right and 156 left.84 cases had single exposure and 130 cases had bilateral exposure.44 RLNs were exposed at the point where RLN enters to throat,104 RLNs at the posterior lateral of the middle thyroid veins,40 RLNs at the inferior thyroid artery,124 RLNs at the lateral region of superior mediastinum tracheoesophageal groove,and 32 RLNs beside the enlarged lymph nodes.For the 2 cases suffering hoarse voice the day after they underwent thyroid operation in other hospital,suture ligation at the the entrance point was found when they received the reoperation in our hospital.Three of the total 344 RLNs (0.87％ ) had RLN branch injury in the entire group.Conclusion It is possible to reduce RLN injury during the reoperation for thyroid disease if surgeons are familiar with the dissection of RLN under normal or pathological condition,avoid adhesive or scar tissues,and select the appropriate anatomic approach.

Objective To study the effects of resveratrol on acute gouty arthritis.Methods According to random number table,thirty-six Wista rats were randomly divided into six groups:the control group,the model group,the colchicine treatment group,the high dose resveratrol group,the medium dose resveratrol group,the low dose resveratrol group.They were administrated with normal saline,colchicine suspension,high dose resveratrol,medium dose resveratrol,low dose resveratrol for 7 days once daily.Acute gouty arthritis rats models were established on the fourth day by injecting monosodium urate solution (concentration of 25 mg/ml,0.05 ml) into the ankle joint,while 0.05 ml normal saline was injected into the joint of the control rats.Seventy-two hours after the model was established,the synovial fluid were collected from ankle joints for the measuring of the level of IL-1β,CXCL10.NF-κB p65 protein levels were assayed with Western blotting.The joint synovia were fixed for histopathological examination with 10％ fromaldehyde.Variance analysis were used.Results The values of IL-1β in the low,medium and high doses of resveratrol were (9.63±0.71),(7.67±0.48) and (6.66±0.29) pg/ml respectively,being significantly lower than that in the model group (11.85±0.45) pg/ml (P＜0.05).The values of CXCL10 in the medium and high doses of resveratrol were (86.8±2.9) and (89.6±5.5) pg/ml respectively,being significantly lower than that in the model group (110.1±4.0) pg/ml (P＜0.05).The Western blotting results showed that the expression of NF-κB p65 protein in the high dose resveratrol group decreased evidently than that in the model group.The histopathological examination showed that resveratrol might reduce joint edema,decrease inflammatory cells infiltration in acute gouty arthritis.Conclusion The levels of IL-1β,CXCL10 and the expression of NF-κB p65 protein are elevated in acute gouty arthritis.The resveratrol can effectively control acute gouty arthritis attacks,and its efficacy is dose dependent.

To compare the pharmacokinetics of syringin, eleutheroside E and isofraxidin after intravenous administration of each monomer and Ciwujia injection. Twenty-four Sprague-Dawley rats were randomly divided into four groups and intravenously administrated with syringin, eleutheroside E, isofraxidin, and Ciwujia injection, respectively. The concentrations of the three components in rat plasma were determined by LC-MS/MS. DAS 2.0 software was applied to calculate the pharmacokinetic parameters while the SPSS 17.0 software was used for statistical analysis. Significant difference (P < 0.05) was found between each monomer and the injection on the main pharmacokinetic parameters such as AUC, CL and t1,/2. Compared with the injection, the group treated with the syringin has obvious decrease in AUC, and increase in CL while the group treated with eleutheroside E has obvious increase in AUC, and decrease in CL The t1/2 of isofraxidin was prolonged in Ciwujia injection. Pharmacokinetic characters of the ingredients in the injection varied greatly from the monomer. Other constituents in the injection may have an impact on the pharmacokinetic profiles of these three components.
