1.GENETIC POLYMORPHISM OF SIX Y CHROMOSOMAL STR IN CHINESE HUI ETHNIC GROUP

Objective To study genetic polymorphism of 6 Y chromosomal STR in Hui ethnic group living in Ningxia Hui ethnic autonomous region, in order to evaluate their usefulness in forensic science and enrich the Chinese genetic information resources. Methods We investigated 101 unrelated, healthy, male individuals of Hui ethnic group and studied their allelic frequency distribution and haplotype diversity of 6 Y chromosomal STR. Primer for each loci was labeled with the fluorescent by FAM (blue) or TAMRA(yellow). The data of Hui ethnic group were generated co-amplification, GeneScan, genotype, and genetic distribution analysis. Results 31 alleles and 43 phenotype(DYS385) were detected, with the frequencies ranging from 0.0099-0.7129. Out of a total of 101 individuals, 96 showed different haplotypes; 91 were unique; 5 were found 2 times. The haplotype diversity for 6 Y-STR loci was 0.9990. Conclusion The date obtained can be valuable for individual identification, paternity testing in forensic fields and for population genetics because of 6 Y-STR loci high polymorphism.
2.Differentiation of adipose tissue-derived stem cells into myocardial cells and their transplantation for treatment of cardiovascular diseases

BACKGROUND: Adipose-derived stem cells (ADSCs), a kind of adult stem cells, possess plasticity and can be induced into myocardial cells under certain conditions. Autologous ADSCs transplanted into the infarct area can differentiate into myocardial cells and vascular endothelial cells to construct new vessels and thereby improve cardiac pump function. OBJECTIVE: To study the factors that influence ADSCs differentiation and transplantation and the current clinical and laboratory research progress of ADSCs transplantation for treatment of cardiomyopathy.METHODS: A computer-based retrieval was performed in Medline (between January 1990 and April 2010), PubMed database, the China Biological Medicine Database (CBM) (between January 1990 and April 2010), and China National Knowledge Infrastructure (CNKI) with the keywords adipose-derived stem cells, myocardial cells, cell differentiation, cell transplantation, cardiomyopathy treatment.RESULTS AND CONCLUSION: A total of 30 articles, consisting of 6 reviews and 24 randomized controlled trials, were obtained. At present, there have been uniform methods of ADSCs isolation and culture, and ADSCs can be effectively proliferated in vitro, but there have been no direct methods to identify these stem cells. ADSCs differentiation can be induced both in vitro and in vivo, besides, with a characteristic of early differentiation. ADSCs transplantation is a more conductive therapy for myocardial disease compared with bone marrow stem cells (BMSCs) transplantation. Different ADSCs transplantation methods should be carried out in different types of cardiomyopathy. Stem cell labeling technique can help to dynamically monitor implanted in vivo. Transplantion of autologous ADSCs is a new way to treating cardiomyopathy. However, for successes in clinical practice, the method to inhibit tumor cells-promoting characteristics is needed to ensure long-term safety of the patients receiving ADSCs transplantation.
3.Performance Testing of Field Blood Transportation Vehicle during Line-haul

Objective To test the performance of field blood transportation vehicle from the aspects of heat preservation,refrigeration and oil consumption.Methods Two vehicles without power supply were stowed with maximum load,one with red cell suspension,and the other with frozen plasma.With the environmental temperature higher than 35 ℃ or lower than 25 ℃,related time lengths were detected respectively.Results The holding time of the vehicles was from 1.5 h to 7 h,refrigerating time was from 15 min to 138 min,and a full tank of gasoline was exhausted around 5~7 hours.Conclusion During line-haul,field blood transportation vehicle has to be equipped with additional cooling agents,in case the refrigeration equipment out of work for the reason of gasoline exhaustion.
4.Efficacy observation of acupuncture for dry eye syndrome of lung-yin deficiency pattern

Objective: To observe the clinical efficacy of combining two needling manipulations, Er Long Xi Zhu (two dragons playing with a pearl) and Guo Yan Re (heat produced to reach the eyes), in treating dry eye syndrome (DES) of lung-yin deficiency pattern. Methods: Fifty-six eligible DES patients of lung-yin deficiency were randomized into an observation group and a control group, with 28 cases in each group. Same acupoints were selected in the two groups: Cuanzhu (BL 2), Sizhukong (TE 23), Taiyang (EX-HN 5) and Fengchi (GB 20) were chosen as the major points and Feishu (BL 13) and Chize (LU 5) as the adjuvant. Er Long Xi Zhu and Guo Yan Re needling manipulations were applied in the observation group while twirling reinforcing manipulation was used in the control group. Treatment was conducted once per day in both groups, for two sets of 15 consecutive days at a 2-day interval. Changes in the symptom score, tear break-up time (BUT) and tear production were observed afterwards, and the clinical efficacy was also compared between the two groups. Results: The total effective rate was 92.8% in the observation group, higher than 71.4% in the control group, and the between-group difference was statistically significant (P<0.05). After treatment, the symptom score, tear BUT and tear production showed significant improvements in both groups (all P<0.05); the symptom score, BUT and tear production in the observation group were significantly different from those in the control group (all P<0.05). Conclusion: Given the same acupoint selection, combining Er Long Xi Zhu and Guo Yan Re needling manipulations can produce more significant clinical efficacy than twirling reinforcing manipulation in treating DES of lung-yin deficiency pattern.
5.AngioJet rheolytic thrombectomy for the treatment of acute deep venous thrombosis of lower extremity

Objective To discuss the efficacy and complications of using AngioJet rheolytic thrombectomy in treating acute lower extremity deep vein thrombosis (DVT).Methods The clinical data of 22 patients with acute lower extremity DVT,who were treated with AngioJet rheolytic thrombectomy during the period from February 2015 to August 2016,were retrospectively analyzed.The improvement of clinical symptoms and the thrombus clearance rate were calculated to evaluate the curative effect.The procedure-related complications were documented.Results The clinical symptoms were relieved immediately after operation in all 22 patients.The thigh circumference difference between the affected side and the healthy side decreased from preoperative (4.5±0.6) cm to postoperative (1.0±0.4) cm,the difference in change was statistically significant (P<0.05).The mean used dose of urokinase was (0.18±0.03) million unit and the average duration of thrombolysis was (4.2±0.7) hours.Complete removal of DVT (>90%) was achieved in 19 patients,most removal of DVT (50%-90%) in 2 patients,and partial removal of DVT (<50%) in one patient.After treatment,6 patients developed transient hemoglobinuria,which was relieved after hydration with fluid infusion on the same day.No serious complications such as pulmonary embolism or hemorrhage occurred.Conclusion For the treatment of acute lower extremity DVT,AngioJet rheolytic thrombectomy is safe and effective with less complications.
6.The Metabolomic Study of Learning and Memory Function of APP/PS1 Double Transgenic Mice Improved by Gouteng San

Objective:The non targeted high-throughput urine metabolomics technology was used to study the pathogenesis of APP/PS 1 double transgenic mice and the mechanism of action of Gouteng san.Methods:5-month-old APP/PS 1 double transgenic mice were test with Morris water maze for spatial learning ability.Then we employed the non targeted high-throughput urine metabolomics technology to study the pathogenesis of APP/PS1 double transgenic mice based on the metabolic network.The focus investigation of the key pathways and the observation of the treatment by Morris water maze and metabolic level have been used after spatial learning ability damaged confirmed.Results:The comparison between APP/PS1 double transgenic mice and normal mice suggested that a significant longer was existed in former,which was call-back by Gouteng san.With the non targeted high-throughput urine metabolomics analysis and pathway focused analysis,we found certain signals from metabolic profiling,which was identified to be 6 biomarkers associated with learning and memory function by mass spectrometry analysis or authoritative database.Respectively,they were taurine,pteroylglutamic acid,neopterin,glutaurine,2-oxoglutarate and dihydroneopterin.They were mainly related to taurine metabolism and folate metabolism and represented an effective callback.Conclusion:Gouteng san possess a favorable effect on learning and memory ability of APP/PS1 double transgenic mice,6 biomarkers may be a potential target for the pathogenesis of APP/PSI double transgenic mice and provide experimental basis for the study of Gouteng san.

Objective To investigate the effects on structure of madibles and femurs in ovariectomized rats. Methods Ovariectomy was performed on female SD rats, which were killed 8 weeks later. The mandibles and femurs were taken out as samples to be analyzed by bone histomorphometry and to calculate trabecular area(Tb.Ar), trabecular width(Tb.Wi), trabecular separation(Tb.Sp). 8. Effect of Celastrus orbiculatus extract on growth and apoptosis-related protein expressions of human gastric adenocarcinoma SGC-7901 cell xenografts in nude mice

To explore the effects of Celastrus orbiculatus extract (COE) on the expressions of apoptosis-related proteins and growth of human gastric adenocarcinoma SGC-7901 cell xenografts in nude mice. Thirty-five mice with subcutaneous xenografts of a gastric cancer cell line (SGC-7901) were randomly divided into five groups: a negative control group, a positive control group (xeloda), and low-dose, medium-dose, and high-dose COE groups (10, 20, and 40 mg/kg, respectively). There were seven mice in each group. Tumor volume, tumor weight, and body weight were measured to draw up tumor growth curves and the tumor weight inhibition rates were calculated. The apoptosis rates were measured by TUNEL staining method and the expressions of p53, Bcl-2, and Bax proteins were detected by immunohistochemical staining and Western blotting assay. The COE could inhibit the growth of SGC-7901 xenografts in a dose-dependent manner. The tumor inhibitory rates were 33.3%, 46.8%, and 57.7% when treated with 10, 20, and 40 mg/kg COE, respectively. TUNEL assay showed that COE presented with apparently more apoptosis than the negative control group. The results of immunohistochemical staining showed the expressions of p53 and Bax proteins had a trend of up-regulation, while the expression of Bcl-2 protein had a trend of down-regulation in nude mice after treatment with COE. Compared with the negative control group, the expressions of p53 and Bax were significantly up-regulated, while the expression of Bcl-2 was down-regulated in the high-dose COE group. Western blotting analysis showed the similar results to immunohistochemical staining. COE could significantly inhibit the growth of SGC-7901 cell xenografts in nude mice, which might be related with the up-regulation of the expressions of p53 and Bax and down-regulation of the expression of Bcl-2.
9.Electroencephalogram Non-linear Analysis on Menopause Depression Subjects

ObjectiveTo investigate the changes of non-linear dynamics characteristics of EEG of the patients with menopause depression under eyes closed state.MethodsEEG were recorded in 18 patients with menopause depression and 18 healthy volunteers under eyes closed state. Approximate entropy (ApEn) was calculated for all subjects.ResultsApEn of the menopause depression group at eyes closed state increased significantly compared with that of the healthy control group.ConclusionDynamic non-linear analysis is more appropriate for the study of mental functions changes of menopause depression.
10.Effects of Different Soil Water Content on the Yield and Quality of Rehmannia Glutinosa

This study aimed to elucidate the effects of different soil water contents on the yield and quality of R.glutinosa.Different soil water contents were adjusted in different periods of growth of R.glutinosa.The yield,content of water extract,catalpol,verbascoside and polysaccharide were determined and analysed by the grey pattern recognition after harvests.The impacts of soil water content from the most important to the least important were medium moisture content (M2),high moisture content (M3),low moisture content (M1) and blank (M4).In the cultivation of R.glutinosa,the soil water content should be remained in the range of 40%-50% at seedling stage,while 50%-60% at the stage of root formation and tuber enlargement,and 20%-30% during harvesting,which can significantly improve the yield and quality of R.glutinosa.
