Objective To investigate the effect of co-expression of neuropilin-1(Nrp-1)and transforming growth factor-β(TGF-β)on regulatory T cell(Treg)-mediated immunosuppression. Methods CD4+CD25+Tregs were isolated from the spleens of male Balb/c mice. CD4+CD25+Tregs were blocked with various doses of Nrp-1 antibody(Ab-Nrp-1,0.5,5,10 μg/ml)for 24 hours with anti-CD3/CD28 and lipopolysaccharide(LPS)stimulation, and expression of Nrp-1 and TGF-βwas determined by flow cytometry. Meanwhile,CD4+CD25+Tregs were cultured with different doses of Ab-Nrp-1 for 1 hour,and co-cultured with CD4+CD25-T cell for 12,24 and 48 hours respectively,the proliferative activity of CD4+CD25-T cells was analyzed by microplate reader. Results Compared to control group,the expressions of Nrp-1 and TGF-β were significantly increased under anti-CD3/CD28 and LPS stimulation(both P<0.05),and treatment with Ab-Nrp-1 markedly inhibited the expression of TGF-β in a dose-dependent manner(P<0.05). The normal Treg had the potential to inhibit the proliferation of CD4+CD25-T cells(P<0.05),while various doses of Ab-Nrp-1 had the ability to reverse the immunosuppressive function of CD4+CD25+Treg in a dose-and time-dependent response manner,5μg/ml has the strongest ability,expecially after 24 hours. Conclusion Treg cell plays an important role in mediating immunosuppressive response via membrane associated TGF-β,and co-expression of Nrp-1 can markedly promote the immunosuppressive function.
2.Effect of compound total extract ginseng and tall gastrodia tuber on content of monoamine transmitters in brain tissue of rats with vascular dementia

BACKGROUND: During vascular dementia, ischemia, hypoxia, energy expenditure, abnormal metabolism of neurons, decrease of generation of Adenosine Triphosphate and disorder of ionic environment in and out of cells are observed in brain tissue, which can cause abnormal release of monoamine transmitter.OBJECTIVE: To probe into effect of shenma yizhi capsule on content of monoamine transmitter in brain tissue of rat models with vascular dementia induced by multiple cerebral infarction.DESIGN: Randomized controlled study.SETTING: Pathological and Physiological Department o Liaoning Basic Medical Institute.MATERIALS: Totally 96 Wistar rats of either gender,aged 8-12 months, weighing 270-500 g, were selected. METHODS: The experiment was completed in the Pathological and Physiological Department of Vocational-technical College, Liaoning College of Traditional Chinese Medicine from April to July 2001. All rats of either gender were divided into 6 groups with 16 in each group. Wistar rats in 5 groups were injected with cruor embolus in internal carotid artery to make animal model of vascular dementia induced by multiple cerebral infarction.After modeling, rats were randomly divided into 3.2 g/kg, 1.6 g/kg and 0.8 g/kg shenma yizhi capsule groups (shenma yizhi capsule was extracted from ginseng and tall gastrodia tuber with 2.7 g raw materials and provide by Xiyuan Hospital of Chinese Academy of Traditional Chinese Medicine),positive control group and dementia control group. Animals without modeling were regarded as normal control group. Rats in each dosage group were perfused with the corresponding dosage of shenma yizhi capsule; rats in positive control group were perfused with 1 mg/kg hydergine dihydroergotoxine; rats in dementia control group and normal control group were perfused with the same volume of saline solution. One week after modeling, rats were medicated once a day for 6 weeks. Content of monoamine transmitter was measured with high performance liquid chromatograpy.MAIN OUTCOME MEASURES: Content of monoamine transmitter such as levarterenol, adnephrin, dopamine, indoleacetic acid, homovanillic acid and 5-serotonin.RESULTS: Nine rats died because of self-body quality and environmental change, and totally 87 animals entered the final analysis. Content of levarterenol of rats in dementia control group was lower than that in normal control group [(0.40±0.23), (0.70±0.14) ng/g, t=2.712, P < 0.01]; content of levarterenol of rats of 3.2 and 1.6 g/kg dosage groups was higher than that of dementia control group [(0.57±0.09), (0.58±0.19), (0.40±0.23) ng/g,t=2.211, P < 0.05], but was lower than that of normal control group. Content of levarterenol of rats of 0.8 g/kg dosage group was lower than that of normal control group [(0.48±0.23) ng/g, t=2.213, P < 0.05], but was higher than that of dementia control group and positive control group [(0.41 ±0.19) ng/g]. Differences of other neurotransmitters were not significant.CONCLUSION: Content of levarterenol in brain tissue of rats of dementia control group is decreased obviously, but shenma yizhi capsule can increase content of levarterenol in brain tissue of rats. The mechanism of shenma yizhi capsule on treating vascular dementia is possibly related with increasing content of levarterenol in brain tissue.
3.Regulation of estrogen, interleukin-6 and interleukin-8 in ovarian cancer cells

Objective To discover the reciprocal regulation and its molecular mechanism of estro-gen, IL-6 and IL-8 in ovarian cancer cells. Methods Based on our previous studies, the effect of 17β-estradiol (E2) on the expression levels of IL-6, IL-8 and their respective receptors was investigated. Mean-while, the effect of IL-6/IL-8 on estrogen receptor (ER) expression and estrogen-dependent transcriptional activation was analyzed. Gene expression profile analysis revealed that CAOV-3 and OVCAR-3 cells, which express ER, IL-6 and IL-8 receptors, were suitable models for this study. Results We found that E2 not only enhanced IL-6/IL-8 secretion via NF-κB signaling pathway, but also modulated IL-6 and IL-8 receptors expression. Tamoxifen (Txf), an ER antagonist, completely abolished E2-stimulated IL-6/IL-8 expression. On the other hand, in the absence of estrogen, both cytokines increased ERα expression, decreased ERβ ex-pression, and activated estrogen-dependent transcriptional activation, which was completely blocked by Txf. Pretreatment of OVCAR-3 with p38 MAPK, MEK1/2 or ErbB2 MAPK inhihitors, respectively, IL-6-media-ted ER activation was blocked, while IL-8-indueed ER activation was blocked by Src inhibitor. Conclusion These data suggest that estrogen, IL-6 and IL-8 may form a mutual amplifying signaling which contributes to the growth and development of ovarian carcinoma.
4.Endoscopic ultrasonography analysis of 420 cases with eminence lesions in the gastrointestinal tract

Objective To evaluate the diagnostic value of endoscopic ultrasonography(EUS) in the eminence lesions of gastrointestinal tract.Methods The clinical data of 420 patients with gastrointestinal eminence lesions subsequently received EUS were analyzed.Results In 420 cases,235 cases lesions were located in the stomach,129 cases in the esophagus,29 cases in the duodenum and 27 cases in the colon.Occurred in the wall itself of the digestive tract lesions was 85.00% (357/420),and 73.10 %(307/420) of submucosal neoplasm,and gastrointestinal stromal tumor,leiomyoma or leiomyosarcoma was seen [60.24% (253/420)].Lesions was not found in the digestive tract wall external oppression and lesions accounted for 14.52%(61/420),external lesions and infiltrated digestive tract was 0.48%(2/420).Conclusions EUS is an ideal diagnostic modality in distinguishing different layers of the wall of the gastrointestinal tract,and thus of important value in precise localization of lesions beneath the mucosa.And be able to hint the nature of the lesion.Meanwhile,this may be associated with colonic mucous membrane structure.
5.A new recombination type of B/C genotype was discoved in hepatitis B virus

Objective To study the recombination type of B/C genotype in hepatitis B virus.Methods The PCR was applied to amplifed the whole genes of HBV through the serums of four chronic HBV carriers who come from Jinghong distict,Yunnan province.The whole HBV genomes were ligated with pMD18-T vector and trasformed to E.coli JM109.After the positive colones were picked up,the HBV genotypes and recombinated sites were discoved through sequenced the acquired positive colones.Results All the acquired sixteen HBV sequences from the four HBV carriers were genotype B which were combinated with genotype C in some region.There are two ways of the combinations.For the first one,a 496 bp fragment from genotype C taked place the genotype B at the place of nt1825 to nt2320 of precore C/C region.For the second way,a 695 bp fragment of genotype B taked place at the both sites of nt822 to nt1020 of P gene region and nt1825 to nt2320 of precore C/C region.Conclusion A new recombination type of B/C genotype in hepatitis B virus was reported for the first time.The new Bj subgenotype was combinated with genotype C not only at the region of precore C/C but also at the place of P gene region.
6.Effect of Ganciclovir in Young Children Hospitalized with Severe Rotavirus Enteritis and Longitudinal Change of Rotavirus Excretion

Objective To study the effect of injection ganciclovir in infants with rotavirus disease.Methods According to age (6 months to 2 years) and typical clinical symptoms in combination with etiologic evidence of rotavirus, 76 patients within 2 days after onset were selected as study subjects. These young children were randomly assigned to two groups according to the hospitalized order.Treated group received intravenous administration of ganciclovir 5~10 mg/(kg?d) once daily for 3 days while control group didn′t receive any antivirus drugs. Rotavirus testing by ELISA on stool samples was performed for every patient on admission and the third day after treatment. Stool sample was collected to a clear box every day in patients with positive results until the reaction was negative.Results The total effective rate after treatment was 88.1% and 61.8% in treated group and the controll group, respectively. Objective To explore the influence of different dose of Helicobacter pylori on the expression of transforming growth factorβ(TGF-β1) and B7-H1 in the infected gastric mucosal epithelial cell and the bacterial factors which influence the expression of TGF-β1.To confirm that H.pylorican induce the expression of TGF-β1 and B7-H1 to inhibit the host immune function in gastric mucosal epithelial cell.Methods (1) We investigated the expression of TGF-β1 of human gastric mucosal epithelial cells infected with different concentration(1.0 × 109 CFU/ml,4.0 × 109 CFU/ml,8.0 × 109 C FU/ml) of H.pylori(NCTC 11637) in different time-point(0 h,0.5 h,1 h,1.5 h,2 h,4 h,8 h,12 h),and compared with the expression of TGF-β1 between the deactivated H.pylori group and activated H.pylori group.The blank group is the gastric mucosa epithelial cells which does not infect H.pylori.To detect expression of TGF-β1 in infected cell culture supernatant by enzyme-linked immunosorbent assay(ELISA) and the expression of B7-H1 mRNA by in situ hybridization.(2)At the same time,the middle concentration of deactivated H.pyloriand in vitro gastric mucosal cells were incubated for 2 h and 12 h,to detect expression of TGF-β1 in the cells and cell culture supernatant.(3)In vitro gastric mucosal cells were incubated with H.pylori bacterial supernatant and sedimentation by ultrasonic crushing and centrifugation and with H.pyloribacterial supernatant and sedimentation after boiling respectively,to detect expression of TGF-β1 in the cells and cell culture supernatant after 2 h,12 h.Results (1)Compared to the control group,the expression of TGF-β1 was significantly increased after stimulation with different concentration of activated H.pylori in different time-point(P <0.05).The expression of TGF-β1 secretion group has a similar dynamic trend,and the highest expression is the middle concentration group(P <0.05).(2)There was no difference between the middle concentration of deactivated H.pylori group and the same concentration of activated H.pylorigroup(P > 0.05).(3) The expression of TGF-β1 in the H.pylori bacterial supernatant group was significantly increased higher than the blank group and the H.pylori bacterial sedimentation group(P <0.05),and the H.pylori bacterial supernatant group after boiling was significantly lower than the H.pylori bacterial supernatant group(P < 0.05),but there was no difference between H.pylori sedimentation group after boiling and not boiling(P > 0.05).The B7-H1 expression of different concentration groups which the H.pylori and gastric mucosal epithelial cells cocultured 12 h are higher than the blank group(P < 0.05) by in situ hybridization,and the middle concentration group is the highest expression.TGF-β1 and B7-H1 mRNA are positively correlated(r,=0.628,P <0.01).Conclusion H.pylori can induce the gastric mucosal epithelial cells to express the TGF-β1,the factor was the soluble protein in the H.pylori thalline.At the same time,H.pylorican induce the B7-H1 expression increased.In gastric mucosal epithelial cells,TGF-β1 and B7-H1 mRNA are positively correlated.So H.pylori can inhibit the host immune response and participate the process of immune escape by increased the expressions of TGF-β1 and B7-H1.
8.Analysis on prevalence of syphilis infection and its influence factors among male STD attendants

Objective:To analyze the prevalence of syphilis infection and influence factors of syphilis infection among the male STD attendants in Songyuan City of Jilin Province, and to provide theoretical basis for the development and implementation of syphilis intervention policy.Methods:A questionnaire survey and serological tests were conducted among the men who went to STD clinic for treatment in the ZhanDong Dermatology Specialist Hospital or Qianguo County Hospital in Songyuan City from 2011 to 2015.Chi-square test was performed to compare the prevalences of syphilis infection between different groups.Multivariable Logistic regression model was used to find the independent factors of syphilis infection among the male STD attendants.Results:Of all 2 000 male STD attendants who involved in the study,the mean age was (34.50±9.03)years old.218 persons (10.90%)of them had sexual behavior with female sex workers in the last three months,433 persons (21.65%)of them had temporary sexual behavior in the last three months,42 persons (2.10%)of them had homosexual behavior in the last three months.Of the respondents,238 persons (11.90%)had a previous diagnosis of sexually transmitted diseases,86 persons of them had gonorrhea (36.13%),43 persons had syphilis (18.07%),15 persons had genital tract Chlamydia (6.30%),55 persons had genital herpes (23.11%),40 persons had genital herpes (16.81%),and 1 person had combined infection of gonorrhea and syphilis. The prevalence of syphilis of 2 000 male STD attendants was 6.4%.From 2011 to 2015,there was a decrease in the prevalences of syphilis (χ2 =44.25,P <0.001).In the last 3 months, the risk of the male STD attendants who had commercial sex with prostitute,temporary sex behavior,homosexual behavior and a history of STD infection to be infected with syphilis in the past were 3.75 times (OR= 3.75,95% CI:2.46 - 5.71),2.31 times (OR = 2.31,95% CI: 1.56 - 3.41),2.97 times (OR=2.97,95%CI:1.33-6.64)and 1.69 times (OR=1.69,95%CI:1.07-2.67)than those who did not have those behaviors,respectively. Conclusion:There is a significantly decreasing tendency in the prevalence of syphilis among the male STD attendants in Songyuan City in the past 5 years.The high risk sexual behaviors are the main influence factors of syphilis infection among them.

Objective To observe the cough-preventing and asthma-relieving effect of Shaonianhong Fanxing Anti-asthmatic Syrup(SFAS).Methods The cough-preventing effect was observed in the experiment of cough induced by ammonia in mice;the phlegm-removing effect was observed in the experiment of eliminating phlegm with phenol red in mice;and the and-asthmatic effect was observed in the experiments of histamine inducing asthma and isolated tracheal smooth muscles of cavies.Results SFAS Can prolong the coughing time induced by ammomia,enhance the excretion of sputum,extend the delitescence period of ohosphoric acid hismmine induced asthma and restrain the contraction of isolated tracheal smooth muscles.Conclusion SFAS has phlegm-removing,cough-preventing and anti-asthmatic effects.

The genus Nodulisporium, is known to produce secondary metabolites with structural diversity. A new alkaloid, 2-hy- droxy-1,1-dimethyl-1,2,3,9-tetrahydro-4H-carbazol-4-one(1), was isolated from the extract of a fungal strain Nodulisporium sp. fermented with rice, together with three known phenols, tyrosol(2), hydroxytyrosol(3), and hydroxytyrosol acetate(4). Their structures were identified by detailed spectroscopic analyses.
