Journal of International Oncology 2013;(6):460-462
Arsenic trioxide (As2O3)is the main component of traditional Chinese medicine arsenic, which was initially used in the treatment of blood system diseases. In recent years, studies have found that it also has a good effect on a variety of solid tumors, especially gastric cancer. It has been confirmed that As2O3 can inhibit proliferation and metastasis of gastric cancer cells, induce apoptosis and protective autophagy, inhibite lymphatic formation and angiogenesis, affect the expression of telomere-related proteins in gastric cancer cells.
2.Effects of heme oxygenase-endogenous carbon monoxide on mortality during septic shock in rats
Chinese Journal of Emergency Medicine 2009;18(2):144-148
Objective To investigate the effects of hemo oxygenase-endogenous carbon monoxide (HO-CO) on mortality of septic shock in rats. Method Eighty healthy Sprague-Dawley rats were randomly divided into four groups (n=20) :group-C,group-Z, group-SS and group-LZ. MAP was monitored continuously, and deaths within 7 days were recorded. Evans blue in lung, ALT, AST, Cr and BUN in plasma, SOD and MDA in kidney, lung and liver, and HO-1 mRNA,HO-2 mRNA,HO-1 protein and HO-2 protein in kidney, lung, femoral artery and liver were measured.Data were analyzed usiny ANOVA. Results ① Mortality in group-SS was lower than that in group-LZ (P<0.05). ②MAP in group-LZ was greater than that in group-SS (P<0.05). ③ Plasma ALT, AST, Cr and BUN,and MDA contents of liver, kidney and lung, and Evans blue content of lung in group-SS were lower than those in group-LZ (P<0.05). Contrarily, the plasma levels of COHb and SOD activity in liver, kid-ney and lung in group-SS were greater than those in group-LZ (P<0.05). ④ HO-1 mRNA and HO-1 protein levels in liver, kidney, femoral artery and lung tissues in group-LZ were lower than those in group- SS (P<0.05). There were no significant difference in HO-2 rnRNA and HO-2 protein found between one another of four groups (P>0.05). Conclusions The up-regulation of HO-1 protein followed by increase CO can reduce the mortality during septic shock, while the hypotension, which is partly attributed to HO-1 protein and CO,matters little to mortality.
3.Protective effects of allopregnanolone against different seizure models in mice
Chinese Journal of Clinical Pharmacology and Therapeutics 2000;5(2):97-100
Aim To examine the protective effects of allopregnanolone against seizure on different animal models. Methods The protective effects of allopregnanolone against maximal electrical seizure (MES) and picrotoxin-induced seizure were studied in C57 mice 15 minutes after vehicle or drug intraperitoneal administration. Results In the MES test, we found that pretreatment with the phenobarbital or allopregnanolone produced a dose-dependent protective effect against seizure. The potency (ED50 value) of phenobarbital in the MES test was 2.40 mg·kg-1, with 95% confidence interval range from 1.22 to 4.72 mg·kg-1. The potency (ED50 value) of allopregnanolone in the MES test was 0.086 mg·kg-1, with 95%confidence interval range from 0.037 to 0.201 mg·kg-1, which was significantly higher than that of phenobarbital (P<0.01). The combination study of half ED50 values of phenobarbital and allopregnanolone resulted in a 80% of protective effect in MES test, which was higher than 50% produced by either phenobarbital or allopregnanolone at their ED50 values respectively. This result indicated that there was a synergism between phenobarbital and allopregnanolone in their anticonvulsant activities. In the picrotoxin test, we found that pretreatment with the allopregnanolone also produced a dose-dependent protective effect against picrotoxin-induced seizure. The potency of allopregnanolone in the picrotoxin seizure test was 0.123 mg·kg-1, with 95% confidence interval range from 0.058 to 0.263 mg·kg-1.Conclusion Allopregnanolone(ip) could protect different seizures in a dose-dependent manner, had a higher potency than phenobarbital, and had synergism with phenobarbital in the MES test.

5.CT features and clinicopathologic analysis of eosinophilic lymphogranuloma in head and neck region
Journal of Practical Radiology 2015;(3):376-378,383
Objective To explore the CT,clinical, and pathological features of eosinophilic lymphogranuloma(ELG)in head and neck region. Methods CT,clinical, and pathological data of 9 cases of ELG in head and neck region, confirmed by biopsy were retrospectively an-alyzed.Results Parotid gland was lonely involved in 3 cases, and cervical lymph nodes were lonely involved in 2 cases, and they were both involved in 4 cases. All of the patients suffered from painless nodule or mass in head and neck region with peripheral blood eosinophilia. On CT examination, nodular lesions of parotid gland were clear or slightly blurred defined with moderately or obviously homogeneous enhancement, and diffuse mass lesions in parotid gland were ill defined with involvement of surrounding tissue, and heterogenous progressive enhancement. The involved cervical lymph nodes were enlarged with clear boundaries, and moderately or obviously homogeneous enhancement. The pathological findings of ELG included the marked infiltration of eosinophilia, lymphocytic proliferation, lymphatic follicles, variable degrees of fibrosis and vascular proliferation. Conclusion ELG in head and neck region has some CT features, and diagnosis could be suggested combining with the clinical manifestations and peripheral blood eosinophilia. The final diagnosis depends on histopathological examination.
6.Effect of antioxidants on secondary conformational transition of aged amyloid ?-peptide_( 1-40) by FT-IR quantitative study
Chinese Journal of Pathophysiology 1989;0(06):-
AIM: To investigate the mechanism that antioxidants TA9901, inhibit the formation of amyloid-?-protein(A?) fibril. METHODS: Fourier-transform infrared spectroscopy was used to study the secondary structure changes on aging A? in vitro. RESULTS: A? aged alone for 30 min, the content of ?-pleated sheet and ?-turn were 43.17% and 32.9% respectively. A? aged alone for 7 days, the content of ?-pleated sheet increased abuot 10% and produced a shift of random coil toward ?-pleated sheet. TA9901 induced a significant decrease of the content of ?-turn (23.5%) and ?-pleated sheet (26.4%). VE mainly decreased the ?-pleated sheet content (30.8%). The combination of TA9901 and VE promoted transition of ?-turn (16.7%) toward ?-helix and random coil. CONCLUSIONS: Both of TA9901 and VE can effectively diminish the ?-structural content. TA9901 showed more intensitive inhibition than VE. The effect of TA9901 on the secondary structure of aged A? was associated with the mechanism that TA9901 inhibited A? aggregation and fibril formation.

10.Data quality in clinical trials: the role of blind review.
Acta Pharmaceutica Sinica 2015;50(11):1498-501
Blind review is one of the most important milestones in clinical trials, which connects data management process to statistical analysis. During blind review, data quality should be reviewed and assessed on both data management and statistical aspects. The primary work of data managers in blind review is to ensure the accuracy of data before it is handed over to biostatistics group. Database auditing, listing data reviewing and reconciliation should become a good clinical data management practice. Statisticians, on the other hand, will focus on quality findings related to protocol deviations or protocol violations. To investigate the protocol deviations and/or violations and relevant impacts on data outcomes, it is important to provide the essential basis of data quality through the blind review, and to assess the reliability of study outcomes.
