1.Inhibitation of Grb2 and anti-tumor therapy
Cancer Research and Clinic 2008;20(11):790-792
The growth factor receptor -bound protein-2 (Grb2) is an adapter protein in cells. Over expression of Grb2 is found in many kinds of tumor and plays an important role in tumor proliferation and progression. So it may be a molecular target for anti-tumor therapy.

Objective To observe the effects of arsenic trioxide(As2O3)on the expressions of TNF-?,Fas and bcl-2 in lung adenocarcinoma cells(LAC)and to explore the mechanism of arsenic trioxide inducing apoptosis. Methods The expressions of TNF-?,Fas and bcl-2 in lung adenocarcinoma cells pretreated by arsenious acid were determined by the double antibody sandwich ABC-ELISA method. Results Compared with the control group,As2O3 showed no effects on the contents of bcl-2 in lung adenocarcinoma cells after 72 hours treatment, but increased the contents of TNF-? and Fas significantly, and the effects in different concentration groups had significant differences. The protein expressions of TNF-? and Fas showed a tendency of concentration-dependent increasing. Conclusions The results suggest that As2O3 induces the apoptosis of LAC cells possibly by up-regulating the expression of TNF-? and Fas.

【Objective】 To explore the therapeutic mechanismof arsenic trioxide(AS2O3),the active component of arsenolitumininhibitingtumor.【Methods】Humanlung adenocarcinoma cells(LAC) at logarithmic growth phase were culturedin vitro for 24 hours, and then were cocultured with arsenic trioxide(AS2O3) in the dosages of 0.75,1.0,1.5 and 2.0 mg/Lrespectively.Meanwhile,model control group was set up.The morphological features of LAC in different groups were observed under reverse microscope,growth-inhibitory rate of LAC was examined by methylthiazolyltetrazolium(MTT) assay, andthe expression of p53 gene was detected by enzyme-linked immunosorbent assay(ELISA) method.【Results】AS2O3in different dosages obviously inhibited the proliferation of LACin a time-effect manner, and markedly increased the expression of p53 gene(P

Chinese Journal of Tissue Engineering Research 2014;(7):1109-1114
BACKGROUND:cells under mechanical stimulation can achieve their biological functions by converting mechanical signals into chemical signals through certain signal transduction mechanism. As the fibrous framework throughout a cell, cytoskeleton is one of the critical components in this process.
OBJECTIVE:Through systemical y analyzing the role of the cytoskeleton in mechanical signal transduction, to provide a potential therapeutic target for the clinical treatment of cytoskeleton related diseases.
METHODS:In order to search relevant articles about the mechanics mechanism of signal transduction of cytoskeleton from PubMed and CNKI databases (from 1990 to 2012), a computer-based search was performed, using the key words of“cytoskeleton, microtubules, microfilaments, intermediate filaments, mechanical stimulation, signal transduction”in English and Chinese, respectively. After eliminating literatures which were irrelevant to research purpose or containing a similar content, 48 articles were chosen for further analysis.
RESULTS AND CONCLUSION:Mechanical stimulation plays an important role in cellproliferation, development and apoptosis. With the gradual understanding of the biological function of cytoskeleton, people have found that cytoskeleton is one of the critical components in the process of the mechanical signal transduction. After getting mechanical stimulation, cytoskeleton can be reorganized through Rho, protein kinase C, integrin and mitogen-activated protein kinase signaling pathways, then converting the mechanical stimulation to chemical signals and finishing its biological functions final y.

Objective To study the clinical efficacy of naloxone combined with edaravone on acute alcoholism. Methods One hundred and twenty-five patients with acute alcoholism were randomly divided into the control and treatment groups. The control group accepted conventional treatment and taking naloxone, while the treatment group accepted edaravone on the basis. Before and after treatment, the treatment recovery time, recovery respiratory rate time and symptom disappearance time were recorded. The malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), neuron-specific enolase (NSE), S-100βand vascular cell adhesion points-1 (VCAM-1) were detected. Results After treatment, the recovery time and recovery respiratory rate time of the treatment group were significantly shorter than those of the control group ( <0.05) . In the control group, S-100β was significantly improved after treatment ( <0.05) . NSE, S-100β, VCAM-1 and MDA have significantly decreased ( <0.05) after treatment in the treatment group. Compared with the control group, there were significant differences of S-100β,VCAM-1 and MDA in the treatment group before and after treatment (<0.05) . While SOD and GSH have significantly increased ( <0.05) after treatment and showed significant differences compared with the control group ( <0.05) . Conclusion Naloxone and edaravone combination therapy can improve oxidative stress and nerve damage, which help early rehabilitation in patients with acute alcoholism.

Objective To understand the knowledge, attitudes and impact factors related to voluntary counseling and testing(VCT)for HIV/AIDS among migrants in Minhang district of Shanghai.Method Using a convenience sampling strategy,74 migrants,27 key informants and 20 community service providers were selected to participate in in-depth interviews and focus group discussions. Results Most migrants were young, less educated and poorly aware of HIV prevention.Only one third of the migrants heard of VCT,but few of them knew where to seek VCT or had any personal VCT experience.Nearly half of the interviewees took a negative attitude towards those who participated in VCT service. The main factors influencing the participation in VCT service among the migrants were the knowledge of VCT,having risk behavior and perception, supportive environments and availability of HIV/VCT services. Conclusions There is an urgent need to promote extensive health education about HIV/VCT,adopt flexible and available approaches to VCT service and provide appropriate care following the HIV test among migrants.

Chinese Journal of Radiation Oncology 2017;26(5):598-602
Myocardial fibrosis is a predominant pathological change of radiation-induced heart disease (RIHD) in late stage. It often occurs several or more than ten years after radiotherapy and can lead to myocardial remodeling, impaired cardiac function, and heart failure. At present there is no effective method to prevent or reverse the development of radiation-induced myocardial fibrosis. Many cells, cytokines, and other factors are involved in the development and progression of myocardial fibrosis in RIHD and some of them have been validated. But most investigators focused on the pathological changes and related mechanisms in early stage, and myocardial fibrosis was just regarded as an endpoint event. The definitive mechanisms of myocardial fibrosis in late stage remain unclear. This paper reveiws the studies about general mechanisms of myocardial fibrosis in RIHD and summarizes the roles of microcirculation dysfunction, mast cells, several cytokines, hypoxia, oxidative stress, and renin-angiotensin system, and points out the future research direction of the pathogenesis of myocardial fibrosis in RIHD.It provides new ideas for discovering the potential targets for clinical intervention of myocardial fibrosis in RIHD.

Objective To investigate the expression of snrvivin,an anti-apoptosis gene,in breast cancer tissues and its relationship with clinical parameters of breast cancer patients. To investigate the role of survivin in oncogenesis of breast cancer and its relation with prognosis of breast cancer. Methods Western blot was conducted to detect survivin expression in cancerous tissues and adjacent tissues of 50 breast cancer patients.20 normal breast tissue samples were used as control. Results Survivin was expressed in 38 of the 50 breast cancer tissues (76.0%)and in 11 of 50 breast adjacent normal tissues(22.0%)while no expression was found in normal tissues. The positive protein expression of survivin was related to axillary lymph node metastasis of breast cancer while it was not related to expression of PR,ER,Her-2 or ages of patients. Conclusion Survivin may play a role in breast tumorigenesis and is an important predictive parameter in breast cancer patients.
