1.Systematic Review of Aspirin for Type 2 Diabetes Mellitus

Objective To assess the efficacy and safety of aspirin for type 2 diabetes mellitus(T2DM).(Methods)Electronic database searching was performed on Medline,Cochrane Library and CBM,and the data from the beginning of the database to October 2005 were included.Randomised and quasi-randomised trials concerning aspirin treatment for T2DM were selected and assessed for the methodological quality,and the extracted data were performed Meta-analysis by statistical software RevMan4.2.8. Results Fourteen randomised and quasi-randomised trials met the inclusion criteria,including 7 papers of aspirin treatment for T2DM,3 papers of aspirin treatment for cardiovascular complication of T2DM and 4 papers of aspirin treatment for type 2 diabetic retinopathy.Compared with placebo treatment,aspirin showed significant positive effects on lowering blood glucose(SMD,0.73;95%CI,(-1.11 ~-0.36;)P=0.0001).In diabetic patients,aspirin treatment was associated with a significant reduction in the total cardiovascular events(RR,0.78;95%CI,0.68 ~ 0.98;P=0.03).Aspirin treatment neither lowered(nor increased) the risk of the development of diabetic retinopathy(RR,1.02;95% CI,0.97 ~ 1.07;P=0.54).(Conclusion Aspirin treatment) can be a choice for T2DM,especially for the patients who have evidence of cardiovascular disease or have high risks for cardiovascular disease.
2.Construction and Expression of Recombinant Wild Lipoprotein Lipase Gene Plasmid

Objective To study the construction of recombinant wild lipoprotein lipase(LPL) gene plasmid and its expression in COS-1 cells. Methods The LPL cDNA was isolated from the human epiploon adipose tissue by means of RT-PCR.The LPL cDNA was ligated into the pcDNA3.1Zeo(+).The recombinant pcDNA3.1Zeo(+)-LPL cDNA was identified by endonucleases,PCR and DNA sequencing.COS-1 cells were transfected with the recombinant LPL gene plasmid using Lipofectamine 2000~(TM).The LPL mass in cells and the culture medium were determined by a Markit-M LPL Kit.Spectrophotometry was used to measure the LPL activity. Results The LPL gene was ligated into the pcDNA3.1Zeo(+) plasmid identified by endonucleases and PCR.The sequence of the LPL gene was the same as the sequence of the Gene Bank identified by DNA sequencing.Wild pcDNA3.1Zeo(+)-LPL(cDNA) plasmids was transformed into the COS-1 cells. Conclusion The recombinant plasmid pcDNA3.1Zeo(+)-LPL cDNA could be constructed and successfully transformed into the COS-1 cells.
3.Study on glucolipid metabolism and insulin sensitivity in lipoprotein lipase gene knockout heterozygous

The levels of triglyceride(TG)and free fatty acid(FFA)in serum,liver,skeletal muscle,and pancreas of lipoprotein lipase gene knockout heterozygous(LPL+/-)mice and C57 mice were determined.Intraperitoneal glucose tolerance test(IPGTT)was performed to evaluate insulin sensitivity and β-cell function.The results showed that the Iipid content in 16 weeks LPL+/- group did not increase significantly.The TG and FFA contents in 28 weeks LPL+/- group were significantly higher than those in control and 16 weeks LPL+/-group(all P<0.05).In 50 weeks LPL+/- group,FFA levels in serum and pancreas,and TG content in pancreas increased significantly compared with other three groups(all P<0.05).The IPGTT result showed that the blood glucose levels increased from 15 to 120 min,not at 0 and 5 min.The blood glucose levels during 30-120 min increased significantly in 50 weeks LPL+/- group compared with other three groups(P<0.05).Fasting insulin(FINS),homoestasis assessment of insulin resistance(HOMA-IR)and pancreatic β cell function also increased gradually with age.FINS and HOMA-IR in 28 weeks LPL+/- group were higher than those in control and 16 weeks LPL+/- group.These results suggest that LPL is a key enzyme in lipid metabolism and plays a crucial role in the development of insulin resistance and diabetes.
4.Application of Sulfur Hexafluoride Micro-bubble in the Treatment of T2WI High Signal Uterine Fibroids with High Intensity Focused Ultrasound

Objective To evaluate clinical value of ultrasound contrast agent the sulfur hexafluoride micro-bubble in the treatment of T2WI high signal uterine fibroids with high intensity focused ultrasound (HIFU).Methods A total of 36 patients with T2WI high signal uterine fibroids were randomly divided into experimental group and control group.Eighteen patients in experimental group received HIFU treatment with sulfur hexafluoride micro-bubble before,during and after the operation while another eighteen in control group received HIFU treatment with sulfur hexafluoride micro-bubble only after the operation.We recorded operation time (min),changing time of massive gray (s),irradiation time (s),therapeutic dose (J),incidence of complications and ablation rate (%).Results Operation time,changing time of massive gray,irradiation time,and therapeutic dose in experimental group were (101.51±43.02) min,(856.42±451.14) s,(1077.37±546.23) s,and (496581.81 ± 267192.31) J,respectively while those in control group were (261.34 ± 85.53) min,(1 833.32 ± 642.67) s,(1 890.21 ± 1 268.43) s,and (784 608.31 ± 357 621.45) J,respectively.Significant differences were found among those parameters between the two groups (P <0.05).Tumor ablation rate in experimental group was 83.3% while that in control group was 55.5%,which was of significant difference between the two groups (P<0.05).One case of muscular stiffness was found in experimental group while one case of second-degree burn and two cases of muscular stiffness were found in the control group.The incidence of complications was 1/18 (5.5%).No severe complication like bowel perforation or bladder perforation was found in the two groups.Conclusion The effect of ultrasound contrast agent sulfur hexafluoride micro-bubble can shorten operation time,improve tumor ablation rate and reduce complications and it can play a guiding role in clinical work.
5.Short-and long -term outcomes of pneumonectomy for non -small cell lung cancer

Objective To assess the mortality,complications and major morbidity of pneumonectomy for non -small cell lung cancer(NSCLC)and to establish the importance of various prognostic factors.Methods The data of 64 consecutive patients who underwent pneumonectomy for NSCLC were retrospectively reviewed.Results The 30 -day mortality rate was 7.8%(5 /64).Complications developed in 29.7%(1 9 /64)and overall 5 -year survival was 1 9.0%.Pathological stage Ⅲ(P =0.030)and right pneumonectomy(P =0.01 0)were independent risk factors of an adverse outcome.Survival was not significantly influenced by histological types (P =0.1 25)or curability (P =0.587).Conclusion Pneumonectomy is associated with acceptable overall morbidity and mortality.However,the patients with pathological stage Ⅲ or right pneumonectomy require special consideration.Pneumonectomy should be performed only in selected patients.
7.Recent Advances in Vaccines and Drugs Against the Ebola Virus.

The Ebola virus belongs to the Filovirus family, which causes Ebola hemorrhagic fever (mortality, 25%-90%). An outbreak of infection by the Ebola virus is sweeping across West Africa, leading to high mortality and worldwide panic. The Ebola virus has caused a serious threat to public health, so intensive scientific studies have been carried out. Several vaccines (e.g., rVSV-ZEBOV, ChAd3-ZEBOV) have been put into clinical trials and antiviral drugs (e.g., TKM-Ebola, ZMAPP) have been administered in the emergency setting to patients infected by the Ebola virus. Here, recent advances in vaccines and drugs against the Ebola virus are reviewed.

8.Analysis of the curative effect of minimally invasive surgery in the treatment of rectal cancer

Objective To study the curative effect of minimally invasive surgery in the treatment of rectal cancer.Methods 100 patients with rectal cancer were researched.They were randomly divided into observation group and control group,50 cases in each group.The control group received traditional abdominal radical operation,the observation group was treated with minimally invasive surgery.The operation effect was compared between the two groups.Results The operation time,length of stay and feeding time after operation in the observation group were shorter than those in the control group[(78.45±12.34)min vs.(98.24±25.31)min,(3.12±0.43)d vs.(7.53±1.12)d,(1.23±0.30)d vs.(3.56±0.57)d](t=4.967,25.992,25.578,all P<0.05).The amount of bleeding during operation of the observation group was less than that of the control group[(11.23±2.31)mL vs.(26.58±4.04)mL](t=23.323,P<0.05).The incidence rate of postoperative complications of the observation group was significantly lower than that of the control group[6.00%(3/40) vs.20.00%(10/50)](x2=4.332,P<0.05).After surgery,the quality of life in the observation group was higher than that in the control group[(57.82±6.84)points vs.(48.23±4.50)points](t=8.282,P<0.05).After 1 year of follow-up,there were no significant differences in metastasis rate,recurrence rate and survival rate between the two groups[2.00%(1/50) vs.6.00%(3/50),0.00%(0/50) vs.4.00%(2/50),98.00%(49/50) vs.92.00%(46/50)](x2=1.042,2.041,1.895,all P>0.05).Conclusion The effect of minimally invasive surgery for patients with rectal cancer is significant,it is worthy of promoting.
9.Establishment of Rabbit VX2 Soft Tissue Tumor Model and Treatment with Percutaneous Ethanol Inj ection under the Guidance of Magnetic Resonance Imaging

Objective Establish rabbit VX2 soft tissue tumor model,and treat it with percutaneous ethanol injection(PEI)under the guidance of magnetic resonance imaging.Make ready for the therapeutic evaluation with functional magnetic resonance imaging. Methods Fifteen healthy New Zealand white rabbits were included in this study.0.2 mL tumor tissue suspensions were injected into the rabbits’posterior limb.14 days later,all rabbits were underwent conventional MRI examination.PET were performed to all the tumors under the guidance of MRI in the next day of the examination.T2 WI was used as guidance and monitoring means.MR com-patible puncture needle with lateral hole was stabed into the lesion center,and inj ected anhydrous ethanol according to the volume of tumors’diameter (1 mL/cm )slowly.the tumors signal characteristics,morphological feature and pathological feature were ob-served pre and post-operation.Results All of the 1 5 rabbits were established soft tissue tumor model successfully;the success rate is 100%.The tumors were oval or round,3-4 cm in diam.MRI scanning showed low signal on T1 WI and high signal on T2 WI be-fore PEI.PEI was performed to all the tumors under the guidance of MR successfully with 3.5 mL ethanol injected into the tumors in average.T2 WI could monitor the ethanol in dispersion and distribution within the tumors clearly.Histologically,tumors were composed of large,uniform,oval/round cells arranged in solid nests which was intensive in the periphery of tumors.Necrosis tissue was apparent in the center of the tumors.10 days after operation,most tissue in the periphery of tumors was coagulative necrosis , only a few tumor cells left.Ranges of necrosis in the tumors center were obviously increased compared with pre-operation.Conclusion Rabbit VX2 tumor of soft tissue model is suitable for the therapeutic evaluation of tumor .It is an animal model which has the characteristic of simple to operate and high rate of suc-cessful.MR T2 WI can monitor the ethanol in dispersion and distribution within the tumors clearly.It is a good guidance and monitoring imaging method of tumor ablation.
10.The Establishment of an Anti-Trypanosoma Drug Screening System with Leucyl-tRNA Synthetase as an Inhibition Target

Trypanosoma is a human parasite severely affecting poor tropical areas.However,current frontline drugs for Trypanosoma treatment have severe side-effects with decreased effectiveness.Based on the fact that aminoacyl-tRNA synthetase is a bonafide drug target for several microorganisms,including bacteria and fungi,it is plausible that it may also be effective target of Trypanosoma.The Trypanosoma brucei leucyl-tRNA synthetase(tbLeuRS)was cloned,expressed and purified to develop an in vitro enzymatic assay system.The assay conditions were further optimized for the effective screening of tbLeuRS inhibitors thus establishing an anti-Trypanosoma drug screening system targeting tbLeuRS.The results indicated that this system can be employed for the effective screening of anti-Trypanosoma drugs with satisfactory specificity.In addition,this system can also be used for compound optimization,as well as IC50 testing.Using this system a series of compounds are identified that are effective Trypanosoma inhibitors without toxicity to human cells.Therefore,targeting tbLeuRS may represent a new venue for the development of anti-Trypanosoma drugs.
