1.CHEMICAL CONSTITUENTS OF PEUCEDANUM DECURSIVUM

AIM To study the chemical constituents of Peucedanum decursivum. METHODS Various column chromatographies with silica gel and HPLC were employed for the isolation and purification. The structures of the compounds were elucidated on basis of spectral analyses and chemical methods. RESULTS Seven compounds were isolated from the roots of Peucedanum decursivum (Miq.) Maxim. 1 is 3′(S)-hydroxy-4′(R)-angeloyloxy-3′,4′-dihydroxanthyletin, named Decursitin D; 2 is 3′(S)-acetoxy-4′(R)-hydroxy-3′,4′-dihydroxanthyletin, named decursitin F; the others are 3′(S)-acetoxy-4′(R)-angeloyloxy-3′,4′-dihydroxanthyletin (3), Pd-C-IV (4), Pd-C-II (5), (+)-3′S-decursinol (6) and (+)-trans-decursidinol (7). CONCLUSION Compounds 1 and 2 are new compounds, while compounds 6 and 7 were isolated from Peucedanum decursivum (Miq.) Maxim for the first time.
2.CT characteristic findings of pancreatic rupture caused by closed abdominal trauma

Objective To study CT characteristic findings of pancreatic rupture caused by closed abdominal trauma.Methods The clinical and CT data of 6 patients with pancreatic rupture caused by closed abdominal trauma were analyzed retrospectively.CT plain scan and the tripple-phased enhancement scan were performed,multi-planar reformation(MPR)and maximum intensity projection(MIP)were used after CT scanning.Thin slice of CT scan and post-processing technique of imaging were used furthermore.Results The rupture of pancreatic neck and tail were shown in 2 cases,the rupture of pancreatic head and body in 1 case.Main CT findings were such as following:(1)Hematomas in pancreas or in pancreatic surrounding tissues were shown in 6 cases,they were hyperintensity or slight hyperintensity in plain scan and were not enhanced in three-phase dynamic scan of pancreas.(2)Pancreatic splits,which were relative hypointensity after administration of CT contrast agent in the pancreatic phase,were found in 6 cases,they were like slit-shaped or wedge-shaped or pancreas separated completely in situs.In the pancreatic phase,pancreas parenchyma enhanced obviously but hematoma in the splits did not enhanced,and this drew the outline of pancreatic splits clearly.(3)Contrast agent in the pancreatic hematomas,which leaked from vessels of pancreas,were shown in two patients,like dot or nodule or pond,were highly intensity in artery phase,and there were more leaking of contrast agent in the pancreatic phase,whose intensity was same as abdominal aorta in the same slice image.In the delayed phase,the volume of leaking contrast agent enlarged and they had higher intensity while intensity of abdominal aorta in same slice image decreased obviously.(4)Dilation of the Wirsung duct,which was like curved-tube,was shown in one patient.(5)Meanwhile the contusion of liver,kidney or spleen with pancreatic rupture were found in 4 patients.Conclusion The pancreatic rupture has typical CT findings.It is helpful to improve the accuracy rate of diagnosis with reasonable scanning techniques and understanding correctly CT appearances of pancreatic rupture.
3.Construction of systematic evaluation guideline of transformation feasibility of achievements in medical research

The prospective evaluation of the transformation feasibility of the research achievements is an important portion of the evaluation of the medical research achievements.This study explored to set up a system with Delphi method for the evaluation guideline of the hlansformation feasibility of the medical research achievements.And it also offered some evaluating methods and theoretical guidance to carry out the transformation feasibility of the achievements in the medical scientific research.
4.Detecting extended-spectrum beta-lactamases in Escherichia coli and Klebsiella pneumoniae.

Objective To detect ESBLs in E.coli and K.pneumoniae with appropriate method.Methods The suspiciously produced ESBLs were detected by standard screen test among 110 E.coli and 84 K.pneumoniae. ESBLs possessing strains were confirmed by E test, single disc diffusion test with clavulanic acid or sulbatan, and two kinds of double disc synergy test. Results There was a significant difference between the detected rate of ESBLs in E.coli (16/26) and K. pneumoniae (4/20) by E test( P 0.05). The single disc diffusion test with clavulanic acid or sulbatan was applicable to confirm ESBLs in K. pneumoniae (12/20). Conclusions Single disc diffusion test is a sensitive, convenient and inexpensive method to confirm ESBLs in E.coli and K.pneumoniae in clinical laboratory.

Objects To observe the TEM gene characteristic of Acinetinbacter lwoffi strains which were resistant to most kind of antibiotics and to compare their sequences with that of Escherichia coli Methods Two strains of A lwoffi, JN18 and JN70,were isolated from sputum of patients′ who suffered from respiratory infection TEM, SHV, OXA, IMP and CTX M genes were tested by PCR TEM sequences of JN18 and JN70 were detected by ABI automated sequencer and were analysed by DNAStar software to compare the differences with E coli TEM genes that had been published in GenBank Results Detected sequences of A lwoffi JN18 and JN70 strains were 1012 bp and 887 bp, respectively They coded regions of 832 bp and 772 bp for TEM of the two strains that were 98 2% identical In other hand, there were 14 pair bases differently in TEM regions The TEM sequence of JN18 was 98 72% identical to that of E coli TEM on the average, which was over 99% to TEM1D, TEM 70, TEM 76, TEM 77 and TEM 95 of E coli The divergence of TEM genes was 1 26 between A lwoffi JN18 strains and E coli JN70 strain had higher identity (98 93%), which reached 99 5% to TEM1D, TEM1F and TEM84 of E coli As JN18 strain, TEM gene of JN70 had very small divergence (1 06 ) with E coli Conclusion JN18 and JN70 strains of A lwoffi isolated from patients′respiratory tract in Jinan shared most of sequences with E coli TEM76 and 84 However, there were many mutant sites in them
6.Application research of formative evaluation in flipped classroom of college English

Objective To discuss the effect of formative evaluation for flipped classroom of college English teaching. Method We chose 180 clinical medicine undergraduates from a college and divided them equally into experimental class and control class. In the experimental class, the formative evaluation was implemented for the flipped classroom of college English teaching from September 2014 to June 2015. At the same time, the teaching in the control class was carried out according to the basic process of flipped classroom, with no formative evaluation. At the end of the experiment, the students' satisfaction survey, autonomous learning ability and English proficiency test of the two classes were finished. SAS 9.1 software was used for statistical analysis, and t test was used to compare two class measurement information and count data with chi-square test. Result ①Experimental class' satisfaction of formative assessment teaching design for flipped classroom was 94.45%, which was obviously higher than that of the control class (66.66%). The difference was statistically significant (χ2=7.94, P=0.005). (χ2=7.94,P=0.005).②The score at first, fifth dimension and the overall score of independent learning in experimental classes were respectively (55.31± 4.99)、 (53.61±4.93)、 (247.16±22.10), and they were significantly higher than those in the control class (47.18±4.23)、(45.72±4.01)、(225.92±20.78) (t=8.889, P=0.001;t=7.299, P=0.001;t=6.642,P=0.001), and other dimensions' score was also significantly higher than that in comparative classes (P<0.05). ③The pass-ing rate of CET-4, and the number of students from 505 to 544 and more than 545 fraction in the experi-mental class were respectively 79 (87.78%)、19 (21.11%)、10 (11.11%), and they were significantly higher than those in the control class 61(67.78%)、10(11.11%)、0(0). Conclusion Formative assessment can obvi-ously improve the teaching effect of flipped classroom of college English.
7.Studies on the Glucoside Constituents of Shengengzhangyacai (Swertia elongata)

Swertio elongata S. W. Lioa dt T. N. He (Gentianaceae) has been found to be effective in the treatment of liver disease. The present investigation resulted in tho isolation and structure elucidation of four xanthone glycoside, two secoiridoid glycosides and a lignan glycoside in the plant. According to the chemical transformation, spectral (UY, IR, 1H and 18CNMR, MS) properties and comparison with reference samples, the structures of four xanthone glycosides were established as:4-?-D-glucopyranosyl-1, 3, 6, 7-tetrahydroxyxanthone (Ⅶ), 2-?-D-glucopyranosyl-1,3,6, 7-tetrahydroxyxanthone (Ⅵ), 8-O-?- D- glucopyranosyl-1,3, 5-trihydroxyxanthone (Ⅳ) and ?-O-?-D-glucopyranosyl-1, 5-dihydroxy-3-methoxyxanthone (Ⅴ). The structures or two secoiridoid glycosidcs were identified as swertiamarin (Ⅱ) and desacetylcentapicrin (Ⅰ). The structure of lignan glycoside was identified as (+) hydroxypinoresinol-1-?- D-glucosid. (Ⅲ).
8.Experimental therapy with angiogenesis inhibitor TNP-470 on human ovarian cancer transplanted subcutaniously in nude mice

Objective To study the effect of an angiogenesis inhibitor TNP-470 (TNP )used alone and in combination with cytoxan(CTX) in the treatment of human ovarian cancer transplanted s.c. in nude mice. Methods Human ovarian cancer transplanted s.c. in nude mice model was established, then divided into 5 groups: control group, vehicle group, TNP group,CTX group and TNP+CTX group, different treatments were served from day 8 after transplantation and all mice were sacrificed after 28 days. The weights of the mice and the volumes of the tumors were measured respectively during the therapy time. Moreover, microscopy was done after H&E staining. Results The growth inhibiting rates in the TNP and CTX group were 26.1% and 33.9% respectively; After combined, the rate was increased to 70.5%. There were no obvious decrease in the weight of all treated mice. Conclusions Treatment with TNP is an potentially useful method of antitumor therapy in ovarian cancer, although the inhibition effects were not obvious in small doses. Moreover,TNP could enhance the effectiveness of antitumor drug.
9.Effect of simulated altitude hypoxia on the activity of matrix metalloproteinase-2 in gingival tissue of chronic periodontitis rat model

Objective To observe the changes in activity of matrix metalloproteinase-2 (MMP-2) in gingival tissue of a chronic periodontitis rat model in normoxia and hypoxia condition, and explore the role of MMP-2 in the development of high altitude periodontal diseases. Methods Thirty-two adult SD rats were randomly divided into 4 groups (8 each): normal control group (N group), normoxia periodontitis group (P1 group), hypoxia control group (H group) and hypoxia periodontitis group (P2 group). The chronic periodontitis rat model was reproduced in P1 and P2 group, and the rats were kept either in normoxia condition or hypoxia condition in a hypobaric chamber in which a condition simulating 5000 meter high altitude was established. Rats without periodontitis in N and H group were treated with the same conditions as control. The activation of MMP-2 in gingival tissue of rats in each group was measured by gelatin zymographic analysis. Results The grey values of activated MMP-2 in N, P1, H and P2 group were 13.72?6.22, 37.23?4.40, 14.45?6.51 and 45.44?4.79, respectively. Objective To investigate the relationship between social support,coping style and general well-bing in armymen.Methods 280 armymen were tested by General Well-Being Schedule(GWB),coping style questionnaire and social support scale.Results There was a significant difference between the common soldiers and the other armymen(P<0.05);all the factor scores of GWB were related highly with all the factors of coping style and social support(P<0.05~0.01).Conclusion Coping style and social support in armymen are related to the general well-being.
