1.Comparison and application of rheumatoid arthritis remission criteria

Rheumatoid arthritis ( RA) is a kind of autoimmune disease with high morbidity characterized in synovitis.It has become a key goal for achieving an early RA remission.It is very important for application of RA remission normatively in the treatment of RA.In this paper, comparison and application of various RA remission criteria were reviewed, such as American College of Rheu-matology (ACR), European League Against Rheumatism (EULAR), US Food and Drug Administration (FDA), Outcome Measures in Rheumatoid Arthritis Clinical Trials ( OMERACT) group, ACR /EULAR remission criteria, etc.
2.THE REGULATORY MECHANISM OF SUBSTANCE P ON THE TENSION OF HUMAN UMBILICAL VEIN AND THE CALCIUM ION OF ENDOTHELIAL CELLS

Objective To observe the regulatory mechanism of substance P on the tension of human umbilical vein and the calcium ion of endothelial cells. Methods The tension was recorded by conventional physiological recording methods,and confocal laser scanning microscopy and patch\|clamp technique were used to observe the concentration of intracytosolic free calcium ion and the opening probability of membrane calcium ion channel of cultured human umbilical vein endothelial cells. Results Substance P induced the endothelial\|dependent relaxing of human umbilical vein and the increasing of the concentration of intracytosolic free calcium ion and of the opening probability of calcium ion channel in the cultured human umbilical vein endothelial cells.Conclusion\ Substance P could be function as the activator of the releasing of the intracellular stored calcium ion,and the inflowing of calcium ion from the outside cell bodies to relax human umbilical vein.

4.The application and research of Xueshuantong combined with radiofrequency thermocoagulation on the treatment of low back pain caused by lumbar degeneration

Objective To investigate the value of Xueshuangtong combined with radiofrequency thermocoagulation in the treatment of lumbar degenerative lumbar pain.MethodsFrom January 1, 2014 to December 30, 2015, 60 patients with lumbar degenerative low back pain admitted in Jiashan Traditional Chinese Medicine Hospital.All of them were randomly divided into observation group and control group, and each with 30 cases.All patients received radiofrequency thermocoagulation, while the observation group received Xueshuantong for further treatment.All of the patients were assessed by the international Pain Association recommended the use of visual analogue scale (VAS) and McGill pain questionnaire (SF-MPQ) and CRP when treatment before starting and after 1, 3, 7, 14, 30 and 90d of the treatment.ResultsThere were no significant differences between the two groups in VAS, SF-MPQ and CRP before treatment (P<0.05).However, when compared with the control group, patients in the observation group got significantly lower levels of VAS at 1, 3, 7 and 14d after the operation;significantly lower levels of SF-MPQ at 1, 3, 7, 14 and 30d after the operation;and significantly lower levels of CRP at 14, 30 and 90d after the operation (P<0.05).And we found no serious complications occurred in the two groups.ConclusionThe Xueshuantong combined with radiofrequency thermocoagulation treatment of lumbar facet joint is a safe, effective and inexpensive for the low back pain which caused by lumbar degeneration.
5.Expression of tumor suppressor PTEN protein in central nervous system of adult rats

Objective To investigate the expression characteristics of the phosphatase and tensin homolog deleted on the chromosome 10 (PTEN) in the central nervous system(CNS) of adult rats. Methods Immunohistochemical staining was used to demonstrate the PTEN-like positive immunohistochemical product. Results Expression of PTEN was extensive in the CNS. The distribution of positive cells was strong, moderate or weak and its subcellular localization included the cytoplasm, nuclei and processes. Conclusion The present results identify the expression of PTEN protein in the CNS of adult rats.
7.Dynamic changes and clinical significance of pulmonary function in segmental mycoplasma pneumonia children

Objective To study the dynamic changes and clinical significance of pulmonary function (PF) in segmental mycoplasma pneumonia (SMPP) children and make the dynamic analysis of X-ray features.Methods Eighty-three SMPP children treated from June 2013 to December 2014 in Jinshan Institute of the Sixth People's Hospital of Shanghai were selected and the PF changes at acute phase, recovery phase and rehabilitative period was monitored and all the patients received the imageological diagnosis.The Pulmonary function includes forced vital capacity (FVC), forced expiratory volume in the first second (FEV1), forced expiratory volume ratio(FEV1/FVC), peak expiratory flow(PEF), and maximum expiratory flow (FEF25-75).Pulmonary function and imageological diagnosis of two groups was compared.Results Compare the acute phase and recovery phase and rehabilitative period of 83 SMPP Children, at acute phase, FVC ((1.58±0.49), (1.76 ±0.62),(2.08±0.84) L),FEV1((1.27±0.46),(1.58±0.53),(1.83±0.66) L),FEV1/FVC((79.64±6.61)%,(85.25±7.38)%,(87.24±8.61)%),PEF((2.61±0.84),(3.15±0.92),(3.52±1.06) L/s), FEF25 ((2.29±0.83), (2.86± 0.95), (3.26± 0.98) L/s), FEF50 ((1.51 ± 0.52), (2.12 ± 0.64), (2.26±0.63) L/s),FEF75((0.58±0.42),(0.76±0.46),(1.02±0.42) L/s) and FEF25-75 ((0.61±0.33),(0.87±0.36), (1.01 ±0.41) L/s) of two groups were reduced, especially FEF25, FEF50, FEF75 and FEF25-75.At recovery phase and rehabilitative period ,the indicators were significantly better than those of acute phase (P< 0.05);for SMPP children at acute phase, FVC ((1.51 ± 0.44), (1.31 ± 0.36) L) and FEV 1 ((1.46±0.56), (1.21±0.48) L) in the lesions of multiple parts was significantly lower than that of lesions of single part(P<0.05).At acute phase, X-ray majorly showed the unilateral lesion with increased density and blurring edges in lung lobe or segment.After 14 d, the condition was obvious improved;a few cases still had the blurring textures and thickening conditions after 2 weeks.Conclusion For SMPP children at acute phase,the major and micro airway function is damaged in different extent.It is majorly expressed as restrictive ventilation dysfunction,especially micro airway function.At recovery phase, PF is obviously improved.The damage of major airway function in the multiple parts of lung lobe is more serious than that of lesions in the single part.
8.Optimal condition of directional-differentiation of neurons from retinoic-acid induced MSESPU35 embryonic stem cell lines

BACKGROUND: Neural axon regeneration is one of the difficulties that must be overcome in treatment of injury of central nerve system. Significant therapeutic effects have been obtained in transplantation of neural stem cells (NSCs), embryonic stem cells (ESCs) and Schwann cells. But the bottleneck situation of insufficiency of cell provider has limited the development on it.OBJECTIVE: To observe directional-differentiation of retinoic-acid induced ESCs so as to find optimal condition for neuronal differentiation.DESIGN: Non-randomized controlled experiment was designed.SETTING: Teaching-Research Room of Histology and Embryology, Department of Basic Medicines. Third Military Medical University of Chinese PLAMATERIALS: The experiment was performed in Staff Room of Histology and Embryology, Third Military Medical University of Chinese PLA from January to May 2000. Eighteen Kunming mice in disoestrus were employed, of which. 12 mice were female and 6 mice male. They were placed in same cage at ratio of 2:1 for mating. The date of pregnancy was recorded. MESPU35 ESC line was prepared.METHODS: Removed head. internal organs and four limbs, feeder-layer Feeder-layer adherent culture was used to proliferate MESPU35 ESCs.Classic 4-/4+ method [The embryoid body (EB) grew naturally for 4 days,without retinoic acid added. In the coming 4 days, retinoic acid was added to induce neural EB of high proportion] was applied to induce the directional differentiation of the nerve. EB was cultured with serum of different concentrations. Phase contrast microscope was used to observe nerve-like EB in serum of different concentrations and to count numbers. ②Immunocytochemical technique was used to observe cellular morphological charac ters at various differentiating phase spots (5th. 9th, 14th days) and with retinoic acid at various concentrations. Flow cytometer (FCM) was used to count the proportion of differentiated neurons.MAIN OUTCOME MEASURES: ①Estimated measurement of the length of process and cell body during formation of neural EB after retinoic-acid induced differentiation of MESPU35 ESCs. ② Observation of cell morphology with immunocytochemical staining and proportion of differentiated cells assayed with FCM.RESULTS: ①It was discovered with phase contrast microscope that serum of different concentrations affect neural directional differentiation after EB formation to certain extent. Excessively high and low concentrations of serum reduced the proportion of neural differentiation of EB. The differentiating proportion is high in serum with 5% concentration. ② It is observed with immunocytochemical technique that the proportions of NF200 positive cell and glial fibrillary acidicprotein (GFAP) positive cell in differentiation of MESPU35 ESCs induced by retinoic acid were increased with phase spots in differentiation and increased concentration of retinoic acid. NF200 positive cell is transformed as multipolar neurons from absence of process in morphology. The processes of GFAP positive cell became longer and linked among each other as reticular pattern finally. ③ It was assayed with FCM that the proportion changes of GFAP positive cell and NF200 positive cell manufactured in differentiation were similar to immunocytochemical one.CONCLUSION: Retinoic acid in combination with proper concentration of serum and differentiating phase spots can induce neural-differentiation of MESPU35 ESC at high proportion and its differentiating regulation is in the patterns of concentration dependence and time dependence.
9.Thrombolytic drug infusion via carotid artery treating experimental central retinal artery occlusion

Objective To investigate the therapeutic effects of thrombolytic drug infusion via carotid artery on experimental central retinal artery occlusion (CRAO), and observe the changes of fibrinolytic activity in the systemic circulation. Methods To dissolve the thrombi in 15 cats (30 eyes) with CRAO established by laser irradiating a branch of central retinal artery after intravenous injection of photochemical drugs, urokinase (UK) was directly infused via carotid artery in 5 cats (10 eyes) in group A or intravenously injected in 5 cats (10 eyes) in group B, and isotonic saline solution was intravenously injected in 5 cats (10 eyes) in group C respectively. The patency of the artery was evaluated by fundus fluorescein angiography. Moreover, the changes of fibrinolitic activity in the blood were observed by blood biochemical examination. Results Four hours after UK infusion, the complete repatency proportion was 80% (5 cats 8 eys) in group A, and 50% (4 cats 5 eyes) in group B. There was significant difference between the two groups. 10.Molecular dynamics simulation and analysis of BAFF which is incorporated with p-nitro-L-phenylalanine

In order to study the structure-function relationship in the protein which is incorporated with p-nitro-L-phenylalanine,the method of MD(Molecular Dynamics) simulation was established and successfully used in the analysis of protein which contains p-nitro-L-phenylalanine.The force field of CHARMM can only stimulate protein with natural amino acid in NAMD.Compared with phenylalanine,p-nitro-L-phenylalanine just has one more group of nitro.If the parameter of group of nitro was defined,the protein containing p-nitro-L-phenylalanine can be simulated.CGenFF-paramchem was used to calculate the energy and topological structure of p-nitro-L-phenylalanine' s new bonds (r),angles (θ),dihendrals (φ) and improper angle (ψ).And then the new defined parameter and topology information was input into the related parameter files and topology files in CHARMM.On the basis of correct parameter,NAMD can successfully simulate the modified BAFF(B lymphocyte stimulator) which contains p-nitro-L-phenylalanine.The changes in structure indicated that there might be new B cell epitopes.The temperature distribution of each frame in the process of dynamics stimulation was in accord with normal distribution,which proved the defined force field parameters was feasible.The RMSD of whole protein solution systemis 2.5.Calculate each resides' RMSF in BAFF,the RMSF of p-nitro-L-phenylalanine's residue is 3.7,which is obviously higher than that of the other residues in β-pleated sheet,and close to the loop rings,indicate that there might be variation in the area of p-nitro-L-phenylalanine residue and might produce new comformational epitopes.The results of MD stimulation will guide the immunogenicity experiments of p-nitro-L-phenylalanine modified proteins.
