Objective To determine the incidence of pregnancy complicated with heart disease and assess the current obstetric management principles of pregnancy complicated with heart disease. Methods 428 cases of pregnancy complicated with heart disease were reviewed.54 cases were sur- gically corrected heart disease.Results:The incidence of pregnancy complicated with heart dis- ease,during January,1973 to December,1982 and January,1983 to December,1992,were 1.2% (254/21 221) and 0.9% (174/18 726)respectively.The ratio of rheumatic heart disease and congenital heart disease were 4:1 and 1:2 in two periods.There were 37 cases of heart failure and 4 cases of ma- ternal deaths occured.No heart failure and maternal death occured in cases with surgically corrected heart disease.In the two periods,the cesarean section rate were 16.9%(43/254),and 33.3% (58/74) ,P

This study was aimed to establish an analysis method of amentotaxus biflavone distribution in rats' tissues, including the heart, liver, spleen, lungs, kidney, brain, stomach, large intestine and small intestine, in order to investigate its distribution characteristics in tissues after rats gavaged with amentotaxus biflavone. HPLC-UV was employed to determine contents of amentotaxus biflavone in rats' tissues. The intragastric administration dose of amentotaxus biflavone was 500 mg·kg-1. Rats were sacrificed by cervical dislocation 10 min, 0.5, 1, 2, 4, 8, 12 h after intragastric administration. Tissues of the heart, liver, spleen, lungs, kidney, stomach, large intestine, small intestine and brain were removed and dissected immediately. Distribution of amentotaxus biflavone in each tissue was determined after processing. The results showed that tissue in different range had a good linear range. The lowest detection limit was 0.125 ng. The RSD of intra and inter-day was less than 10%. The absolute recovery rate of amentotaxus biflavone in each tissue was between 75.07% and 89.80%. The relative recovery rate was between 92.00% and 107.00%. Amentotaxus biflavone in each tissue was stable in different tissues in refrigerator of -20° C for 15 days. It was concluded that there were relatively large concentration differences of amentotaxus biflavone in different tissues. After intragastric administration, amentotaxus biflavone was mainly distributed in the stomach, large intestine, small intestine, liver and kidney, and then the heart, lungs and spleen. It also distributed in brain tissues through the blood-brain barrier.

Objective:To investigate the effect of donor lymphocyte infusion (DLI) by intra-bone marrow (IBM) or intravenous (IV) on the incidence of graft-versus-host disease(GVHD) after allogeneic peripheral hematopoietic stem cell transplantation (allo-PBSCT).Methods:Female C57BL/6 mice as recipients received total body irradiation (TBI) on day 0,followed by injection of peripheral hematopoietic stem cells from mobilized male BALB/c with granulocyte-colony stimulating factor (rhG-CSF),and DLI was performed via IV or IBM.The extent of GVHD was compared in recipients received allogeneic IBM-DLI with those received IV-DLI.The percentage of donor-derived cells and CD4~+CD25~+ regulatory T cells (Tregs) was detected by flow cytometry.14 days after DLI,the levels of IL-4 and interferon (IFN)-γ were tested by ELISA.Results:It was found that the frequency and severity of GVHD were reduced in IBM-DLI compared with that of IV-DLI (P＜0.01).7 days after transplantation,the percentage of H-2~d-positive cells was over 95% in all surviving transplanted mice;and 14 days after transplantation,the percentage of Tregs detected as CD4~+CD25~+ was significantly higher in recipients treated with IBM-DLI than those treated with IV-DLI(P＜0.01).Compared with that of the recipients in IBM-DLI group,the level of IL-4 was significantly decreased,while the level of IFN-γ were elevated in group IV-DLI (P＜0.01).Conclusion:IBM-DLI could induce the proliferation of Tregs and the Th polarizing to Th2,resulting in decreasing the incidence and alleviating severity of GVHD after allo-PBSCT.

Objective To optimize the extraction technology for total flavonoids in Selaginella tamariscina.Methods To determine the content of total flavonoids by UV and the content of index constituent of amentoflavone by HPLC.The optimum extraction condition was investigated by orthogonal design and the extraction quantity was regarded as the investigated index.Results The optimum extracting condition was A1B2C2D2 with the extraction quantity of total flavonoids as the investigated index.The optimum extracting condition was A1～3B1～3C2D2 with the extraction quantity of amentoflavone as the investigated index.The optimum extracting condition was A1B2C2D2.That is adding ten times amount of 95% alcohol and refluxing twice,2 h once.Conclusion The optimum technology is stable and feasible for the extraction of S.tamariscina.

Objective To compare the dosemetry between helical tomotherapy (HT) and intensity-modulated radiotherapy (IMRT) for early-stage postoperative breast cancer and provide more valuable evidences to the clinical researches. Methods Clinical trails of dosimetric comparing between HT and IMRT for early-stage breast cancer were obtained from PubMed, Embase, Sciencedirect, CNKI, VIP and Wanfang databases, evaluated and analyzed with the Cochrane Collaboration's RevMan 5.2 software. Results 10 studies were included with a total of 135 patients. Compared to IMRT plans, HT plans provided a significantly better conformity index (P<0.000 1), mean (P<0.000 01) and maximal dose (P=0.003) of the planning target volume (PTV). HT plans had a lower heart maximal dose (P=0.005), V20 (P=0.05), V30 (P=0.003), and ipsilateral lung maximal dose (P=0.003), V20 (P=0.02), as while as had a higher contralateral breast V5 (P=0.01), mean (P=0.05) and maximal dose (P<0.000 01). There was no significantly difference between HT and IMRT plans for homogeneity index of PTV, heart V5, V10, mean dose, ipsilateral lung V5, V10, V30, mean dose, contralateral breast V10, contralateral lung mean and maximal dose (all P >0.05). Conclusion Compared to IMRT plans, HT plans have the dosimetry superiority for early-stage breast cancer with significantly better coverage and dose conformity while maintaining lower doses to high risk organs.

Objective To investigate the effects of PTEN and KAI1 double gene transfection on proliferation,metastasis in AsPC1 pancreatic cancer cells under hypoxic condition.MethodsRecombinant vectors that over expressing PTEN and KAI1 protein which was established previously were double transfected into hypoxic AsPC1 cells.Western blot was performed to measure the expression level of PTEN and KAI1.Then,cell proliferation was detected by MTT,and colony forming assay were used to test the ability of tumor cells forming colonies,and transwell assay was used to evaluate metastatic function.ResultsAfter double gene trnsfection,both PTEN and KAI1 protein expression was significantly up-regulated,which was 2.05 and 1.5 1 folds higher than that of empty vector group; and cell proliferation of hypoxic AsPC1 cells was suppressed significantly (0.5 vs 0.8,P =0.00031 ),number of colony formation was significantly decreased [ (41.67 ± 5.03) vs (86.00 ± 7.81 ),P =0.017) ] ; the capacity of metastasis was significantly decreased (0.68 ± 0.05 vs 1.23 ± 0.03,P =0.0025 ).ConclusionsDouble gene transfection of PTEN and KAI1 could inhibit proliferation and metastatic activity of hypoxic AsPC1 cells,which might indicate that combined gene therapy may play a role in the treatment of pancreatic cancer.

ObjectiveTo investigate the effects of heterogeneous phosphatase and tensinhomologue deleted on chromosome ten (PTEN) on cell cycles,proliferation,invasion,tumorigenicity,metastasis and the expressions of vascular endothelial growth factor (VEGF) and epidermal growth factor receptor (EGFR)proteins in human pancreas cancer cell line ( ASPC-1 ).MethodsASPC-1 cells was transfected with plasmid pE-PTEN containing PTEN,and empty plasmid pE-PTEN transfection was used as control,then ASPC-1-pE-PTEN (A-pE-P) cell and ASPC-1-pE (A-pE) cell was obtained.The expression of PTEN mRNA was determined by RT-PCR. PTEN,VEGF and EGFR proteins were measured by cell immunohistochemical method.Clone formation assay was used to observe the numbers of clone.Transwell was used to test the invasion ability of cells.The growths of tumor were detected by nude mice subcutaneous injection of cancer cells in vivo.Results Compared with ASPC-1,the expressions of PTEN mRNA of A-pE-P increased by 179.3％,and the expressions of PTEN protein were also significantly increased.The expressions of VEGF protein were significantly decreased.The expressions of EGFR protein were not significantly changed.Number of G2/M phase cells was significantly increased from (26.81 ± 1.03)％ to (31.5 ± 1.76)％ (P ＜0.05).The numbers of clone was decreased by 28％ (P ＜0.05).The number of penetrating cells was decreased[(46.3 ±6.6) vs (63.8 ±7.5) per high power field,P ＜0.05].The tumor volumes were significantly reduced [(142.4 ±30.9) vs (202.7 ±43.6) mm3,P ＜0.05].The tumor inhibitory rate was 42.4％.The distant metastases were significantly reduced [(2.0 ±0.7) vs (5.0 ± 1.3),P ＜0.01 ].Conclusions Heterogeneous PTEN can not only inhibit the proliferation,invasion and metantasis of ASPC-1 cells,arrest the cell growth at G2/M phase,but also decrease the expressions of VEGF.
