1.Effect of berberine on the proliferation and apoptosis of lung cancer stem cells and the possible mechanism

BACKGROUND: Previous studies have demonstrated that berberine represses multiple tumors and tumor stem cells, but the effect of berberine on lung cancer stem cells (LCSCs) remains unclear. OBJECTIVE: To explore the effect of berberine on the proliferation and apoptosis of LCSCs and the possible mechanism. METHODS: CD133+ LCSCs were separated from A549 cells by immunomagnetic beads. The effects of different concentrations (0, 2.5, 5, 10, 20, 40 mg/L) of berberine on the proliferation and apoptosis of LCSCs were determined by MTT and flow cytometry analysis, respectively. In order to further affirm the effect of berberine on the proliferation and apoptosis of LCSCs, the expression levels of Ki67, Bax and Bcl-2 protein were detected by western blot. In addition, to investigate the potential mechanism by which berberine exerts regulatory effects on LCSCs, the expression levels of Hedgehog signaling pathway-associated proteins (PTCH1, SHH, Gli-1 and SMO) were determined. RESULTS AND CONCLUSION: After magnetic cell sorting, the content of the CD133+ fraction was enriched up to 84.13%. MTT and flow cytometry assays showed that berberine inhibited proliferation and promoted apoptosis of LCSCs in a concentration-dependent manner. Western blot analysis showed that the expression levels of Ki67, Bcl-2, PTCH1, SHH, Gli-1 and SMO proteins of LCSCs cultured in the medium with 20 mg/L berberine were dramatically decreased compared to the control, while the expression level of Bax protein was markedly increased compared to the control. 2.Analysis of lymphocyte subsets and interleukin-2 receptor in patients with severe fever with thrombo-cytopenia syndrome in Zhoushan island

Objective To analyze the characteristics of lymphocyte subsets and interleukin-2 re-ceptor (CD25) in patients with severe fever with thrombocytopenia syndrome (SFTS) in Zhoushan island. Methods Automated blood analyzer , automatic biochemical analyzer and flow cytometry were used to meas-ure hematological parameters , biochemical parameters and lymphocyte subsets in patients with SFTS before and after treatment .The dynamic changes and correlation analysis were statistically analyzed .Results The counts of white blood cells (WBCs), platelets, total T cells and CD4+T cells in patients with SFTS before treatment were significantly lower than those in healthy subjects .The counts of WBCs and platelets were sig-nificantly increased after ten days of treatment , and the level of CD25+cells was higher than that in control group, but the level of total T cells and CD 4+T cells were still lower than those in healthy subjects ( P<0.05).Patients died of SFTS had significantly lower counts of total T cells , CD4+T cells, CD8+T cells and CD25+cells than cured patients (P<0.05).The level of CD25+cells and B cells were positively correlated with WBC level (P<0.05).Platelet level was positively correlated with the percentage of CD 4+T cells (P<0.05).The level of alanine aminotransferase and creatine kinase were positively correlated with the number of CD3+T and CD8+T cells (P<0.05).Conclusion SFTS virus infection affected the percentage of body′s immune cells, especially the level of CD4+T cells.The imbalanced immune system was correlated with the counts of WBCs , platelets and various enzymes in serum and it might be one of the factors causing extensive damage of multiple organs and tissues .
4.Effect of Intensive Training and on Motor Function of Cerebral Infarction Patients

Objective To investigate the effects of intensive training and on the activity of daily living (ADL) and motor function in cerebral infarction patients. Methods 52 cerebral infarction patients with early rehabilitation were divided into an experiment group (n=26) and a control group (n=26). All patients were treated with the conventional training. Meanwhile, the patients in the experimental group were also treated with intensive training. The Modified Barthels Index (MBI) and brief-style Fugl-Meyer assessment (FMA) were used to evaluate the ADL and motor function before and 1 month after treatment. Results The scores of MBI and FMA were significantly different between the two groups 1 month after treatment (P<0.05). Conclusion Intensive training can improve the ADL and motor function more than conventional training in cerebral infarction patients.
5.Effect of Inflatable Shoulder Harness on Hemiplegia Subluxation of Shoulder Joint

Objective To observe the therapeutical effect of inflatable shoulder harness on hemiplegia subluxation of shoulder joint.Methods 68 stroke patients with hemiplegia subluxation of shoulder joint were divided into two groups: the experiment group (n=34) and the control group (n=34). All the patients accepted routine rehabilitation for subluxation of shoulder joint, meanwhile, the experiment group wore inflatable shoulder harness. The interval between acromion and caput humeralis were measured according to the X-ray before and after treatment, as well as the Fugl-Meyer Assessment and Visual Analogue Scale (VAS) of shoulder pain. Results After 8 week of treatment, the rate of diaplasis or improvement was not different significantly between these groups (P<0.05). Compared to the control group, the incidence and VAS score of shoulder pain, and the score of Fugl-Meyer Assessment improved significantly after the treatment (P<0.05). Conclusion On the basis of routine rehabilitation, inflatable shoulder harness may reduce the incidence and grade of shoulder pain, improve motor function of hemiplegic upper limb further in hemiplegia subluxation of shoulder.
6.Clinical observation of combined use sodium ozagrel and ultrasonic therapy for acute cerebral infarction

Objective To observe the clinical effect of Ozagrel sodium with ultrasonic therapy in patients with acute cerebral infarction (ACI). Methods Eighty-three ACI patients were randomly divided into 2 groups: ultra-sortie therapy group(n=42) received Ozagrel sodium and ultrasonic therapy, whereas control group(n = 41) only ac-ecpted the Ozagrel sodium for injection. Both two groups were under conventional treatment. The level of laboratory in-dex and the neurological impairment were detected before and after treatment. And the clinical effect were detected in the end of the treatment. Results The neurological impairment and the clinical effect defected after therapy in both groups, had statistical difference(P < 0. 05), especially in the ultrasonic therapy group. Conclusion Ozagrel sodium with ultrasonic therapy in patients with ACI could promote recovery of the neurological function, thus increase clinical effect.
7.EML4-ALK and EGFR mutation status and survival analysis in Uygur with stageⅣNSCLC

Objective To investigate the relationship between the echinoderm microtubule associated protein like 4-anaplastic lymphoma kinase (EML4-ALK) and epithelial growth factor receptor (EGFR) mutation status and overall survival (OS) in Uygur patients with stageⅣnon-small cell lung cancer (NSCLC) who did not accept tyrosine kinase inhibitor treat-ment. Methods Totally 97 tissue samples were collected from Uygur patients with stageⅣNSCLC who did not accept tyro-sine kinase inhibitor treatment. EML4-ALK fusion gene and EGFR mutation status were detected by using FISH and ARMS methods. The survival rates were analysed. Results In 97 tissue samples, EML4-ALK fusion genes were found in 6 (6.2%) samples, EGFR mutations were found in 26 (26.8%) samples. The survival analysis showed that there was no significant dif-ference in OS between EML4-ALK fusion gene group and no EML4-ALK fusion gene group (P=0.941). There was no signifi-cant difference in OS between EGFR mutation group and wild-type EGFR group (P=0.607). The values of median OS were 17.7 months, 17.3 months and 16.2 months for EGFR mutant group, EML4-ALK positive group and EML4-ALK negative+EGFR wild-type group, and thre was no significant difference between them (P=0.915). Conclusion Excluding the thera-peutic influence in TKIs, EML4-ALK fusion gene and EGFR mutation status of tumor tissue can not be used as an indepen-dent factor in assessing the prognosis in Uygur patients with stageⅣNSCLC.
8.Injury related behavior of middle school students:a case study

Objective To explore the behavioral patterns of middle school students and provide basis for unintentional injury prevention in these students.Methods 171 injury students and 171 normal students as controls were evaluated by Youth Self-Report(YSR),and mean scores were compared between the two groups by t Test.Logistic regressions were performed to analyze the relationship between injury students and behavioral problems.Results The rate was higher in injury group(15.79%) than in control group(8.19%).The score of injury group of boys was higher than thar of control group(t=2.942,P=0.004);the score of injury group of girls was higher than thar in control group(t=3.766,P=0.000).Multi-factor Logistic regression analysis showed that attention-deficit(OR=5.376),the problem of thinking(OR=3.672),social problems(OR=2.204) and friendly fellows' action was a risk factor.Conclusions There is a significant effect of the behavior of students and fellow students on the injury to students.
9.Effects of myofibrillogenesis regulator on myocardial hypertrophy

AIM: To investigate the effects of myofibrillogenesis regulator-1 (MR1) on myocardial hypertrophy. METHODS: Three stem-loop structures of rMR1 mRNA were selected as targets to establish RNA interference carriers. After transient transfection with plasmids, cultured cardiomyocytes of neonatal were used to perform RT-PCR for choosing the first target to carry out RNA interference blocking MR1 gene. In order to observe the effect of MR1 gene silence on myocardial hypertrophy induced by angiotensin Ⅱ (AngⅡ), the radiation intensity of tritium-leucine ([3H]-Leu) was used to label the cardiomyocytes. 10.Application research of cone beam CT in dowel preparation for the mandibular first molar.

OBJECTIVETo measure the root anatomic topography of mandibular first molar using cone beam CT (CBCT), calculate the proper space for dowel preparation.
METHODSThe roots of 121 mandibular first molars from 71 patients were scanned by CBCT, the images from cross section, sagittal plane, and coronal plane were got respectively. The dowel preparation spaces according to established three dimensional images and dowel preparation requirements were analyzed.
RESULTSThree dowels could be prepared. The dowel length for distal-root was (7.19 +/- 0.91) mm, dowel diameter was (0.80 +/- 0.12) mm on the level of 4mm above root apex. The dowel length for mesio-root was (6.92 +/- 1.02) mm, and dowel diameter was (0.71 +/- 0.08) mm on the lever of 4 mm above root apex for buccal side and (0.71 +/- 0.07) mm for lingual side. Dowel should be prepared cone-shaped.
CONCLUSIONMany factors affect dowel preparation, individualized design is needed. CBCT can provide reliable radiographic evidence.
