1.Changes of dendritic cell subsets and cytokine before and after dexamethasone treatment in ITP patients

Objective To investigate the changes of dendritic cell subsets and CD 80,CD86 expression in peripher-al blood in patients with immune thrombocytopenia (ITP), and to evaluate the changes of serum interleukin -2(IL-2), serum interleukin-4(IL-4), serum interleukin-10(IL-10) and interferon-γ(IFN-γ) before and after dexam-ethasone treatment, overall, further analyzing the relationship between them and dexamethasone .Methods Collec-ting blood samples of 60 ITP patients and 10 normal controls with heparin anticoagulation , and distribution of den-dritic cell subsets of ITP and normal controls was detected by flow cytometry , and the changes of serum IL-2, IL-4, IL-10 and IFN-γwere detected by enzyme linked immunosorbent assay (ELISA).Results The percentage of DC2 was increased in ITP patients compared with the control group (P <0.05) and there was no statistically significont difference after dexamethasone treatment ; the expressions of CD80 on DC1 and DC2 were increased compared with the control group(P <0.05), as well as the expression of CD86 on DC2 and the percentage of them decreased after treatment.The serum IL-2, IFN-γlevels before the dexamethasone treatment were higher than the normal control group(P <0.05),then decreased after treatment.However, the serum IL-4, IL-10 levels were lower than the nor-mal control group before the treatment (P <0.05), increased after treatment.Conclusion The disorder of the number and function of DCs and the changes of the serum IL -2, IL-4, IL-10 and IFN-γlevels may play important roles in the pathogenesis of ITP.Moreover, there is a probable key relationship between them and the effects of dexamethasone treatment.

3.Effect of PE Related miR-18a on Expression of Estrogen Receptor Alpha in HTR-8 Cells

Objective:To study the effect of miR-18a on expression of estrogen receptor alpha in human normal trophoblast cells(HTR-8).Methods:The synthesized miR-18a (miR-18amimics,miR-18a inhibitor and blank carrier) were transfected into HTR-8 cells.Experiment included experimental group 1,pre-miRNA un-transfection group(NC group) and experimental group 3.RT-PCR detected the expression of miR-18a in the three groups.RT-PCR and Western-blot detected the expression of ERα mRNA and protein in the transfected HTR-8 cells.Results:According to the results of RT-PCR,the expressions of miR-18a in experimental group 1 (0.880 ± 0.060) was significantly higher than that of NC group(0.407 ±0.019) (P <0.05),and the expressions of miR-18a in experimental group 3(0.160 ±0.014) was significantly lower than that of NC group(0.407 ±0.019) (P<0.05);After transfected for 48 hours,there was no significant difference between the expression of ERα mRNA between experimental group 1 (0.249 ±0.003) and NC group(0.313 ±0.010) (P>0.05),the expression of ERα mRNA in transfected cells in experimental group 3 (0.823 ± 0.023)was significantly higher than that of NC group (P < 0.05);Western-blot results showed that the expression of ERα protein of experimental group 3(1.030 ±0.006) was significantly higher than that of NC group(0.960 ±0.008) (P < 0.05);the expression of ERα protein of experimental group 1 (0.660 ±0.013)was significantly lower than that of NC group (0.960 ± 0.008) (P < 0.05).Conclusions:in human normal trophoblast cells,miR-18a may play regulatory role in the expression of ERα both in mRNA and protein levels.
4.Application of case analysis and problem discussion in teaching the course of introduction to clinical medicine for law majors

Objective To explore the efficacy of case analysis and problem discussion in teaching the course of introduction to clinical medicine for law majors and to discuss their influence on cultivating students' ability and quality.Methods Totally 58 students were randomly divided into three groups:trial group A(n =20),trial group B (n =16) and control group (n =22).Trial group A was further randomly divided into six groups.Teaching contents included iron deficiency anemia and acute leukemia.Trial group A and trial group B adpoted case analysis and problem discussion while control group practiced traditional classroom teaching.Results of theoretical test were processed through one-way analysis of variance and the least significant difference,using SPSS 11.5 software.Self-evaluation of teaching effects on students'specialized learning,logic thinking and knowledge of diseases were analyzed by Chi-square test and Fisher's exact test.Influences of different teaching methods on self learning ability,summarizing ability,team cooperation consciousness and lateral thinking were evaluated after lecture.Results Exam results showed no difference between trial group A and control group (P =0.564),the students in trial group A had a significant higher score than those in trial group B (P =0.004).100% and 75.0% students in trial group A considered the course of introduction to clinical medicine helpful concerning knowledge of diseases and students' specialized learning,higher than those of 77.3% and 40.9% in control group,with statistical differences(P =0.049,P =0.026).Positive evaluation rates of self learning ability,summarizing ability,team cooperation consciousness and lateral thinking were 100%,70.0%,80.0%,60.0% in trial group A ;75.0%,68.8%,62.5%,81.3% in trial group B;22.7%,36.4%,4.5%,27.3% in control group.Except for summarizing ability,students in trial group A and B were significantly better than those in control group(P<0.05).Conclusions The teaching method of case analysis and problem discussion meets the needs of versatile legal personnel training and it can stimulate students' study interest as well as improve their professional quality and ability.
5.Detection of chlamydia and mycoplasma in urogenital tract infection and the analysis of drug sensitivity of mycoplasma

Objective To investigate the status of Ureaplasma urealyticum (Uu) ,Mycoplasma hominis (Mh) and Chlamydia trachomatis(Ct) in patients with urogenital tract infection ,and to analyze the drug resistance of mycoplasma .Methods The myco‐plasma and its drug sensitivity of urogenital tract specimens were detected by culture‐identification‐susceptibility integration kit .Ct was detected by using immunochromatographic method .Results Among 497 cases of specimens ,there were 194 cases of simplex Uu infection (39 .0% ) ,9 cases of simplex Mh infection (1 .8% ) ,73 cases of Uu and Mh mixed infection (14 .7% ) ,and 54 cases of Ct infection (10 .9% ) .There were 38 cases of Ct and Uu mixed infection (7 .6% ) in the Ct positive specimens .The isolated myco‐plasmas were sensitive to doxycycline ,josamycin and minocycline ,but resistant to other drugs in varying degrees .Conclusion There are both simplex infection and mixed infection of chlamydia and mycoplasma in the patients with urogenital tract infection . The clinical use of antibiotics should be reasonable based on pathogen detection and drug susceptibility test results .
6.Three different regimens of CF/5-FU,DDP in treatment of patients with advanced gastrointestinal adenocarcinoma

Purpose:To study the efficacies and adverse effects of three different regimens in treatment of patients with gastrointestinal adenocarcinoma, which consisted of two doses of CF(20 mg/m 2, 200 mg/m 2) and two methods of administration of 5-Fu (continuous intravenous infusion and two hours per day intravenously) and DDP 20 mg/m 2. Methods:We selected 36 cases with GI adenocarcinoma and randomizedly divided them into A and B groups, who were hospitalized from January 1999 to November 2000. At the same time, 29 cases hospitalized from April 1997 to December 1998 with the same disease were designated as C group (control) . A group: CF 200 mg/(m 2.d) for 5 days, 2 hours intravenously ; 5-Fu 3 g/m 2 , continuous intravenous infusion during 5 days; DDP 20 mg/(m 2.d) for 5 days, 2 hours IV. B group: CF 20mg/(m 2.d) for 5 days, 2 hours, IV.; the dosage and usage of 5-Fu and DDP as A group. C group: CF 200 mg/m 2, usage as A, B groups; 5-Fu 600 mg/(m 2.d) for 5 days, 2 hours; dosage and usage of DDP as A, B groups. One cycle of these regimens consisted of three weeks and they were evaluated after three cycles. Results:There were 8 cases of gastral cancer and 12 of rectal cancer in A group whose response rates were 50% and 41.7% respectively. B group had 7 cases of gastric cancer and 9 cases of rectal cancer, their response rates were 42.9% and 44.4%. C group consisted of 14 cases of gastric cancer and 15 cases of rectal cancer, whose response rates were 35.7%and 40%, respectively. Adverse effects included hematological, renal and bladder and gastrointestinal toxicity. A group was similar to B group. C group had more renal and bladder toxicities(27.5%) and incidence of phlebitis was 34.5%. Conclusions:Combination regimen of low dose of CF, continuous intravenous infusion of 5-Fu and DDP is of better efficacy, tolerance and relative economy, especially suitable for to the elderly patients.

Objective:To detect the biological activity of the expression products of the human ?-NGF cDNA in CHO cell line and purified the seperated protein.Methods:Human ?-NGF was transfected with lipofectamine reagent into CHO cell line.The biological activity was analyzed by objection with microscope and the method of MTT.The pure protein was proved by SDS-PAGE analysis.Results:The protein in the culture supernatant of the positive CHO cells transfected with ?-NGF gene could stimulate the growth of PC12 cell line and go into BBB.Conclusion:The target gene expressed successfully in the transfected CHO cell line and had good biological activity. 8.High-frequency ultrasound guided venipuncture of forearm superficial veins

Objective To assess the value of high-frequency ultrasound guided venipuncture of forearm superficial veins. Methods Forty patients with failure of direct venipuncture were given high-frequency ultrasound guided venipuncture. After the direction and depth of superficial veins were identified by high-frequency ultrasonography, a re-puncture was performed. Real-time ultrasonic guidance was employed in case of necessity. Results High-frequency ultrasound identification was used in 11 patients, and real-time ultrasonic guidance was used in 29 patients. All the procedures were successfully completed on one session. Conclusions High-frequency ultrasound guided venipuncture of superficial veins is a simple method without complications, being worthy of wide clinical application.
9.Effects of some drugs for resolving phlegm on blood rheological property in rats

In order to study on the theory of " phlegm and blood stasis having same source" and effects of drugs for resolving phlegm on blood rheology, effects of Ban Xia, Gua Lou, Zhe Bei Mu and Shi Chang Pu on whole blood viscosity, erythrocyte deformation index and plasma fibrinogen content in normal rats. Results indicated that the drugs.for resolving phlegm had functions of decreasing whole blood viscosity, inhibiting hemoagglutination (except Gua Lou) and increasing capability of erythrocyte deformation; at high shearing rate, Shi Chang Pu and Zhe Bei Mu could slightly increase whole blood viscosity: Shi Chang Pu increased slightly plasma fibrinogen content. It is suggested that resolving phlegm also can removing blood stasis, and the mechanism of the drug for resolving phlegm in decreasing blood viscosity is related with decrease of blood lipids and anti - peroxidation of lipids.
