1.Study on Relationship of Human Papillomavirus Genotyping and Recrudescence for Female Patients with Condyloma Acuminatum in Xi'an

Objective To determine the genotypes of human papillomavirus (HPV) and to estimate their recurrence of condyloma acuminatum (CA) among female in Xi'an.Methods HPV genotype was detected with human papilloma virus genotyping kit in 63 female patients with CA.Follow-up was carried out to observe the recurrence of CA in these patients after CO2 laser treatment.Results Of the 63 specimens,61 (96.83%) were positive for HPV,and a total of 16 HPV subtypes were identified.The predominant genotypes were HPV6 (59.2%),11 (22.95%),42 (8.20%),43 (8.20%),52 (8.20%),16 (6.56%) and 18 (4.92%) respectively.43 cases of single HPV infection was accounted for 70.49% (43/61),mixed infection in 14 cases was accounted 22.95 % (14/61).61 cases of patients with laser treatment,observing the recurrence of cuses after 3-month later,it was found that the recurrence rate in higher risk group and mixed infection group was higher than that in low-risk group.Conclusion HPV6,11 is main HPV infection type of the female patients in Xi'an,the recurrence of CA is higher in patients with high risk HPV than in those with low risk HPV and mixted infection.Mixted infection of HPV and high risk HPV is a risk factor for recurrence of female genital warts in Xi'an region.

[Summary] SIRT3 is a member of the silent information regulator 2 ( Sir2) family, located in the inner mitochondrial membrane, with a strong effect of deacetylation. SIRT3 not only modulates energy metabolism, cell apoptosis, tumor growth, anti-aging etc. , but also plays an important role in the field of cardiovascular diseases. In this review, we summarize recent findings related to SIRT3 with metabolic syndrome and cardiac hypertrophy, to provide a theoretical basis for the further study on the potential role of SIRT3 in cardiovascular diseases.
3.Drug Resistance of Pathogens in Blood Culture Specimens in Shaanxi Antimicrobial Resistant Investigation Net from 2013 to 2015

Objective To explore the Gram-positive bacterial drug resistance in blood culture in 2013 to 2015 from the members of Antimicrobial Resistant Investigation Net of Shaanxi province,and guide the clinicians touse antimicrobial drugs rationally.Methods All the objective bacterial isolates were collected and identified susceptibility date by software WHONET 5.6.Results 8 824 Gran positive bacterial isolates and their antibacterial susceptibilitydata were collected.The top five populations of Gram-positive bacterial isolates were Staphylococcus epidermidis,Staphylococcus hominis,Staphylococcus haemolyticus,Staphylococcus aureus and Enterococcus feacium.The isolating rates of methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-resistant coagulase negative Staphylococcus(MRCNS) were 33.9 ~ 54.9% and 72.1 ~88.6 % respectively.No vancomycin,Linezolid and teicoplanin resistant Staphylococcus isolates were found.There were 0.9 ~2% E.faecium vancomycin-resistant isolates.Conclusion The composition of blood culture from 2013 to 2015 was not changed,The rate of MRSA and MRSE showed downward trend.But it was severe that the situation of bacterial drug resistance in blood culture in Shaanxi province.Should fully use bacterial drug resistance surveillance results for supervision and administration,and take effective measures for controlling the spread of resistant isolates.

Sirt3 is a kind of nicotinamide adenine dinucleotide (NAD+)-dependent deacetylases.Sirt3 is localized in mitochondria and has been involved in a wide range of mitochondrial biological functions, such as nutrient oxidation、ATP generation, reactive oxygen species (ROS) detoxification and mitochondrial homeostasis.Sirt3 plays an important role in the occurrence and development of cardiovascular diseases.Increased expression of Sirt3 gene has been associated with extended lifespan of humans.
5.Relationship between lifestyle interventions and glycemic level in adults with impaired glucose tolerance: A systematic review and meta-analysis

Objective To determine the efficacy of lifestyle interventions in adults with impaired glucose tolerance (IGT).Methods MEDLINE (Pub Med),EMBASE,Science Citation Index (SCI),and Cochrane Database were retrieved for articles about the relations of lifestyle intervention and IGT.Searches were limited to English language publications.The RCTs outcome evaluated in this study included 2 h plasma glucose level and fasting plasma glucose,meta-analysis was carried oat by Stata 11.0 among articles for the inclusion and exclusion criteria.The difference of effects was expressed as standardized mean deviation(SMD).Results A total of 10 RCTs studies met the inclusion criteria.Lifestyle interventions were associated with a decline in 2 h plasma glucose and fasting plasma glucose level (SMD =-0.67,P<0.01 ;SMD =-0.33,P<0.01),but high heterogeneity was identified in this meta-analysis.Conclusion Physical,dietary and both combined interventions can reduce 2-h plasma glucose and fasting plasma glucose levels in adults with impaired glucose tolerance.As high heterogeneity was identified in this meta-analysis,more high quality research is needed.
6.Correlation between apolipoprotein E gene polymorphism in Yu patients and cerebral infarction in Taishun county of Zhejiang province

Objective To explore the relationship between apolipoprotein E gene polymorphism and cerebral infarction in Taishun county.Methods Determination of 112 cases of Taishun Siqian cerebral infarction patients and healthy persons 88 cases of ApoE gene polymorphism,and the cerebral infarction patients were admitted to the hospital for different period of time after the NIHSS score.Results The cerebral infarction group and the control group were ApoE -/3 genotype most were 70.5%(79 /112)and 63.6%(56 /88),both had significant difference(P >0.05);The cerebral infarction group and the control group in terms of ApoE -up to 3,respectively 82.1%(92 /112)and 75.6%(66.5 /88),both had no significant difference (P >0.05).The cerebral infarction group epsilon 4 to 9.8%(11 /112),and was significantly higher than that of the control group 4.0%(3.5 /88);E2 was 6.2% (7 /112), which was significantly lower than that of the control group [19.3% (17 /88)],the differences were significant (χ2 =6.189,7.970,all P <0.05).Carrying ApoE -4 of the cerebral infarction patient each time period at the time of admission,admission 7d and 14d NIHSS scores were not carrying epsilon 4 patients increased significantly (t =7.853,6.185,5.165,allP <0.05);and at each time point carrying epsilon 2 gene in patients with and without carry-ing epsilon 2 patients NIHSS scores had no significant difference (P >0.05 ).Conclusion Cerebral infarction patients ApoE gene polymorphism and disease progression and prognosis are closely related,ApoE -4 Taishun Siqian cerebral infarction patients predisposing factors.At the same time,the detection of apoE genotype and NIHSS score is helpful to the prognosis of the patients.
7.Gene cloning, expression of a feruloyl esterase A and purification of its hydrolysis products.

To express feruloyl esterase A from Aspergillus oryzae in Pichia pastoris expression system and study its hydrolysis function, explore the conditions and effects of purification for ferulic acid extracts by macroporos resin. Using the total RNA from A. oryzae CICC 40186 as the template, we amplified coding sequence AorfaeA encoding a mature feruloyl esterase A (AorFaeA) by RT-PCR technique. Then, the coding sequence AorfaeA was successfully expressed in Pichia pastoris GS115 mediated by an expression plasmid pPIC9K. The purified recombinant AorFaeA (reAorFaeA) showed one single band on SDS-PAGE with an apparent molecular weight of 39.0 kDa. The maximum activity of reAorFaeA to methyl ferulate, measured by high-performance liquid chromatography (HPLC), was 58.35 U/mg. Then, reAorFaeA was used to release ferulic acid from de-starched wheat bran in the presence of xylanase. The purification tests for ferulic acid from the enzymatic hydrolysate were carried out with preselected macroporous resins. The results showed that macroporous resin HPD-300 had much higher adsorption and desorption capacities. Ferulic acid could be quantitatively recovered by 50% of the eluent concentration at a flow speed of 1 mL/min. Under the purification condition, the recovery ratio of ferulic acid was 92%, and the content of ferulic acid was increased from 0.13% in the raw material to 10.55%. This work exploits the breakdown of ferulic acid by recombinant enzymeand provids a good strategy to its "green production".

8.Study on Preparation and Quality Standard of Ethacridine Lactate Solution

OBJECTIVE: To prepare ethacridine lactate solution and to evaluate its quality.METHODS: The solution was prepared with ethacridine lactate as principal components and sterilized by steam sterilization.HPLC method was used for the determination of ethacridine.Calesil ODS column was used with mobile phase consisted of methanol-acetonitrile-0.05% sodium laurysulfonate (pH=3.0,20 ∶ 20 ∶ 60) at detection wavelength of 270 nm.The column temperature was set at 30 ℃ and injection volume was 10 ?L.The effect of sterilization on the content of preparation was determined.RESULTS: The preparation assumed as yellow transparent solution.The linear range of ethacridine was 5~50 ?g?mL-1(r=0.999 9) with an average recovery of 101.47% (RSD=1.32%,n=9).The content of ethacridine in the solution was not changed after sterilization.CONCLUSION: The quality of prepared ethacridine lactate solution is up to the standard.

Aim To study the effect of EA on the injury induced by hypoxia/reoxygenation in primary cultures of rat hippocampal neurons.Methods Rat hippocampal neurons in primary culture were used,and a apoptosis model was induced by hypoxia/reoxygenation.MTT assay and LDH releasing rate were used to detect the cell viability.The apoptosis rate of hippocampal neurons was analyzed by Hoechst 33258 staining,flow cytometry with AnnexinV-FITC and PI staining.Western blot was used to detect the protein expression of AKT and p-AKT.Results Compared to control group,three hours of hypoxia followed by sixteen hours of reoxygenation induced hippocampal neuronal apoptosis.EA could raise the neuronal viability and reduce apoptosis rate and the damage degree of rat hippocampal neurons.EA could increase the expressing of p-AKT.Conclusions EA has protective effects on damaged neurons,and the mechanism may be related to activating the PI3K-AKT signal transduction pathway.
10.Effects of astragalosides on the expression of BDNF,TrkB and p75NTR mRNA against focal cerebral ischemia-reperfusion injury

Aim To observe the neurological protective effects of astragalosides(AST) on focal cerebral ischemia-reperfusion(I/R) injury in rats and to explore its possible mechanism.Methods Male SD rats received right middle cerebral artery occlusion for 120 min,and were decapitated 1,3,7,and 14 days after reperfusion.AST(40 mg?kg-1) was orally administered after I/R.Neurological deficit score was daily determined,the expressions of BDNF and p75NTR mRNA were detected by RT-PCR,and the expression of TrkB mRNA was detected by real-time PCR.Results AST reduced the neurological deficit score on days 3,increased the expression of BDNF mRNA on days 3,7 and 14,decreased p75NTR mRNA and increased TrkB mRNA on days 3 and 7.Conclusions AST improves the neurological deficits after I/R in rats.The mechanism may be related with increasing BDNF,and TrkB mRNA,and decreasing p75NTR mRNA.
