1.Influence of different culture methods in DNA methylation status of breast cancer MDA-MB-23 1 cells

Objective:To explore the influence of different cell culture methods in the genome-wide DNA methylation status of breast cancer MDA-MB-231 cells,and to clarify the relationship between genome-wide DNA methylation status and cell growth environment and the role of genome-wide DNA methylation status in the occurrence and development of tumor.Methods:The MDA-MB-231 cells were cultured with 2D and 3D cell culture models and mouse orthotopic transplantation model (Ti model)and collected, then DNA was extracted by DNA extraction kit and the genome-wide DNA methylation status of MDA-MB-231 cells after cultured with three different culture methods was detected by DNA methylation chip,then the value of beta,DiffScore and Delta_Beta of the CpG loci of each gene were calculated by applying Genomestudio software, and the differential methylation genes were screened by Genomestudio software and GO and Pathway analysis of these genes were performed in DAVID on-line analysis tool.Results:All 480 genes of the MDA-MB-231 cells showed significant differences in the degree of methylation in 3D and 2D models (P<0.05);86 448 genes in 3D and Ti models (P<0.05);90 005 genes in Ti and 2D models (P<0.05).The differential methylation genes in 3D and 2D,3D and Ti,and Ti and 2D models were enriched on the multicellular organismal development term and cell differentiation term (P<0.05);also on MAPK signaling pathway,cell adhesion molecules (CAMs),and regulation of actin cytoskeleton (P<0.05). Conclusion:There are differences in genome-wide DNA methylation status of MDA-MB-231 cells cultured in 2D, 3D cell culture and Ti models.
2.Survey of characteristics in negative energy balance in different-aged patients with metabolic syndrome

ObjectiveTo study the characteristics in negative energy balance in different-aged patients with metabolic syndrome (MS) and come out with nursing countermeasures.Methods Twenty-four hour dietary data reviewing and the international physical activity questionnaire were used to investigate energy intake and consumption among 384 MS patients.The fat content,fat percentage and lean body mass were measured by the human body composition analyzer.Body mass index (BMI),waist/height,fat percentage,lean body mass,energy intake and energy consumption were compared between different ages and sexes.Results In the group aged over 51 years,the waist/height of female MS patients was significantly smaller than their male counterparts (P<0.05).For all groups of different ages,the percentage of fat in the female patients was significantly larger,their lean body mass was less(P<0.01), their intake of energy was significantly lower than that of the males (P<0.05).The intake of energy among the male patients at the group aged 21~30was significantly larger than the males of other age groups and so it was with the energy intake of the males aged 21-70 years as compared to the males aged 71~80 years (P<0.05).Conclusion Education on disease knowledge should be strengthened among healthy people so as to realize the negative energy balance based on the rate of waist/height,body composition,energy intake and energy consumption,which is of great significance for the prevention and control of metabolic syndromes.
3.Biofilms produced by Haemophilus influenzae in vitro and antibiotics sensitivity changes

Objective To establish a bioiflm (BF) models of Haemophilus inlfuenza in vitro, and to observe the changes of antibiotic susceptibility after the BF fromation. Methods Thirty strains Haemophilus inlfuenzae isolated from adenoids of children with adenoidal hypertrophy and cultured in a 96-well plate. The BF was identiifed by crystal violet staining and scanning electron microscopy (SEM). The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and the minimum bioiflm bacteria bioiflm clear concentration (MBEC) of ampicillin (AMP), ceftriaxone (CRO), levolfoxacin (LVFX) and azithromycin (AZM) were individually detected. Result All of 30 strains of Haemophilus inlfuenzae formed various BF. After BF is formed, the increase of MBEC for different antibiotics was inconsistent with the increase of MIC and MBC. The difference was statistically signiifcant (MBEC/MBC, H=91.54;MBEC/MIC, H=87.91;all P<0.001). The MBEC of AMP was the highest, up to 100 times than the MBC and MIC. The MBEC of CRO was dozens of times than the MBC and MIC. The MBEC of LVFX and AZM were most close to those of MBC and MIC. Conclusion After the formation of BF, resistance to antibiotics of Haemophilus inlfuenzae is enhaced. LVFX and AZM showed more favorable effect on Haemophilus infuenzae BF.
4.Application of continuous nursing based on IKAP model in patients with coronary heart disease

Objective:To explore the application effect of continuous nursing based on information, knowledge, attitude and practice (IKAP) model in patients with coronary heart disease.Methods:Using the convenient sampling method, a total of 100 patients with coronary heart disease who were hospitalized in Department of Cardiovascular Medicine in Jiaozuo People's Hospital were selected as the research objects from January 2019 to January 2020. According to the random number table method, the patients were divided into the intervention group and the control group with 50 cases in each group. The patients in the control group were given routine nursing, while patients the intervention group were given continuous nursing based on IKAP mode on the basis of routine nursing.Scores of the MOS Item Short From Health Survey (SF-36) , Self-rating Anxiety Scale (SAS) , Self-rating Depression Scale (SDS) and Treatment Compliance Questionnaire were compared between the two groups before and after nursing intervention.Results:After the intervention, scores of each dimension of SF-36 and Treatment Compliance Questionnaire of the intervention group were higher than those of the control group, and the differences were statistically significant ( P<0.05) .The SAS and SDS scores of patients in the intervention group were lower than those in the control group after intervention, and the differences were statistically significant ( P<0.01) . Conclusions:Continuous nursing based on IKAP model can effectively improve the quality of life and treatment compliance of patients with coronary heart disease and relieve the adverse mood of patients, which is worthy of clinical promotion and application.
5.Segmentation adjustment method of three capacity nutrients ratio of metabolic syndrome patients

Objective To explore a method of subsection adjustment for the proportion of three capacity nutrients in patients with metabolic syndrome (MS).Methods Totally 334 MS patients who were diagnosed in the physical examination center and the department of endocrinology and nutrition of Xi'an Central Hospital Affiliated to Xi'an Jiaotong University of School of Medicine were enrolled in this study.According to calculator random digital method,the patients were divided into intervention group (n=168) and the control group (n=166).The energy of two groups were calculated according to the following formula,y =13.5-0.025x1 + 0.215x2-0.006x3+0.342x4-0.268x5+0.623x6 (x1:age,x2:activity intensity index,x3:waist circumference,x4:environmental temperature,x5:BMI,x6:sex).The limit of daily energy was set to 5.02-7.53 MJ.The proportion of the three major nutrients for energy was adjusted of different energy segments of intervention group.When the daily energy was 5.02 MJ,the protein energy ratio was 30%,and the energy increased by 0.42 MJ per time while the protein energy supply ratio was reduced by 2.5%.The energy ratio of carbohydrate was 40%,and the energy increased 0.42 MJ per time while the energy ratio increased by 2.5%.The energy ratio of fat was 30% on various stages of energy supply.The energy proportions of the three major nutrients in the control group were as follows:protein 15%,fat 25%,and carbohydrate 60%.Results After the intervention by 6 months,the 2-hour postprandial blood glucose of intervention group decreased from (9.22±5.57) mmol/L to (6.05±4.68) mmol/L,the hemoglobin A 1 c decreased from (6.79 ± 1.12) % to (5.56± 1.32) %,and the triglyceride decreased from (3.14±1.73) mmol/L to (1.72±1.17) mmol/L.There was significant difference compared with those in control group (P =0.000,0.027,0.034).Conclusion The proportions of the three major nutrients in MS can remarkably improve the glucose and lipid metabolism in MS patients.
6.Relationship between the initial change of thyroglobulin and outcome in differentiated thyroid carcinoma patients with pulmonary metastases after 131I treatment

Objective To investigate the relationship between the initial change of thyroglobulin (Tg) and clinical outcome in differentiated thyroid carcinoma (DTC) patients with pulmonary metastases after 131I treatment. Methods A total of 69 DTC patients with pulmonary metastases from January 2010 to December 2015 were retrospectively analyzed. Patients were divided into 3 groups according to the variation of Tg: groupⅠ(Tg declined≥ 50%), groupⅡ(Tg declined<50% or Tg increased<10%), and group Ⅲ (Tg increased ≥10% ). The follow-up time was (49.2 ± 9.3) months. Clinical outcomes were divided into remission, stable disease and progressive disease according to the serum test and imaging results. Results The percentage of group Ⅰ, Ⅱ, Ⅲ patients was 44.9% (31/69), 40.6% (28/69), and 14.5% (10/69) respectively. Results of follow-up showed 19.4% (6/31) patients achieved remission and 80.6% (25/31) had stable disease in groupⅠ. There were 10.7% (3/28) patients with remission, 60.7% (17/28) with stable disease and 28.6% (8/28) with progression disease in groupⅡ. All patients showed progressive disease in groupⅢ. The clinical outcome was related to the variation of Tg after 131I treatment (P<0.01). Conclusions Initial Tg after 131I treatment could be a predictor to the outcome of patients. The increased Tg level indicates a high possibility of 131I refractory disease.
7.Pathogen spectrum of viral encephalitis and meningitis in children in Shijiazhuang city and surrounding areas in 2018

Objective:To investigate the pathogenic characteristics of viral encephalitis or meningitis in children living in Shijiazhuang city and surrounding areas, and to study the relationship between pathogenic and clinical findings.Methods:A total of 132 cerebrospinal fluid specimens were randomly collected from hospitalized children diagnosed with viral encephalitis or meningitis (January 2018 to December 2018) in the Department of Neurology of Hebei Children′s Hospital in Shijiazhuang city and surrounding areas. The nucleic acids of four viruses in cerebrospinal fluid were detected by real-time quantitative PCR. SPSS 21.0 software was used for statistical analysis.Results:Among the 132 cases, 78 were boys and 54 were girls, with a sex ratio of 1.44∶1. However, in the gender composition of children in each age group, there was no significant difference (χ 2=3.901, P=0.272). Of the 132 children, 121 had signs of fever, 109 had symptoms of headache, 92 had symptoms of vomiting, 17 had abnormal electroencephalogram(EEG), and 15 had abnormal head magnetic resonance imaging(MRI). 132 cerebrospinal fluid specimens were tested for pathogenic pathogens, and 80 of them were successfully detected. There was 1 case of human herpesvirus type I(HHV-I), 2 cases of varicella-zoster virus (VZV) and 77 cases of enterovirus(EV). The age group of 1~3 years′s EV detection rate was 66.67%, it is the highest, but overall, the EV detection rate, there was no significant difference among the four age groups (χ 2=3.147, P=0.369). The detection rate of EV in summer and autumn were 65.52% and 70.83%, respectively, which were significantly higher than those in spring and winter (33.33% and 0.00%), and there was a significant difference (χ 2=22.504, P=0.000). There was no significant difference in the positive rates of fever, headache and vomiting between EV-positive and non-EV-positive children ( P>0.05). There was no significant difference in the incidence of abnormal EEG and abnormal head MRI between EV-positive and non-EV-positive children ( P>0.05). Conclusions:In 2018, EV was the main pathogen of viral encephalitis and meningitis in children in Shijiazhuang city and surrounding areas, and EV detection rate was high in summer and autumn.

Group A Streptococcus (GAS) is a very important pathogen, especially for children.On a global scale, GAS is an important cause of morbidity and mortality.But the burden of disease caused by GAS is still unknown in China and also has not obtained enough attention.For this purpose, the expert consensus is comprehensively described in diagnosis, treatment and prevention of GAS diseases in children, covering related aspects of pneumology, infectiology, immunology, microbiology, cardiology, nephrology, critical care medicine and preventive medicine.Accordingly, the consensus document was intended to improve management strategies of GAS disease in Chinese children.
9. Sub-chronic aluminum exposure inhibits synaptic plasticity in hippocampus of rats via inhibiting PI3K/AKT/mTOR signaling pathway

OBJECTIVE: To study the effect of sub-chronic aluminum exposure on synaptic plasticity in the hippocampus of rats and to explore the mechanism of phosphatidylinositol 3 kinase(PI3 K)/protein kinase B(AKT)/rapamycin target protein(mTOR) signaling pathway. METHODS: Specific pathogen free adult healthy male SD rats were randomly divided into control group and low-, medium-and high-dose groups based on body weight, with 10 rats in each group. Rats were treated with maltol aluminum solution at the concentrations of 0, 10, 20 and 40 μmol/kg body weight by intraperitoneal injection, 5 days per week for 3 months. After the exposure, rats were weighed. Morris water maze was used to test the learning and memory ability, and the two-electrode binding technique was used to record the long-term potentiation(LTP) amplitude in the hippocampus CA1 area of rats. The protein expression of PI3 K, AKT and mTOR in rat hippocampus tissues was detected by Western blot. RESULTS: After the exposure, the body weights of rats in the medium-and high-dose groups were lower than that of the control group(P<0.05). The results of the positioning navigation experiment showed that the escape latencies of the rats in the medium-and high-dose groups were shorter than that in the control group during the 2 nd to 4 th days of the experiment(P<0.05). The results of space exploration experiments showed that there was no statistical difference on the target quadrant retention time and the number of crossing the platform among the 4 groups(P>0.05). At 1, 30, and 60 min after high-frequency stimulation, the LTP amplitudes in the hippocampus CA1 area of the aluminum-treated groups were lower than that of the control group at the same time point(P<0.05), and the LTP amplitudes of hippocampus CA1 area of rats decreased with the increase of maltol aluminum exposure dose(P<0.01). The relative expression of PI3 K, AKT and mTOR protein in the hippocampus tissues of the aluminum-treated groups was lower than that of the control group(P<0.05), and the relative expression of the above three proteins decreased with the increase of the maltol aluminum exposure dose(P <0.01). CONCLUSION: Sub-chronic aluminum exposure could lead to dose-dependent inhibition of hippocampus synaptic plasticity in rats, thereby impairing the spatial learning ability of rats. This process may be related to inhibition of PI3 K/AKT/mTOR signaling pathway by aluminum.
10.Vitamin C alleviates aging defects in a stem cell model for Werner syndrome.

Werner syndrome (WS) is a premature aging disorder that mainly affects tissues derived from mesoderm. We have recently developed a novel human WS model using WRN-deficient human mesenchymal stem cells (MSCs). This model recapitulates many phenotypic features of WS. Based on a screen of a number of chemicals, here we found that Vitamin C exerts most efficient rescue for many features in premature aging as shown in WRN-deficient MSCs, including cell growth arrest, increased reactive oxygen species levels, telomere attrition, excessive secretion of inflammatory factors, as well as disorganization of nuclear lamina and heterochromatin. Moreover, Vitamin C restores in vivo viability of MSCs in a mouse model. RNA sequencing analysis indicates that Vitamin C alters the expression of a series of genes involved in chromatin condensation, cell cycle regulation, DNA replication, and DNA damage repair pathways in WRN-deficient MSCs. Our results identify Vitamin C as a rejuvenating factor for WS MSCs, which holds the potential of being applied as a novel type of treatment of WS.
