1.Cloning and screening novel gene with phage fusion antibody

Objective The cDNA expression library,which was constructed with human colorectal cancer cell HRT-18,was screened with the phage antibody CH209 in order to find novel antigen genes of colorectal carcinoma.Methods Total RNA was extracted from cell HRT-18 and mRNA was isolated from total RNA and then double strand cDNA was synthesized by SMART technique.cDNA was ligated into ? TripIEX2 vector and then was packaged into ? phage in vitro.The primary library was titrated and the percentage of recombinant clones were determined.The length of cDNA inserts was tested for ligation efficiency.The library was screened with phage fusion antibody CH209 and the sequences of the reacted clones were determined.Results The primary library consisted of 6.5?10~(6)pfu/mL,and the percentage of recombinant clones was 97%.The length of cDNA inserts was 0.5-2.0kb.The titer of the amplified cDNA library was 1.1?10~9 pfu/mL.Ten positive clones were obtained and derived from ten different genes.Five of these genes were high homologous to genes known in GenBank,and there were also three genes with low homology to genes known in GenBank.The remainder two genes might be novel genes by matching in GenBank with BLAST software.Conclusion The quality of the constructed cDNA library from human CRC cell HRT-18 is excellent.Ten positive clones were obtained and two of them may be novel genes.
2.HRCT Findings and Pathological Analysis of Bronchioloalveolar Carcinoma

Objective To analyse the HRCT manifestations of bronchioloalveolar carcinoma.Methods HRCT manifestations ofbronchioloalveolar carcinoma confirmed by pathology in 20 cases were retrospectively analysed.Results The most focus of disease located under the pleural membrane with irregular from,the lesions might appeared as patch,nodus and star-like.The ground glass,vacuolus and honeycomb sign had higher incidence in this tumor.Conclusion The ground glass and honeycomb sign usually point out the diagnosis of bronchioloalveolar carcinoma.
3.Concentration Determination of Indigo in Rats'Plasma by LC-MS/MS and Its Pharmacokinetics Study

OBJECTIVE:To establish a method for the concentration determination of indigo in rats'plasma,and study the pharmacokinetic characteristics in rats in vivo. METHODS:18 rats were randomly divided into low-dose,medium-dose,high-dose groups,6 in each group,which were intragastrically administrated 10,20,40 mg/kg of indigo solution. The sample blood 0.3 mL was taken from eye socket before administration and 0.083,0.25,0.5,0.75,1,2,4,6,8,10,12,16,24,48,72 h after ad-ministration,separating the plasma,then LC-MS/MS was used to determine the plasma concentration of indigo after methanol pre-cipitation. The column was Agilent Poroshell EC-C18 with mobile phase consisting of methanol-5 mmol/L ammonium acetate solu-tion(95:5,V/V)at a flow rate of 0.4 mL/min;multiple reaction monitoring was conducted for the quantitative analysis,with ion pair of 263.1-218.8 (indigo) and 237.2-194.1 (carbamazepine,internal standard). Pharmacokinetic parameters were calculated by DAS 3.0 software. RESULTS:The linear range of indigo was 0.5-100 ng/mL(r=0.9999),intra-day and inter-day RSDs were low-er than 9%(n=5);matrix effects of low,medium and high does quality control samples were (98.25 ± 3.71)%,(102.23 ± 2.64)%,(102.29±3.79)%(n=5). The pharmacokinetic parameters of indigo in low-dose,medium-dose,high-dose groups were tmax of(8.6±1.1),(9.2±0.8)and(9.5±0.8)h;cmax of(30.9±8.6),(44.9±10.1),(96.1±17.4)ng/mL;t1/2 of(14.9±2.1), (16.3±2.9),(15.3±3.7)h;AUC0-72 h of(366.6±83.4),(694.9±105.8),(1223.42±108.7)ng·h/mL,respectively. CONCLU-SIONS:The method shows high sensitivity,good specificity,and can be used for the content determination of indigo in plasma samples of rats. The pharmacokinetics of indigo in rats in vivo fits non-compartment model.
4.Pharmacokinetics of loratadine in loratadine, paracetamol and pseudoephedrine sustained-release tablets in healthy volunteers

AIM: To study the pharmacokinetics of loratadine in healthy volunteers after single and multiple oral administrations of loratadine, paracetamol and pseudoephedrine (LPP) sustained-release tablets.METHODS: Twenty-four volunteers were randomized into two groups which included six men and six women in each group. In the single dose design, volunteers received either one or two tablet (s) of LPP. After 1 wk wash out period, volunteers of one tablet group participated in multiple dose design in which each volunteer received one tablet of LPP twice per day for six consecutive days. The concentrations of loratadine in plasma were determined by HPLC-MS method and the pharmacokinetic parameters were calculated. RESULTS: In the single dose design, main pharmacokinetic parameters of one and two tablet group were as follow: cmax were ( 1.5 ±groups were similar to each other. The obtained multi-dose pharmacokinetic parameters were as follows: AUCssrespectively. D (F) was (3.3 ± 0.8) %. The pharmacokinetics of loratadine was linear. There were no significant difference in pharmacokinetics between single-dose and multi-dose. CONCLUSION: The release and absorption of loratadine in experimental tablet are close to those in loratadine tablet and not affected by the other two components, pseudoephedrine and paracetamol, in LPP sustained release tablet.
5.Study on Quality Standard for Zhimiling Suppository

Objective To establish the quality standard of Zhimiling Suppository (Cortex Phellodendri, Radix Sophorae Flavescentis, Catechu, Borneolum Syntheticum, Alumen). Methods Catechu, Borneolum Syntheticum, Alumen in the preparation were identified by physics and chemstry method. Berberine Hydrochloride was determined by HPLC. Results The standard curve for Berberine Hydrochloride was linear in the range of 0.080 2~0.802 ?g, the average recovery was 100.4%, RSD was 0.85% (n=6). Conclusion The methods are simple, accurate and reproducible.
6.Clinical effects of a renew radiofrequency ablation on uterine myomas and disemboweling them from uterine after RFA

Objective To assess the effects on volume of uterine and the dominant fibroid by a re-new radiofrequency ablation ( RFA ) on uterine myomas and disemboweling them from uterine after RFA . Methods Three hundred and eight -three patients were treated by RFA and followed up at 1, 3, 6 and 12 months,165 of them by usual RFA , 163 by improved RFA.To them which myomas diameters were >5cm, 55 cases were added by disemboweling the myomas out of the body after one month of improved RFA .But 66 were not disemboweled.The pre-and postoperative uterine and myoma volumes were measured by 3D ul-trasonography .Results The volume of uterine and the dominant fibroid were reduced in usual and im-proved RFA groups , especially in improved group ,but the difference was no significantly ( P >0.05 ) .In the groups of which myomas diameters were >5cm , the median reduction rates of uterine and myoma vol-ume was more significantly in disemboweling group than un -disemboweling one after 3 and 6 months ( P <0.05 , P <0.01 ) .The overall effective rates were 90.90%,93.25%,100%and92.42%respectively in u-sual and improved RFA groups( P >0.05),and in disemboweling and un -disemboweling groups ( P >0.05).But the obvious effective rates have significant difference in disemboweling (81.82%)and un-dis-embowelin(41.94%)group ( P <0.01).Conclusion Disemboweling myomas from uterine after RFA can increase the clinical effects significantly .
7.Clinical analysis of 24 cases of aortic dissection during pregnancy

plus stented elephant trunk implantation (Bentall+Sun′s surgery), aortic root replacement (Bentall surgery), stent implantation, thoracic and abdominal aorta replacement. The aortic operation time of the 19 patients were 5 gestational weeks to 1 month after delivery. The relation between aortic operation and the termination of pregnancy: 4 patients underwent aorta surgery after termination of pregnancy, 9 patients had cesarean section and aorta surgery at the same time, 6 patients underwent aorta surgery before cesarean section. ②5 patients did not receive arota surgery, 2 patients of type A dissection and 1 patient of type B dissection died before the surgery;2 cases of type B dissection underwent conservative treatment. The termination time of pregnancy was 6-37 gestational weeks, with the average of (26 ± 10) weeks. (3)Maternal and fetal outcomes:20 patients survived after treatment (83%,20/24) and 4 patients died (17%,4/24). 10 cases were live births, including 4 full-term infants and 6 preterm premature infants. The birth weight of the neonates was 1 080-3 800 g, with the average of (2 302±764) g. Three of them were very low birth weight infants and 1 was low birth weight infant;3 neonates had mild asphyxia. The neonates were followed up for 0.5 to 10 years, with the average time of (1.4 ± 1.7) years. So far the infants′ development was good.Conclusions Pregnancy with aortic dissection is pernicious. Early identification, prompt diagnosis and prompt interventing of the vascular surgery are necessary to the safety of mother and fetus.
8.Clinical Literature Analysis of Influence on Si-shen Decoction on Biological Markers of Knee Osteoarthritis

This study was aimed to analyze the domestic relevant documents since 1991 involving Si-shen Decoc-tion and its individual component drugs with knee osteoarthritis biomarkers. This article took medical journal databases of CBMdisc, CNKI, SWIC and Wanfang as sources of information, and searched respectively with key words such as knee osteoarthritis and biomarkers, Si-shen Decoction or Leguminosae, Dendrobium,Achyranthes bidentata, Polygala tenuifolia Willd, Lonicera Japonica and biomarkers such as matrix metalloproteinase, cartilage oligomeric matrix protein, collagen type II C-telopeptide. Bibliometric analysis was given on the acquired relevant documents. The results showed that among 340 documents about knee os-teoarthritis and biomarkers, IL-1, TNF-α and MMPs are mostly used to respond the index of cartilage degenera-tion. Among 565 documents about Si-shen Decoction and its individual component drugs with biomarkers, they are mostly concentrated on the index of TNF-α, IL-1 and oxygen free radical, and the research methods are relatively mature. It was concluded that the bibliometrics method has certain significance in the effect of Si-shen Decoction in the treatment of knee osteoarthritis. More study is required to provide evidence in the development of Si-shen Decoction into a specific prescription for the treatment of knee osteoarthritis.
9.The safety and feasibility of a full-implanted cortical electrical stimulator with low-intensity stimulation in local cerebral infarction rats

BACKGROUND:Cortical electrical stimulation has achieved good effects in treatment of stroke through animal and clinical experiments.
OBJECTIVE:To observe the effects of a ful y implanted cortical electrical stimulation device with long time, low intensity and various frequencies stimulation protocols on the neurological function recovery in a rat model of local cerebral infarction.
METHODS:The cerebral infarction model was established through middle cerebral artery occlusion in 60 Sprague-Dawley adult male rats. Forty rats with 1-3 points by Bederson scale were detected with magnetic resonance imaging, which was used to confirm cortex infarction and to identify a location for implantation of stimulating electrode over the peri-infarct cortex. Twenty-three rats with cortex infarction were randomly divided into cortical electrical stimulation group (CES group, n=13) and no stimulation group (NS group;n=10). The device was implanted on 6 days after middle cerebral artery occlusion, and the stimulation was given for 16 days. The stimulation program consists of two sessions lasting half an hour each in the morning and in the afternoon respectively. Stimulator delivered biphasic charge balanced pulses (pulse width=200μs) with various frequencies of 50 Hz, 20 Hz and 5 Hz within 10 second blocks and then repeated. The rats of NS group were implanted with the device, but received no electrical stimulation. The behavioral tests, includingforelimb use asymmetry test and foot fault test were performed at 2 and 16 days after implantation. Final y, al of the devices were taken out to test if they were normal y working and al of the rats were sacrificed for hematoxylin-eosin staining, which can reflect the structure of peri-infarct cortex and cellmorphology.
RESULTS AND CONCLUSION:There was only one stimulator in CES group cannot normal y work, and the remaining 22 ones worked wel . The skin covered the implanted stimulator was slightly ulcerated in one rat, and the incisions of the other rats were healed wel . Hematoxylin-eosin staining showed clear and intact structure in peri-infarction cortex (i.e., electrodes were implanted at the cortex), neurons arranged in neat rows, with abundant neuronal cytoplasm and clear nucleolus. The glial cells have complete structures, and there was no edema in the intercellular spaces. Foot-fault and forelimb use asymmetry tests showed the improved neurological function in rats of CES group than that of NS group. We designed a ful-implanted cortical electrical stimulator used in cerebral ischemic rats, and established an implanted method with long time, low intensity and various frequencies pulsed electrical stimulation. The results indicated the stimulation pattern in our study is safe and effective, and it can significantly promote functional recovery in local cerebral infarction rats.
