1.Study on the changes of cardiac reserve function during normal vaginal delivery

Objective To investigate the trend of cardiac reserve function during the normal labor.Methods Sixty-three cases were chosen randomly from hospitalized maternal women in the First Affiliated Hospital of Chongqing Medical University from 2010 June to December ( six months).The digital technique of heart sound signal processing was used to analysize cardiac reserve function parameters including the heart rate ( HR),the ratio of the amplitude of the first heart sound to the second heart sound and the ratio of diastolic to systolic duration (D/S) of pregnant women.Results ( 1 ) Comparisons of cardiac reserve function between uterine contractions and relaxations during labor:①Latent phase of labor ( cervix dilation < 3 cm ):HR was ( 87.3 ± 14.0 ) beats/min in uterine contractions and ( 82.8 ± 12.5 )beats/min in uterine relaxations,the ratio of D/S was 1.14 ±0.27 in uterine contractions and 1.21 ±0.22 in uterine relaxations,the comparisons of the above two were statistically significant,P <0.05 ; But the ratio of S1/S2 was 2.19 ±0.82 in uterine contractions and 2.28 ±0.81 in ueterine relaxations,the comparison was not statistically significant,P > 0.05.② During early active stage of labor ( cervix dilation 3-6 cm):HR was (89.3 ± 15.4) beats/min in uterine contractions and (83.1 ± 13.5) beats/min in uterine relaxations,the ratio of D/S was 1.09 ± 0.30 in uterine contractions and 1.20 ± 0.27 in uterine relaxations,the comparisons of the above two were statistically significant,( P <0.05 ) ;But the ratio of S1/S2 was 2.42 ± 1.08 in uterine contractions and 2.29 ±0.83 in ueterine relaxations,the comparison was not statistically significant ( P >0.05 ); ③During late active stage of labor (cervix dilation 6-10 cm),HR was (95.4 ± 18.7 ) beats/min in uterine contractions and (86.2 ± 15.6) beats/min in uterine relaxations,the ratio of D/S was 1.01 ±0.25 in uterine contractions and 1.18 ± 0.25 in uterine relaxations,the comparisons of the above two were statistically significant,( P < 0.05 ) ; But the ratio of S1/S2 was 2.61 ± 1.26 in uterine contractions and 2.67 ± 1.19 in ueterine relaxations,the comparison was not statistically significant ( P > 0.05 ).④ The second stage of labor (cervical dilation ≥ 10 cm ):HR was (109.4 ± 19.7 ) beats/min in uterine contractions and (93.5 ± 16.7 ) beats/min in uterine relaxations,the ratio of D/S was 0.89 ± 0.23 in uterine contractions and 1.14 ±0.26 in uterine relaxations,the ratio of S1/S2 was 3.66 ± 1.37 in uterine contractions and (2.81 ± 1.07 ) in uterine relaxations,the comparisons of all were statistically significant (P<0.05).(2)Comparison of cardiac reserve function in uterine relaxations of each stage of labor:①Maternal heart rate gradually increased from latent stage of labor to the second stage of labor,and decreased postpartum,the comparison was statistically significant ( P < 0.05 ) ;② The ratio of S1/S2 of maternal gradually increased from latent stage of labor to the second stage of labor,and decreased postpartum,the comparison was statistically significant( P <0.05 ) ;③ The ratio of D/S gradually decreased from latency to the second stage of labor,and increased postpartum,the comparison was statistically significant( P <0.05 ).(3) Comparison of cardiac reserve function in uterine contractions of each stage of labor:① Maternal heart rate gradually decreased from latent stage of labor to the second stage of labor,the comparison was statistically significant( P < 0.05 ) ;② The ratio of S1/S2 of maternal gradually increased from latent stage of labor to the second stage of labor,the comparison was statistically significant( P < 0.05 ) ; ③ The ratio of D/S gradually decreased from latency to the second stage of labor,the comparison was statistically significant (P <0.05).Conclusions The maternal cardiac reserve function decreased in uterine contractions than relaxation during labor; With the progress of labor,the maternal cardiac reserve function declined,especially in the second stage of labor,and recovered in postpartum stage.
2.Effects of (Val8)GLP-1-Glu-PAL on motor ability and dopaminergic neurons of Parkison's disease mice

Objective To investigate the protective effects of (Val8)GLP-1-Glu-PAL to dopaminergic neurons on PD mice induced by MPTP.Methods 56 male C57BL/6 mice which are 10-12 weeks old were randomly divided into Control,model (MPTP),muti-injection (Val8) and therapy (MPTP + Val8) groups by random number table method.Mice in MPTP group were received MPTP IP.treatment (30mg/kg · d),the control group were treated with 0.9% saline with the same volume,Val8 group were injected with (Val8) GLP-1-Glu-PAL (25 nmol/kg),and MPTP+Val8 group were going to received (Val8)GLP-1-Glu-PAL 1 h after treatment of MPTP,all the groups were treated for 8 consecutive days.Behavior test were processed 2h after drug IP injection,including swimming test and rotarod test.Mice were sacrificed immediately 1h after the final trail of behavior test in 8th day,brains were withdraw for IHC assay which tested the number TH positive neurons in SNpc area.Results The classical PD behavior symptom were induced by MPTP,from 1 st to 8th,the swimming score(1.715±0.143 and dropping latency(68.048±7.823) were both decreasing compared with control.The differences were statistically significant (P<0.05).The positive dopaminergic neurons were significantly lower in SNpc area (P<0.01).Compared with MPTP group,(Val8) GLP-1-Glu-PAL could statistically improved the behavior deficit,the swimming score (1.120±0.143) and dropping latency(20.546±7.823) in the MPTP+Val8 group,and differences were statistically significant (P<0.05).Meanwhile,the TH positive neurons were significantly elevated (P<0.01).Conclusion The results suggest that (Val8)GLP-1-Glu-PAL has protective effects for MPTP induced mice PD dopaminergic neurons.
3.Correlation of serum acute amyloid A level and activity of Behcet's disease

Objective·To investigate correlation of serum acute amyloid A (A-SAA) level and activity of Behcet's disease (BD).Methods·Blood samples from patients with active BD (n=40),remission BD (n=15),Takayasu disease (TA,n=12),rheumatoid arthritis (RA,n=25),systemic lupus erythematosus (SLE,n=25) and healthy donor (HD,n=25) were collected.Serum A-SAA,ESR,and CRP levels were detected and compared among groups.The correlation between A-SAA and its downstream IL-8 or International Society for Behcet's disease (ISBD) disease activity scores was analyzed as well.Results·Serum A-SAA level was significantly increased in the patients with active BD [(115.70±87.78) mg/L],as well as patients with RA and TA,which levels were (68.72±61.50) mg/L and (96.25±87.41) mg/L respectively,but remained unchanged in SLE patients.Moreover,serum A-SAA level was found correlated with ISBD score as well as IL-8 level.Conclusion·A-SAA can work as a detection index for evaluating BD activity.
4.Clinical Observation on Heat Sensitive Moxibustion in Treating 30 Cases of Simple Obesity with Spleen-kidney Deficiency Type

Objective To examine the clinical effect of heat sensitive moxibustion in treating simple obesity with spleen-kidney deficiency type.Methods Ninety simple obesity patients with spleen-kidney deficiency type were randomized into a acupuncture group,a heat sensitive moxibustion group and a combination group,with 30 cases in each group.The heat sensitive moxibustion group was given moxibustion by self-made heat sensitive moxa stick.Heat sensitization acupoints were sought at patients' lumbosacral portion and hypogastrium.Two or three heat sensitization acupoints were selected each time.Patients were treated for 5 times each week,each time for 45min.In the acupuncture group,acupoints of Zhongwan (RN12),Shuifen (RN9),Qihai (RN6),Guanyuan (RN4),Mingmen (DU4),Yaoyangguan (DU3),bilateral Tianshu (ST25),Daheng (SP15),Daimai (GB26),Pishu (BL20),Weishu (21) and Shuidao (ST28) were selected.Seven to ten acupoints were selected each time.Even reinforcingreducing method was used.Needle was retained for 30min.During needle retention,manipulating the needles once every 10min.Patients were treated by 5 times each week.The combination group was given both acupuncture and heat sensitive moxibustion.The methods were the same as above.One month was a course.Each group was treated for 3 courses.Patients' body weight,waistline and height were recorded before and after treatment.Body mass index (BMI),body fat percent and waist to height ratio were calculated before and after treatment.The clinical effect was evaluated.Results After treatment,body weight,BMI,body fat percent and waist to height ratio in each group decreased (P < 0.05).After treatment,body weight,BMI,body fat percent and waist to height ratio in the combination group was lower than those in the acupuncture group and the heat sensitive moxibustion group (P < 0.05);while the difference between the acupuncture group and the heat sensitive moxibustion group was not significant (P >0.05).The total effective rate in the acupuncture group was 73.3%,with 76.7% in the heat sensitive moxibustion group,and 86.7% in the combination group.The clinical effect in the combination group was superior to that in the acupuncture group and in the heat sensitive moxibustion group.Conclusion Heat sensitive moxibustion therapy might be effective in treating simple obesity with spleen-kidney deficiency type.Moreover,the clinical effect seemed better if combined with acupuncture.
5.Observation and nursing effect of pediatric pneumonia with dyspneic cough adjunctively treated with orientation of traditional Chinese medicine drug therapy and multiple frequency vibration therapeutic instrument

Objective To evaluate the nursing effect of orientation of traditional Chinese medicine drug therapy combined with multiple frequency vibration therapeutic instrument in treating pediatric pneumonia with dyspneic cough. Methods A total of 277 children (pneumonia with dyspneic cough syndrome of wind-cold invading lung, wind-heat invading, lungphlegm-heat congesting lung) were enrolled in this study. They were divided into the orientation of traditional Chinese medicine drug therapy combined with multiple frequency vibration therapeutic instrument group (lung therapy and sputum excretion group,100 cases), the orientation of traditional Chinese medicine drug therapy group (lung therapy group, 94 cases) and the multiple frequency vibration therapeutic instrument group (sputum excretion group, 83 cases) by random digits table method.The main symptoms improvements, therapeutic evaluation, the comfortable level of the children, family satisfaction degree of the 3 groups were evaluated before and after 5 days of treatment. Results After 5 days, the main symptoms score was (7.22±2.74) points in the lung therapy and sputum vibration group, and (8.98±3.40), (8.78±2.91) points in the lung therapy group and sputum excretion group, there were significant differences among 3 groups, F=9.756, P<0.01. The nursing effect in the lung therapy and sputum vibration group was better than that in the other two groups,and there were significant differences among 3 groups, Z=10.376, P<0.01. There were no significant differences among 3 groups in the comfortable level of the children,χ2=1.868,P>0.05. The family satisfaction degree in the lung therapy and sputum vibration group was higher than that in the other two groups, and there were significant differences among 3 groups, χ2=9.102, P<0.05. Conclusions The nursing effect of orientation of traditional Chinese medicine drug therapy combined with multiple frequency vibration therapeutic instrument has more advantages in treating pediatric pneumonia with dyspneic cough.
6.Influence of neuregulin-1 on pacing threshold of low voltage area in rats after myocardial infarction

Objective:To explore the influence of neuregulin- 1 (NRG-1)intervention on pacing threshold of low voltage area in rats after myocardial infarction.Methods:Myocardial infarction model of rat was established via an-terior descending coronary artery ligation.The 34 rats with myocardial infarction were randomly and averagely di-vided into two groups:NRG-1 group (received NRG-1peritoneal injection)and control group (received peritoneal injection of normal saline of the same volume).After two weeks,pacing thresholds were tested in low voltage area related with infarct in rats after myocardial infarction,and Cx43 (an integral membrane protein of the connexin family)expression was measured in low voltage area.Results:After drug intervention two weeks,compared with control group,there was significant reduction in pacing threshold [(1.466±0.503)V vs.(0.7167±0.194)V,P =0.002]in NRG-1 group;there was significant rise in Cx43 expression [(0.30±0.15)vs.(0.95±0.20),P <0.001] in low voltage area in NRG-1 group.Conclusion:Neuregulin-1 significantly reduces pacing threshold of low voltage area in rats after myocardial infarction,the mechanism may be related to increase Cx43 expression and improve e-lectrical conductivity of myocardial cells.
7.Pathological evaluation of immune system in drug safety study

The immune system is a complex system involving multiple organs,and it is vulnerable to age,gender,environment and other factors.For a variation normal physiological range,it is a great challenge to evaluate drug-induced immunotoxicity in preclinical safety study.Histomorphologic assessment of the immune system is a recognized cornerstone in the identification of immunotoxicity at present.In this paper,the principles of pathological evaluation for immune system,and pathological evaluation for important immune organs including thymus,spleen,lymph nodes are discussed briefly,so that it is intended to assist toxicity pathologists in the accurate and consistent characterization of intended and unintended drug-induced alterations of the immune system.
8.Effect and its mechanism of Coptis chinensis on oxidative hemolysis of erythrocytes in mice induced by acetylphenylhydrazine.

OBJECTIVETo investigate the effect of Coptis chinensis on oxidative hemolysis of erythrocytes in mice.
METHODAcetylphenyhydrazine (APH)-induced oxidative hemolysis of erythrocytes in mice were used. The contents of free hemoglobin of blood plasma, indirect bilirubin of serum, reticulocytes of blood, and malondialdehyde (MDA) of erythrocytes were measured. The activity of superoxide dismutase (SOD), reduced glutathione hormone (GSH), and glucose-6-phosp hate dehydrogenase (G-6-PD) of erythrocytes were also determined and the total-antioxygen capability (T-AOC) of blood was analyzed.
RESULTThe levels or amount of free hemoglobin of blood plasma, indirect bilirubin of serum, reticulocytes of blood and MDA of erythrocytes were higher in APH (0.03 g x kg(-1))-induced mice than normal mice. The activity or content of SOD, GSH and G-6-PD was lower in APH-induced mice than in normal mice. Primaquine (0.058 g x kg(-1)) could aggravated the degree of elevated hemolysis of erythrocytes in APH-induced mice. C. chinensis (0.6 g x kg(-1) could deprssed significantly the elevated levels of indirect bilirubin in serum. The levels of free hemoglobin of blood plasma, indirect bilirubin of serum, reticulocytes of blood, the production of SOD and GSH and T-AOC were also decressed by C. chinensis (0.6 g x kg(-1)).
CONCLUSIONC. chinensis suppressed t he degree of hemolysis of erythrocytes in APH-induced mice due to the suppression of the production of lipid peroxidation and increasing of the activity of antioxidase of erythrocytes.

9.Changes and significance of Th17 and Treg cells as well as their associated cytokines in peripheral blood of patients with chronic hepatitis C and cirrhosis

Objective:To study the changes and significance of Th17 and Treg cells as well as their associated cytokines in pe-ripheral blood of patients with chronic hepatitis C and cirrhosis in order to investigate their role in the pathogenesis of chronic hepatitis C and hepatitis C cirrhosis. Methods:Flow cytometry and ELSIA assay were used to detect the expression ratio of Th17 and Treg cells and the serum levels of IL-10,TGF-β,IL-6 and IL-17 in peripheral blood of healthy human and patients with chronic hepatitis C and hepatitis C cirrhosis. Than we analyzed the differences of the above detection index between the healthy and patients with chronic hepatitis C and hepatitis C cirrhosis. Results:The expression ratio of Th17 cells (1. 33%±0. 30%) in peripheral blood of patients with chronic hepatitis C and IL-6[(8. 10±2. 42)ng/L] and IL-17[(16. 70±4. 73)ng/L] serum levels were significantly higher than those in the healthy (Th17:1. 14%±0. 19%) and IL-6[(1. 72±6. 70)ng/L],IL-17[(12. 29±1. 88)ng/L],P<0. 05;The expression ratio of Treg cells in peripheral blood of patients with hepatitis C cirrhosis and chronic hepatitis C patients was significantly higher than that in the healthy (6. 21%±0. 76%,5. 89%±0. 85% vs 5. 51%±0. 59% ),P<0. 05,The ratio of Th17/Treg in patients with cirrhosis was significantly lower than that in patients with chronic hepatitis C and the healthy(0. 19±0. 02 vs 0. 22±0. 03,0. 21±0. 03),P<0. 05;the levels of IL-1[(16. 21±3. 76)ng/L] and TGF-β[(5. 15±0. 83)ng/L] in peripheral blood of patients with hepatitis C cirrhosis were significantly higher than those in chronic hepatitis C[(14. 36±2. 78)ng/L;(4. 47±0. 87)ng/L] and the healthy[(14. 01±3. 01)ng/L;(4. 43±0. 98)ng/L)],P<0. 05. Conclusion:Th17 and Treg cells and their related cytokines involved in the process of chronicity of hepatitis C and cirrhosis,Th17 and Treg cells and their related cytokines in the treatment and prevention of chronicity of hepatitis C and cirrhosis may have important significance.
10.Toxicological pathology in preclinical drug safety evaluation

Toxicological pathology is a morphological elucidatet ion of tissue damage caused by drug toxicity,including damage type,location,severity grade and prognosis.It provides objective and accurate data to support preclinical drug safety evaluation.Therefore,toxicological pathology plays an important role in drug safety evaluation,and the pathologic data or conclusion often determines the termination or continuation in many drug development projects.In this paper,the characteristic of toxicological pathology,standardized management,the development of new techniques including in situ hybridization,laser scanning cytometry and laser capture microdissection,and related problems in pathology are discussed briefly.
