Objective By the designing and clinical application of score table and focus nursing sheet to brain trauma patients,paramedics can observe state of illness individually,optimize the nursing care in a further performance,and ensure the security of brain trauma patients more profitably.Methods To design the score table and focus nursing sheet according to the disease species and symptoms,contrast to the previous nurse observing table in noting time,error rate,exam results and surgeon satisfaction degree.Results By evaluating 130 cases in this test,the score table and focus nursing sheet were superior to the previous nurse observing table.Conclusions With the score table and focus nursing sheet,nurses can observe the illness and note the progress more scientifically and conveniently,the concept of individuation and specialization are worthy of recommending.
3.Repair of knee articular cartilage defects with bone marrow-derived mesenchymal stem cells and bioprotein gel compound in rabbits

Objective To investigate the role of bone marrow-derived mesenchymal stem cells (BMSCs) compounded with bioprotein gel (Bg) in repair of knee articular cartilage defects of rabbits.Methods Bilateral femoral condylar cartilage defect models (3.5 mm in diameter and 3 mm in depth) were established in 36 healthy New Zealand rabbits.Animals were divided into three groups of 12 rabbits each according to the random number table:BMSCs-Bg group,Bg group and blank control group.Effect of repair was evaluated by anatomic observation and histological examination at 4,8,12 and 16 weeks after operation and by articular cartilage histological hemi-quantitative scoring system.Results Hyaline-like cartilage was noted in BMSCs-Bg group at 4 weeks postoperatively,got close to the normal at 8 and 12 weeks and showed no disintegration at 16 weeks.Only fibrous tissue or fibrocartilage was observed in other 2 groups.In BMSCs-Bg group histology scores were (4.5 ± 0.1) points,(4.9 ± 0.1) points,(5.5 ± 0.1) points,and (6.2 ± 0.1) points respectively at 4,8,12 and 16 weeks,higher than those in other 2 groups (P < 0.05).Besides,the histology scores were higher in Bg group than in blank control group but the differences were insignificant.Conclusion BMSCs-Bg compound can well repair the articular cartilage defect in a short-term period,when Bg contributes to cartilage generation as a good cell carrier.
4.The packaging and purification of recombinant adeno-associated virus loaded with anti-amyloid ? peptide single-chain antibody gene

Objective To produce and purify recombinant adeno-associated virus(AAV) loaded with anti-amyloid ? peptide single-chain antibody gene for gene therapy of Alzheimer's disease.Methods The plasmid pSNAV2.0-Abeta-scFv was used to transform BHK-21 cell by Lipofectamine 2000 for stable package cell line establishment.The helper virus HSV1-rc/?UL2 was used to transfect package cell for anti-amyloid ? peptide single-chain antibody gene loaded recombinant adeno-associated virus production.The chloroform-PEG/NaCl-chloroform extraction and ion-exchange chromatography were employed for recombinant AAV purification.SDS-PAGE and PCR amplification were adopted for purified virus identification.The final virus physical titer was determined by digoxin-labeled DNA probes.The effect of gene therapy was tested with transgenic mice by water maze test.Results The purity of recombinant adeno-associated virus with our target gene reach up to 98% after stable cell package and serial purification.The physical titer of the final virus was 1?1012vg/mL.The latency of treated mice in water maze test were reduced significantly.Conclusion The adeno-associated virus carrying anti-amyloid? peptide single-chain antibody gene was produced by HSV1system and purified.The animal behavior test demonstrated the recombinant AAV waseffective in the gene therapy of Alzheimer's disease.

Patrolling monocyte (PMo) is a subset of monocytes.Its main function is patrolling vascular endothelium and removing cellular debris from the microvasculature.Studies have revealed that PMo can inhibit the growth of tumor cells,recruiting NK cells to kill tumor cells.PMo can prevent tumor cells metastasis to the lung,which plays a role of immunosurveillance.PMo may be a target for cancer immunotherapy.

Objective To evaluate the prevention and treatment effect of double-tube-drainage (DTD) method on anastomotic fistula(AF) after surgery of rectal carcinoma. 7.Advance of Exercise Prescription in Aging Sarcopenia and Osteoporosis (review)

Aging is associated with loss of muscle (sarcopneia) and bone (osteoporosis), resulting in an impaired capacity to perform daily activities, furthermore might progress to the extent that an older person loses his or her ability to live independently. Recent studies showed that exercise had remarkable beneficial effects on the musculoskeletal system, including prevention and treatment of these syndromes. Based on the recent research achievement, this paper reviewed exercise prescription in aging sarcopenia and osteoporosis, the result revealed resistance training is the most effective measure to prevent and treat sarcopenia and osteoporosis. In addition, long periods of walking or jogging for over half a year can increase leg muscle size in older adults.
8.Establishment of a miniature pig model of mandibular osteoradionecrosis.

OBJECTIVETo establish a miniature pig model of mandibular osteoradionecrosis (ORN).
METHODSA single dose of irradiation (IR25 Gy, 28 Gy) was delivered via an electronic linear accelerator to the right mandible of 24 miniature pigs by utilizing three dimensional conformal radiotherapy (3D-CRT). The first molar of the mandible was extracted 2 months after radiation. Mandibular ORN was diagnosed through gross observation, X-ray film, CT, and histopathologic examination.
RESULTSAt 3-4 months after radiation, all animals suffered from mandibular ORN after dental extraction. The symptoms of ORN were more severe in the 28 Gy group than in the 25 Gy group.
CONCLUSIONA miniature pig model of mandibular ORN can be established through 25 Gy irradiation + dental extraction.

9.EphA2/ephrinA1 expression in human malignant gliomas and its relationship with angiogenesis

Objective:To investigate the expressions and significance of tyrosine kinase receptor EphA2 and its ligand ephrinA1 in human malignant gliomas and their correlation with tumor angiogenesis. Methods:The expressions of EphA2, ephrinA1, and CD105-stained microvessel density (MVD) were detected via immunohistochemical assay in 62 glioma tissues and 8 normal brain tissues. The correlation between EphA2 and ephrinA1 expression and microvessel counts in the glioma tissues were assessed. Results:Immunohistochemical staining results revealed that variable levels of EphA2 and MVD expression were significantly higher than that of the normal brain samples. Statistical difference was observed in EphA2 and MVD expressions between human gliomas and normal brain samples (P<0.01). The positive rate of EphA2 and MVD expressions was significantly higher in high-grade gliomas (WHO III-IV) than that in low-grade gliomas (WHO I-II) (P<0.01). EphrinA1 was expressed at low levels in most malignant gliomas, and the increased ephrinA1 expression was associated with lower-grade histology. MVD was significantly positively correlated with EphA2 expression (r=0.713, P<0.01) and significantly negatively correlated with ephrinA1 expression (r=-0. 772, P<0.01). EphA2 was significantly negatively correlated with ephrinA1 expression (r=-0.912, P<0.01). Conclusion:Specifically over-expressed EphA2 and its low-expressed ligand ephrinA1 in malignant gliomas may be closely correlated with the invasion and malignant degree of gliomas. Cooperation is involved in the angiogenesis and has an important function in the initiation and progression of gliomas.
10.Adenosine-stress myocardial perfusion imaging for post-operation follow-up after percuteneous coronary intervention in the elderly patients

ObjectiveTo evaluate the value of adenosine-stress myocardial perfusion imaging (MPI)in post-operation follow-up after the percutaneous coronary intervention(PCI) in the elderly.MethodsThere were patients (53 cases) underwent adenosine-stress MPI prior to and 6 months after PCI. Major adverse cardiovascular events (MACE) were followed. Left ventricular ejection fraction (LVEF) was analyzed in 40 cases. The image of the left ventricular myocardium was divided into 16 segments to calculate total defect score using a 4-grade assessment.Myocardial scintigrams were evaluated by calculating the summed adenosine-stress score and coronary angiography assessed by calculating the summed score of coronary artery stenosis.Results There was a significant correlation between the scores of coronary artery stenosis and adenosine-stress MPI scores (r=0.675,P<0.01). Adenosine-stress MPI scores were obviously lower after PCI than before(2.2± 2.3 vs. 4.5±1.9,t= 7.348, P<0.01); LVEF was significantly greater after PCI than pre-operation[(58.3±10.5 ) % vs. (53.8 ± 10.3) %, t = 3.497, P<0.01] and the effective rate for myocardial perfusion were 86.84 %. Conclusions Adenosine stress MPI is a good noninvasive diagnostic method with high accuracy and plays an important role for evaluating recovery of myocardial perfusion and left ventricle function in the elderly patients after PCI.
