1.IL-6 gene rs1800796 polymorphism and febrile seizures susceptibility:a Meta analysis

Objective To systematically evaluate the association between IL-6 gene rs1800796 polymorphism and febrile seizures(FS)susceptibility.Methods The related literatures on IL-6 gene rs1800796 polymorphism and febrile seizures(FS)susceptibility were retrieved from PubMed,Web of Science Embase,Cochrane Library,CNKI,WanFang,VIP and CBM Databases by computer.OR and its 95%CI were taken as the pooled effects.The RevMan 5.2 software was adopted to conduct the statistical analysis.The publication bias analysis was performed by using the STATA12.0 software.Results Seven independent case-control studies were included according to the inclusion and exclusion standard,involving 516 accumulated cases and 528 controls.The results showed that IL-6 gene rs1800796 polymorphism had a significant association with FS susceptibility in Chinese population(GG+CG vs.CC:OR=2.22,P=0.05;G vs.C:OR=2.44,P<0.01;GG vs.CC:OR=3.69,P=0.03;GG vs.CG+CC:OR=3.43,P<0.01).The subgroup analysis was conducted according to possible important confounding factors,and the results showed that this gene polymorphism had a significant association with FS susceptibility in Chinese population(GG+CG vs.CC:OR=3.32,P<0.01;G vs.C:OR=3.23,P<0.01;GG vs.CC:OR=7.27,P<0.01;GG vs.CG+CC:OR=5.17,P<0.01:CG vs.CC:OR=2.56,P=0.02).But in other populations,except recessive model(GG vs.CG+CC:OR=2.40,P<0.01),all other genetic models did not display obvious correlation.The subgroup analysis was conducted by FS diagnostic standard,and the results showed that this gene polymorphism had a significant correlation with infantile FS onset in diagnosing FS by the Chinese standard(GG+CG vs.CC:OR=4.57,P<0.01;G vs.C:OR=4.36,P<0.01;GG vs.CC:OR=12.75,P<0.01;GG vs.CG+CC:OR=8.60,P<0.01:CG vs.CC:OR=3.40,P<0.01).Conclusion IL-6 gene rs1800796 polymorphism may be associated with infantile FS susceptibility and allele G may be a risk factor for FS onset.
2.A Meta-analysis of the Risk of Secondary Infection of Tocilizumab in the Treatment of COVID-19

Objective Meta-analysis was conducted to assess the risk of secondary infection caused by tocilizumab(TCZ)in the treatment of Corona Virus Disease 2019(COVID-19),in order to provide an evidence-based basis for the safety of tocilizumab in patients with COVID-19.Methods Cochrane Library,PubMed,Web of Science,CNKI,SinoMed and Wanfang databases were searched in computer to collect randomized controlled trial and cohort study of treating COVID-19 with tocilizumab from December 19,2019 to December 30,2022.A meta-analysis of the results of each study was performed using RevMan 5.4.1 software.Results A total of 1691 references were screened and eighteen studies involving 3933 patients were included.The incidence of secondary infection in the tocilizumab with the standard treatment group and standard treatment group was 19.14%(331/1729)and 12.11%(267/2204),respectively.Meta-analysis showed that the tocilizumab + standard treatment group had a higher incidence of secondary infection than the standard treatment group[RR = 1.35,95%CI(1.05,1.74),P = 0.02].The results of the subgroup analysis showed that the risk of secondary infection with different doses of tocilizumab was different.The incidence of secondary infection was significantly higher in the subgroup with doses of 400~800 mg/d tocilizumab than in the standard care group[RR = 1.48,95%CI(1.19,1.84),P = 0.0004].The incidence of secondary infection in subgroups with doses of≤400 mg/d tocilizumab was also significantly higher than that in the standard treatment group[RR = 1.87,95%CI(1.28,2.72),P = 0.001].However,there was no statistical significance between the subgroup 6~8 mg/kg tocilizumab and the standard treatment group.Conclusions Tocilizumab may increase the risk of secondary infection in patients with COVID-19 compared with standard treatment,and the benefits and risks of tocilizumab should be carefully evaluated before clinical administration.Moreover,large and high-quality studies are needed for further evaluation.
3.Characteristics and trends of adolescents health risk behaviors in Shanghai from 2004 to 2019

Objective:
To analyze the trend of adolescent health risk behaviors in Shanghai, and to provide reference for the comprehensive intervention of middle school students health risk behaviors.
Methods:
Based on the health risk behavior questionnaire of Chinese Center for Disease Control and Prevention, the questionnaire was adapted according to the actual monitoring needs. It was divided into junior high school version and senior high school version. From 2004 to 2019, using the method of multistage stratified cluster random sampling, 59 209 middle school students who completed the questionnaire in 6 surveys were selected for analysis.
Results:
From 2004 to 2019, in the 7 days prior to the survey, 9.2%-50.6% of middle school students drank a glass of soda more than or equal to once a day and 54.2%-76.1% of middle school students reported eating dessert twice or more. Within 7 days, 48.3%-60.7% of middle school students had≥60 min of exercise per day for less than or equal to 3 days, and 16.1%- 35.2% of middle school students reported too much extracurricular activities, and the reporting rate increased year by year with the annual percent change ( APC ) of 5.15%( t =9.28, P <0.01). The reporting rate of long time online learning was 6.0%-13.6%, which showed an upward trend among high school students, with the APC of 5.35%( t =3.14, P <0.05). The reporting rate of middle school students pedestrian safety problems decreased from 69.1% in 2004 to 27.6% in 2019, with the APC of -6.28%( t =-8.18, P <0.01), but the reporting rate of cycling safety problems have increased in recent years. The reporting rate of intentional injury behaviors decreased by year such as fighting, bullying, etc. The reporting rate of initial smoking age ≤13 years old decreased, but attempted drinking behavior increased in junior middle school students, and the APC was 1.61%( t =3.48, P <0.05). A total of 1.6%-3.4% of middle school students had an Internet addiction behavior tendency. The detection rate of Internet addiction tendency was increasing in high school students and girls, and APC was 6.59% and 10.29% respectively( t =6.37, 8.62, P <0.01).
Conclusion
From 2004 to 2019, unintentional injury behavior, intentional injury behavior and substance addiction behavior of middle school students in Shanghai have improved. However, unhealthy diet and physical inactivity are still high. In the follow up. It need to focus on adverse diet and lack of physical activity behavior and take comprehensive intervention measures to control them.
4.Trends in the prevalence of Internet addiction among adolescents from 2004 to 2019 in Shanghai

Objective:
To analyze the trends of Internet addiction among adolescents in Shanghai, so as to provide theoretical evidence for the prevention and control of Internet addiction in adolescents.
Methods:
A multistage stratified cluster random sampling method was conducted, and 92 171 junior and senior high school students in Shanghai were selected for questionnaire survey in 2004-2019. Evaluation of Internet addiction, loneliness, academic pressure and depression among middle school students.
Results:
In 2004-2019, the rate of Internet addiction among middle school students in Shanghai was 4.3%, which without significant differences by year( t =1.8, P >0.05). The detection rate of Internet addiction in boys (5.3%) was higher than that in girls (3.4%) ( χ 2=186.8, P <0.01), and was highest in secondary vocational school (7.7%), followed by senior high school (4.6%) and junior middle school students (2.8%) ( χ 2=746.5, P <0.01). The prevalence of Internet addiction in girls, senior high school students, secondary vocational school students, students aged 16-20, students with loneliness or high academic pressure showed an increasing trend in 2004-2019, with the average annual increase rate ( APC ) of 6.62%, 5.45%, 2.13%, 3.36%, 4.09% and 3.60% respectively ( P <0.05).
Conclusion
Internet addiction among adolescents in Shanghai showed an increasing trend in some groups from 2004-2019, but without significant differences by year in the overall groups. Comprehensive prevention and control programs are needed for adolescent Internet addiction, especially for those with mental health problems.
6.Trend of diet related behaviors among secondary school students in Shanghai, 2008-2019

Objective:
To understand the prevalence and trend of diet related behaviors among secondary school students in Shanghai, and to provide a scientific basis for developing appropriate health education strategies.
Methods:
Anonymous questionnaire surveys were conducted among 71 291 students selected by multi stage stratified cluster sampling method from junior high schools,senior high schools and vocational schools in Shanghai in the year of 2008,2012,2015,2017,2019. The questionnaire adopted was "Shanghai adolescent health related behavior questionnaire". Test for trend test were applied for the analysis of the prevalence of their dietary behaviors.
Results:
The proportion of middle school students nutritional status within the normal range in each survey year were 73.40 %, 72.60%, 68.06%, 68.73%, 67.53%, respectively, while the rate of overweight and obesity increased from 13.17% in 2008 to 23.73% in 2019. The proportion of "eating fried food" decreased significantly by year, from 12.05 % to 4.23% (male, APC=-7.94%, t =-4.33, female, APC= -9.29%, t=-7.71, P <0.05) respectively. And the proportion of "eating dessert" also decreased significantly by year, from 31.66% to 11.14% (male, APC=-8.41%, t =-6.37, female, APC=-8.67%, t=-6.96, P <0.05). In the five surveys, the percentages of healthy diet related behaviors of secondary school students were 12.81%-21.82%, no significant upward trend was observed in the whole( P >0.05). The proportion of diet-related behaviors of nutrition unbalance and easily leading to obesity decreased significantly from 35.47% to 17.00%(APC=-6.71%, t= -5.00 , P <0.05). The proportion of diet related behavior of nutrition unbalance and not easily leading to obesity increased significantly from 39.15% to 60.16%(APC=3.46%, t=6.18, P <0.05).
Conclusion
Overweight and obesity among secondary school students in Shanghai are increasing during the past decade, and the proportion of healthy dietary behaviors is low. The knowledge of rational diet should be publicized among these students in order to develop healthy dietary behaviors.

Objective To explore the biological function and downstream mechanism of ETS1 in glioma. Methods Bioinformatics and immunohistochemistry were used to analyze the differential expression characteristics of ETS1 in gliomas; qRT-PCR was employed to detect the expression level of ETS1 mRNA and lncRNA X-inactive specific transcript (XIST). CCK-8 and 5-ethyl-2′-deoxyuridine experiments were conducted to detect cell growth. Western blot was used to detect the expression of apoptosis-related proteins (Bax, Bak, Bcl-2). PROMO database was utilized to predict the binding sites between ETS1 and XIST promoter. Objective To evaluate the association between site polymorphism of glutathione S-transferase omega1 (GSTO1) gene (rs4925),arsenic (+3) methyhransferase (As3MT) gene (rs11191439) and susceptibility to endemic arsenic poisoning.Methods All related studies in published papers up to March 1,2018 on association between site polymorphism of As3MT gene,GSTO1 gene and susceptibility to endemic arsenic poisoning were collected by searching PubMed,Embase,CNKI,Wanfang and VIP database.The data were screened according to the inclusion and exclusion criteria,and extracted,the quality of included studies was evaluated.The pooled odds ratios (OR) with 95% confidence intervals (95%CI) were calculated using Review Manager 5.3 software.Publication bias and sensitivity analysis were also assessed.Results A total of 7 case-control studies involving 989 cases and 1 212 controls were included in the Meta-analysis.Individuals carrying at least one allele A at the rs4925 locus of the GSTO1 gene might increase the risk of susceptibility to arsenic poisoning compared with individuals carrying wild homozygous type [AA + CA vs CC,OR (95%CI) =1.37 (1.13,1.65),P < 0.01].There was no statistically significant relationship between susceptibility to arsenic poisoning in individuals with at least one allele C at the rs1 1191439 locus of the As3MT gene compared with individuals carrying wild homozygous type [CC + TC vs TT,OR (95%CI) =1.12 (0.69,1.81),P > 0.05].Conclusions GSTO1 gene rs4925 polymorphism is significantly associated with susceptibility to arsenic poisoning,and the A allele is a risk gene for susceptibility to arsenic poisoning.The As3MT gene rs1 1191439 locus polymorphism is not associated with susceptibility to arsenic poisoning.
9.Integrating Network Pharmacology Based on UPLC-Q-Exactive/MS Technology to Explore the Mechanism of Chaihu Guizhi Decoction in the Treatment of Secondary Bacterial Pneumonia Caused by Influenza

Objective To study the mechanism of Chaihu Guizhi Decoction(CGD)in the treatment of influenza and staphylococcus aureus co-infection.Methods The co-infection model of influenza and staphylococcus aureus was established and CGD was used to intervene.The chemical components of CGD were qualitatively analyzed by UPLC-Q-Exactive/MS technology.The potential action targets of chemical components in CGD and the related targets of influenza Staphylococcus aureus co-infection were mined by network pharmacology method.The"component target disease"network was constructed.Core targets were selected according to degree ranking.Core action pathways were enriched by KEGG analysis and GO annotation analysis.The core target was verified by RT-qPCR,and the interaction between the core component and the key target was verified by molecular docking.Results CGD could significantly improve the decrease of body weight and thymus index(P<0.05)caused by co-infection.The lung index(P<0.05),relative amount of MmRNA expression(P<0.05)and bacterial load(P<0.05)were decreased,and the survival rate was improved.51 chemical constituents were identified from CGD.Through network pharmacological analysis,107 related targets corresponding to CGD treatment of bacterial pneumonia secondary to influenza were excavated.TNF,AKT1,ALB,VEGFA,MAPK3,PTGS2,STAT3,EGFR and other targets with strong correlation,mainly involved Fc epsilon RI signal pathway,GnRH signal pathway,NF-κB signal path,etc.Molecular docking study showed that the main active component of CGD,including oroxyloside,baicalein and wogonin have strong affinity with TNF,PTGS2 and EGFR targets.Compared with co-infection model group,in CGD group TNF-α、EGFR and PTGS2 increased significantly(P<0.05).Conclusion The main active ingredient of CGD is oroxyloside,baicalein and wogonin.TNF-α,PTGS2,EGFR and other targets to played a role in the treatment of influenza staphylococcus aureus co-infection.
10.Comorbidity of common illnesses and associated factors among primary and middle school students in Shanghai in 2021

Objective:
To understand the current status and associated factors of comorbidity of common illnesses among primary and middle school students in Shanghai, so as to provide a reference for improving the pathogenic environment and preventing the occurrence of comorbidity of common illnesses.
Methods:
From September to November 2021, a stratified cluster sampling method was used to select 30 331 middle school students from 112 schools in 16 districts of Shanghai, each with 2 primary schools, 2 junior high schools, 2 high schools and 1 vocational high school. Elementary school students, their health status and influencing factor questionnaires were analyzed.
Results:
The detection rate of two or more common diseases such as myopia, overweight and obesity, high blood pressure, and abnormal spinal curvature among primary and secondary school students in Shanghai was 38.91%, of which 29.64% of primary and secondary school students had two diseases at the same time, and 9.19% had three diseases, 0.09% had 4 diseases. The detection rate of comorbidity was higher in boys (44.12%) than in girls (33.40%), and higher in junior high school students (40.40%) than in high school students (38.58%) and primary school students (38.01%). The differences were statistically significant (χ2=366.44, 50.33, P<0.01). Multifactor Logistic regression analysis showed that girls sometimes supervise themselves and often remind themselves that their sitting and standing postures were negatively correlated with the common comorbidities of primary and secondary school students (OR=0.64, 0.93, 0.90); junior middle school students and watching TV/d ≥1 h, using computers for ≥1 h/d, and using mobile electronic devices for >1 h/d were positively correlated with the comorbidity of common diseases among primary and secondary school students (OR=1.07, 1.10, 1.06, 1.10) (P<0.05).
Conclusions
The prevalence of comorbidity of common illnesses among primary and middle school students in Shanghai is not optimistic, which might be associated with combined effects of various external environmental factors. Targeted intervention should be implemented to effectively prevent the occurrence of comorbidities among students.
