1.Correlation between uncoupling protein 2 gene polymorphism and diabetic retinopathy

Objective To investigate the association between a 45-bp insertion/deletion(Ins/Del) polymorphism in the 3′-untranslating region(3′-UTR) of its exon 8 in uncoupling protein 2(UCP-2) gene and diabetic retinopathy(DR).Methods A total of 183 patients with diabetic mellitus(DM) were selected,51 with DR,132 without DR.UCP-2 gene polymorphism was confirmed by electrophoresis after PCR,then genotype and allele gene frequency were compared.Results No significant difference of genotype frequencies for 3′-UTR Ins/Del polymorphism of UCP-2 was found between control group and DR group,and no difference in two alleles frequencies.Conclusion No relationship of a 45-bp Ins/Del polymorphism in the 3′-UTR of exon 8 in the UCP-2 gene was found with DR.
2.Clinical Observation on the 50 Cases of Refractory Ascites Resulted from Hepatocirrhosis Treated by Xiaogu Granule plus Ultrafiltration and Reinfusion of Ascites

Objective To observe the effect and machanism of Xiaogu Granule (Granules for treating tympanites) plus ultrafiltration and reinfusion of ascites on refractory ascites resulted from hepatocirrhosis. Methods The 100 cases of refractory ascites due to hepatocirrhosis were randomized into two groups,with 50 cases in each. Both groups were administered basic treatment + ultrafiltration and reinfusion of ascites. The experimented group was added Xiaogu Granule and the control groups was added placebo. Before treatment, 3 and 6 months after treatment, the portal vein diameter (PVD), splenic vein diameter (SVD), blood flow quantity of portal vein (PVQ) and of splenic vein (SVQ), and the level of serum alanine aminotransferase, total bilirubin, endotoxin, nitrous oxide (NO) and endothelin (ET) of the two groups were measured respectively. Uncoupling protein 2 (UCP2) gene polymorphism was assayed by PCR in 201 diabetic patients with various degress of albuminuria. The frequency of 3′-UTR 45 bp insertion/deletion genotypes of UCP2 gene in these diabetic patients does not show any significant difference.
4.Association between uncoupling protein 2 gene polymorphism and macroangiopathy in diabetes mellitus

Objective To investigate the effects of uncoupling protein 2 (UCP-2) gene a 45 bp insertion/deletion (Ins/Del) polymorphism in the 3'-untranslated (3'-UTR) of its exon 8 on macroangiopathy in diabetes mellitus.Methods A total of 182 patients were selected,80 cases with macroangiopathy( A group), 102 cases without macroangiopathy(B group) ,UCP-2 gene polymorphism was confirmed by electrophoresis after PCR with 3% agarose,then compared genotype and allele gene frequency. Results The 3'-UTR Ins/Del polymorphism of UCP-2 gene in A group ( II:6. 25% 、ID: 18. 75% 、DD:75.00% ) and B group( II:9. 80% 、ID:23.53%、 DD: 66. 67% ) had no significant difference ( P > 0. 05 ), and there was also no difference of alleles frequencies in two groups ( I: 15.63 %、 D: 84. 37 % )and(I:21.57% 、D:78.43%)(P >0.05). Conclusion No relationship of the 3'-UTR a 45bp Ina/Del polymorphism in exon 8 of the UCP-2 gene was found with macroangiopathy in diabetes mellitus.
5.Influence of multidrug resistance gene 1 C3435T genetic polymorphism on the eradication of gastric ulcer with Helicobacter pylori infection

Objective To study the influence of multidrug resistance gene (MDR)1 C3435T genetic polymorphism on the eradication of gastric ulcer with Helicobacter pylori (Hp) infection.Methods A total of 106 gastric ulcer patients with positive Hp were randomly divided into two groups by lot with 53 cases each.One group was assigned with 20 mg esomeprazole,0.5 g clarithromycin,1.0 g amoxicillin twice one day(EAC group),and the other group was assigned with 20 mg omerprazole,0.5 g clarithromycin,1.0 g amoxicillin twice one day (OAC group).The therapy of two groups was one week.Hp was detected at least 4 weeks after the end of treatment.MDR1 C3435T genetic polymorphism was determined by polymerase chain reaction-restriction fragment length polymorphism assay.The influence of MDR1 C3435T genetic polymorphism on the eradication of Hp was recorded and analyzed.Results The Hp eradication rate was 84.9% (45/53) and 77.4% (41/53) in EAC group and OAC group,and there was no significant difference between two groups (P > 0.05).There was no significant difference in the Hp eradication rate in patients with different MDR1 C3435T genotypes in two groups (P > 0.05).The Hp eradication rate was 66.7%(16/24),86.3%(44/51),83.9%(26/31) in TT,CT,CC genotype,and there was significant difference(P< 0.05).The Hp eradication rate in patients with TT genotype was lower than that in patients with CT,CC genotype,and there was significant difference (P< 0.05).Conclusion There is significant relationship between the effect of gastric ulcer with Hp eradication and MDR1 C3435T genetic polymorphism,and the Hp eradication rates of patients with TT genotype are more lower.
6.Relationship between uncoupling protein 2 gene promoter-866 G/A polymorphism and Chinese diabetic nephropathy

Objective To investigate the relationship between uncoupling protein 2 (UCP-2) gene promoter -866 G/A polymorphism and Chinese diabetic nephropathy (DN). Methods A total of 182 patients with type 2 diabetic mellitus were selected and divided into DN group (90 cases with DN ) and NCD group (92 cases without DN ). Genomic DNA was detected, and UCP-2 genotype and allele gene frequency was confirmed by polymerase chain reaction-restrictive fragment length polymorphism, then compared.Results The genotype frequency of UCP-2 gene promoter-866 G/A polymorphism was distributed in DN group[GG 14.44%( 13/90),GA 31.11%(28/90),AA 54.44%(49/90)],and distributed in NCD group[GG 29.35% ( 27/92 ), GA 32.61% ( 30/92 ), AA 38.04% ( 35/92 )], and there was significant difference between two groups ( χ2 = 7.28 , P < 0.05 ). There was also significant difference in allele gene frequency between DN group and NCD group[G 45.65% (84/184) vs. 30.00% (54/180),A 54.35% (100/184) vs. 70.00% (126/180)]( χ2 = 9.47, P < 0.05 ). Conclusion There is correlation between the UCP-2 gene promoter-866 G/A polymorphism and Chinese DN, and the incidence of DN with A/A genotype is increased.
