2.Use of translational medicine in the early diagnosis of xenobiotic-induced intrauterine growth retardation.
Acta Pharmaceutica Sinica 2011;46(1):30-4
Translational medicine is an emerging idea in current medical research area. Typically, for the purpose of bridging the gap between basic and clinical research, it not only emphasizes the urgency and necessity to break the traditional working formats, including single subject centered research team and limited cooperation among different scientific groups, but also highlights a more close and frequent interaction between basic scientist and clinician. In order to reach this goal, the theory and method of systems biology should be employed. This paper mainly focused on a central issue that how to carry out an investigation on early clinical diagnosis of xenobiotic-induced intrauterine growth retardation (IUGR) by using research concept of translational medicine and method of systems biology. Briefly, a hypothesis of common mechanism of IUGR was first proposed and subsequent validation was performed via integrating--omics (e.g. genomics, proteomics, cytomics, metabonomics/metabolomics) and molecular biology techniques. Metabonomics was further utilized to explore IUGR biomarker and establish preliminary forecasting model by bioinformatics and computational biology, which is available for early diagnosis of IUGR and make a complement to current evaluation criteria.

Objective To investigate the feasibility of repairing spinal tracts with peripheral nerve graft combing neurotrophic factors in rats following complete spinal cord transection. Methods One hundred and twenty-one male Wistar Rats were transection at T9 level of spinal cord, and randomly divided into five groups. Group A with spinal cord transection was underwent acidic fibroblast growth factor (aFGF) treatment and peripheral nerve grafts (n=25); Group B: spinal cord transection was underwent aFGF treatment only (n=25); Group C: spinal cord transection was underwent peripheral nerve grafts only (n=25); Group D: spinal cord transaction only (n=25); and Group E: sham control (laminectomy only, n=21). The locomotor behavior of all rats was analyzed by the BBB open field locomotor test over the six months of survival time. Motor evoked potentials (MEP) were used to evaluate axon growth across the damage site. Biotinylated Dextran Amine (BDA) and retrograde tracing with fluorogold were used to evaluate the presence of axons through the damage site after treatment. Results The presence of anterograde BDA labeling of corticospinal tract axons at the graft site and fluorogold retrograde labeling of neuron populations was found in motor cortex and in red nucleus, reticulospinal nuclei, raphe nuclei, and vestibular nuclei in Group A. The average latency and amplitude of MEP were improved significantly in Group C. The mean of BBB scores showed significant improvement in Group A. Statistical analysis indicated that Group A had significant improvement compared to Group BC and D at 6 months post-surgery (P

PurposeCurrent conventional imaging methods X-ray, CT, MRI and ultrasound for rheumatic diseases are mostly local site imaging, rather than systemic display, thus is difficult to show the whole picture of the diseases, this study aims to explore the18F-FDG PET/CT findings of rheumatic diseases and its preliminary clinical value. Materials and Methods18F-FDG PET/CT imaging findings and clinical data of 18 patients with rheumatic diseases confirmed by clinical diagnosis were retrospectively analyzed, and 18F-FDG uptake of lesions was observed. Results Among all the 18 patients with rheumatic diseases: four cases were with rheumatoid arthritis (RA), 5 cases with takayasu arteritis (TA), 2 cases with adult onset Still’s disease (AOSD), 1 case with polymyalgia rheumatica (PMR) and 6 cases with ankylosing spondylitis (AS). Findings of PET/CT: RA showed increased FDG uptake in multiple bigger joints such as shoulder joint, elbow, hip, wrist joint, and part palm finger joints, with no bone abnormalities in CT; TA displayed as diffusely increased uptake of FDG the aorta and its major branches vascular wall while CT showed multiple arterial wall irregular thickening with dot or strip calcification, and in one case, the vascular wall FDG uptake decreased obviously after one year treatment, the SUVmax of left clavicle dropped from 6.8 to 3.2; AOSD showed diffusely increased FDG uptake in spleen and bone marrow, while CT had no obvious spleen and bone abnormalities; PMR showed increased uptake of FDG in the atlantoaxial joint, shoulder joint, sternoclavicular joint, hip joint, pubic symphysis, ischial tuberosity and lesions around lumbar spinous process; only 1 case of 6 AS patients showed left acetabular articular surface hyperplasia and sclerosis with increased FDG uptake, while CT showed spinal multiple vertebral bone hyperplasia and sclerosis as bamboo change in all of 6 AS cases, with normal FDG metabolism. Conclusion18F-FDG PET/CT is helpful for the diagnosis of rheumatic diseases, because it can be used to display the lesion extent, reflect its activity, and also to evaluate the effect of the treatments.
