Objective: To observe effective of antitumor immunity elicited by heat shock protein70(HSP70) associated peptide complexes isolated from hepatoma (HCaF), a tumor model of no MHC Ⅰmolecule expression in mice. Methods: Specific active immunization and adoptive cellular immunization assay was adopted to observe the immunoprotective effect elicited by HSP70 associated peptide complexes purfied from the HCaF. Results: Mice immunized with HSP70 associated peptide complexes were protected from HCaF challenge. The immunoprotective effect in female mice was better than the male groups. Adoptively transferred immune spleen cells of mice which had been immunized with HSP70 associated peptide complexes could be elicited immunity against HCaF challenge, and the mice free of tumor can resist repeated challenge, the maximal challenge quantities exceeded 1?10 7 HCaF cells. Mice immunized once with spleen cells pulsed by HSP70 associated peptide complexes could also be elicited certain immunity against HCaF challenge. Conclusions: HSP70 associated peptide complexes derived from the HCaF can elicit no MHC Ⅰ molecule restrictive immunoprotective effect against HCaF and could be continuously enhanced by repeated challenge with alive HCaF cells. Adoptively transferred immune spleen cells of mice which had been immunized with HSP70 associated peptide complexes could provide protection against HCaF cells challenge.

Chinese Journal of Clinical and Experimental Pathology 2001;(2):136-138
Purpose To investigate relationship between androgen receptor and apoptotic index and to further understand the tumor biology of prostate cancer. Methods Fifty-six patients with histologically proven prostate cancer and 20 cases with benign prostatic hyperplasia(BPH) were collected. Androgen receptor(AR) were stained by 2H12 monoclonal antibody using immunohistochemical method. The apoptotic index (AI) was determined by the terminal deoxynucleotidyl transterase-mediated dUTP biotin nick end labeling (TUNEL) technique on serial sections of formalin fixed, paraffin embedded tissues. Results AR was not found significantly difference (P>0.05) between prostate cancer and BPH. The association of AR with AI according to Gleason score were not observed in prostate cancer. AI were significantly higher in prostate cancer compared to BPH (P<0.05).AI were also significantly higher in AR-positive prostate cancer than in AR-negative prostate cancer(P<0.05). Conclusion AR expression can induce prostate cancer cells to become apoptosis, and may be a useful predictor in functional classification and endocrine response of prostate cancer.

Objective To establish a simple and practical superparamagnetic immunochromatographic test strip for rapidly monitoring human serum level of CA72-4. Methods Water-soluble carboxylated super-paramagnetic nanoparticles were prepared with a modified one-step hydrothermal synthesis method. Magnetic probe was prepared by immobilizing specific antibody (mAb1) onto the surface of nanoparticles. Following with optimization and assembly of the test strip , we evaluated sensitivity , specificity , stability of this method for serum CA72-4 detection. Results The optimized test strip provided not only the qualitative results, but also the high sensitivity quantitative detection through stable magnetic signal. The detection limit was 0.83 IU/mL. One hundred clinical samples ( 70 positive and 30 negative ) were measured to assess these test strips with high sensitivity (99%) and high specificity (93%). The test strip and magnetic signal possessed high stability. Conclusion A rapid and quantitative detection of CA72-4 by the test strip was accomplished. This method is rapid, sensitive and quantitative, possessing great potential in large sample screening or in-home testing.
