1.Biological Property and Tissue Distribution of Doxorubicin Albumin Microspheres in Rats

OBJECTIVE:To investigate the biological property of doxorubicin albumin microspheres(DR-HAS-MS) and its tissue distribution in the rats.METHODS:Pancreatin degradation method was applied to determine the biodegradation property of DR-HAS-MS as well as the correlation between the cross linking agent concentration and the degradation time and between the setting time and the degradation time.The dialysis method was applied to study the in vitro drug dissolution/ release characteristics of DR-HAS-MS vs.DR solution.DR concentrations in different tissue of rats at different time following intravenous DR HAS-MS vs.DR solution(at a dose of 2.5 mg?kg~(-1))were determined.RESULTS:The degra dation time increased with the increase of cross linking agent concentration and cross - link time.The optimal setting time was 90 min.Within 6 hours the dissolution ratio of DR from DR solution was 100%,while the releasing ratio of DR from DRHAS -MS suspension was only 80%within 168 hours;DR concentrations in the liver and the spleen in the DR HAS-MS group were significantly higher than in DR solution group(P

Objective:To investigate the expression of matrix metalloproteinase(MMP) and tissue inhibitor of matrix metalloproteinase(TIMP)in mesangial proliferative glomerulonephritis(MsPGN) rat renal tissue and the effects of nourishing kidney and activing blood recipe of traditional Chinese medicine.Methods:Chose 54 male rats and separated them into control group, MsPGN group and treated group with nourishing kidney and activing blood recipe of traditional Chinese medicine.Chose six rats per group at the time of the second,the fourth and the eighth week.The sections were treated with HE and PAS stains and the routine pathologic study was carried.Immunohistochemical staining was used to check the expression of MMP-2,TIMP-2, membrance type metalloproteinases-1(MT1-MMP) and transforming growth factor-?1(TGF-?1).Flow Cytometry was used to check the protein expression of MMP-2,TIMP-2,MT1-MMP and TGF-?1 in rat renal tissue.Results:In MsPGN group,following with the disease progressing,the glomerulus grew graduatly,ECM became more and more,the basement membrane of glomerulus was thicker.The immunohistochemical staining showed the expression of MMP-2 and MT1-MMP was lower compared with control group,but in treated group the level was weaker,the expression of TIMP-2 and TGF-?1 was higher compared with control group,but in treated group the level was weaker too. 4.Research on the development of database for the monitoring of radiation dose in cardiovascular interventional procedures

Objective:A database was designed and developed for effective utilization monitoring data of radiation dose to patients and occupational operators in cardiovascular interventional procedures. Methods:To complete a command reception of the client program, data display and other functions, compiled database software using MS Visual Basic level programming language. Using MS Access database completed data storage, management and maintenance. Results:The database was designed according to technological indicator acknowledged;the relevant help file was set up. The database includes two sub-libraries professional database and patient database. Professionals and patients can be recorded separately input, record view and record query. Conclusion:The data platform for monitoring data of radiation dose in cardiovascular interventional procedures was provided, which can provide basic data for constituting normalization and technical standard of dose limited of radiation dose to patients and occupational operators in cardiovascular interventional procedures.
5.Tooth enamel electron spin resonance dosimetry of people living in the area with lime tobacco custom

Objective To study the effect of the custom of long term chewing lime tobacco on human tooth enamel electron spin resonance (ESR) dosimetry. Methods A total of 20 enamel samples from lingual parts of adults teeth collected in Bombay, India were obtained by mechanical method. Some enamel samples from Japanese adults were extracted and 10 mixed samples were prepared. Enamel samples were exposed to different doses of 60Co γ-rays several times, and ESR spectra were measured after exposure. Results ESR background signals of 9 Bombay samples were found 1.5-3.3 times higher than those of Japanese mixed samples. The γ-ray dose responses of dosimetric signal with higher background level were a little lower, and the average sensitivity was (0.42 ± 0.03 )mGy, which was close to that of Japanese mixed samples. Conclusions The average level of background signals of Bombay samples was much higher than that of other non-chewing tobacco area, which was possibly caused by tobacco lime, the main component in chewing tobacco productions, and it would help to explore its special influences on ESR, and improve dose reconstruction in accuracy.
6.Primary Study on Fingerprint of Nutmeg in Different Areas

Objective To establish the fingerprints analysis of the methanol extracts of nutmeg,and study quality uniformity of nutmeg in different areas.Methods A ZORBAX EclipseXDB-C18 (4.6 mm?150 mm,5 ?m) column was used.The mobile phase consisted of methanol-acetonitrile-water (25∶35∶40),the flow rate was 1 mL/min,the column temperature was 30 ℃,the detective wavelength was 270 nm.Dehydrodiisoeugenol was used as reference compound.Results Fingerprint of nutmeg was established,consisted of l7 common peaks.The similarity of fingerprints was over 0.9.Conclusion The fingerprints of nutmeg in different areas have no differences.This method is accurate,reliable and provides a scientific basis for the quality control of nutmeg.
7.Method and Complications of Direct Fetal Blood Sampling

Objective To study the indications of percutaneous ultrasound-guided direct fetal blood sampling; the methods of fetal blood assessment; and the relationship between the different puncture techniques and the complications. Methods Direct fetal blood sampling were performed on 90 pregnant women whose gestational age were from 16 to 36 weeks for medical indications under the guide of ultrasound. None stress test were performed before and after the puncture. Ulrtrasonographies were done at 2 hours and 24 hours after the procedure. Results There were no severe maternal or fetal complications such as fetal loss and placental abruption. The incidence of bradycardia was 35.6%(32/90), all recovered within 60 seconds; the incidence of bradycardia happened within 4 hours after the puncture was 2.25(2/90). There was a positive relationship among the incidence of bradycardia, the volume of blood taken and the place of punctures,r=0.27 and 0.36 respectively. There was one case with placental changing happened after 2 hours of the puncture, the placenta changing incidence was 1.1%. No emergency cesarean section was done due to the complications of the procedure. All the umbilical cord were normal when the babies were born. Conclusions Direct fetal blood sampling is a safe technique for both fetus and the mother under the guide of real time ultrasound. The data come from fetal blood analysis give reliable informations about chromosome, congenital infections, the status of fetal metabolism, endocrine system and hematologic system directly, which make this technique unreplacable for intrauterine diagnosis.
8.Effects of glucosidorum tripterygii tororum on cytokine productions in graft-versus-host disease mice

AIM: To observe the effect of glucosidorum tr ipterygii tororum (GTT) on cytokine productions in acute graft-versus-host disea se (aGVHD) mice. METHODS : C 57 BL/6 mice were exposed to radiation delivered by a linear accelerator . To es tablish a aGVHD model, the cell suspensions, which were obtained from bone marro w and spleen of the BALB/C mice, were transplanted to the radiated C 57 BL/6 mice. The recipients were treated with GTT, GTT+CsA and CsA+MTX. The serum conc entrations of IL-2, TNF-?, IL-4 and IL-10 were determined by ELISA. RES ULTS: The survival rate on day 11 in GTT group (9/10) was higher than in allogeneic bone marrow transplatation (allo-BMT) group (8/19). The concentratio ns of IL-2 and TNF-? in GTT group were significantly lower, but the concentrati on of IL-10 was remarkably higher than that in allo-BMT group ( P 0 0 5). CONCLUSION: GTT inhibited aGVHD development by regulating th e production of cytokines in the host.
9.CLONING OF ZNRD1 AND EFFECT OF ITS ANTISENSE VECTOR ON ACCUMULATION OF ADRIAMYCIN IN DRUG-RESISTANT GASTRIC CANCER CELLS

The aim of this study was to construct antisense of ZNRD1 encoding gene, and to transfect it into SGC7901/VCR cells, to seek measures to overcome multidrug resistance of gastric cancer cells. ZNRD1 cDNA amplified by PCR was confirmed by DNA sequencing, and then inserted into the multiple cloning site of the expressing vector pcDNA3 1 + with molecular cloning technique. The recombinant vector was identified by endonuclease digestion. Antisense recombinant vector was transfected into SGC7901/VCR cells using lipofectamine. Flow cytometric analysis was used to detect adriamycin (ADM) accumulation in SGC7901/VCR cells. The results showed that a fragment was obtained by PCR, and its sequence was consistent with ZNRD1 cDNA reported in the literature. After antisense recombinant vector having been transfected into SGC7901/VCR cell, ADM concentration in such cells was increased. The above results indicated that the antisense vector ZNRD1 could enhance the intracellular accumulation of ADM in SGC7901/VCR cells, which might be of potential treatment value.
10.Analysis of related factors of postoperative delirium in elderly patients

Objective To explore related influencing factors for postoperative delirium in elderly patients, to provide a reference for the prevention and treatment. Methods Fifty-four patients with delirium after surgery were used as observa?tion group, and a total of 150 subjects with no delirium after surgery during the same period were selected as the control group. Data of age, gender, malnutrition, disorders of water and electrolyte metabolism, postoperative mechanical ventilation, postoperative hypoxemia, severe infection and postoperative pain degree, and the combination of basic diseases were com?pared and analysed between two groups. The binary logistic regression analysis was used to analyse the influencing factors of postoperative delirium. The outcome and prognosis were observed and analyzed in observation group. Results The average age was significantly higher in observation group than that of control group (P<0.05). The percentages of postoperative hy?poxemia and severe infection were significantly higher in observation group than those of control group ( P<0.05). Patients with higher age, postoperative hypoxemia and severe infection were risk factors for postoperative delirium. In observation group,1 case died of lung infection, 1 case died of multiple organ failure, the remaining 52 patients were improved and dis?charged from hospital after three months. Conclusion For patients with higher age, postoperative hypoxemia and severe in?fection are the risk factors for occurrence of postoperative delirium. More attention should be paid to clinical preoperative and postoperative periods.
