1.Synergistic antitumor effects via apoptosis of quercetin in combination with cisplatin in the prostate cancer cells

Objective To study the synergistic antitumor effects of quercetin and cisplatin in human prostate cancer PC3 cells and LNCaP cells.Methods Twelve h after PC3 cells or LNCaP cells were seeded,different dose of quercetin (10 μmol/L,20 μmol/L,40 μmol/L,80 μmol/L,160 μmol/L) or cisplatin (0.01 μmol/L,0.05 μmol/L,0.1 μmol/L,1 μmol/L,10 μmol/L),or quercetin (20 μmol/L) + cisplatin (0.01 μmol/L,0.05 μmol/L,0.1 μmol/L,1 μmol/L,10 μmol/L) were added for48 h and then the antiproliferative effects were detected with MTT assay.After incubated with quercetin (20 μmol/L) or cisplatin (0.05 μ mol/L),or quercetin (20 μ mol/L) + cisplatin (0.05 μmol/L) for48 h,cell cycle distribution and apoptosis of PC3 cells or LNCaP cells were detected by flow cytometer,PI and Annexin V staining.Protein expression was detected by Western blotting.Results After treatment with quercetin or cisplatin alone,the IC50 were (0.99 ± 0.13) μmol/L,(0.75 ± 0.09) μmol/L and (91.60 ± 6.10) μ mol/L,(72.90±4.70) μ mol/L for LNCaP cells or PC3 cells,respectively;The IC50 were (0.11±0.06)μ mol/L,(0.07±0.02) μmol/L for quercetin + cisplatin treatment (Compared with quercetin,P<0.01 ;Compared with cisplatin,P<0.05.After treatment with cisplatin or quercetin + cisplatin for 48 h,the S phase percent of LNCaP cells or PC3 cells were (22.4±2.7)%,(31.2±2.4)% and (20.1±1.6)%,(31.0±2.5)%,respectively,(Compared with control,P<0.05,however,treatment with quercetin alone has no significant difference (Compared with control,P>0.05).After treatment with cisplatin or quercetin + cisplatin for 48 h,the apoptotic percent of LNCaP cells or PC3 cells were (14.8 ± 1.9) %,(39.6 ± 3.1) % and (11.5± 1.2) %,(34.1 ±3.3) %,respectively,(compared with control,P < 0.05,however,treatment with quercetin alone had no significant difference (compared with control,P>0.05).After treatment with quercetin alone for 48 h,the activation of caspase-3,caspase-8 and caspase-9 were slightly increased,the expressions of Bax and p21 were up-regulated,the expressions of Bcl-2 and CDK2 were down-regulated.Furthermore,these effect of cisplatin and quercetin + cisplatin were significantly enhanced (compared with quercetin,P<0.05;compared with quercetin,P<0.01,respectively.Conclusions The combination modality with quercetin and cisplatin has a better treatment effect in vitro not only in androgen-dependent LNCaP cells but also in androgen-independent PC3 cells.
2.Anterior cingulate and amygdala volume changes in first-episode, medication naive females with major depressive disorder: a voxel-based morphometric MRI study

Objective To research the changes of regional gray matter volumes in first-episode, medication naive females with major depressive disorder. Methods Magnetic resonance imaging and voxel-based mor-phometry were used to compare regional gray matter volumes between 14 medication-naive female participants with a first episode of major depressive disorder ( MDD) ( age( 29. 5 ±6.8) years, number of education years(11.4 ± 2. 7) years, duration of iIIness(5.4 ±5. 2 ) months, H AMD score(27.6 ±5.5)) and 14 female healthy comparison participants (HC) (age(29.5 ±6.9)years,number of education years( 12. 2 ±3. 3)years,HAMD score(2. 1 ± 1.9)). Results Volumes of bilateral anterior cingulate cortex ( ACC) and right amygdala were significantly smaller in the MDD group than those in the HC group. Conclusion These results suggest that volume reductions in ACC and amygdala in females with MDD are present at illness onset.
3.Formula Optimization of Aspirin Chitosan-Sodium Alginate Microcapsules and Drug Releasing Mechanism Study

OBJECTIVE:To prepare aspirin chitosan-sodium alginate microcapsules(ACSPM)and to investigate its optimal formula and releasing mechanism.METHODS:The formula of ACSPM was optimized by the orthogonal design with entrapment ratio as index,and then ACSPM was prepared,with its release rate determined as well.The releasing mechanism of aspirin from the microcapsules was established by equation fitting of releasing kinetic model.RESULTS:The prepared microcapsules were uniform in size and contents.The optimized formula was as follows:the concentration of sodium polymannuronate and chitosan were 3.0% and 1.0%,respectively,and the proportion of polymannuronate to aspirin was 1∶ 4.The in vitro drug release was in line with both Higuchi equation and Peppas equation.CONCLUSION:This preparation technology was simple and the drug releasing mechanism of the preparation was chiefly characterized by drug diffusion including bulk erosion non-Fickian process.
4.Preparation Technology of Ketoconazole Calcium Alginate Gel Beads

OBJECTIVE:To investigate the preparation technique and optimal formulation of ketoconazole calcium alginate gel beads.METHODS:Ketoconazole calcium alginate gel beads were prepared by droplet method.The orthogonal experiment was carried out with encapsulation efficiency(EE)and drug loading(LD)as indexes to optimize the optimal formulation of the calcium alginate gel beads.The encapsulation efficiency and drug loading amount for the optimized formulation were determined,and the optimized formulation was compared with crude drug in respect of the in vitro drug release behavior.RESULTS:The result showed that the optimal formulation was as follows:2.0% Na-alginate and 0.3mol? L-1 CaCl2 with the ratio of Na-alginate:ketoconazole at 2∶ 1.The average EE and LD were(90.53? 2.32)% and(31.51? 2.08)%,respectively,and the slow-release was better as compared with that of the crude drug.CONCLUSION:The preparation procedure is simple,feasible,stable and reproducible.
5.Preparation of Breviscapine Chitosan-alginate Microcapsula

Objective To investigate the preparation technique and optimal formulation of Breviscapine Chitosan-alginate Microcapsula. Methods Breviscapine Chitosan-alginate Microcapsula was prepared by coacervate technology. The orthogonal test design by adopting the standard of drug encapsulation efficiency and drug loading was applied to obtain the optimal formulation of the microcapsula. Results The result showed that the optimal formulation was that Na-alginate 25 mg/mL, chitosan 2 mg/mL, CaCl2 0.2 mol/L, and Na-alginate-Breviscapine 1∶1. Conclusion The preparation procedure is simple, feasible, stable, and repeatable.

First, this article systematically introduces the process of PBL in the organ system integrated course which is conducted in Nanshan College of Guangzhou Medical University. Second, this article discusses the teaching results and reflection so as to provide reference for the continuous localization reformation of PBL and further medical curriculum integration in college and university.
7.Research on serum homocysteine level and its relationship with oxidative stress in elderly patients with obstructive sleep apnea/hypopnea syndrome and the inner mechanism

Objective To investigate the association between serum homocysteine (Hcy) level and oxidative stress in elderly patients with obstructive sleep apnea/hypopnea syndrome (OSAHS). Methods The 123 patients from January 2007 to June 2010 were recruited and divided into the elderly OSAHS group (n=55) and the non-elderly OSAHS group (n=68). One hundred healthy subjects were selected and divided into the elderly control group (n=52) and the non-elderly control group (n=48). The PSG variables were recorded and the levels of Hcy, malondialdehyde (MDA) and glutathione (GSH) were detected after sleep in all subjects. Serum Hcy level was measured by cyclophorase, MDA and GSH levels were measured by spectrophotometer. Results There were statistical differences in Hcy concentration between four groups (F=28.26, P=0.000). The Hcy concentration was higher in elderly OSAHS group than in other groups, and was higher in elderly control group than in non-elderly control group, and was higher in non-elderly OSAHS group than in non-elderly control group. There were statistical differences in MDA and GSH concentrations between the four groups (F=5.56, P=0.002;F=4.79, P=0.004). The concentrations of MDA and GSH were higher in elderly OSAHS group than in other groups. Multiple regression analysis indicated a statistically significant relationship of Hcy concentration with age, MDA, GSH and AHI (β=0.07, 0.50, 0.23 and 2.00). Conclusions The concentration of Hcy increases and oxidative stress enhances with aging, especially in the elderly patients with OSAHS. The mechanism of high level of serum Hcy in elderly patients with OSAHS may be the high oxidative stress.
8.The clinical application of imaging-pathology fusion guided targeted cryoablation for localized prostate cancer

Objective To investigate the value of imaging-pathology fusion guided targeted cryoablation for localized prostate cancer.Methods Between June 2014 and February 2017,64 patients undergoing targeted cryoablation for localized prostate cancer were retrospectively collected and analyzed.The average age was 77.8 years old,ranging 45-87 years old.The preoperative valne of PSA ranged from 5.1 to 19.8ng/ml,with mean of 15.3 ng/ml,including 6 case of cT1c,35 cases of cT2a,and 23 cases of cT2b,and 35 cases of Gleason score 6,8 cases of 3 + 4 and 21 cases of 4 + 3.All patients had good continence and 11 cases had good erectile function before operation.All the patients received MRI or contrast-enhanced ultrasonography examinations before biopsies,and the sites and range of cryoablation were determined by MRI or contrast-enhanced ultrasonography and biopsy pathological results.Results All the patients underwent the surgery successfully.The average postoperative hospital stay was 1.6 days,ranging 1 to 3 days,including 22 cases discharged within 24 h after surgery.The operative duration ranged from 85 to 152 min,with mean of 99.6 min.The satisfaction rate of the patients was 100%.Seven patients had Clavien Ⅰ or Ⅱ postoperative complications,and recovered after symptomatic treatment.The follow-up duration ranged from 0.5 to 32.2 months,with median of 11.8 months.All patients were alive without new metastasis.Eight patients developed biochemical recurrence,5 of those underwent intermittent endocrine therapy,resulting in stable PSA levels.Conclusion Imaging-pathology fusion guided targeted cryoablation was safe and effective for localized prostate cancer with the advantages of few complications,high satisfaction rate and quick recovery.
9.Diagnostic value of confocal laser endomicroscopy in predicting the grading of gastric intestinal metaplasia

Objective To evaluate the efficacy of confocal laser endomicroscopy(CLE)for the assessment of the grading of gastric intestinal metaplasia(GIM)in vivo.Methods Patients with known GIM underwent CLE(Pentax EC-3870K).The presence of GIM was indentified immediately by the endoscopist during the procedure.The updated Sydney Classifycation System as reference,GIM was subdivided as mild,moderate and severe according to the area of intestinal metaplasia glands and the number of goblet cells.The histological evaluation remained the gold standard for the final diagnosis of GIM.The presence of the dysplasia and the expression of the Ki67 were examined.Results A total of 151 GIM positive areas were found in 58 patients with mild in 92,moderate in 34.and severe in 25 by CLE. One hundred and forty-six GIM areas were examined histopathologically with positive rate of 96.7%(mild in 82,moderate in 36 and severe in 28).The sensitivity and specificity of CLE were 90.2%and 73.9%in diagnosis of mild GIM,69.4 oA and 92.2%in moderate GIM,71.4% and 95.9%in severe GIM.The kappa coefficient of CLE criteria and the histopathological grading for mild,moderate and severe GIM were 0.65,0.63 and 0.70,respectively.The more severe the GIM,the higher the ratio of incomplete GIM,the ratio of dysplasia and the stronger expression of Ki67.Conclusions CLE may offer an instant and reliable diagnosis for GIM with high accuracy.It is helpful in grading of GIM.
10.Effect of livin-modified BM-MSCs transplantation on cardiac function fol-lowing acute myocardial infarction in a rat model

[ ABSTRACT] AIM:To study the effect of livin gene-modified bone marrow mesenchymal stem cells ( BM-MSCs) transplantation on the cardiac function following acute myocardial infarction in a rat model and the expression of livin , caspase-3, caspase-7 and caspase-9 in the livin gene-modified BM-MSCs.METHODS: The MSCs were obtained by the whole bone marrow culture method , and the apoptosis of the MSCs after infection with adenovirus vector carrying enhanced green fluorescent protein ( EGFP) gene and livin recombinant vector ( rAd-livin) were detected by flow cytometry .The ex-pression of livin, caspase-3, caspase-7 and caspase-9 was detected by Western blot .After permanent left anterior descend-ing artery occlusion , the rats were randomized to receive intramyocardial injection of DMEM without cells ( vehicle group ) , or containing MSCs ( MSCs group ) , MSCs ( EGFP ) ( rAd-control/MSCs group ) or MSCs ( livin ) ( rAd-livin/MSCs group).Left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP), the maximum in-creased rate of left ventricular pressure ( -dp/dtmax ) and the maximum decline rate of left ventricular pressure ( +dp/dtmax ) were recorded for evaluating the cardiac functions .RESULTS: The apoptosis of rAd-livin/MSCs was significantly decreased as compared with MSCs and rAd-control/MSCs (P<0.05).Meanwhile, the expression of caspase-3, caspase-7 and caspase-9 was significantly downregulated as compared with the other 2 groups ( P<0.05 ) .The cardiac function in rAd-livin/MSCs group was significantly improved as compared with DMEM group , and those in the other 2 groups got the similar results, but the function in rAd-livin/MSCs group was better improved .Meanwhile, the number of surviving cells in rAd-livin/MSCs group was significantly improved as compared with the other 2 groups .CONCLUSION:The apoptosis of MSCs is decreased after rAd-livin transfection, and the expression of caspase-3, caspase-7 and caspase-9 is also significant-ly downregulated while the expression of livin is significantly upregulated .Transplantation of livin-modified BM-MSCs by lentiviral vector results in better prognosis for treating myocardial infarction by enhancing cell survival .
