1.Effects of the aqueous extract of dendrobium nobile lindl on NF-κB and IL-6 expression of renal tissue in diabetic rat model

Objective To observe the effects of dendrobium nobile Lindl(DNL) on Nuclear factor(NF)-κB and interleukin(IL)-6 expression of renal tissue in diabetic rat model .Methods The diabetes mellitus rat model was induced by streptozotocin .All rats were divided into four groups at random :the normal control group(group N) ,diabetic control group(group diabetes mellitus) ,drug group one(group DNL1) ,drug group two(group DNL2) ,and administrated for 12 weeks .The levels of the fasting plasma glucose , urea nitrogen ,serum creatinine ,urine creatinine ,24 hours urinary protein were determined .NF-κB and IL-6 protein expression levels were measured by Western-Blot .IL-6 mRNA expression levels were measured by Real-time fluorescence quantitative PCR . Results Compared with group N ,the blood sugar ,urea nitrogen ,24 hours urinary protein of group diabetes mellitus were signifi-cantly higher(P<0 .01) ,creatinine clearance was significantly reduced (P<0 .01) ,the NF-κB、IL-6 protein expression levels and IL-6 mRNA level increased significantly (all P<0 .05) .Compared with group diabetes mellitus ,the blood sugar ,urea nitrogen ,24 hours urinary protein of group DNL1 and group DNL2 were significantly decreased(P<0 .05) ,creatinine clearance increased significantly (P<0 .05) ,the NF-κB、IL-6 protein expression levels and IL-6 mRNA level decreased significantly(all P<0 .05) .Conclusion The DNL could protect the kidney of diabetic rats ,reduce the levels of NF-κB 、IL-6 protein and IL-6 mRNA expression in renal tissue of diabetic rats .
2.Aorta-pulmonary septal defect and aortic origin of the right pulmonary artery with interruption of the aortic arch: a clinical analysis of 5 cases

Objective Reviewing the experience in the diagnosis and treatment of cases with aorta-pulmonary septal defect and aortic origin of right pulmonary artery complicated with interruption of the aortic arch. Methods Reviewed clinical data from medical records for5 patients (4 boys and 1 girl, age from 1 to 18 years on admission) who had been treated in Wuhan Asia Heart Hospital between 2005 and 2009. Results All cases had type A interruption of the aortic arch ( according to Celoria and Patron's Classification) , type Ⅱ aorta-pulmonary septal defect( according to Mori Classification) and aortic origin of right pulmonary artery complicated with interruption of the aortic arch. Large patent ductus arteries were observed in all cases,and no cardiac abnormality such as ventricular septal defect was found. All patients received echocardiography and spiral CT examination, and 3 of them over 10 years of age received cardiac catheterization and angiography for the evaluation of the pulmonary artery pressure. Two patients, at the age of 14 and 18 years, had severe pulmonary hypertension and declined to receive the operation. One of the 2 cases had hemoptysis and was given prostacyclin ( Iloprost solution for inhalation, a drug for pulmonary artery hypertention ) for reducing the pulmonary pressure. The patient now has an improved cardiac fu0ction without recurrent hemoptysis. One 14-year-old case gave up the operation because of the financial problem and failed to communicate with us after discharge. Radical surgery was performed in the remaining three cases, one had serious infection in the lung and died 11 days after the operation, in one case ( 2 years old) pulmonary artery hypertension has been persisted even though drug therapy was given for a long time and was found at a poor cardiac function 18 months after the operation. One patient recovered well under routine drug therapy and has been followed-up. Conclusion Aorta-pulmonary septal defect and aortic origin of right pulmonary artery with interruption of the aortic arch ( with intact ventricular septum) are different from the classic triad of inter ruption of the aortic arch, consisted of the interruption of the aortic arch, patent ductts arteriosus and ventricular septal defect.It should be considered as a special kind of cardiovascular malformation syndrome. Echocardiography can be used for the preliminary diagnosis of aorta-pulmonary septal defect and aortic origin of right pulmonary artery with interruption of the aortic arch. The final diagnosis and decision for surgery should be based on the combined information from cardiac catheterization,cardiovascular angiography and spiral CT examination. Pulmonary artery pressure and resistance are the most important factors,because they have great effects on the duration of surgery and the outcome. A radical operation is necessary for all cases, but the mortality rate is high. The prognosis would be great if only the patient can survive successfully during the peri-operative period, a period with a poor cardiac function, potential lung infection and the pulmonary artery hypertension.
3.Strengthen the scientific management based on the demand oriented to promote the discipline construction of the hospital

Objective To explore the scientific management based on the needs of the clinical medical practitioners and the comprehensive promotion of the discipline construction of the hospital.Methods Totally 150 clinical professional technical staff and 33 academic leaders were investigated to explore the research needs and suggestions of them by questionnaire and expert interview.Results Among141 subjects,78.7% (111/141) medical staff thought that the major factors of restricted themselves to scientific research were the clinical heavy task and pressure and no time,and 58.2% (82/ 141) of them want to do research but lack of scientific research methods.Their favorite knowledge and skills were the research methods for 75.9% (107/141),and the knowledge about the research proposal design for 58.9% (83/141).The discipline construction evaluation of our hospital was divided into the key disciplines,the dominant disciplines,the developing of disciplines and the supporting of disciplines.And each discipline of level has its different needs.Conclusions To enhance of the scientific management of the scientific research base on the practical demands of the various disciplines can comprehensively promote the construction of the disciplines of the hospital.
4.The effects of natural static magnetic fields on the proliferation of three human hepatocarcinoma cell lines

Objective To observe the effects of a natural static magnetic field (NSMF) on the proliferation of human hepatocarcinoma cells. Methods Three human hepatocarcinoma cell lines ( BEL-7402,HEP G-2 and QGY-7701 ) were cultured under standard conditions and then exposed to NSMFs of 0.1 Tesla at 600 Hz,0.2 Tesla at 250 Hz,0.2 Tesla at 400 Hz or 0.2 Tesla at 500 Hz for 30 minutes daily for 3 or 6 days.Flow cytometry and the XTT cell proliferation assay were used to check apoptosis and the proliferation of cells all groups. Results Only exposure to 0.2 Tesla magnetism at 400 Hz induced apoptosis of the BEL-7402 cell line.None of the NSMFs induced apoptosis in the other 2 hepatocarcinoma cell lines. Conclusions Different reactions of the three hepatocarcinoma cell lines to the same NSMF implies that each cell line has its own features,and that individualized NSMF treatment protocols should be adopted for better clinical outcomes.
5.Study on the safety margin of esphagus in the radical gastrectomy of adenocarcionma of esophagogastric junction

Objective To discuss decision of the safety margin of esophagus in the radical gastrectomy of adenocarcionma of esophagogastric junction.Methods The length of esophageal resection margin of intraoperative,postoperative and pathological large slice under the microscope of 50 cases adenocarcionma of esophagogastric junction underwent radical gastrectomy were measured,and the proximal invasive length of esophagus in large pathological slice were also measured,and the relationship between it and pathological factors was analyzed.Determined the minimum safety cut edge distance.Results In 46 of 50 patients,proximal intramural spread of the tumor was observed.The invasive length of tumor from 0.1 cm to 3.5 cm.The invasive length of tumor was <1.0 cm with 11 patients,1.0-1.5 cm with 19 patients,1.5-3.0 cm with 15 patients,in only 1 patient was the distance of spread 3.5 cm.Invasive length of adenocarcionma of esophagogastric junction had a significant difference between the groups in different pathological factors(P<0.001).Conclusion The length of 3.5 cm is the safety margin of esophagus in the radical gastrectomy of adenocarcionma of esophagogastric junction(SiewertⅡ,Ⅲ type) by abdominal incision.

Objective To observe the effect of antisense hypoxia inducible factor-1?(HIF-1?) on (chemosensitivity) of human pancreatic cancer cell line BxPC-3 under hypoxia. Objective To investigate the anti-migratory and anti-invasive effect of somatostatin receptor type 2(SSTR2) gene transfection mediated by adenovirus in human pancreatic carcinoma cell and the mechanisms involved in this effect.Methods The full length human SSTR2 cDNA was introduced into pancreatic cancer cell line BXPC-3 by adenovirus-mediated transfection,and stable expression of RNA and protein of SSTR2 were detected by RT-PCR and Westen-blot.The Matrigel coated Transwell was used to detect the migratory and invasive ability of SSTR2expressing cells,Adv-GFP control cells and mock control cells.Furthermore,the expressions of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2) were detected by RT-PCR method in these cells.Results The stable expression of SSTR2 was detected in BXPC-3 cells transfected by Adv-GFP-SSTR2.A dramatic decrease of BXPC-3 expressing SSTR2 cell(migrated) through a Matrigel-coated filter was observed,as compared with Adv-GFP control cells and mock control cells(P

Objective To explore the expression and clinical significance of EphA2 and E-cadherin in (pancreatic) cancer tissue and cells.Methods The expression of EphA2 and E-cadherin in 56 pancreatic(carcinomas) and 23 adjacent noncancer tissues were studied by immunohistochemistry,and their relationship to clinicopathological characteristics were analyzed.RT-PCR was performed to explore the expression of EphA2,BXPC-3,PC-3 and PANC-1 in pancreatic carcinoma cell lines.Results In pancreatic carcinomas tissuse showed increased EphA2 expression and reduced E-cad expression which compared with adjacent noncancer tissues.The expression level of EphA2 had a significant positive relationship with tumor differentiation degree,lymphatic metastasis and clinical stage.However,the expression level of E-cadherin had negative relationship with both the tumor clinical stage and lymphatic invasion.Furthermore,a significant negative relationship(between) the expression of EphA2 and E-cadherin was observed.The expressions of EphA2 were higher in high-invasive cell lines PXPC-3 and Panc-1 than in low-invasive cell line PC-3.Conclusions The(expression) and/or abnormal function of EphA2 and E-cadherin may together be involved in the development and progression of pancreatic cancer;the combined measurement of these two proteins may be useful for(determination) of metastatic potency of pancreatic carcinoma.
10.Dynamic changes of mouse cardiac functions in early stage of sepsis induced by CLP

AIM:To detect the changes of cardiac functions of septic mice in the early stage of sepsis. METHODS:Health male Kunming mice were used in the study. The techniques of 2D,Mmode and Doppler echocardiography were applied to evaluate the cardiac functions before cecal ligation and puncture(CLP) as baseline and at time points of 12 h,24 h,36 h,48 h and 168 h after CLP. The mice survived for 168 h(7 d) were considered as survivals. RESULTS:Compared to the baseline at the time point of 24 h after CLP,the blood volumes of heart return decreased significantly in the early stage of sepsis induced by CLP. LVEDV reduced by 46%. Notable compensatory responses of the hearts in septic mice were observed,especially the systolic functions,in which LVEF and LVFS increased by 27% and 39% ,respectively. However,the compensatory responses of diastolic function were weaker than the systoles. E/A ratio and EDT decreased by 30% and 25% respectively at the time point of 24 h. CONCLUSION:The strong compensatory cardiac functions are one of the factors for supporting the septic animal to survive. Protection of the cardiac functions especially the diastoles is important in the treatment of septic patients.
