In the paper,we will deeply research sorts of ethical problems in medical genetics and their origins through a view of the human dual evolutions,and bring forward the viewpoint that any ethical problem can be originated from the conflict between human biological evolution and cultural evolution.Based on fully reasoning the relation between ethical problems in medical genetics and human evolution,the authors put forward a series of corresponding strategies to solve the ethical problems in medical genetics.
2.Effect of Electroacupuncture on Hypothalamic and Hippocampal Expressions of Ghrelin and Ghrelin Receptor mRNA in Functional Dyspepsia Rats

Objective To investigate the effect of electroacupuncture on hypothalamic and hippocampal expressions of ghrelin and ghrelin receptor (GHS-R) mRNA in functional dyspepsia (FD) rats. Methods Eighty SD rats were randomly allocated to normal control, model control, medication and electroacupuncture groups, 20 rats each. A rat model of FD was made by tail clamp modeling method. The medication group received an oral gavage of cisapride solution and the electroacupuncture group, electroacupuncture treatment. The expressions of ghrelin protein and GHS-R mRNA in the hypothalamus and hippocampus were examined by Western blot and real-time PCR, respectively, in all the four groups. Results Compared with the model control group of rats, the expression of ghrelin protein in the hypothalamus and the expression of GHS-R mRNA in the hypothalamus and hippocampus increased in the medication group (P<0.05) and the expressions of ghrelin protein and GHS-R mRNA in the hypothalamus and hippocampus were up-regulated in the electroacupuncture group (P<0.05). Compared with the medication group of rats, the expressions of ghrelin protein and GHS-R mRNA in the hippocampus were up-regulated in the electroacupuncture group (P<0.05). Conclusions Electroacupuncture and medication can improve gastrointestinal functions and correct an imbalance in the brain-gut axis through the hippocampus-hypothalamic pathway by regulating hypothalamic and hippocampal ghrelin protein and GHS-R mRNA contents and activating hypothalamic neurons and hippocampal excitatory synapses.
3.Hemodynamic changes of brain in newborns with intracranial hemorrhage

Objective To explore the hemodynamic changes in the brains of newborns with intracranial hemorrhage.Methods Totally,61 newborns,born in Anyang Maternal and Child Health Hospital of Henan Province,with intracranial hemorrhage diagnosed by ultrasound in early neonatal period,and 50 healthy newborns,also born in the same hospital from December 1,2010 to June30,2012,were selected.Intracranial hemorrhage newborns were graded according to the severity of hemorrhage and divided into mild (n=45) and severe groups (n =7).The peak systolic flow velocity (Vs),end diastolic velocity (Vd),mean flow velocity (Vm) and resistance index (RI) in the middle cerebral artery (MCA) and anterior cerebral artery (ACA) of brain were measured by Doppler ultrasound.Ttest was applied for statistical analysis.Results Among the 61 neonates with intracranialhemorrhage,12(19.7%) were grade Ⅰ,42(68.9%) were grade Ⅱ,6(9.8%) were grade Ⅲ,and one (1.6%) was grade Ⅳ.The Vs,Vm and Vd of MCA and ACA in the mild and severe intracranial hemorrhage group were significantly lower than those in the control group,the differences were statistically significant [MCA:(55.1±9.1) cm/s,(53.0±6.5) cm/s vs (60.1± 10.3) cm/s;(34.2±6.1) cm/s,(32.5±5.2) cm/s vs (38.2±6.9) cm/s; (17.1±4.8) cm/s,(15.3± 4.0) cm/svs (20.2±5.3) cm/s.ACA:(41.3±11.7) cm/s,(39.4±9.2) cm/s vs (45.3±9.8) cm/s;(25.2±5.8) cm/s,(23.3±4.9) cm/s vs (28.1±5.9) cm/s;(15.0±3.9) cm/s,(13.2±3.1) cm/s vs (15.9±3.8) cm/s,all P<0.05].But the RI values were significantly higher in the control group,the differences were statistically significant (MCA:0.70 ± 0.10,0.77 ± 0.07 vs 0.62 ± 0.10 ; ACA:0.67±0.06,0.73±0.08 vs 0.61±0.05;all P<0.05).TheVs,Vm and Vd of MCA and ACA in the severe intracranial hemorrhage group were significantly lower than in the mild hemorrhage group,while the RI values were significantly higher,and the differences were statistically significant (all P<0.05).Conclusions Newborns with intracranial hemorrhage have significant hemodynamic changes in the brain and more prominent changes could be found in those with severe intracranial hemorrhage.
4.Bone mineral density of schizophrenia patients and its influencing factors

Objective To explore the bone mineral density (BMD) status and its influencing factors in schizophrenia patients in order to provide basis for risk assessment in psychiatric nursing.Methods A total of 1,139 hospitalized schizophrenia patients were recruited and assigned into the medication group (n=652) and the non-medication group (n=487) according to previous antipsychotic drug history.T-score and Z-score of BMD were determined using Sunlight Omnisense 7000S Bone Densitometry.Blood calcium,blood phosphorus and serum prolactin levels were measured using fasting blood of ulnar vein.Results Differences in age,BMD,milk intake,level of activity,level of smoking,history of fall,history of fracture,serum Ca2+ and PRL were statistically significant between two groups(P<0.05);there were significant differences in BMD rank distributions among schizophrenia patients with different courses of disease and lengths of taking antipsychotics (P<0.001);multiple linear regression showed that influencing factors of BMD with statistical significance were courses of disease,lengths of taking antipsychotics,serum Ca2+,serum PRL,milk intake,level of activity,and level of smoking.Conclusion The BMD was lower in the medication group than that in the non-medication group,and the development of osteoporosis was correlated to various factors.Clinical nurses should master high-risk factors thoroughly and adopt intervention measures in a timely manner.
5.Observation of ultraweak luminescence of human hepatocarcinoma HepG2 cell

Objective:To explore the characteristics of the ultraweak luminescence of human hepatocarcinoma cells(HepG2). Methods: The intensity of the ultraweak luminescence of HepG2 cells and human hepatocytes(QZG)was examined by an IFFM-D chemiluminescence instrument.The intensity of the ultraweak luminescence of HepG2 cells and human hepatocytes(QZG)were affected by different concentration of Luminol and H_(2)O_(2). Results: In 10~(-4)mol/l Luminol and 0.3% H_(2)O_(2),the intensity of the ultraweak luminescence of HepG2 cells was significantly higher than that of QZG cells(P)0.05). Conclusion:The intensity of the ultraweak luminescence of HepG2 cells is higher than that of QZG cells.The intensity of the ultraweak luminescence is affected by cell number and the concentration of Luminol and H_(2)O_(2).The ultraweak luminescence reflect functional status of tumor cells.It can become an sensitive index of physiology and metabolism of tumor cells.
6.Effect of angiotensin II on cord blood CD34+ cells expansion in vitro.

In order to investigate the influence of angiotensin II on hematopoietic system, CD34+ cells in cord blood were purified, and the effects of angiotensin II in combination with various cytokines on their growth and differentiation were studied by cell culture in vitro. It was found that angiotensin II in suspending medium could stimulate both BFU-E and CFU-GM expansion. The number of BFU-E and CFU-GM was increased with the increases of angiotensin II concentrations during a certain range. In addition, the expansion fold of CFU-GM was increased from 2.3 +/- 0.8 times to 7.8 +/- 2.3 times when angiotensin II was added in the presence of SCF+G-CSF+GM-CSF+IL3 cytokines mixture. Similarly, the expansion fold of BFU-E was increased from 3.1 +/- 1.8 times to 9. 2 +/- 2.3 times with angiotensin II in the presence of SCF+EPO+TPO+IL-3. In the semi-solid medium, angiotensin II could stimulate CFU-GM expansion but had no effect on the growth of BFU-E. In conclusion, angiotensin II had some stimulating effects on cord blood hematopoietic progenitors expansion in vitro in the presence of other cytokines.

7.Optimization of the Formulation of Azithromycin Sustained-release Vaginal Suppository

OBJECTIVE:To optimize the formulation of azithromycin sustained-release vaginal suppository(ASVS).METHODS:The ASVS was prepared using S-40 as suppository base,HPMC as sustained-release material and glycerine as humectant.The formulation of ASVS was optimized by orthogonal design with accumulated release percentage and hardness as indexes and the amount of 8% HPMC and glycerol as factors;meanwhile,a verification test and the fitting of in vitro drug release model were performed.RESULTS:The optimized formulation ASVS was as follows:azithromycin 6 g,8% HPMC 23.52 g,glycerol 29.40 g,ethanol absolute 3 g,ethylparaben 0.59 g,and S-40 294 g.Three batches of suppositories prepared under the optimized formulation reached a mean content of 99.5%,with hardness up to the standard,showing good reproducibility and homogenicity in drug release in vitro.The accumulative release rates of all samples were greater than 98% at 180 min,and the release dynamics in vitro were in line with Higuchi equations.CONCLUSION:The optimized ASVS is feasible in formulation,stable and reproducible in preparation technology and up to the standard for sustained-release preparation.
8.MR imaging of anteromedial and posterolateral bundles of anterior cruciate ligament of knee joint

ObjectiveTo improve the visualization of anteromedial and posterolateral bundles of anterior cruciate ligament (ACL) and investigate the optimal MRI plane for the bundles at a 3.0 T MR scanner.MethodsMR images of 149 knee joints were reviewed retrospectively.Display rates of AMB,PLB and their different parts (the top portion,the middle portion and the low portion) on MR different planes including axial,sagittal and coronal planes were analyzed and their differences were compared with the x2 section method.ResultsThere was no statistical difference in the display rates of two bundles of ACL between axial plane ( 115/149,77.2% ) and coronal plane (103/149,69.1% ) (x2 =2.4606,P >0.0125 ).Statistical differences were found between axial and sagittal plane,coronal plane and sagittal plane (21/149,14.1% ) ( x2 =119.5138,92.8695 respectively,P <0.0125 ).There was a statistical difference for the top portion of ACL between axial plane ( 104/149,69.8% ) and coronal plane,sagittal (0/149,0)and coronal planes ( 7/149,4.7% ) ( x2 =135.081,159.7526 respectively,P < 0.0125 ),between sagittal and coronal planes (x2 =7.1684,P < 0.0125 ).For the middle portion of ACL,there was no statistical difference between axial plane ( 108/149,72.5% ) and coronal plane (94/149,63.1% ) (x2 =3.0120,P > 0.0125 ),while statistical differences were found between axial and sagittal plane,coronal planes and sagittal plane ( 10/149,6.7% ) ( x2 =134.7454,104.2173 respectively,P < 0.0125 ).For the low portion of ACL,there was no statistical difference between axial plane ( 103/149,69.1% ) and coronal plane (101/149,73.8% ) (x2 =0.8065,P >0.0125),while statistical differences were detected between axial and sagittal plane,coronal planes and sagittal plane ( 18/149,12.1% ) ( x2 =100.5300,115.9132,P < 0.0125 ). The different parts of ACL displayed low intensity on different MR planes and normal morphology.ConclusionsACL can be displayed on conventional MR planes at a 3.0 T MR scanner to some extent.Axial and coronal planes might be the optimal MRI planes for ACL and its two bundles.
9.Application of endoscopy combined with 125I seed implantation in the pancreatic cancer surgery in elderly patients

Objective To observe the application of endoscopy combined with 125I seed implantation in the pancreatic cancer palliative surgery in elderly patients.Methods A total of 78 elderly patients with pancreatic cancer who can not be treated with radical operation were divided into observation group and control group (n=39 each group).All patients were treated with stent intervention.Patients in the ation group were treated with 125I seed implantation guided by CT or ultrasonangraphy additionally.The clinical symptoms were compared between groups.The survival time and complications of the patients were recorded during the follow-up.Results The clinical symptoms of patients in both two groups were greatly improved (x2 =0.06,P>0.05).Compared with the control group,the pain remission was better and the incidence of pain caused by pancreatic cancer was lower in the observation group (46.2% vs.76.9%,x2 =7.80,P<0.05).There was a significant difference in the average of survival time between the observation group and control group [(13.7±3.4) months vs.(8.4±3.8) months,t=3.73,P<0.01],but no statistical difference was found in the long-term survival rate between groups.There was no significant difference in the incidence of complications between the two groups (10.3% vs.7.7%,x2=0.15,P>0.05).Conclusions Endoscopy combined with 125Ⅰ seed implantation is an effective treatment for elderly patients with advanced pancreatic cancer,which can effectively relieve the clinical symptoms,proiong the survival time,reduce the pain and complications.
10.Short tandem repeat analysis for diagnosing complete hydatidiform mole and coexisting fetus: a casereport and literature review

Objective To investigate the parental origin for a rare case of complete hydatidiform mole and coexisting fetus and to discuss its diagnosis and differential diagnosis.Methods Tissues from the fetus,mole and placenta were collected and pathology analysis and chromosome analysis were done.The DNA from the fetus,mole and parents' peripheral blood leukocytes was amplified with five short tandem repeat (STR) markers (D4S2460,D18S488,D21S2039,DXS1205 and DYS219) at the same time to confirm the parental source of the hydatidiform.Results (1) Casereport:A 27-year-old woman,gravida 1,para 0,was found high risk for neural tube defects at 20 weeks of gestation.At 24+5 weeks of gestation,ultrasound examination demonstrated a normal fetus,a normal placenta and a huge mass with a multicystic appearance attached to the placenta with an obvious demarcation.The fetus died at 26 weeks of gestation.Serum human chorionic gonadotropin-β(β -hCG) level decreased obviously during the first two weeks after artificial induction,but elevated at the third week,and β-hCG titers fell to normal after 2 courses of chemotherapy.Fetus autopsy showed no structure abnormality.Histopathologic examination of the hydatidiform showed swelling of chorionic villi with hyperplasia of the trophoblast and formation of central cisterns suggesting of a twin pregnancy consisting of a complete hydatidiform mole and coexisting fetus.(2) Genetic analysis:The karyotype analysis of the normal placental villi was 46,XY; the cell cultures of fetal cartilage tissue and hydatidiform were failed.STR analysis showed that the fetus was diploid from biparental source;the mole was androgenetic source.And the mole had locus both from Y and X chromosome of the father,so it was heterozygous.It was suggested that this case was derived from one single oocyte fertilized with three spermatozoas.Conclusions STR analysis could be used to confirm the diagnosis of complete hydatidiform mole and coexisting fetus and to find the pathogenetic rnechanism.
