Objective:
To establish a rapid detection method for Salmonella based on the combination of enzymatic recombinase amplification (ERA) and lateral flow chromatography (LF), so as to provide technical support for the on-site detection of Salmonella.
Methods:
Specific ERA primers and probes were designed based on the highly conserved flagella gene fimY in Salmonella. The primers were screened using capillary electrophoresis, and the probes were designed according to the amplification range of the screened primers. The amplification temperature and time were optimized to establish the amplification method, and the product was detected using LF strips. A standard strain of Salmonella was used to verify the sensitivity, 10 other gut bacteria were used to to verify the specificity and sensitivity, and the nucleic acid of the actual Salmonella strains was amplified to verify the detectability.
Results:
After screening for Salmonella-specific primers using capillary electrophoresis, the minimum detection concentration was 5 copies/μL under the amplification temperature of 37 ℃ and reaction time of 20 minutes. This method had a positive amplification result for Salmonella nucleic acid, and the amplification results of 10 other gut bacteria were all negative, with good specificity.
Conclusion
This method provides a possibility for on-site point of care testing of Salmonella infection.

Objective To investigate the value of ultrasound diagnosing for hydropneumothorax. Methods In a prospective double-blind randomized concurrent controlled trial. 213 patients doubted pneumotborax were exam-ined with CT, senography and conventional radiography. Results In 213 cases, hydropneumothorax diagnosed in 30 hemithoraces of 30 patients by CT,29 hemitboraces by ultrasound and 22 hemithoraces by X-ray. The sensitivity, nega-tive predictive value, accuracy by ultrasound and X-ray were 96.7% vs 73.3% ,99.8% vs 98.0% ,99.8 vs 98.1% respectively(P<0.05), the specificity and positive predictive value of both ultrasound and X-ray were 100%. Ultra-sound surpassed the X-ray in detecting pneumothorax ( McNemar test P<0.025 ). Conclusion If ultrasound is served to detect pneumothorax, it can make up the defects of the methods commonly used cuxrently.

Bone morphogenetic proteins (BMPs) are members of the transforming growth factor-β (TGF-β) superfamily, acting as important regulators in embryogenesis and bone and cartilage formation and repair. The complexity of BMPs action has early been unveiled by generous cell and molecular biology approaches and transgenic animal studies.Genetic engineering allows for the mass production of BMPs for clinical use of bone repair, but the desirable scaffold material is also needed to ensure controlled release and maximum biological activity of BMPs at the surgical site.Currently, BMP-2 and BMP-7 have been approved by United States Food and Drug Administration for clinical therapy in long bone fracture, spinal fusion and oral and maxillofacial surgery. The results are satisfactory, but the side effects have been also widely concerned. The functions and clinical applications of BMPs in bon repair are reviewed.

They were control group (NC group), low fluoride group (LF group), high fluoride group (HF group), NC + rapamycin (RAP) group, LF + RAP group, HF + RAP group, NC + chloroquine (CQ) group, LF + CQ group, and HF + CQ group. The NC group drank tap water (fluoride concentration was 0.5 mg/L), LF group drank fluoride water (fluoride concentration was 5.0 mg/L), HF group drank fluoride water (fluoride concentration was 50.0 mg/L); NC + RAP group, LF + RAP group and HF + RAP group were fed with corresponding drinking water, respectively, for 3 months, and then RAP (1.5 mg/kg) was intraperitoneally administered for 10 d; NC + CQ group, LF + CQ group and HF + CQ group were fed with corresponding drinking water, respectively, for 3 months, and then CQ (60 mg/kg) was intraperitoneally administered for 10 d. Bone and 24-hour urine samples of rats in each group were collected to detect the contents of bone fluoride and urine fluoride; liver histomorphological changes were observed through hematoxylineosin staining; protein and mRNA expressions of LC3B, P62 and Beclin1 in liver were detected by immunohistochemistry and real-time fluorescence quantitative PCR, respectively. Results:Compared with the NC group [(0.03 ± 0.00) mg/kg, (0.34 ± 0.08) mg/L], the contents of bone fluoride [(3.86 ± 0.08) mg/kg] and urine fluoride [(1.11 ± 0.16) mg/L] in HF group were higher ( P < 0.05). In the NC group, the lobule structure of liver tissue was clear, the hepatic cords were arranged in order, and the cell structure was normal. There were different degrees of hepatocyte edema in LF and HF groups. After intraperitoneal injection of RAP, compared with the corresponding fluoride group, the morphology of hepatocytes did not change significantly. After intraperitoneal injection of CQ, compared with the corresponding fluoride group, the liver cells showed obvious edema, and the degree of edema aggravated with the increase of fluoride concentration. Compared with the NC group, the protein expressions of LC3B and Beclin1 in HF group were higher ( P < 0.05), and the protein expression of P62 was lower ( P < 0.05). After intraperitoneal injection of RAP, the protein expressions of LC3B and P62 in LF + RAP group was lower than that in LF group ( P < 0.05); Compared with HF group, the protein expressions of LC3B and Beclin1 in HF + RAP group were lower ( P < 0.05). After intraperitoneal injection of CQ, protein expression of P62 in LF + CQ group was higher than that in LF group ( P < 0.05); Compared with HF group, protein expression of P62 in HF + CQ group was higher ( P < 0.05). Conclusions:Early (3 month) fluoride intake could promote autophagy and induce edema of hepatocytes in rats, and RAP had similar effects. CQ may induce liver injury by inhibiting autophagy of hepatocytes.

2 mmx3 mm experimental windows were prepared in adamant slippery surfaces of 7 fresh uprooted permanent teeth. The teeth surfaces in the windows area were demineralized to create artificial caries mould of early stage by aciding the experimental teeth surfaces of 0, 12, 24, 48, 72, 96, 120 hours with demineralized liquid (pH 4.5) in vitro. The demineralized changes on the experimental teeth surfaces were detected by dental Optical Coherence Tomography (OCT) system, which were newly developed by our research group recently, and the detecting results were compared with clinical digital photomicrography and scanning electron microscopy on the same tooth sample, to checkout the efficacy and feasibility of dental OCT for early quantification detecting of artificial enamel demineralization in vitro. The dental OCT system can safelydetect early enamel demineralization of micron level and noninvasively obtain fine resolution quantification information both in surfaces view and sectional view; OCT could accurately detect surface demineralization changes on the experimental windows of artificial dental caries as early as after 12 hours aciding treatment, earlier than the visual inspection and clinical digital photomicrography. OCT could obtain both superficial view and sectional view of quantificational demineralization in early enamel caries homeochronously, and had high correlation to the results of ultramicromorphological changes detected by scanning electron microscopy. Dental OCT system developed by our group could accurately detect early artificial dental caries atraumaticly with high sensitivity and safety. Moreover, it can obtain quantification data in micron level without damaging the experimental teeth samples.
