Modern Clinical Nursing 2015;(4):29-31
Objective To analyze the clinical effects of dexmedetomidine on postoperative agitation in children patients . Method Forty-four children undergoing surgeries in our hospital between March 2011 and March 2014 were treated with intravenous injection of 10 mL dexmedetomidine with a dose of 0.5μg/kg in 15 minutes after general anesthesia. Result The occurrence rate of postoperative restlessness was only 6.82%(3/44). Conclusion The use of dexmedetomidine combined with specific clinical nursing intervention for children undergoing surgery can effectively reduce the occurrence of postoperative restlessness , which ensures the clinical therapeutic effect and life safety.

Objective:To study the influence of Sinapine on the formation of microvessels in chick embryo chorioallantoic membrane(CAM).Methods: Sinapine was purified by the procedure of pressurized-solvent extraction from Semen Sinapis Albae. The inhibition effect of Sinapine on the chorioallantoic vessel was detected by the chick embryo chorioallantoic membrane model. Results: Compared with physiological saline group, large areas of blood vessels in Sinapine groups(5.0g/L and 10.0g/L) had heavy inhibition of angiogenesis on CAM,accompanied by the decreased density of vessels(P0.05).Conclusions: Sinapine can prevent the formation of microvessels of CAM and the effects depend on the concentration.

ObjectiveTo investigate the effects of leukocyte-depleted allogeneic blood transfusion on perioperafive cellular immunity in children.MethodsThree hundred and fifty-nine ASA Ⅰ or Ⅱ children aged 3 month-14 yr,weighing 5-74 kg requiring allogeneic blood transfusion during operation were randomly divided into 2 groups:control group (group C,n =163) and leukocyte depletion group (group D,n =196).In group D allogeneic blood was filtered with a leukocyte filter before being transfused during operation.Blood samples were collected from peripheral vein before blood transfusion, and at 2 and 6 d after blood transfusion for determination of levels of CD3+,CD4+,CD8+,and CD56+ by flow cytometry.CD4+/CD8+ ratio was calculated. The volume of allogeneic blood transfused during operation, the duration of operation, postoperative drainage, antibiotics administration and hospital stay and incidence of postoperative infection were recorded.ResultsThe levels of CD3+,CD4+,CD56+ and CD4+/CD8+ ratio were significantly increased at 6 d after blood transfusion while the duration of postoperative drainage, postoperative antibiotics administration and hospital stay and incidence of postoperative infection were significantly decreased in group D compared with group C.ConclusionLeukocyte-depleted allogeneic blood transfusion is helpful in improving the postoperative cellular immunity in children.

To optimize the water extraction process for glycyrrhiza. Methods: HPLC was used to determine the con-tents of glycyrrhizic acid and liquiritin. The comprehensive index included the contents of glycyrrhizic acid and liquiritin and the yield of dry extract. The water amount and the extraction time were selected as the independent variables, and the comprehensive index was set as the dependent variable. Design-expert 8. 06 software was used to fit multivariate linear or quadratic multinomial models for the experimental values. Response surface methodology was used to optimize the water extraction process. The prediction was carried out through comparing the observed and predicted values. Results:The regression coefficient of binomial fitting complex model was as high as 0. 979 7. The optimum conditions of extraction process were as follows:12-fold amount of water, extracting 3 times with 90 min for each time. The deviation between the observed and predicted values was -1. 72%. Conclusion: Central composite design-response surface methodology is convenient and highly predictive in optimizing the water extraction process for glycyrrhiza, which can be applied in the further membrane separation and purification.

Objective To observe changes of the pancreas after limb ischemia reperfusion, and the protective effect of taurine (Tau) postconditioning. Methods Twenty-four male SD rats were randomly divided into control group (Control), limb ischemia reperfusion (LIR) group, and taurine postconditioning (Tau) group (n=8 for each group). Plasma contents of xanthine oxidase (XOD), malondialdehyde (MDA), reactive oxygen species (ROS), superoxide dismutase (SOD), pancreatic amylase (AMS), pancreatic lipase (LPS) and trypsin were detected in three groups. Morphological changes of the pancreas were observed using optical microscopy. The expressions of anti-C/enhancer-binding protein homologous protein (CHOP) in pancreas were analyzed by immunohistochemistry assay. TUNEL staining was performed to estimate the apoptosis of pancre-atic cells. Results Compared with the control group and Tau group, plasma contents of MDA, XOD, ROS, AMS, LPS, and trypsin were significantly increased in LIR group, but the level of SOD was significantly lower in LIR group ( P<0.05). HE staining showed that acinar structure was disrupted , pancreatic lobule gap widened, stromal vascular dilatation and conges-tion were observed in LIR group. The perivascular infiltration of inflammatory cells appeared, islet cells were damaged with irregular islet. The immunohistochemical analysis showed the increased expression of CHOP in pancreas in LIR group than that of Tau group. And the pancreatic apoptosis was enhanced in LIR group detected by TUNEL staining. Conclusion Tau-rine postconditioning can ameliorate pancreatic injury following limb ischemia reperfusion, which may be related to the inhi-bition of oxidative stress, down-regulation of CHOP expression, thereby reducing endoplasmic reticulum stress-induced apoptosis.

Objective To discover the influence of thiomersalate exposure on the behavior and humoral immunity of premature rats. Methods According to common thiomersalate dose of vaccine used in human, the surface conversation method was used to change the dose to 1,2,3,4 times for rats(32.8,65.6,98.4 and 131.2 μg/kg),and the thiomersalate was separately injected to the gluteus maximus of premature rats respectively, which were delivered on the 20th day of gestation by cesarean for the injection on day 1 (premature).Simultaneously, saline was injected to the control group. On day 44 to 48 after injection, the Morris water maze test was used to evaluate the spatial learning and memory abilities of rats and on the post-injection day 49,immunoglobulin content in rat blood and cerebrospinal fluid was measured. Results The spatial learning abilities of rats in 131.2 μg/kg group and the memory abilities in 65.6 μg/kg,98.4 μg/kg and 131.2 μg/kg group were significantly weaker than those in 9 g/L saline group(all P <0.001).The immunoglobulin content in blood and cerebrospinal fluid changed in different groups exposed to thiomersalate. The levels of IgG,IgA,IgE,IgM in 98.4 μg/kg group and 131.2 μg/kg group rose significantly higher than those in 9 g/L saline group(all P < 0.01).Analysis of rectilinear correlation between the immunoglobulin and learning abilities of rats revealed that IgG,IgA,IgE levels in the blood were correlated with the spatial learning abilities (r =0.36,0.47,0.50,all P < 0.05) and the levels of IgG,IgA,IgE,IgM in blood were rectilinear correlated with the memory abilities of rats (r =-0.39,-0.43,-0.49,-0.38,all P < 0.05).In rats'cerebrospinal fluid,IgG,IgA,IgE,IgM levels were rectilinear correlative with both the spatial learning and memory abilities (r =0.48,0.59,0.54,0.41,all P<0.05;r=-0.39,-0.61,-0.57,-0.44,all P<0.05).Conclusion With the dose ofthiomersalate increased, the learning and memory and humoral immunity are sustained damaged for premature rats.
