1.Research progress on tumor targeting of splice variants

Splice variants are transcripts that exhibit various functions or activities and are obtained from alternative splicing of precursor mRNA. An increasing number of tumor-specific splice variants are being discovered and their roles in cancer development and progression are being investigated. In this study, the expression patterns and activities of tumor-specific splice variants were charac-terized. The results suggest that a variant-specific gene therapy that targets cancer cells would be more specific and would have minimal effect on healthy tissues. Using antisense oligonucleotides, mRNA that contributed to cancer cell survival and altered splicing patterns was downregulated in a clinical trial. The results provide a new appreciation for the gene therapy of cancer. Identification of novel tu-mor-specific splice variants and constant improvement of therapeutic strategies by modifying splicing patterns can lead to the develop-ment of more effective and safer cancer therapies.
2.Summary and analysis on external quality assessment results of clinical immunology during 2009 - 2013

Objective To perform the summary and analysis on the external quality assessment(EQA) results of the clinical im‐munology during 2009 - 2013 to find out the possible problems existing in the laboratory work of the clinical immunology for con‐tinuously improve the quality of clinical immunological detections and ensuring the accuracy of clinical immunological test results . Methods The results of the external quality assessment in the clinical Immunology participating in the National Center for Clinical and Laboratory and the Guangxi Center for Clinical Laboratory during 2009 - 2013 were performed the statistical analysis and sum‐mary .Results 600 samples were detected during these 5 years .The performance was worst in 2012 ,the annual coincidence rate of the results was 98% ;and which in other 4 years was 100% ;each 1 sample of result in the two items of anti‐HCV and anti‐TP was unconformity during these 5 years ,the total coincidence rate was 98 .6% ;the total coincidence rates of HBsAg ,HBsAb ,HBeAg , HBeAb ,HBcAb and anti‐HIV were 100% .Conclusion By participating in the external quality assessment of the clinical immunolo‐gy and retrospectively analyzing the results ,the possible problems would be found and some effective improvement measures could be formulated in time for improving the quality of examinations and ensuring to provide reliable laboratory data for clinic .
3.The changes of carbonic anhydrase activity and its mRNA expression in rats cochlea after noise exposure.

OBJECTIVE:
To investigate the changes of carbonic anhydrase (CA) activity and its mRNA expression in rats cochlea after noise exposure.
METHOD:
Twenty-four healthy Sprague Dawley rats were randomly allocated into 4 groups including 1 control group without contact of noise and 3 experimental groups exposed to a narrowband white noise of 4 kHz, 110 dB SPL 4h/d for 1 day, 1 week and 3 weeks, respectively. ABR thresholds were tested for each group at the time points described above. An immunohistochemical method was used to detect the expression of CA in the cochlea tissue. Differences in CAII mRNA expressions with and without noise exposure were examined using RT-PCR.
RESULT:
The thresholds of ABR in noise exposed cochlea were increased compared with the control cochlea (P < 0.01). CA activity and the CAII mRNA expression in noise exposed cochlea were decreased compared with the control cochlea (P < 0.01). The increase of ABR thresholds and decreases of CA activity and the CAII mRNA expression showed a time dependent pattern as the extension of duration exposed to noise.
CONCLUSION
Noise exposure could downregulate the CA activity and CAII mRNA expression in the cochlea. CA was proably involved in the pathogenesis of noise-induced hearing loss.

4.Photodynamic therapy inducing apoptosis of human lung adenocarcinoma cell line A549

Objective:To observe the effect of photodynamic therapy(PDT)on apoptosis of human lung adenocarcinoma cell line A549.Methods:Human lung adenocarcinoma cell line A549was used and the optimal parameters of PDT were se-lected by MTT colorometric assay.The experiment was divided into2groups:control group and PDT group.Twenty-four hours after PDT(HPD concentration10mg/L,laser dosage10J/cm 2 ),cell cycle and apoptosis rate were estimated by flow cytometry;agarose gel electrophoresis and TUNEL were used to observe the DNA fragments and apoptosis index(AI).Re-sults:The killing effect was saturated when HPD was10mg/L and laser was10J/cm 2 .Under the same laser(10J/cm 2 ),A549cells were exposed to6different duty time fabrications and the outcomes showed that their D 492 had no statistical differ-ence.Agarose gel electrophoresis of DNA showed DNA ladder,a biochemical hallmark of apoptosis in the PDT group.Flow cytometry showed that PDT group had G 0 /G 1 stage arrest.The apoptosis rate was(18.443?7.122)%in PDT group vs (0.301?0.361)% in control group(P
5.Analysis of Polycystic Ovary Syndrome Prescriptions in Our Hospital

OBJECTIVE:To provide reference for rational use of polycystic ovary syndrome(PCOS)drugs. METHODS:The utilization of PCOS drugs in our hospital during Jan.-Dec. 2013 were analyzed in respects of consumption sum,DDDs,sorting and so on. The rationality of prescription was analyzed according to guidelines and drug package inserts and so on. RESULTS:There were 1 551 prescriptions,involving 76 kinds of drugs,and their total consumption sum was 181 072.90 yuan. Main types of drugs were hormone related drugs (accounting for 81.37% of prescription frequency,73.51% of consumption sum) and Chinese patent medicine (accounting for 15.41% of prescription frequency,19.43% of consumption sum). Top 5 hormone drugs in the list of DDDs were Ethinylestradiol and Cyproterone acetate tablets,Metformin hydrochloride enteric-coated tablets,Progesterone soft cap-sule,Dydrogesterone tablets and Progesterone soft capsules. Top 5 Chinese patent medicines in the list of DDDs were Kuntai cap-sule,Yougui capsule,Huanshao capsule,Jiawei xiaoyao capsule and Nvjin capsule. Single drug accounted for 72.08% in prescrip-tion. The proportion of irrational drug use was 1.69%. CONCLUSIONS:The application of drug in patients with PCOS is reason-able generally. Some prescriptions are unreasonable,mainly manifesting as incomplete clinical diagnosis,irrational usage and dos-age. Pharmacists should strengthen prescription verification and intervention.
6.Application of fiberoptic bronchoscopy in neonates with weaning failure

Objective To evaluate the safety and efficacy of fiberoptic bronchoscopy in the diagno-sis of 41 neonates with weaning failure.Methods A retrospective study on the results of fiberoptic broncho-scopy was performed in 41 neonates with weaning failure.Results A total of 41 neonates of throat abnor-malities were examined by fiberoptic bronchoscopy,including 38 cases with laryngomalacia,8 cases with seri-ous hyperemia and swelling of the vocal cords,2 cases with scar stenosis of subglottic,and one case in each of congenital laryngeal web,glottic insufficiency and congenital epiglottis cyst;trachea-bronchial abnormali-ties,including 31 cases with tracheomalacia,2 cases with tracheal stenosis,4 cases with partial bronchial ste-nosis,1 cases with bridging bronchus and 1 cases with tracheoesophageal fistula.Thirty-eight neonates under-went fiberoptic bronchoscopy to obtain bronchoalveolar lavage fluid and culture,bacterial culture positive in 15 cases(39.4%),of which 5 cases of Klebsiella pneumoniae,3 cases in each of Pseudomonas aeruginosa and Acinetobacter baumannii,2 cases of Staphylococcus aureus,1 case in each of Escherichia coli and Strep-tococcus mitis;fungal culture positive for 6 cases(15.7%),of which 3 cases of Aspergillus fumigatus,1 case in each of Candida glabrata,Candida albicans and Candida parapsilosis.The most common complications of fiberoptic bronchoscopy were hypoxia and heart rate transient decline(26 cases,63.4%),3 cases fever af-ter operation.Conclusion Fiberoptic bronchoscopy can not only find congenital tracheal deformity and tra-cheal mucosa lesions,but also complete the etiology from deep discharge.Fiberbronchoscopy is safe and relia-ble given adequate preparation and if it is performed by skilled personnel.
7.An analysis of cognitive status for elderly people with subjective memory complaint in seven community of Shapingba district in Chongqing city

Objective To explore the cognitive status of community-dwelling older adults with subjective memory complaints (SMC).Methods A total of 262 older adults with SMC were included based on convenience sampling.Face-to-face interview was conducted for demographic information collection and cognitive assessment.Statistical analyses Were undertaken using SPSS version 19.0.Results The participants with SMC had a low level of general cognitive function,with a mean score of 22.18±4.21 on the MoCA; the two cognitive domains of abstraction' and deIayed memory had the lowest scores,with an average score of 0.85±0.82 and 2.91± 1.50 respectively,and the average scoring rate lower than 60.00%.About 54.20% of the older adults with SMC presented with cognitive impairment.The scores ofabstraction were low in the participants with and without cognitive impairment.In addition,the scores of visuospatial/executive function, language and delayed memory were low in the participants with cognitive impairment.The cognitive impairment of persons with SMC may be associated with age and marital status.The people over the age of 70 years and widowed ones are more likely to suffer from cognitive dysfunction.Conclusion The older adults with SMC have a low level of general cognitive function,and cognitive imPairment detection rate is higher.Advanced age and widowhood are important factors affecting their cognitive function.
8.Unilateral nasal cavity and paranasal sinus neoplasm in adolescent.

The clinical data of two adolescents with unilateral nasal cavity neoplasm were analyzed. Both were male, one was 14-year-old and the other was 17. Both were complained of unilateral nasal obstruction and nasal discharge; The course of disease of the former was one month ,and that of the latter was three month; both were found neoformation in unilateral nasal cavity in medical examination. Occupying lesion in unilateral nasal cavity and sinus were demonstrated in CT scan of two cases. Endoscopy surgery were operated in both cases, incisal opening in labiogingival groove was added in the latter. Pathology of the former was capillary hemangioma and the latter was rhabdomyosarcoma. No findings indicating recurrence has been observed in two cases under 6 months follow-up. Recognition at nasal cavity and paranasal sinus neoplasm in adolescent is essential so that proper management can be initiated.

9.Influence factors of intravenous urokinase therapy for acute cerebral infarction

Objective To investigate the baseline factors that influenced prognosis after intravenous thrombolytic therapy. Methods All 82 patients received intravenous urokinase (UK) for acute ischemic stroke. The modified Rankin scale score at 90 days was dichotomized into good outcome (mRS 0-1) and poor outcome (mRS 2-6) as the primary outcome measure. Multivariable logistic regression method was used to analyze independent predictors of good outcome. Results The mRS was good in 36.6% and poor in 63.4%. Univariate analysis indicated that age, time from stroke onset to treatment, atrial fibrillation, pretreatment ESS score, baseline serum glucose, early ischemic changes on baseline CT scans and dose of UK were shown correlated significantly with mRS. However, the multivariate logistic regression did select the ESS score, time to treatment, dose of UK and serum glucose as the independent predictors of good outcome. Conclusion Pretreatment ESS score, time to treatment, dose of UK and baseline serum glucose should be independently predictive of outcome in patients with acute cerebral infarction treated by intravenous UK.
10.Analysis of onset age and cutaneous lesions in 54 patients with tuberous sclerosis

Objective To analyze the features of cutaneous lesions and age of onset of tuberous sclerosis(TS).Methods A total of 54 TS patients were recruited;their clinical data were analysed.Results The occurance of facial angiofibromas,periungual fibromas,hypomelanotic macules and shark macules was 72.5%,26%,70%and 48%,respectively,and the median age of onset of these lesions was 7,14,0.2 and 6 years,respectively,with a peak at 1-9,10-19,<1 and 1-9 years,respectively.Conclusions Cutaneous lesions are commonly seen in TS,and most of them develop early in life.
