1.Study on the Anti-inflammatory Effect of Biota orientalis

AIM The purpose is to investigate the Anti-inflammatory mechanism of Biota orientalis. METHODS Using rat leukocytes and rabbit platelets as experimental material the biosynthesis of leukotriene, 12-hydroxyheptadecatrienoic acid(12-HHT) and 12-hydroxyeicosatetraenoic acid(12-HETE) in the cells was determined with HPLC. RESULTS Biota orientalis was shown to inhibit the biosynthesis of leukotriene B4(LTB4) and 5-hydroxyeicosatetraenoic acid(5-HETE) with IC50 of 0.40 and 0.41 mg/ml crude herb, respectively. It was also displayed to inhibit biosynthesis of 12-HHT in the platelets. CONCLUSION Biota orientalis leaves contain anti-inflammatory components and the anti-inflammatory mechanism was related with inhibiting metabolism of arachidonic acid.

Aim To study the effect of resveratrol on the expression of FAK and the level of phosphorylated FAK, and to investigate its possible mechanism of resveratrol on the metastasis-associated ability of human highly metastatic ovarian carcinoma HO-8910PM cells line. Methods MTT assay was used to examine cytotoxicity of resveratrol on HO-8910PM cells after 24 h treatment; The expression of FAK protein and the level of phosphorylated FAK were assessed by Western blotting analysis. Results MTT assay showed that resveratrol had weak cytotoxicity on HO-8910PM cells after 24 h treatment, its IC50 value was 163.40?2.48 ?mol?L-1. The expression of FAK and the level of phosphorylated FAK decreased obviously in HO-8910PM cells treated with 25~100 ?mol?L-1 resveratrol for 24 h. Conclusion The mechanism of resveratrol inhibiting the metastasis-associated ability of human highly metastatic ovarian carcinoma HO-8910PM cells line involves the expression of FAK and the level of phosphorylated FAK.
4.The anti-inflammatory effect of total flavonoids isolated from Platychadus orientalis and its mechanism

AIM To investigate the anti-inflammatory eff ec t of total flavonoids isolated from Platychadus orientalis and its mechanism . METHODS The inflammatory models in mice and swelling of rats hind paws were induced by dimethylbenzene and carrageenan respectivel y. The biosynthesis of leukotriene and ?-glucuronidase release in the cells we re measured by HPLC and fluorescence spectrophotometer respectively. RES ULTS Total flavonoids isolated from Platychadus orientalis remarkably inhibited the ear edema induced by dimethylbenzene in mice at the dose of 12.5 ~50.0 mg?kg -1 and the swelling of rats hind paws induced by carrageenan at the dose of 25.0~50.0 mg?kg -1 . Total flavonoids at final concentrati on of 5.0~125.0 mg?L -1 inhibited 5-HETE and LTB 4 synthesis by 30.5%~ 87.3% and 27.8%~84.3% respectively. In comparision with controls, Total flavono ids at final concentration of 5.0~45.0 mg?L -1 inhibited ?-glucuronidas e release by 13.4%~32.2%. CONCLUSION Total flavonoids isolated f rom Platychadus orientalis has an anti-inflammatory effect which may be rel ated to inhibiting 5-HETE and LTB 4 production and ?-glucuronidase release.
5.Effect of Cx43 on acquired gefitinib-resistance mechanisms in human NSCLC HCC827 cells

Aim To explore the effect of connexin 43 ( Cx 4 3 ) on acquired gefitinib-resistance in human non small cell lung cancer ( NSCLC ) . Methods HCC827 GR, a gefitinib-resistant ( GR) NSCLC cell lines from their parental cells was established by gradually in-creasing the concentration of gefitinib. Gefitinib effica-cy in HCC827 and HCC827 GR cells was detected by MTT assay. Expression of Cx43 mRNA in HCC827 and HCC827 GR cells was determined by RT-PCR. The protein expressions of Cx43 and phospho-Akt ( p-Akt) in these cells were detected by Western blot. The func-tional gap junction intercellular communication ( GJIC ) was measured by parachute assay. The cellular locali-zation of Cx43 protein was evaluated by immunofluores-cence staining. Results MTT assay showed less ge-fitinib cytotoxicity in HCC827 GR cells than that in their parental cells with IC50 of (10. 84 ± 0. 021) μmol ·L-1 versus (0. 07 ± 0. 019) μmol·L-1 , respective-ly. Moreover, both mRNA and protein expressions of Cx43 in HCC827 GR cells were significantly lower than those in HCC827 cells ( P<0. 05 ) . However, the p-Akt protein in HCC827 GR cells was obviously higher than that in HCC827 cells ( P<0. 05 ) . Furthermore, treatment with LY294002 caused a significant reduced p-Akt expression, but a significant increased Cx43 ex-pression in HCC827 GR cells. Moreover, no detecta-ble GJIC was found in HCC827 and their GR cells with or without RA ( a well-defined GJIC enhancer ) treat-ment. Immunofluorescence staining clearly showed that Cx43 protein accumulated in the cytoplasm of HCC827 and their GR cells. Conclusion The down-regulation of Cx43 expression in cytoplasm of HCC827 GR cells may contribute to the acquired gefitinib resistance in NSCLC cells by GJIC-independent activation of PI3 K/Akt signaling pathway.
6.Effect of luteolin on metastasis of ovarian carcinoma cell line

AIM: To explore the effect of luteolin on the metastasis of ovarian carcinoma HO-8910PM cells in vitro, and to elucidate its mechanisms. METHODS: The in vitro invasion and mobility of HO-8910PM were measured by Boyden chamber assay after exposure to luteolin for 6 h. Gelatinase activity was tested by gelatin zymography assay. The expression of tissue inhibitor of metalloproteinase-1 (TIMP-1), nm23 mRNA and that of ERK2 protein were tested by RT-PCR and Western blotting, respectively. RESULTS: Luteolin significantly inhibited in vitro invasiveness and mobility in HO-8910PM cells in a dose dependent manner as measured by Boyden chamber assay. Luteolin inhibited the MMP-9 secretion from HO-8910PM cells. Luteolin did not affect the mRNA expression of TIMP-1 and nm23. Luteolin also decreased ERK2 expression. CONCLUSION: Luteolin dose-dependently inhibited the metastasis of HO-8910PM cells in vitro. Inhibition of MMP-9 secretion and ERK2 expression may be involved in the anti-metastasic process of luteolin.
7.The impact of telmisartan on angiotensin converting enzyme 2 mRNA expression in monocyte-derived macrophages of diabetic hypertensive patients

Objective To investigate the effects of telmisartan on the expression of angiotensin converting enzyme2 (ACE2) mRNA in monocyte-derived macrophages of hypertensive patients companied with diabetes.Methods 62 essential hypertensive patients companied with diabetes were randomly divided into two groups:regular treatment group,and telmisartan group.Then the content of ACE and ACE2 in serum was detected by ELISA,and the expression of ACE mRNA and ACE2 mRNA in monocyte-derived macrophages of patients was detected by RT-PCR before and after having been treated.Results (1) After having been treated for 4 weeks and 12 weeks,the blood pressure of the patients in two groups were decreased significantly,Comparing with regular group,telmisartan group seemed to have more obvious therapeutic effect (P < 0.05) ; (2) After having been treated for 12 weeks,glycosylated hemoglobin diseased in both group,but there was no significant difference between the two group (P > 0.05) ; (3) In telmisartan group,the content of ACE2 in serum was increased after having been treated for 12 weeks than that in regular treatment group,[(23.9 ± 8.2) U/L vs (16.3 ± 8.9) U/L,P < 0.05] ; and the expression of ACE2 mRNA in monocyte-derived macrophages in telmisartan group was obviously increased after 12 weeks comparing with regular treatment group (0.73 ±0.06 vs 0.51 ±0.04,P <0.01).Conclusion The role of telmisartan in decreasing blood pressure and it's advantage to the metabolism of glucose are partly related with the up-regulation of ACE2 mRNA.
8.Effects of fluoride on bone mass of tibia and lumbar in BALB/c mice

Objective To investigate the effects of fluoride on trabecular bone of the tibia and lumbar in BALB/c mice.Methods Totally 64 four-week-old BALB/c mice were randomly divided into 4 groups by weight,16 per group:control group (treated with distilled water) and 3 sodium fluoride (NaF) exposure groups (treated with NaF at 25,50 and 100 mg/L F-),respectively.At 12 weeks,mice were killed and blood,two hind limbs and lumbar were collected.Bone fluoride content and incidence rates of dental fluorosis were determined.Serum content of alkaline phosphatase (AKP) and acid phosphatase (ACP) were detected by micro enzyme labeled method.The ultrastructure of osteoblasts and osteoclasts in lumbar were observed via transmission electron microscope.The pathological changes of the trabecular bone of the tibia and the lumbar were observed under optical microscope,the percentage of trabecular area (%Tb.Ar) was measured with Image-Pro Plus (IPP) software.Results Bone fluoride contents of low,middle and high fluoride groups [(1 828.62 ± 102.93),(3 308.27 ± 185.63),(4 933.36 ± 301.16) mg/kg] were higher than that of the control group [(775.23 ± 92.56) mg/kg,all P < 0.05].The incidences of dental fluorosis in the 4 groups were 0(0/16),47%(7/15),93%(14/15) and 100%(16/16),respectively;the difference was statistically significant (x2 =27.23,P < 0.05).In middle and high fluoride groups,serum AKP [(18.30 ± 1.99),(24.50 ± 3.14) king unit/100 ml] and ACP [(11.97 ± 1.73),(11.31 ± 1.46) king unit/100 ml] were significantly higher than those of control [(14.63 ± 1.21),(9.07 ± 1.47) king unit/100 ml,respectively,all P < 0.05].Under the electron microscope,osteoblast had developed organelles in each fluoride group,rough endoplasmic reticulum,Golgi body,and mitochondria were abundant,and nucleolus was obvious in the osteoblast.Osteoclast was rich in mitochondria,ruffled border clear and distributed phagocytic vacuoles in low fluoride group and middle fluoride group.Compared with the control group (17.03 ± 3.73),HE staining of tibia %Tb.Ar in high fluoride group (28.79 ± 8.26) was significantly increased (P < 0.05).The lumbar spine %Tb.Ar in low,middle and high fluoride groups (15.87 ± 2.59,18.28 ± 0.89,21.99 ± 1.81) were higher than that of the control group (12.06 ± 1.76,all P < 0.05].Conclusions BALB/c mice could be used as a model of skeletal fluorosis.Osteoblast and osteoclast are activated in BALB/c mice with skeletal fluorosis.Bone formation is more obvious than bone resorption and bone mass is increased.What is more,bone mass has increased more significantly in the lumbar spine of mice.

Objective Though analyzing the epidemiological trend of human Brucellosis in Qinghai Province in 2012, to provide a advice for Brucellosis prevention and control. Methods Brucellosis investigation was carried out on key populations aged 7-60 years who were keeping in touch with livestock in Ping’an County, Haiyan County, Tianjun County, Dari County, Jiuzhi County, Henan County, and three or four towns were selected as investigation sites in each county; then serological test was done with the methods of the rose bengal plate agglutination test(RBPT), standard tube agglutination test(SAT) and Coombs; diagnosis was based on “Diagnostic Criteria for Brucellosis”( WS 269-2007 ) . Results A total of 4 253 people in the 6 counties were investigated;the positive rate of RBPT was 2.92%(124/4 253); the positive rate of SAT was 0.85%(36/4 253), while the Coombs was 0.05%(2/4 253), totally infected people was 124, the infection rate was 2.92%(124/4 253); the number of patients was 71, the prevalence rate was 1.67%(71/4 253); and new cases were 68. Conclusions Prevalence of human Brucellosis in Qinghai Province is active in some local areas. We should strengthen health education of occupational population, improve people’s ability of prevention and control, and control prevalence of Human epidemic situation.
10. A survey on awareness of digestive system injury caused by corona virus disease 2019 in gastroenterologists

Objective:
To investigate awareness of digestive system injury caused by corona virus disease 2019 (COVID-19) in gastroenterologists.
Methods:
From February 21 to 23 in 2020, the electronic questionnaire was sent out to explore the condition of the basic knowledge of COVID-19 and knowledge of digestive system injury caused by COVID-19 grasped by gastroenterologists. Chi-square test was used for statistical analysis.
Results:
A total of 2 216 gastroenterologists from 31 provinces, autonomous regions and municipalities nationwide completed the survey. 99.7% (2 209/2 216) of gastroenterologists stated that they had read the COVID-19 diagnosis and treatment guidelines. The percentage of physicians who well knew the diagnostic criteria of suspected and confirmed cases of COVID-19 was 34.9% (774/2 216) and 39.4% (874/2 216), respectively. The percentage of physician who gave the right answer of COVID-19 detectable methods and lung imaging was 68.4% (1 516/2 216) and 71.6% (1 586/2 216), respectively. The percentage of correct answer of digestive system injury caused by COVID-19 in residents, attending physicians, associate chief physicians and chief physicians was 30.9% (134/433), 33.9% (234/691), 32.4% (213/657) and 34.9% (152/435), respectively, however there were no statistically significant differences among physicians of different level (χ2=6.60,
