1.HISTOCHEMICAL STUDIES ON BASIC SUBSTANCES AND ENZYMES CONCERNED WITH ENERGY METABOLISM IN THE ENTERIC PLEXUS OF GUINEA PIG

The present study deals with the histochemistry of intramural plexus in stomach, duodenum, ileum, caecum, proximal colon, distal colon and rectum of normal adult guinea pig, including the reaction of Mg~(++)-ATPase, Ca~(++)-ATPase, CCO, SDH, G6PDH, carbohydrate, protein, nucleic acid and lipid in the enteric neurons. The reactions of all the enzymes mentioned above are observed under the light microscope and estimated semiquantitatively, among which, Mg~(++)-ATPase and Ca~(++)-ATPase are measured quantitatively with microphotometer (Leitz MPV II). Both results of light microscope, and microphotometer were analysed statistically. It was found that the above mentioned enzyme activities in the enteric neurons, suggesting that the neurons carry out active metabolism with these concerned enzymes. There were significant differences in some enzyme activities between myenteric plexus and submucous plexus, the former showed higher activity in SDH, while the latter in LDH, G6PDH, and two kinds of ATPase. It is suggested that there are some differences in. the major metabolic manner of carbohydrate, energy metabolism and functional activity between, two plexus. Besides, there were marked difference in enzyme activities among myenteric plexus of different part of digestive tract, the highest activity was showed in duodenum and proximal colon, the lowest was in caecum. These results indicate that enteric neurons differ greatly in functional state, but whether or not the variation of enzyme activities is related with the different types of neurons is unknown.
2.Application prospects of human-originated hepatocellular carcinoma antibodies

Non-surgical therapy is important to liver cancer treatment. Actinotheraphy and chemotherapy, because of their less specific for cancer tissue and serious side effect, badly affect the patients' life qualities, sometimes even endanger the patients lives. Hepatocellular carcinoma specific antibodies have been the focus of liver cancer biotherapy. After 20 years of study since 1975, it has been demonstrated that antibodies derived from mouse could not efficiently stimulate the functions of human reactor, but could induce human anti-mouse antibody responses which could bring about serious side effects. The application of DNA recombination technique provides a good method for resolving the immune selection problem. The techniques of humanoriginated antibodies and phage displaying will lead to development of innovative strategies to manage liver cancer. In our country, the antibody products aimed at cancer all originated from mouse while the study of human antibodies is still at its primary stage. Non-human antibodies has been forbidden in clinical trails by FDA in America since 2001. It can be speculated that human antibody products will domminate the future market rapidly.
3.Transendosepoic Miniature Ultrasonic Probe in the Diagnosis of Gastric Submucosal Tumor

In order to evaluate the use of transendoscopic miniature ultrasonic probe (TEMP) in the diagnosis of gastric submucosal tumor, 18 patients were investigated. The results obtained by TEMP were compared with those of gastroscopy, and endoscopical ultrasonography (EVS). The result showed that among those 18 patients, gastric submucosal leiomyomas, were common and accounted for 88.9%. Most of them were intramural. The diagnostic accuracy of TEMP for the size of tumor and gastric submucosal tumor was 88.9% and 100% respectively. Which were much higher than those of gastroscopy (33.3% and 61.1% respectively). The sonogrophic characteristics of gastric submucosal tumors in TEMP imaging were related to the nature of the tumors, hut the common manifestations were that all the tumors were hypoechoic under the hyperechoic mucosal layer with clear bell shaped origin, and usually appeared as round or nearly round intragastric eminence. It is concluded that the selective use of TEMP has great value in the diagnosis of gastric submucosal turmor, especially for those lesions which were small and difficult to be detected by EUS.
4.Reliability and validity of 3D CT imaging in preoperative evaluation of tibial plateau fractures

Objective To compare the diagnostic values of 3D CT and 2D CT imaging for tibial plateau fractures. Methods A retrospective study was performed to evaluate 21 tibial plateau fractures. Four experienced orthopaedists were asked To make assessmem of fracture classifications and characteristics using 3D CT and 2D CT images.Agreement test Was performed to evaluate interobserver and intraobserver reliability for fracture classification and criteria validity for fracture characteristics(intraoperative findings were taken as the golden standard).Furthermore,MeNemar test was used to compare the agreement of 3D CT results and 2D CT results respectively with the golden standard for fracture characteristics.Results When 3D CT images were used.interobserver reliability and intraobserver reliability for fracture elassiftcation increased to"almost perfect agreement".The agreement of golden standard with 3D CT results for fracture characteristics Was significantly higher than that with 2D CT results. Condmiou Since 3D CT imaging can improve the reliability and accuracy of preoperative assessment oftibial plateau fractures,it is helpful and valuable for tibial plateau fractures.
5.Research on the application of PBL and Scenario simulated teaching in pathological experiment

The study was o investigate the effectiveness of problem-based learning and scenario simulated teaching in Pathological experiment course.Clinical common,multiple cardiovascular diseases related experiments were selected for reform.The case was in advance handed out to the second year students of clinical medicine,stomatology,imaging medicine and otherspecialty,requiring them to complete the data access,group discussion,PPT production,diagnoses and treatment scenarios simulation video shooting tasks.Every group showed their PPT and video in class.They were asked between groups.Summative evaluation was given before class.Curriculum effectiveness questionnaire was issued and recycled,and according to students' performance teacher gave their scores and reward.The study showed that the comprehensive application of the above teaching methods and means could effectively improve students' learning enthusiasm and initiative,cultivate their medical humanistic spirit,and help to improve the teaching quality of pathology experiment.
6.CRISPR-mediated downregulation of PD-1 expression on T cells

Objective To investigate the feasibility of using clustered regularly interspaced short palindromic repeats ( CRISPR)-mediated genome editing to downregulate the expression of programmed cell death protein 1 (PD-1) on primary T cells by using a lentivirus delivery system. Methods Lentivirus vec-tors pLentiCRISPR A1-A6 containing different PD-1 genomic DNA sequences as single guide RNA ( sgRNA) for Cas9 targeting were constructed individually. The lentivirus vectors were tranduced into primary CD4 T cells. Flow cytometry analysis was performed to detect the expression of PD-1 for evaluating the knockout ef-ficiency. Results The lentivirus vectors pLentiCRISPR A1-A6 carrying six different target sites were con-structed and respectively tranduced into primary CD4 T cells. The expression of PD-1 accompanied with the activation of T cells. Co-expression of CD25 and PD-1 was observed on activated T cells. All of the six sites could be targeted by Cas9, of which A2 and A6 sites were more efficient in knocking out the gene encoding PD-1 with a rate of 19% and 29%, respectively. Conclusion This study suggests that it is feasible to knock out the expression of PD-1 on primary T cells by using CRISPR.
7.Clinicopathological features of eight cases with tubular aggregate myopathy

Objective To investigate the clinicopathological features of tubular aggregate myopathy.Methods Eight patients as experimental group were diagnosed with tubular aggregate myopathy in Department of Neurology,People' s Liberation Army General Hospital,between March 2000 and March 2013.The data were retrospectively analyzed.Enzyme histochemical techniques and electron microscopy were taken to observe the muscle structures.Results The detection rate of tubular aggregate was 0.374% (8/2 137).All of the 8 patients with tubular aggregate myopathy were male.Five patients presented with episodes of muscle weakness,while 3 patients presented chronic progressive muscle weakness.The main clinical features of all patients were muscle weakness.The creatine kinase level was mildly elevated in 4 patients,while it was normal in the other 4 patients.Electromyogram showed myogenic damage in 5 patients and normal in 3 patients.All of the 8 patients denied family history.By light microscopy,hematoxylin-eosin staining showed that tubular aggregates were multiple basophilic subsarcolemmal substance.Tubular aggregate stained red with modified Gomori trichrome stain,reacted intensely for nicotinamide adenine dinucleotidetetrazolium reductase,and remained unstained on oil red O,periodic acid Schiff,sudan black B,acid phosphatas,adenosine triphosphatase.By electron microscopy,tubular aggregates were densely packed tubules predominantly in the subsarcolemmal region.Conclusion Enzyme histochemical staining and electron microscopy show special features of tubular aggregate myopathy,are the critical techniques for the diagnosis of the disease.
8.Determination of Astragaloside IV in Radix Astragli and Its Preparations by Online SPE-Liquid Chromatography with Charged Aerosol Detector

A novel method for quantification of astragaloside IV in Radix Astragli was developed by using online SPE liquid chromatography coupled with corona charged aerosol detection( CAD) . The sample solution
was loaded into Acclaim Polar AdvantageⅡC18(50 mm×4. 6 mm, 3 μm) which was selected as online SPE column. Then the cleaning process was done by using the Right one of dual gradient pumps with Methanol-water as mobile phase. Acclaim C18(150 mm×4. 6 mm, 5 μm) was selected as analytical column with acetonitrile-water as mobile phase at a flow rate of 1. 0 mL/min. The eluate containing the target from SPE column was transferred into the analytical column by heart-cutting mode. The temperature of nebulizer of corona CAD was set at 30 ℃ and the nitrogen pressure was 241. 3 kPa. The baseline separation of astragaloside IV from matrix components has been achieved. There was a good linear correlation in the range of 4 . 0-80 mg/L for astragaloside IV and the correlation coefficient was 0 . 9998 . The standard addition average recovery of astragaloside IV in Radix Astragli was 97 . 6%. It has been validated that astragaloside IV in Radix Astragli and its preparations can be quantified rapidly and accurately with this method.
9.Grading of cerebral glioma with susceptibility weighted imaging evaluation of bleeding

Objective To explore the relationship between the amount of bleeding and the histopathologic grade of cerebral glioma with susceptibility weighted imaging (SWI), so that to assess the diagnostic value of SWI in grading cerebral glioma. Methods Totally 30 patients with cerebral glioma underwent SWI. The area of hemorrhage of every slice displayed in SWI was measured with software. The rate and amount of bleeding were compared between high and low grade tumors. Results There was no statistical significance in the incidence of detected bleeding on SWI (P=0.064), though it was higher in highly malignant group (80.00%) than that in low grade group (46.67%), while there was statistical significance in the amount of bleeding detected on SWI (Z=-2.275, P=0.026) between highly malignant group and low grade group. Conclusion The amount of tumor hemorrhage displayed in SWI is valuable for the preoperative grading of cerebral glioma.
10.Characterization of one novel mutation of the low density lipoprotein receptor gene in patients with familial hypercholesterolemia-one case report

Objective To analyze the genetic variation of low density lipoprotein receptor(LDLR)gene in patients with familial hypercholesterolemia(FH).Methods Polymerase chain reaction(PCR)was used to amplify the promoter region and the 18 exons of LDLR gene family with the patients' genomic DNA as templates.DNA sequencing of the PCR production was used to find mutations in patients with FH.Results A novel missense mutation(G→T)in exon 4 was identified by DNA sequencing in the proband and his mother,and the mutation was characterized at nucleotide site of 385 in exons of coding region,with tyrosine substitute for aspartate at amino acid site of 108.Conclusion Characterization of the novel mutation provides an example of the genetic basis of LDLR causing FH.
