AIM To investigate the anti-atherosclerosis mechanisms of arecoline. METHODS Fifty-eight male rats were assigned to control, model, arecoline 1 mg?kg -1 and arecoline 5 mg? [FQ(12。46,X-WZ]-kg -1 groups randomly. The rats in model and arecoline-supplied groups were fed with hypercholesterol diet. The following variables were measured: HE staining of rat aorta; serum level of TC, LDL-CHO, HDL-CHO, IL-8, ET-1 and NO; expression of eNOS protein on rat aorta; expression of MCP-1, ICAM-1, CXCR-2 and eNOS mRNA on rat aorta. RESULTS Arecoline increased plasma level of NO and the expression of eNOS protein and mRNA, decreased plasma level of IL-8 and the expression of ICAM-1, MCP-1 and CXCR-2 mRNA on rat aorta. CONCLUSION The anti-atherogenic effects of arecoline seems to be closely involved in increasing plasma level of NO and eNOS protein and mRNA expression, decrease in plasma IL-8 level and down-regulation of the expression of ICAM-1, MCP-1 and CXCR-2 genes.-

AIMTo investigate the anti-atherosclerosis mechanisms of the novel compound PPVP. METHODSThirty-six male rats were a ssigned to control, model and PPVP 5 mg?kg -1 groups randomly. The rats in model and PPVP 5 mg?kg -1 groups were fed with hypercholesterol diet for inducing atherolcerosis model. The following variables were measured: HE stainin g of rat aorta; serum level of TC, LDL-CHO, HDL-CHO, IL-8, ET-1, PGI 2, TXA 2 and NO. RESULTSPPVP increased serum level of NO, decreased plasma level of TXA 2 and inhibited the over expression of IL-8. CONC LUSIONThe anti-atherogenic effect of PPVP seems to be closely involved in increasing serum NO level, decreasing plasma level of TXA 2, and inhibiting over expression of IL-8. The result also suggests that the anti-atherogenic ef fect of PPVP in high cholesterol-fed rat may not due to the regulation of plasm a lipid profile, plasma level of ET-1 and PGI 2.

BACKGROUND:Cerebral white matter demyelination is outstanding in the images of delayed encephalopathy after acute carbon monoxide (CO) poisoning. Since Nogo-A and Nogo-receptor are expressed in onoligodendrocytes and neurons respectively, we infer that Nogo-A system is involved in brain injury after acute CO poisoning and related to delayed encephalopathy after acute CO poisoning. Endogenous CO is a gaseous messenger, which is the metabolic product of hemeoxygenase. There is no report about the CO effect on Nogo-A system til now. OBIECTIVE: To in vitro culture oligodendrocytes using endogenous CO, inhibit the activity of hemeoxygenase system using zinc protoporphyrin-IX (ZnPPIX) and observe the variation of Nogo-A in oligodendrocytes at mRNA and protein levels. METHODS: Rat oligodendrocytes cultured in vitro were divided into control, CO, ZnPPIX groups. Cels in the CO and ZnPPIX groups were treated with 1% CO directly, In the ZnPPIX group, 10 μmol/L ZnPPIX was added into the culture medium before CO treatment. The expressions of Nogo-A mRNA and protein at 6, 24, 48 hours after culture were compared. Differences in the peak levels of Nogo-A mRNA and protein between CO and ZnPPIX groups were detected using RT-PCR and immunohistochemistry respectively. RESULTS: The expression levels of Nogo-A mRNA and protein were significantly higher in the CO group than the control group and reached the peak at 24 hours of culture. Compared with the CO group, oligodendrocytes cultured with ZnPPIX showed higher expressions of Nogo-A mRNA and protein at 24 hours of culture. These findings suggest that except the influence of hypoxia occurring in CO poisoning, exogenous CO increases the expression of Nogo-A in cultured oligodendrocytes in vitro, and the heme oxygenase system can inhibit the expression of Nogo-A mRNA and protein.

Chinese Pharmacological Bulletin 2016;32(6):795-800
Aim Toinvestigatethepro-angiogenic effects of Danshensu derivative ADTM and explore its underlying possible signaling pathway using zebrafish embryosasinvivomodels. Methods Theangiogenesis activities of ADTM were determined in experimental models of normal and VEGFR tyrosine kinase inhibitorⅡ(VRI )-induced vascular defective zebrafish embry-os.Embryos were treated with various concentrations (50,100,200 μmol · L-1 ) of ADTM for indicated time. The diameter and the numbers of endothelial cells of zebrafish SIVs were evaluated, respectively. In VRI model, the number of intact and defective ISVs in each zebrafish embryo was counted. The total RNA of zebrafish embryos was extracted and transcriptional profiling was analyzed by deep sequencing.Quantita-tive real-time PCR(qPCR)was performed to 4 genes selected from transcriptional profiling to validate the data collected from transcriptome analysis. Results ADTMsignificantlyincreasedsubintestinalvessels (SIVs)diameter in a concentration-dependent manner in normal zebrafish as well as restored VRI-induced blood vessels defect in VRI-exposed zebrafish. The transcriptome data analysis demonstrated that 19 signif-icantly changed genes were mapped to insulin signaling pathway. The qPCR data are in good agreement with those obtained by deep sequencing and support the consistency between the two methods for determining relative expression levels in the zebrafish model.Con-clusion Inzebrafishmodel,ADTMexhibitsthe effects of angiogenesis and blood vessel restoration. The underlying mechanism may be involved in the acti-vation of insulin signaling pathway.

Objective:To evaluate the rapid nucleic acid amplification detection of Mycoplasma pneumoniae (MP)-DNA and MP-RNA in the diagnosis of MP infection and therapeutic values in children. Methods:Patients who were diagnosed with pneumonia were enrolled from the Department of Respiration, Children′s Hospital of Capital Institute of Pediatrics from January 2018 to December 2018.Specimens were detected using the MP and Macrolide-Resistant isolates Diagnostic Kit (PCR Fluorescence Probing, Jiangsu Mole Bioscience Co., Ltd.) and MP Diagnostic Kit (Isothermal RNA amplification, Shanghai Rendu Biotechnology Co., Ltd.).Results:Among them, 42.1%(840 cases) of the 1 994 cases were positive for MP-DNA, and the macrolide associated gene mutations were detected in 96.0% (806/840 cases) of them, while 33.9% (551 cases) of 1 624 cases were positive for MP-RNA.Seven hundred and fifty-eight specimens were simultaneously detected by adopting MP-DNA and MP-RNA, and the positive rate was 43.1% (327/758 cases) and 36.7% (278/758 cases), accordingly, which were inconsistent (Kappa=0.604) in 613 (80.9%, 613/758 cases) cases, with significant differences ( χ2=6.60, P=0.01). Part of the specimens were rechecked with the interval of 7 days: MP-RNA was negative in 70.1% (47/67 cases) specimens and MP-DNA was negative in 36.1% (22/91 cases) specimens ( χ2=33.20, P<0.01). Conclusions:The positive detection rate of MP was at a high level in 2018, in Beijing, China. The results of MP-DNA and MP-RNA are consistant. But RNA detection can help to diagnose MP in the early stage, and monitor the survival of MP and its efficiency.
