1.Pharmacokinetics and Bioavailability of Pulse Controlled-release Tablets of Ketoprofen in Dogs

OBJECTIVE: To study the pharmacokinetics and bioavailability of pulse controlled-release tablets of ketoprofen(KRT) in dogs.METHODS: A total of 6 dogs were randomly assigned to receive KRT(trial group,n=3) or KP enteric-coated capsules(control group,n=3) 150 mg,then a crossover trial was conducted 1 week later.Plasma samples were taken at different time points and the plasma concentration of ketoprofen in dogs was determined by HPLC.The pharmacokinetic parameters of the two KP preparations were computed and the correlation between absorption percentage in vivo and release rate in vitro was compared.RESULTS: The pharmacokinetic parameters of KRT versus those of its control preparation were as follows: tmax:(5.00?0.41) vs.(3.75?0.00) h;Cmax:(15.40?1.94) vs.(20.73?1.72) ?g?mL-1;AUC0~t(101.33?17.49) vs.(93.21?25.03) ?g?h?mL-1.The relative bioavailability of the trial preparation was 108.71%.The test preparation showed a good correlation(r=0.987) between absorption percentage in vivo and release rate in vitro.CONCLUSIONS: The KRT and KP enteric-coated capsules were bioequivalent.
2.Study of Acupuncture Zhongzhu(SJ3) and Waiguan(SJ5) by Functional MRI(fMRI)

Objective To observe the distribution of the activated cerebral regions after acupuncture stimulation of Zhongzhu(SJ3) and Waiguan(SJ5) by functional magnetic resonance imaging(fMRI) technique.Methods A total of 8 cases of volunteer healthy subjects were recruited for this study.Before and during manipulation of the needle,cerebral images were sampled separately by fMRI scanner and analyzed by AFNI software.Results During manipulation of the needle inserted in the unilateral Zhongzhu(SJ3) and Wauguan(SJ5),the highly-activated cerebral structures were cingulated gyrus,temporal gyrus,cerebellum,occipital gyrus,etc.Conclusion The effect of acupuncture analgesia was probably produces by the interaction of multiple brain structures of functional connectivity rather than through the activation of a single brain region.
3.Effect of simvastatin and atorvastatin on the expression of p 27 protein in mesangial cells of rat under high glucose

Objective To explore the effect of simvastatin and atorvastatin on the level of p 27 protein in mesangial cells of rat under high glucose .Methods Cultured rat mesangial cells in vitro were divided into four groups :normal glucose(5 .6 mmol/L glu-cose) ,high glucose(30 mmol/L glucose) ,high glucose(30 mmol/L glucose) plus simvastatin(10 μmol/L) ,high glucose(30mmol/L glucose) plus atorvastatin(10 μmol/L) .After treatment for 24 ,48 ,72 120 hours ,the cells were collected .The protein level of p27 was detected by Western blotting and the mRNA level of p27 was detected by RT-PCR .Results Compared to the normal glucose group ,the levels of p27 protein and mRNA in the high glucose group had a significant increase(P<0 .05) .Compared to the high glucose group ,the level of p27 protein and mRNA in the high glucose plus simvastatin or atorvastatin group both had a significant decrease(P<0 .05) .Conclusion By modulating the expression of P27 in mesangial cells ,statins could become the independent lipid-decreasing drug to prevent kidney .
4.Relationship between cofilin-1 expression and implantation capacity in eutopic endometrium of patient with endometriosis

Objective To investigate the relationship between expression of eofilin-1 and in vivo implantation capacity of eutopic endometrium of endometriosis.Methods Eutopic endometrium of 20 cases with stage Ⅲ or Ⅳ endometriosis were obtained by laparoscopic or laparotomy surgery (endometriosis group) matched with 20 cases of eutopic endometrium from patients with cervical cancer in situ (control group) in Department of Obstetrics and Gynecology,ShengJing Hospital.All cases' eutopic endometrium were collected and injected into abdominal cavity of nude mice to establish endometriosis animal model, then the successful rate of animal model and volume of endometriosis lesion were calculated.The expression and positive rate of eofilin-1 protein were measured by western blot and immunohistochemistry staining.Results The mean volume of endometriosis lesions (2.38 ± 0.22)mm~3 in endometriosis group was significantly bigger than (0.36 ± 0.08) mm~3 in control group (P < 0.05).The successful rate of establishing endometriosis model was 95% (19/20) in endometriosis group and 5% (1/20)in control group, which reached statistical difference(P <0.05).The expression and positive rate of cofilin-1 protein in eutopic endometrium and the expression of cofilin-1 protein in endometriosis lesion of animal model were 0.82±0.06, 90% (18/20), 0.85 ± 0.03 and 0.21 ± 0.03, 20% (4/20), 0.22 ± 0.02 in control group, which reached statistical difference(P <0.05).The successful rate of establishing endometriosis model with positive cofilin-1 in endometrium 86% (19/22) was significantly higher than 6% (1/16) of model's endometrium with negative cofilin-1 expression(P <0.05).Conclusions Implanting capacity and cofilin-1 expression level of eutopic endometrium of endometriosis were more intensive than that of normal endometrium.High cofilin-1 expression was probably related with implanting capacity of eutopic endometrium of endometriosis.

9.Influence of ulinastatin on inflammatory response and cellular immune function in patients with acute respiratory distress syndrome

Objective To observe the influence of ulinastatin (UTI) on curative effect,inflammatory response and cellular immune function in patients with acute respiratory distress syndrome (ARDS).Methods 96 patients with ARDS were randomly divided into two groups:the control group (48 cases) were treated with conventional treatment,and the UTI group (48 cases) were added with UTI injection based on the control group treatment.The changes of oxygenation index,vital signs,inflammatory factors and T lymphocyte subsets of patients in the two groups were compared before treatment and after treatment in 7 days,recorded the duration of mechanical ventilation time,ICU treatment time and mortality rate in ICU in the two groups.Results After treatment in 7 days,the vital signs (RF,HR,MAP),inflammatory factors (TNF-α,IL-6,IL-8,PCT,CRP,WBC),liver function (AST,ALT),renal function (BUN,Cr) and APACHE Ⅱ score,MODS score of UTI patients in the UTI group were lower than those in the control group (P < 0.05 or P < 0.01).Oxygenation index (PaO2/FiO2) and cellular immunity (CD4+,CD4+/CD8+) in the UTI group were higher than those in the control group (P < 0.01).Mechanical ventilation time,ICU treatment time in the UTI group were shorter than those in the control group (all P < 0.05),the mortality rate of ICU in the UTI group was lower than that in the control group (P < 0.05).There was no significant difference in the incidence of adverse reactions between the two groups (P > 0.05).Conclusions UTI can more effectively improve oxygen metabolism,vital signs for ARDS,liver and kidney function,reduce the APACHE Ⅱ score and MODS score,shorten the mechanical ventilation time and ICU treatment time,reduce the mortality rate of ICU.Its mechanism may relate to clearance of inflammatory factors and regulate cellular immune function of UTI.
10.Association of language dysfunction and age of onset of benign epilepsy with centrotemporal spikes in children

Objective To assess language function in children with benign epilepsy with centrotemporal spikes in children(BECTS) and its association with the age of epilepsy onset.Methods We assessed language function in 63 children with BECTS and 30 age and sex matched controls by the subtest of CWISC.Results Children with BECTS performed significantly worse on all language tasks as compared to controls and overall better language function was positively correlated with older age of the child.Early age at seizure onset demonstrated significant negative correlation with language dysfunction,age below 6 years being related to the lowest performance scores(P < 0.05).There was no relationship between the language function and the laterality of epileptic focus,seizure treatment status,or the duration of epilepsy(P > 0.05).There was significant relationship between the language dysfunction and onset age of epilepsy (P < 0.05).Conclusion Children with BECTS have language difficulties that are more pronounced in younger age group.Despite better language functioning in older children with BECTS,their verbal abilities remain inferior to those of children without epilepsy.Early age at seizure onset is a significant factor predicting worse language functioning in children with BECTS.
