China Pharmacist 2014;(11):1843-1845
Objective:To develop a simple and rapid HPLC method to determine the concentration and pharmacokinetics of gliqui-done in human plasma. Methods: Acetonitrile was used to precipitate plasma proteins directly and gliquidone was detected by an HPLC-fluorescence assay. The mobile phase consisted of acetonitrile-20 mmol·L-1 biphosphate potassium (80∶20, v/v, pH 2. 5). The flow rate was 1. 0 ml·min-1 . The excitation wavelength was 315 nm and the emission wavelength was 410 nm. Results:The line-arity of gliquidone in plasma was good from 0. 034 to 2. 2 μg·ml-1 and the quantitative lower limit was 0. 034 μg·ml-1 . The range of method recovery was 99. 23%-108. 11%, and that of extraction recovery was 85. 78%-88. 77%. The intra-and inter-day precision was both less than 10%. Both specificity and stability met the requirements. Gliquidone in plasma was determined using the above method after a single oral dose of 60 mg gliquidone tablets in 20 healthy volunteers, and then the pharmacokinetic parameters were ana-lyzed. The variation coefficients were close to or more than 50%. Conclusion:The method is simple, rapid and sensitive, and suit-able for the detection of gliquidone in a large quantity. Individualized administration of gliquidone should be considered due to the obvi-ous individual difference in pharmacokinetics.

Objective To evaluate the therapeutic response of transcatheter hepatic arterial chemoembolization (TACE) for primary HCC using 18 F-FDG PET/CT.Methods Twenty-four HCC patients (21males,3 females; age range:40 to 75 years, mean age:58.1 years) with high uptake of 18F-FDG on pretreatment PET/CT and with follow-up 18F-FDG PET/CT 1-2 months post TACE were retrospectively analyzed.A lesion was regarded as positive for HCC if it had higher FDG uptake than the surrounding liver parenchyma. All patients were followed for 3 months by clinical symptoms,AFP and 18F-FDG PET/CT.x2 test was used to analyze the difference between the diagnostic value of 18F-FDG PET/CT and AFP.Results On follow-up 18F-FDG PET/CT,9 patients had lower 18F-FDG uptake than the surrounding liver parenchyma, indicating no residual tumor; while 11 patients with residual tumor showed higher uptake of FDG than the liver background though the uptake was lower than that of baseline PET/CT.Two patients with isometabolic FDG uptake were confirmed of having residual tumor during follow-up.One case showed increased 18F-FDG uptake more than that of baseline, indicating disease progression. One case was false positive for residual tumor due to over attenuation correction in the presence of lipiodol. The sensitivity and accuracy for the detection of residual tumor by 18F-FDG PET/CT and AFP were 100% (14/14) vs 71.4% (10/14),and 95.8%(23/24) vs 70.8%(17/24) (x2 =6.56 and 4.18,both P<0.05),respectively.Conclusion 18F-FDG PET/CT is useful to evaluate the efficacy of TACE for 18F-FDG-avid HCC and it may provide important information for further treatment planning.

China Oncology 2014;(8):568-574
Background and purpose:RASSF10 acts as a kind of tumor suppressor in various tumor tissues, but researches in cardiac adenocarcinoma has not been reported. This study aimed to detect the methylation status and expression ofRas-association domain family 10 (RASSF10) in gastric cardia adenocarcinoma (GCA), and explore its role in occurrence and development of GCA.Methods:Methylation speciifc polymerase chain reaction (MSP), reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry method were respectively used to detect methylation status, mRNA expression and protein expression ofRASSF10 in 81 GCA tissues and corresponding normal tissues. Results:The promoter methylation frequency ofRASSF10 in GCA tissues (64.20%, 52/81) was signiifcantly higher than that in corresponding normal tissues (20.99%, 17/81,P<0.05), and was closely correlated with TNM stages, differential degree and lymph node metastasis (P<0.05). RASSF10 mRNA expression in GCA tissues (0.57±0.05) was significantly lower than that in corresponding normal tissues (0.78±0.02,P<0.05), and was closely correlated with TNM stages and lymph node metastasis (P<0.05). Protein expression of RASSF10 in GCA tissues (31.10%, 26/81) was signiifcantly lower than that in corresponding normal tissues (71.60%, 58/81,P<0.05), and was closely correlated with TNM stages, differential degree and lymph node metastasis (P<0.05). The promoter methylation frequency ofRASSF10 in GCA tissues was inversely related to its protein expression. Conclusion:Inactivation of RASSF10 caused by aberrantmethylation in the promoter region may be closely correlated with the GCA tumorgenesis.

A new on-column conductivity detection for microchip capillary based on the accessional solution conductor between separation channel and electrodes was designed. In this detection, the carrier electrolyte solution was chosen as conductor to eliminate large drifts of the detection signals, that because different composition or concentration can lead to transport processes(diffusion and electromigration) between the carrier electrolyte solution and mediator. The separation channel incorporated a dual-T and a crux-channel, dual-T was used for sample introduction, and the crux-channel was used for detection. The sensing electrodes connected to separation channel through the crux-channel and obtained the potential different of conductivity. This method avoids integrating the sensing electrodes directly within the separation channel and prevents any direct contact of the electrodes with the sample. It not only avoids electrode reactions but also is convenient for washing and replacing electrodes. In addition, the signals, contrary to contactless conductivity detection, can be detected in the low excitation voltage(2.5-4.0 V) and frequency(700-1700 Hz) range. In the buffer solution of 15 mmol/L MES-His(pH 5.8), the detection limits was 0.5 and 0.1 μmol/L for K+ and Na+.
