1.A study on the relevance of eosinophil counts in induced sputum and fractional concentration of exhaled NO and lung functions in asthmatics

Objective To investigate the correlations between eosinophil counts in induced sputum and lung function (FENO) and evaluate these parameters in medication adjustment in patients with asthma.Methods Sixty-five outpatients with mild to moderate persistent asthma ( mild,32 ; moderate,33 ) from January to August 2008 were enrolled in the study.All were treated with combined medications comprising inhaled corticosteroids plus long-acting β2 agonists for 1 year.Lung function (FEV1% and PEF% ),eosinophil counts in induced sputum,FENO,and Asthma Control Test (ACT) scores were obtained at regular follow-up intervals.Twenty-one healthy volunteers served as controls,and lung function,eosinophil counts in induced sputum,and FENO were also obtained.Results Sixty-three subjects completed 1-year or longer follow-up.Lung function of 63 subjects recovered quickly in the early days and improved slowly during the following 6 months.FENO decreased from (61 ± 25 ) nmol/L at baseline to ( 32 ± 19 ) nmol/L by the third month (q =7.32,P<0.05) and to (22 ± 12) nmol/L by the sixth month,which showed significant difference from normal controls [ ( 13 ± 8) nmol/L; q =6.63,P < 0.05 ].Eosinophil counts in induced sputum of the asthma group at baseline were (0.093 ±0.023) × 109/L and decreased to (0.032 ±0.011)× 109/L by the third month,which was significantly different from baseline and normal controls [ (0.005 ±0.003) × 106/ml; q =5.49,P <0.05 and q =5.87,P <0.05,respectively].FENO showed a significantly positive correlation with eosinophil counts in induced sputum in the first 6 months (r1 =0.612,r2 =0.558,r3 =0.675; all P<0.05) and a negative correlation with FEV1 (r1 =-0.537,r3 =-0.658,r6 = -0.623,r9 =-0.537,r12 =-0.597 ; all P <0.05 ) at any time point of the study.The ACT score of 63 subjects at baseline was 14 ±3,and the scores after treatment for 1,3,6,9,and 12 months were 18 ±5,19 ±7,23 ±2,24 ± 1,and 24 ± 1,respectively; at the same time,significant difference was found ( F =5.72,P < 0.05).Effectiveness was found according to the ACT score only 1 month after treatment.Conclusion The parameters of FENO and eosinophil counts in induced sputum were sensitive in the detection of airway inflammation and may be useful in evaluation of the efficacy of treatment and adjustment of medication regimens.
2.Comparison of in vitro isolation and culture of retinal pigment epithelial cells in rabbit eye and pig eye

Background Culture of retinal pigment epithelium(RPE) cells is very important for establishment of proliferative vitreoretinopathy (PVR) model,prevention and treatment of PVR as well as RPE cell transplantation.Isolation of animal RPE cells by trypsinization is a critical step.ObjectiveThe present study is to establish the methods of isolation and culture of retinal pigment epithelium (RPE) cells in rabbit and comparied with that of pig RPE culture.MethodsRPE cells were isolated by trypsinization in pigmented rabbit and pig and cultured in DMEM containing 20% fetal bovine serum.Cultured RPE cells were identified by immunochemistry.The fourth generations of cells were used in this experiment.Morphology and characteristics of cultured RPE cells from rabbit and pig were examined and compared under the light microscope.ResultsIsolated RPE cells from pig were obtained by once trypsin digestion,but two times of trypsinization were needed in rabbit RPE cells isolation.The differentiation in response to trypsinization was related to anatomic difference between the two types of cells .The adherence time of pig RPE cells was 24 hours ,however,the rabbit RPE needed 48-72 hours after culture.Proliferation and vitality of cultured cells were gradually attenuated and melanin decreased after several times subculture.The morphology of culture RPE cells was obviously different between rabbit and pig because species difference.Immunohistochemistry demonstrated the positive response of RPE cells for keratin.ConclusionRPE cells can be acquired from both rabbit and pig by trypsinization and culture.The culture process of RPE cells of pig is simpler than that of rabbit.Cells within the fourth generations are suitable for experimental application.
3.Evaluation of the treatment effect of psychotherapy on vitiligo

Objective To investigate the role of psychotherapy plays in the treatment of vitiligo.Methods Psychological health questionnaire(SCL-90)was used in the study to evaluate the psychological well being of 120 vitiligo patients.66 subjects suffering from psychological problems were divided randomly into groups Ⅰand Ⅱ while 54 subjects free of psychological problems randomly into groups Ⅲ and Ⅳ.Psychotherapy was applied together with drug and phototherapy to those subjects from groups Ⅰ and Ⅲ while groups Ⅱ and Ⅳ having no psychotherapy accompanied.After 3 months of treatment,evaluation was done on the treatment effect with psychological testing once again.The correlation between the improvement of psychological conditions of the subjects and the lesion development was then evaluated by"P"value of the four groups.Results Significant difference was achieved between group Ⅰ and group I1I(P<0.05).There was no significant difference between groupsⅢand IV(P>0.05).There was significant difference between groups Ⅰ and Ⅱ.There was no significant difference between groups Ⅱ and Ⅳ.Conclusions The psychotherapy is helpful in improving the early treatment effect and quality of life in vitiligo patients.
4.Effect of apoptosis of vesicular stomatitis virus on tumor cells of nasopharyngeal carcinoma in vivo.

OBJECTIVE:
To investigate apoptosis induced by vesicular stomatitis virus (VSV) in HNE-1 cancer cells of human nasopharyngeal carcinoma by models of nude mice-BALB/c in vivo.
METHOD:
HNE-1 cells are collected from culture bottle and infected infra right-side post-back epithelium of nude mice-BALB/c (6 x 10(6) cells/0.1 ml/each mice) to create HNE-1 tumor models of nude mice-BALB/c. When the diameter of HNE-1 tumors is 5 to 8 millimeter, HNE-1 tumor models are treated with VSV (1 x 10(8) pfu /ml) or Saline. By Hoechst 33258-staining under fluorescence microscope, induction of apoptosis by VSV in HNE-1 tumor models are recorded and studied, compared with that by Saline in HNE-1 tumor models in vivo.
RESULT:
Compared with control group of saline, apoptosis of HNE-1 cancer cells of human nasopharyngeal carcinoma increase apparently in the remaining tumor cells of nude mice treated by VSV (P < 0.01).
CONCLUSION
The present study suggests that the treatment with VSV could augment the apoptosis cells of human nasopharyngeal carcinoma in vivo.

5.Analysis on post-traumatic stress disorder and its relative factors of emergency department nurses after suffering from workplace violence

Objective To describe the general condition of workplace violence against emergency department nurses in five hospitals of Guangzhou;to investigate the status of the post-traumatic stress disorder in the emergency department nurses after suffering from workplace violence;to analyze the relative factors of post-traumatic stress disorder.Methods 143 emergency department nurses from 5 hospitals in Guangzhou were investigated by general information questionnaire,workplace violence questionnaire,PCLC and SSRS.The investigation data were analyzed.Results 86.7% of emergency department nurses suffered from workplace violence during the past 1 year;the most popular style was non-physical violence.The emergency department nurses suffered from negative emotional experience,such as grievance,chagrin,low work passion,not focused spirit.The scores of PCL-C of emergency department nurses who had suffered from workplace violence were obviously higher than those who hadn't.21.8% of the emergency department nurses who suffered from workplace violence in the past one year had certain degree of the signs of PTSD,12.1% had obvious signs of PTSD.The influencing factors of PTSD:degree of hurt,objective support and availability of social support.Conclusions The situation of workplace violence which the emergency department nurses were faced with was more and more grave.The emergency department nurses who had suffered from workplace violence were in different degree of PTSD.The more social supports the nurses get,the better mental health status they will possess.
6.Screening of the pORF3 binding protein of HEV in human liver cells by using yeast double hybridization system

HEV is a positive strand RNA virus containing 3 open reading frames(ORFs), while the function of its binding protein pORF3 is poorly understood. In our study, the interactive protein of the pORF3 from human liver cDNA library was screened by the SOS recruitment system, and the construction of the pSOS bait-ORF3 plasmid was identified. The expression of the pORF3 was analyzed by Western blotting, and the detection result of the self activation of bait protein was obtained as expected. Totally, 8 putative interacting proteins had been successively screened. Some of these cellular binding proteins of pORF3, especially the important immunological pathway kinase p110δ, may advance our understanding of the role in ORF3 protein of HEV, and the possible implications of these candidate proteins for the further research are discussed.
7.The Analyses of Auditory Brainstem Pathway Encoding of Speech Sounds in Healthy Post -Menopause Women

Objective The aim of this study was to compare the basic characters of the speech - evoked audi‐tory brainstem response (speech - ABR) in healthy young women with that in post - menopause women and to get the changes of the probable factors for the auditory brainstem pathway encoding of speech sounds in healthy post -menopause women .Methods Speech - ABR of forty - seven normal hearing subjects including twenty - seven young women and twenty post - menopause women were recorded .The speech syllable /da/ as stimulus sound was trans‐mited to right ear by the insert earphones in speech - ABR test .Results The response waves of speech - ABR in post - menopause women were similar to those in young women ,which containal the onset responses (peak V and A) ,the transition (peak C) ,the frequency following responses (peak D ,E and F) and the offset response (peak O) .Except wave C ,the latency of wave V (6 .99 ± 0 .34ms) ,wave A (8 .32 ± 0 .49 ms) and wave O (48 .86 ± 0 .50 ms) in post - menopause women were evidently longer than those of in young women (the waveV :6 .60 ± 0 .25 ms ,wave A :7 .56 ± 0 .31 ms and wave O :47 .88 ± 0 .31 ms) ,while the amplitude of wave A ( - 0 .16 ± 0 .06 μV ) and O ( - 0 .12 ± 0 .06 μV ) in post - menopause women were obviously lower (modulus) than those of in young women (the wave A : - 0 .27 ± 0 .08 μV and O : - 0 .18 ± 0 .07 μV)(P< 0 .05) .In the FFR of speech - ABR ,the latency of wave D (23 .27 ± 0 .67 ms) ,wave E (31 .88 ± 0 .58 ms) and F (40 .61 ± 1 .22 ms) in post - menopause women were also longer than those of in young women (the wave D :22 .40 ± 0 .44 ms ,wave E :31 .00 ± 0 .43 ms and F :39 .53 ± 0 .61 ms) .Besides ,the amplitudes of wave D ( - 0 .12 ± 0 .04 μV ) and F ( - 0 .08 ± 0 .06 μV ) in post -menopause women were visibly lower (modulus) than those of in young women (the wave D : - 0 .17 ± 0 .08 μV and F : - 0 .16 ± 0 .07 μV)(P< 0 .05) . ④ The V /A slope also showed very significant difference ,the V/A slope in young women was much bigger than that in post - menopause women (P< 0 .05) .Conclusion The synchronism and phase locking of auditory brainstem pathway to processing and encoding capability in healthy post - menopause women is poorer than those of in young women .It may suggest that the growth of the age and the decrease of hor‐mone will weaken the ability to processing of speech sounds in women .
8.Diagnosis boundary values of metabolic syndrome obesity index for Children and adolescents

Objective: To determine the distribution characteristics of waist circumference (WC), waist height ratio (WHtR) of 6–18 years olds in Guangzhou, and to put forward the WC and WHtR appropriate boundary values for 6–18 years olds on the basis of cardiovascular disease (CVD) risk factor assessment. Methods: We analyzed the height, weight, WC and its metabolic indication data (blood pressure, fasting blood glucose, and blood lipids) of 15 000 children in Guangzhou, aged 6–18, with the receiver-operating characteristic curve (ROC), and explored the best value point of WC and WHtRfor the prediction of cardiovascular diseases. Results: When the WC percent reached P85, and WHtR reached 0.48, the cardiovascular risk factors of fasting blood-glucose, blood pressure, and blood fat were signiifcantly higher. Conclusion: The 85th percentile value of WC and 0.48 of WHtR are the appropriate boundary values in increasing the cardiovascular disease risk factors in Chinese children and teenagers. WC and WHtR as a relatively simple inspection method, can well predict cardiovascular diseases, and be used in the conventional measuring items among students.
9.Effect of apoptosis and anti-tumor of vesicular stomatitis virus on tumor cells of nasopharyngeal carcinoma in vitro.

OBJECTIVE:
To study the result of HNE-1 cancer cells lesion and HNE-1 cancer cells apoptosis caused by vesicular stomatitis virus in vitro.
METHOD:
Firstly, HNE-1 cancer cells of human nasopharyngeal carcinoma were cultured in vitro. Secondly, tumor cells were treated with vesicular stomatitis virus (VSV) in different concentration (0.1 moi, 1.0 moi, 10.0 moi and 100.0 moi), while cells without treatment were used as blank assay. Finally, we observed the lesion of HNE-1 cells treated with VSV for 24 to 48 hours under invert microscope, compared with HNE-1 cells of blank assay. By measurement of MTT reduction assay and Hoechst 33258- staining under fluorescence microscope, the effect of VSV killing HNE-1 cancer cells and the induction of apoptosis by VSV in HNE-1 cancer cells were investigated in vitro.
RESULT:
Compared with blank assay, HNE-1 cancer cells of human nasopharyngeal carcinoma treated with VSV showed phenomenon of lesion under invert microscope. Followed the increase of concentration of VSV, cell survival rate of HNE-1 cancer cells decreased. The research methods of MTT reduction assay and Hoechst 33258 staining under fluorescence microscope defined the effect of VSV killing HNE-1 cancer cells and confirmed that VSV inducing apoptosis of HNE-1 cancer cells in vitro.
CONCLUSION
VSV not only exhibited the anti-tumor activity in HNE-1 cancer cells of human nasopharyngeal carcinoma but also showed the potential power of inducing apoptosis of HNE-1 cancer cells in vitro.

10.Relationship between CD4⁺CD25(High)CD127(low) regularly T cells in the peripheral blood and tumor regression after neoadjuvant therapy in patients with rectal cancer.

OBJECTIVETo observe the proportion change of immune cells in the peripheral blood of patients with rectal cancer after neoadjuvant therapy and to explore the relationship between tumor regression and CD4⁺CD25(High)CD127(low) regularly T cells(Treg cells).
METHODSPatients with rectal cancer who underwent the neoadjuvant therapy before surgery at the Shanxi Cancer Hospital Colorectal Surgery Department from January to December 2013 were prospectively enrolled. These patients were divided into down-staging group and non-down-staging group according to the change of staging in accordance with TNM classification for rectal cancer after neoadjuvant therapy. Flow cytometry was used to analyze the proportions of Treg cells, CD4+T cells, CD8+T cells, NK cells, B cells, and CD4+/CD8+ ratio in the peripheral blood from these patients before and after neoadjuvant therapy.
RESULTSA total of 108 patients were enrolled, including 76 cases in the down staging group and 32 cases in the non-down-staging group. Differences of immune cells proportions between two groups before neoadjuvant therapy were not statistically significant(all P>0.05). In the down-staging group, the proportions of Treg cells, B cells and CD4+/CD8+ ratio were decreased while the proportion of NK cells did not change obviously after the neoadjuvant therapy. Interestingly, in the non-down-staging group, the proportions of B cells and CD4+/CD8+ ratio were decreased while the proportions of Treg cells and NK cells did not change obviously after the neoadjuvant therapy. In addition, after neoadjunvat therapy, the proportion of Treg cells in down-staging group was significantly lower than that in non-down-staging group [(4.4 ± 1.7)% vs. (6.2 ± 1.9)%, P=0.001].
CONCLUSIONFor patients in the down-staging group after neoadjuvant therapy, the proportion of Treg cells in peripheral blood decreases, suggesting that Treg cells may be a valuable biomarker for assessing tumor regression.
