As a member of homeobox gene family,HOXA5 is expressed in many organs and can regulate gene expression,cell differentiation and morphogenesis.Its structure and (or) function abnormalities are closely related to the development of leukemia,breast cancer,brain hemangioma,hepatocellular carcinoma and so on.Researching the relationships between HOXA5 and tumors contributes to cancer diagnosis,treatment and prevention.

Inhibin B(INHB) and anti-Müllerian hormone(AMH) detection have important clinical significance in assisted reproductive technology.INHB for evaluation of male testis function is very meaningful,playing a guiding role in the treatment of oligospermatism.INHB has very high clinical diagnosis value for evaluation of ovarian reserve function,being an important indicator to predict the effect of controlled ovarian hyperstimulation.AMH for evaluation of women ovarian function is significant,and is a diagnosis index of reproductive domain related diseases such as polycystic ovary syndrome and one of the important indices for evaluation of assisted reproductive outcomes.

Objective: To study the cellular immune responses and the anti-tumor effects induced by peptides derived from onco-protein HER2/neu. Methods: Two HER2/neu peptides compatible with H-2Kd binding motif as tumor rejection antigens were synthesized. The ability of inducing peptide-specific CTLs and suppressing the tumor growth in BALB/c mice immunized by HER2/neu peptide was analyzed. Results: HER2/neu peptides could promote the proliferation of BALB/c lymphocytes both in vitro and in vivo. HER2/neu peptide-specific CTLs could be induced from peptide-immu-nized BALB/c mice, which showed specifically killing effects on HER2/neu positive SP2/OHER2 cells but not on HER2/neu negative SP2/O cells. Moreover, the growth of SP2/OHER2 tumor were significantly suppressed in BALB/c mice immunized with HER2/neu peptides. Conclusion: These results suggested that it was feasible to use MHC-I epitopes derived from tumor-specific antigens as vaccines for cancer immunotherapy.

Objective: To develop an HPLC method for the simultaneous determination of 3 ultraviolet absorbers (benzylidene camphor sulfonic acid, camphor benzalkonium methosulfate, 4-methylbenzylidene camphor) in cosmetics.Methods: After extracted by acetonitrile-methanol-ammonium acetate aqueous solution-tetrahydrofur(30∶35∶30∶5, v/v)(for lotion and milk) or acetonitrile-methanol-ammonium acetate aqueous solution(30∶20∶50, v/v)(for cream and powder), the UV absorbers were separated on a Thermo scientific C18 column (250 mm×4.6 mm,5 μm) at 35 ℃ with methanol, acetonitrile and 0.02 mol·L-1ammonium acetate aqueous solution (pH 5.15 adjusted by glacial acetic acid) as the mobile phase with gradient elution at the flow rate of 1.0 ml·min-1, and then analyzed by a DAD detector at the wavelength of 288 nm and 300 nm.Results: Benzylidene camphor sulfonic acid, camphor benzalkonium methosulfate and 4-methylbenzylidene camphor showed good linearity within the range of 12.26-98.06 μg·ml-1(r=0.999 9), 9.31-74.48 μg·ml-1(r=0.999 9) and 6.86-54.88 μg·ml-1(r=0.999 9), respectively.The limit of detection (LOD) and the limit of quantitation (LOQ) were 4.6, 6.4 and 1.6 ng and 17.4, 15.9 and 5.5 ng, respectively.The average recovery was 99.2%(RSD=1.58%), 99.8%(RSD=2.38%) and 99.2%(RSD=2.03%)(n=36), respectively.Conclusion: The method is simple, rapid, reproducible, accurate and reliable, and can be applied in the determination of ultraviolet absorbers in cosmetics.

The literature was analyzed on the improvement of motor sensory function, urination and defecation in myelitis treated with acupuncture and moxibustion and focused on the characteristics of the diagnosis and treatment. The literature on acupuncture and moxibustion treatment for myelitis was collected from CNKI, Wanfang, VIP and PubMed. The analysis included the characteristics of acupoint selection, methods of acupuncture and moxibustion, time of treatment, total treatment period, efficacy, follow-up, safety, etc. Totally, 26 articles were collected. The combined therapy of acupuncture and moxibustion was predominated (12/26, 46. 15%). For the motor and sensory impairment, the acupoints were mainly selected from the four limbs, the yangming meridians of hand and foot and those adjacent to the affected spinal segments and on the governor vessel as well as Jiaji (EX-B 2) points. For urinary impairment, the acupoints were selected mainly from the lower abdominal region on the conception vessel and the lumbosacral region on the bladder meridian. For the intestinal impairment, the acupoints were from the lower limb on the stomach meridian, the lower abdominal region on the conception vessel and the back points on the bladder meridian. The intervention started commonly in the first 3 months after onset. The total treatment period was in the range from 1 to 3 months. The efficacy of acupuncture and moxibustion was 69.19% to 82.56% for the improvement of motor sensory and urination, defecation function. The efficacy in follow-up was stable and the adverse reactions were not reported. It is viewed that on the basis of early diagnosis and active medication, acupuncture and moxibution achieve a certain of efficacy on the impairment of motor sensory function, urination and defecation. A clinical research is expected to further verify the efficacy.

Objective To explore the effects of lentivirus-mediated RNA interference targeting HOXA10 gene on the proliferation, apoptosis and drug resistance of leukemic cell line NB4.Methods NB4 cells were divided into three groups: interference group, negative control group and untreated group.The infection efficiency of lentivirus for NB4 cells was detected by flow cytometry, and the expression of HOXA10 gene of NB4 cells at mRNA and protein level was detected by real-time PCR and Western blot.Cell survival was determined by MTF assay, and apoptosis and necrosis rates were detected by flow cytometry.Western blot was used to detect the influence of down-regulation HOXA10 gene on the multi-drug resistance-1 (MDR-1) protein.Results The ratio of GFP positive cells was up to 90 ％.HOXA10 gene mRNA and protein levels were decreased in interference group compared with control group.The inhibition rate of interference group was (52.12±4.02) ％, the apoptosis rate of interference group was (30.0±2.7) ％, and their differences in the interference group and in control groups (negative control group and untreated group) were significant (P ＜ 0.05).Western blot results showed that interfering HOXA10 gene significantly reduced the resistance gene MDR-1 expression level and reverse the drug-resistant of leukemia cells.Conclusions Lentivirns-shHOXA10 can steadily reduce the expression level of HOXA10, inhibit the leukemic cells proliferation, promote apoptosis and reverse drug-resistant.HOXA10 gene is expected to become a new target for reversing leukemia drug resistance.

Objective:The clinical factors of single-port video-assisted thoracoscopic surgery (SP-VATS) were compared with those of multi-port video-assisted thoracoscopic surgery (MP-VATS). The differences between the two surgical methods and their respective postoperative recoveries were also discussed. Methods:A total of 522 patients who underwent surgical treatment for lung cancer in Tianjin Medical University Cancer Institate and Hospital from January, 2014 to December, 2015 were retrospectively reviewed. Of these cases, 83 underwent SP-VATS and 439 underwent MP-VATS. The two surgical methods were then compared in terms of opera-tive time, operative bleeding, number of lymph node and lymph node cleaning station, pain degree, 24 h postoperative chest drain-age, and in-hospital time after operation. Results:The differences between the patients who underwent SP-VATs and those who under-went MP-VATS in term of gender, age, smoking, tumor diameter, TNM stage, pathological type, and tumor location were not statistical-ly significant. The operative time in SP-VATS group was longer than that in the MP-VATS group (P<0.01), whereas in-hospital time after operation in the former group was shorter than that in the latter (P=0.011). Furthermore, pain degree in the SP-VATS group is lower than that in the MP-VATS group (P=0.041). The differences between the two groups in terms of operative bleeding, number of lymph node and lymph node cleaning station, and 24 h postoperative chest drainage were not statistically significant. Conclusion:SP-VATS can achieve a surgical effect similar to that of MP-VATS but has a prolonged operation time. SP-VATS is beneficial to postoperative re-covery and reduces the degree of pain. Thus, it has great potential for development.

It has been recognized only in the past 15 years that the major pathological mechanism of asthma is inflammation, which has a hallmark of focal leukocyte extravasation. The different recruitment behaviors of inflammatory cells depend on the expression of specific adhesive molecules on both leukocytes and endothelial cells(EC). Intercellular cellular adhesion molecule-1(ICAM-1) plays a major role. In the previous detailed in-vivo study of asthmatic models, we have proved the significant pathological increase of both the leukocyte-endothelium adhesion phenomenon and the ICAM-1 expression in the tissues of asthmatic lung. It may be due to an ICAM-1 accumulation on the microvascular endothelial cells and thus an enhancement of adhesion force during the course of disease. In this study, endothelial cells were obtained from rats by mechanical isolation of lung tissues and in vitro culture on glass. Confluent endothelial cells were incubated with serum collected from rat blood of normal and asthmatic models and were used for in vitro study of endothelial cell adhesion. The effect of pathological serum stimulation was examined on the expression of ICAM-1 of the pulmonary microvascular endothelial cells (PMEC). The expression of ICAM-1 on PMEC was measured by indirect immuno-fluorescence with flow cytometry. We found that the surface expression of ICAM-1 was obviously increased on serum-incubated EC as compared with that on the culture solution-incubated one, and that asthmatic serum increased the expression of ICAM-1 on EC to a peak in 4 hours and then decreased it rapidly,the expression level remained the same in the whole course when EC was treated with culture solution or normal serum.

Objective To investigate the expression of hypoxia-inducible 1 alpha(HIF-lα)and glucose transporter 1(Glut1)in human breast cancer and its relationship to proliferating cell nuclear antigen (PCNA)protein and clinical pathologic factors.Methods Immunohistochemical staining was used to measure the expression of HIF-lα.Glut1 and PCNA in human breast fibroadenoma,usual hyperplasia and breast carcinoma.Results HIF-1α expression was not found in breast fibroadenoma and hyperplastic Iesions.In contrast.the positive rate of HIF-1α was found in the ductal carcinoma in situ 55%(DCIS,11/20)and the invasive breast carcinoma 85%(51/60).Glut1 positivity in breast carcinoma was 58.8%(47/80).The totsl positive rate of PCNA in breast carcinoma was 75%(60/80),that in DCIS was 65%(13/20)and that in invasive carcinoma was 78.3%(47/60).There was a positive correlation between HIF-lα and Glut 1 level (r=0.653,P＜0.01),a positive correlation between HIF-1α and PCNA level(r=0.693,P＜0.01);and also a positive correlation between Glutl and PCNA level(t=0.742.P＜0.01).conclusion The overexpression of HIF-lα and its target gene Glut1 played important roles in carcinogenesis and progression of breast carcinoma and closely correlated with cell proliferation of breast carcinoma and may become a new target for treatment of breast carcinoma.
