1.Effect of thrombospondin-1 on proliferation of pulmonary artery smooth muscle cells

Objective To investigate the effect of thrombospondin 1(TSP-1) on the cell proliferation of cultured rat pulmonary artery smooth cell (PASMCs) in vitro .Methods Rat PASMCs were cultured in vitro ,and treated with different concentrations (10-12 、10-11 、10-10 mol/L) of TSP-1 for 12 ,24 ,48 h .The cell proliferation was quantified by MTT assay .The cell cycle of PASMCs was measured by flow cytometric(FCM) analysis .Results MTT assay showed that TSP-1 promoted the proliferation of PASMCs significantly ,and the effect was concentration-dependent and time-dependent .FCM analysis indicated that TSP-1 increased the percentage of S phase .The percentage of S phase of PASMCs were increased after treated with thrombospondin-1 for 12 h , slight down after 24 h ,while reached a maximal level at 48 h .Conclusion The TSP-1 promotes rat PASMCs proliferation in a con-centration-dependent and time-dependent manner .
2.Effects of Jiantai Liquid on Progesterone Receptor(PR)and PRmRNA in the Endometrium of Mice at Nidation Stage

Objective:To study on effect of Jiantai Liquid on progesterone receptor(PR)in the uterus at the nidation stage in the mouse with disturbance of blastocyst nidation.Methods:The gestational mice were randomly divided into a normal group,a model group and a Chinese drug group.Mifepristone was used for inducing disturbance of blastocyst nidation in the mice,which were interfered with Jiantai Liquid.At the eighth week of pregnancy,the mice were killed and morphological changes of the endometrium were observed,and PR expression in the uterus was detected with immunohistochemical method and PR and PRmRNA expressions were detected with Western blot and PT-PCR methods.Results:The endometrium showed maldevelopment in the model group and normal development in the Chinese drug group,similar to the normal group;the gland,the PR expression range and intensity of PR expression in the interstitial substance in the Chinese drug group increased as compared with the model group(P0.05);expressions of PR protein and gene in the Chinese drug group were significantly higher than those in the model group(P0.05).Conclusion:Jiantai Liquid can promote development of the endometrium and improves blastocyst nidation via regulating expressions of PR and PRmRNA in the uterus in the mice with disturbance of blastocyst nidation.
3.A social survey and analysis of setting up the new subject of physical education in Chinese Medicine colleges

Objective To investigate into social cognition of new subject physical education in Chinese Medicine colleges and to find out existing issues and institutes strategy to solve them. Methods Questionnaire and analysis of documents and telephonic interview were adopted. Results Society needs a large number of graduates who have solid knowledge of health protection and a good command of methods of preseving health and keeping fit and at the same time requires them to go in for teaching athletics, training and organizing games. New subject Physical Education has some advantages,and there are also lots of difficulties. Conclusions Course setting should be sptimized, practical teaching should be strengthened and publicized to ensure that the cultivation of talents meets the need of society.
4.Evaluation of left ventricular contraction pattern and function with area strain

Objective To evaluate left ventricular (LV) myocardial contraction patterns and function by analyzing the distribution of peak value of segmental area strain and time sequence to the peak value.Methods 31 healthy subjects were involved.Real-time three dimensional full volume images of LV were acquired and analyzed with EchoPAC.LV volume and function parameters,as well as segmental and global systolic area strain and time to peak value were measured.The distribution of peak segmental area strain and time sequence to the peak value,interobserver and intraobserver variability of area strain parameters,as well as the relationship between global area strain and ejection fraction were evaluated.Results The peak segmental area strain was lower in the basic inferior and apical segments,and the peak value was reached earlier in the basic inferior,anteroseptal,and septal segments.Intraobserver and interobserver reliability of area strain parameters were good.Furthermore,a negative correlation was observed between global area strain and ejection fraction (r =-0.688,P =0.000).Conclusions The area strain measured by 4D strain can be used to evaluate the LV myocardial motion pattern and function.
5.B-type natriuretic peptide and risk of type 2 diabetes mellitus

Objective To explore the association of NH2-terminal pro-B-type natriuretic peptide ( NT-proBNP) with the risk of type 2 diabetes.Methods One hundred and twenty-six impaired glucose regulation( IGR) participants from Diabetic Identification Center of Tianjin Metabolic Diseases Hospital were included.NT-proBNP was measured in plasma samples collected from participants at baseline condition.Results At baseline, NT-proBNP was inversely associated with body mass index, waist circumference, fasting glucose, insulin and low-density lipoprotein-cholesterol( LDL-C) levels.During a follow-up of 2 years, 51 participants reported a new diagnosis of diabetes from OGTT.Baseline quartiles of NT-proBNP were inversely associated with diabetes risk, even after multivariable adjustment.Theadjustedrelativerisksfordiabeteswere1.0(reference),0.83(95%CI0.74-0.96),0.78(95%CI 0.68-0.90), 0.74 (95%CI 0.64-0.87) for the 1st, 2nd, 3rd, and 4th quartiles of baseline NT-proBNP, respectively ( P<0.01 ) .Conclus ion In IGRpopulation , lowlevels of NT-proBNP were associated with a significantly increased risk of type 2 diabetes.
6.Expression and clinicopathological significance of OPN and CD44v6 in lung cancer.

BACKGROUNDThere have been a lot of studies about surface adhesion molecule (CD44) in recent years, however study on osteopontin (OPN) is still few. The aim of this study is to investigate the levels of OPN and CD44v6 in lung cancer and their correlation.
METHODSOPN and CD44v6 expression were detected in 78 lung cancer tissues by immunohistochemistry.
RESULTSThe positive rate of OPN and CD44v6 expression in non-small cell lung cancer (NSCLC) was 58.46% and 66.15% respectively, but no positive case in small cell lung cancer. The expression of OPN and CD44v6 in NSCLC was closely related to TNM stages (P < 0.01), but not to cell differentiation (P > 0.05). There was a remarkably positive correlation between the expression of OPN and CD44v6 (r=0.255, P < 0.05).
CONCLUSIONSThe co-expression of OPN and CD44v6 may be involved in progression and metastasis of lung cancer. The interrelationship of OPN and CD44v6 was coordinative with development and metastasis of lung cancer. It might be helpful to predict the metastasis and prognosis of NSCLC.
7.Study of left ventricular diastolic function in patients with DDD pacemaker implantation for sick sinus ;syndrome during atrioventricular conduction and right ventricular apex pacing mode

Objective To analyze the left ventricular (LV) diastolic function parameters of patients with sick sinus syndrome (SSS) and DDD pacemaker implantation during atrioventricular conduction (AVC) and right ventricular apex pacing(RVAP)mode.Methods Forty-six consecutive patients with SSS who had undergone DDD pacemaker implantation were studied.Fifty volunteers were selected as control group.Changing from AVC to RVAP mode,the acute effect on echocardiographic left ventricular diastolic function parameters,including mitral inflow PW parameters,mitral annulus tissue Doppler imaging parameters,left atrial volume index,and LV∕apical∕basal untwist parameters were measured.And all subjects were classified into normal diastolic function or three degrees of LV diastolic dysfunction.Results Compared with control group,both AVC and RVAP mode of patients with DDD pacemaker implantation resulted in the worsening of LV diastolic function as shown by(1) prolonged deceleration time of E wave, decreased descending slope of E wave,as well as decreased early diastolic velocity at the septal mitral annulus,(2) the decrease of LV∕basal∕apical untwist velocity,(3) the increase in the degree of diastolic dysfunction.Conclusions In patients with DDD pacemaker implantation,both AVC and RVAP mode are associated with the deterioration of LV diastolic function,which is particularly obvious in RVAP mode.
8.Study of left ventricular function in patients with DDD pacemaker implantation during atrioventricular conduction and right ventricular apex pacing mode

Changing from AVC to RVAP mode ,the acute effect on echocardiographic parameters ,including LVEF , parameters of aortic/pulmonary artery pulse wave Doppler ,and parameters of left ventricular twist by speckle tracking imaging were measured respectively . Pacemaker implantation duration and cumulative ventricular pacing proportion ( Cum% VP) were recorded . The relationships of pacemaker parameter and above left ventricular function parameters were analyzed . Results Compared with control group ,values of peak rotation in LV apex and LV twist were significantly lower during AVC and RVAP mode . The value of peak rotation in LV base showed no significant difference between three groups .Apical‐basal rotation delay during RVAP was significantly longer than those during AVC and in control group respectively ( P <0 .05) . LVEF during RVAP decreased statistically ( P< 0 .05 ) ,but showed no difference during AVC , compared with the control group . The peak LV twist related positively with LVEF ,and negatively with Cum% VP . Conclusions RVA pacing decreases left ventricular function , which is independent of asynchrony contraction patterns caused by pacing . LVEF ,apical rotation and LV twist are more sensitive to demonstrate the LV dysfunction in patients with pacemaker implantation . LV twist related negatively with Cum% VP .
9.Effects of sarcopenia on physical performance and nutritional status in elderly patients with gastrointestinal tumors

Objective:To investigate the effect of sarcopenia on physical performance and nutritional status in elderly patients with gastrointestinal tumors.Methods:a total of 120 eligible in-patients were enrolled from gastrointestinal surgery department of a Grade A tertiary hospital in Ningbo city from September 2021 to January 2022. General clinical data were collected, nutritional risk was screened using Nutritional Risk Screening 2002 (NRS 2002), physical performance was assessed by Short Physical Performance Battery (SPPB), skeletal muscle index at the third lumbar vertebra level (L3 SMI) was calculated using abdominal CT scan, and grip strength/muscle strength, gait speed by 6 Meter Timed Walk Test and calf circumference were measured. Subjects were divided into sarcopenia and non-sarcopenia group according to the diagnostic criteria of the Asian Working Group for Sarcopenia (AWGS). Impacting factors of sarcopenia and the correlation between muscle mass and physical performance in elderly patients with gastrointestinal tumors were analyzed.Results:The overall prevalence rate of sarcopenia in elderly patients with gastrointestinal tumors was 28.3% (34/120) and it was 28.9% (22/76) and 27.3% (12/44) in males and females respectively. There were statistically significant differences in age, tumor stage, nutritional risk, body mass index, calf circumference, L3 SMI, grip strength/muscle strength, gait speed and SPPB score between patients with and without sarcopenia (P<0.05). Muscle strength/grip strength, L3 SMI and gait speed were closely correlated with the occurrence of sarcopenia in elderly patients with gastrointestinal tumors (P<0.05). The L3 SMI was positively correlated with physical performance as assessed with SPPB in both groups.Conclusions:Sarcopenia is a common complication in elderly patients with gastrointestinal tumors with multiple influencing factors. Timely nutritional intervention and exercise intervention should be incorporated into the treatment of elderly gastrointestinal tumor patients with sarcopenia in order to improve nutrition status.
10.Complement C3a receptor 1 high-expression progresses malignancy in glioma through nuclear factor-kappa B signaling pathway

Objective:To investigate the complement C3a receptor 1 (C3AR1) expression in glioma and its mechanism in progressing malignancy.Methods:(1) The C3AR1 mRNA expression data and clinical information were obtained in 607 glioma patients from The Cancer Genome Atlas (TCGA) database and 656 glioma patients from Chinese Glioma Genome Atlas (CGGA) database; the differences in C3AR1 mRNA expression were analyzed among gliomas with different World Health Organization (WHO) grading. The overall survival and disease-free survival were compared between high and low C3AR1 mRNA expression patients obtained from TCGA database by Gene expression profiling interactive analysis (GEPIA). Gene body (GO) function analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of C3AR1 related differentially expressed genes were performed by DAVID database. Correlation of C3AR1 mRNA expression with immune cell infiltration was analyzed using TIMER online website. (2) The brain tissues from 3 non-tumor patients and 9 glioma patients surgically resected in Department of Neurosurgery, First Affiliated Hospital of Xinxiang Medical University from January 2019 to September 2021 were collected; the C3AR1 protein expression was detected by Western blotting. (3) The in vitro cultured U87 and U251 cells were divided into negative control group and C3AR1 knockdown group ( C3AR1 being knocked down by lentivirus transfection); and CCK-8 assay, plate cloning assay and Transwell assay were used to detect the proliferation rate, number of colony formation and number of membrane penetrating cells. Western blotting was used to detect the nuclear factor-κB (NF-κB) signaling pathway protein expressions. Results:(1) In TCGA database, the C3AR1 mRNA expression in gliomas of WHO grading II, grading III and grading IV increased sequentially, with significant differences ( P<0.05). In CGGA database, the C3AR1 mRNA expression in glioma of WHO grading IV was statistically higher than that in gliomas of WHO grading II and grading III ( P<0.05). GEPIA showed that the overall survival and disease-free survival in the low C3AR1 mRNA expression group were statistically higher than those in the high C3AR1 mRNA expression group ( P<0.05). GO function analysis and KEGG pathway enrichment analysis revealed that C3AR1 related differentially expressed genes were more enriched in such biological processes and signaling pathways as calcium homeostasis, membrane structural valves, proton transmembrane transporter protein activity, chemokine signaling pathway and NF-κB signaling pathway. TIMER showed that C3AR1 mRNA expression in glioblastoma and low-grade glioma was positively correlated with infiltration degrees of B cells, CD4 + T cells, neutrophils, macrophages and dendritic cells, and C3AR1 mRNA expression in glioblastoma was negatively correlated with infiltration degree of CD8 + T cells ( P<0.05). (2) C3AR1 protein expression in glioma tissues was significantly higher than that in non-tumor tissues. (3) Compared with the negative control group, the C3AR1 knockdown group group had significantly lower proliferation rate, smaller numbers of colony formation and membrane penetrating cells, and lower expressions of NF-κB, phosphorylated (p)-NF-κB, p-NF-κB inhibitory protein (IκB)α, p-I-κB kinase (IKK)α and N-cadherin, and significantly higher E-cadherin expression. Conclusion:C3AR1 is highly expressed in glioma and progresses malignancy through NF-κB signaling pathway.
