1.The establishment of chronic rejection model of rat's aortic transplantation

Objective To establish an aortic transplantation model in the rat. Methods A thoracic aorta to abdominal aorta interposition transplant model in the rat was established. The male Lewis rats were used as donors and the female Brown Norway(BN) rats were used as recipients.Intercepted the thoracic aorta of lewis rats about one and a half centimeter. Transplanted it on the abdominal aorta of BN rats. The activity and weight change of BN rats were observed everyday until 1 week after transplantation. Then observed these index one time a week. The transplanted aortas were harvested at 8 weeks after transplantation. The intimal thickening were observed by pathological. The expression of CD3,CD68 and α-actin on transplanted aorta were observed by immunohistochemistry. Resuts All operations were finished within 2 hours. No recipients died during operation. All recipients could survival more than 8 weeks. The symptoms of bowing back, hair shedding and weight loss was appeared after transplantation. The weight of the rats were upturn at 4 or 5 days after transplantation. About 1 week after transplantation, all situations of the rats were recovery to the lever before transplantation. 8 weeks after transplantation the intimal thickening of the transplanted aortas were observed obviously by pathological. And the expressions of CD3, CD68 and α-actin on aortic allograft were observed. Conclusion The intimal thickening and the expressions of CD3, CD68 and α-actin on aortic allograft were typical expressions of chronic rejection. So this aortic transplantation model is feasible and reliable for studying chronic rejection.
2.Effect of mesenchymal stem cell in transplantation

Mesenchymal stem cell(MSC) is a kind of tissue stem cell having a characteristics of high self-renewal and multipotency. It is tended to be separated in vitro and play an important role in hematopoietic stem cell transplantation and transplantation of other systems. This article described mesenchymal stem cell's function and mechanism on supporting hematopoiesis, enhancing implantation,adjusting immunity and alleviating graft versus host disease (GVHD) in transplantation and also give some advices and experties about its further application.
3.Efficacy Observation of ~(131)I in the Treatment of Hyperthyroidism

OBJECTIVE:To explore the clinical efficacy of 131I in the treatment of hyperthyroidism.METHODS:131I therapy for 784 cases of hyperthyroidism was analyzed retrospectively.The dose of 131I(?Ci) was calculated with formula:?Ci=the weight of thyroid(g)?80-120(?Ci?g-1)/the highest absorbed iodine rate(%).Clinical efficacy was evaluated according to the results.RESULTS:394 cases were relieved completely after once therapy,523 cases after twice therapies and 547 cases after three times of therapies.577 patients were visited at random and 94.80% of them were cured.Early-stage toxic reaction had been found in 46 cases with incidence of 7.97%.Hyperthyroidism crisis had not been developed.Temporary hypothyroidism had been found in 68 cases with incidence of 11.79%,while permanent hypothyroidism had been found in 146 cases with incidence of 25.30%.CONCLUSION:131I has an obvious effect on hyperthyroidism with one disadvantage of high incidence of hyperthyroidism.However,131I is an effective therapy which is safe,economical,simple and convenient.
4.Pilot study of acoustic radiation force impulse imaging for clinical pathological typing of advanced gastric cancer

ObjectiveTo explore the value of virtual touch tissue quantification (VTQ) in the evaluation of clinical pathological typing of advanced gastric cancer.MethodsFifty six patients who had been diagnosed as gastric cancer were examined using acoustic radiation force impulse.According to clinical pathological typing,all cases were divided into highly-moderately differentiated adenocarcinoma (14 cases) and non-highly-moderately differentiated adenocarcinoma (42 cases).A comparison with clinical pathologic results was made after surgery.The correlation of VTQ results and clinical pathological typing of gastric cancer was analyzed.ResultsThe VTQ value of highly-moderately differentiated adenocarcinoma was lower than that of non-highly-moderately differentiated adenocarcinoma [(1.49 ± 0.44) m/s vs (2.12 ± 0.45) m/s],with statistical significance( t =-4.53,P <0.05).According to the maximum area under the ROC curve,the cutoff value of VTQ was 1.795 m/s,the sensitivity and specificity were 78% and 86%,respectively,the Youden's index was 0.64,and with high reproducibility(Kappa =0.81).ConclusionsVTQ could initial estimate the clinical pathological typing of advanced gastric cancer before operation.
5.Evaluation of left ventricular movement in patients with type B Wolff-Parkinson-White syndrome

Objective To investigate the left ventricular (LV) torsion parameters of type B WolffParkinson-White syndrome (B-WPW) and to explore alternation of the left ventricular motion pattern in patients with B-WPW.Methods Thirty-eight patients with B-WPW were studied.And 40 volunteers were selected as control.Two-dimensional speckle tracking imaging (2D-STI) was used to acquire left ventricular torsion parameters,including peak value and time to peak value of LV twist,LV apex rotation,and LV base rotation.Apical-basal rotation delay (RDA-B) was calculated.Biplane Simpson method was used to measure LV end-diastolic volume (LVEDV),end-systolic volume (LVESV) and ejection fraction (LVEF).All above parameters were measured in patients with B-WPW before (B-WPW-B group) and after (B-WPW-A group) radiofrequency catheter ablation and in control group.The QRS width of lead Ⅱ was recorded,and the relationships between all above parameters with the RDA-B or QRS width were analyzed.Results Compared with the contrd group,in patients with B-WPW,the peak value of LV apex rotation (RotA),LV base rotation (RotB) and LV twist (TwistLV) were lower(LV base was more obvious than LV apex)(P <0.05),the time to RotB (TTPB) was shorter (P =0.004),RDAB was longer(P =0.002),the left ventricular enlarged and the QRS prolonged (P =0.000).After radiofrequency catheter ablation,the left ventricular torsion parameters in patients with B-WPW recovered significantly(P <0.05),but still lower than those in the control group(P <0.05).Conclusions The left ventricular motion pattern of patients with B-WPW alters,including the changes of peak value and time to peak value in LV apex rotation,LV base rotation and LV twist,as well as LV myocardium dyssynchrony.The motion pattern of LV in patients with B-WPW is improved in acute stage after radiofrequency catheter ablation but not yet recover totally.
6.B-type natriuretic peptide and risk of type 2 diabetes mellitus

Objective To explore the association of NH2-terminal pro-B-type natriuretic peptide ( NT-proBNP) with the risk of type 2 diabetes.Methods One hundred and twenty-six impaired glucose regulation( IGR) participants from Diabetic Identification Center of Tianjin Metabolic Diseases Hospital were included.NT-proBNP was measured in plasma samples collected from participants at baseline condition.Results At baseline, NT-proBNP was inversely associated with body mass index, waist circumference, fasting glucose, insulin and low-density lipoprotein-cholesterol( LDL-C) levels.During a follow-up of 2 years, 51 participants reported a new diagnosis of diabetes from OGTT.Baseline quartiles of NT-proBNP were inversely associated with diabetes risk, even after multivariable adjustment.Theadjustedrelativerisksfordiabeteswere1.0(reference),0.83(95%CI0.74-0.96),0.78(95%CI 0.68-0.90), 0.74 (95%CI 0.64-0.87) for the 1st, 2nd, 3rd, and 4th quartiles of baseline NT-proBNP, respectively ( P<0.01 ) .Conclus ion In IGRpopulation , lowlevels of NT-proBNP were associated with a significantly increased risk of type 2 diabetes.
7.Propagating and educating (planning, doing, checking, and acting) circulation combined with neuronavigation system in improving perioperative compliance of pituitary tumor patients

Objective To explore the effect of propagating and educating (planning, doing, checking, and acting) circulation combined with neuronavigation system on improving surgical coordination at perioperative nursing in patients with pituitary tumors. Methods One hundred and sixteen patients with pituitary tumors, admitted to and accepted neuronavigation assisted microsurgery in our hospitals from August 2016 to August 2018, were chosen in our study; these patients were randomly divided into experimental group and control group (n=58). Patients from the control group accepted traditional perioperative propagating and educating circulation, while patients from the experimental group accepted perioperative propagating and educating (planning, doing, checking, and acting [PDCA]) circulation. Awareness rate of surgical preparation knowledge, self-rating anxiety scale (SAS) scores, degrees of operation coordination, length of hospital stays, hospitalization cost and postoperative complications were compared between the two groups. Results As compared with patients in the control group, patients in the experimental group had significantly higher knowledge of surgical preparation, statistically lower SAS scores and statistically lower proportion of patients with poor surgical coordination (P<0.05). As compared with those in the control group, the hospitalization cost, length of hospital stays and incidence of surgical complications (5.1% vs. 12.1%) in the experimental group were statistically decreased (P<0.05). Conclusions PDCA circulation combined with neuronavigation system can be used as a kind of brand-new education mode for patients with pituitary adenomas. As compared with traditional perioperative mode, the new mode can significantly improve the awareness rate of surgical preparation knowledge, relieve anxiety level, improve cooperation degree, and reduce the postoperative complications, length of hospital stays, and hospital expenses, which means great clinical significance.
8.The study of osteogenous differentiation of MSCs transfected with different gene.

This experiment is sought to study the contribution of different gene in osteogenous differentiation of bone marrow stromal cells (MSCs) and optimize the seed cells of bone tissue engineering. Firstly, we obtained the full length gene of BMP-2, VEGF165 and bFGF by RT-PCR, and cloned into the expression vector pcDNA3. 0. After to be transfected, the MSCs cell lines which could express each of the target protein were selected out by G418. RT-PCR and immunohistochemistry were used to confirm the exogenous gene expression in MSCs. MTT showed that almost all of the MSCs which have been transfected with exogenous gene had more strong proliferative potential than the untransfected group did. There was a notable increase of ALP activity in transfected cell compared with the control group. The concentration of OCN in cell culture medium had a increase in some degree except of VEGF group. The outstanding osteogenous differentiation could be observed in BMP-2 and bFGF transfected MSCs. The results showed that the MSCs modified by BMP-2 and bFGF would be more effective in bone tissue engineering field.

9.Growth and osteogenesis characteristics of cultured canine mesenchymal stem cells under osteogenic induction.

To investigate the growth and osteogenesis characteristics of cultured canine mesenchymal stem cells (cMSCs) under osteogenic induction. We found the cMSCs were isolated from adult canine using density gradient separation method. The cMSCs attachment formed soon after seeding and grew into colonies with the appearance of fibroblastic cells. The osteogenic induction compound of Dexamethasone (Dex), beta-sodium glycerphosphate (beta-GP), ascorbic acid (AA) was added to passaged cMSCs and the proliferation and osteogenic differentiation of them was studied. The morphology of cells was observed by light micrograph and transmission electron microscope. The proliferation and growth characteristics of cMSCs were observed during primary and passage cultures through MTT. The differentiation were assayed by alkaline phophatase (ALP) and osteocalcin (OCN). We found the cMSCs have an active proliferative ability in primary and passage culture, and cMSCs under osteogenic induction have the typical characteristic of a secretory cell; the osteogenic induction compound may induce cMSCs to differentiate to osteoblasts. There are higher expression of ALP and OCN in passage 3 cMSCs under osteogenic induction than that of the osteoblasts osteogenic induction condition. Our research suggest the cMSCs in our culture system are mainly undifferentiated osteoprogenitors and can differentiate to osteoblast under osteogenic induction.

10.Research of osteoblast-induced rat mesenchymal stem cells cocultured with beta-TCP/PLLA composite of different ratio.

There are three key factors in tissue engineering: seeding cells, scaffold and their interaction. Although mesenchymal stem cells (MSCs) are potential seeding cells, the problem of what phase MSCs should be used is not yet solved. On the other hand, degradable porous scaffolds which have good mechanics and good biocompatibility are preferred. To choose the optimum seeding cells and the suitable ratio of beta-TCP/PLLA porous scaffold, we observed the phenotype of the male SD rat's osteoblastic MSCs and detected the amount of alkaline phosphatase, osteocalcin and type I collagen secreted by the osteoblastic rMSCs in different phase. About 10, 14 and 20 days after induction, the induced cells came into proliferative phase, matrix synthesis phase and mineralization phase, respectively. Then we chose the suitable cells and seeded them on beta-TCP/PLLA composite scaffolds with different ratios (beta-TCP/PLLA = 1:1; beta-TCP/PLLA = 1:2; and beta-TCP/PLLA = 2:1). Fluorescence microscope, scanning electron microscope and MTT assay were used to observe and to detect the biocompatibility of the scaffolds. The results indicated that all of these materials have biocompatibility to some extent. Cells can grow well on all of the scaffolds. However, scaffold beta-TCP/PLLA = 2:1 seems to be a more suitable tissue engineering scaffold on account of its minimal influence on cell growth and differentiation.
