1.Dynamic observation on splenocytes subsets in mice immunized with the transgenic alfalfa vaccine containing Eg95-EgA31 fusion gene of Echinococcus granulosus

To observe changes on subsets of splenocytes in mice immunized with the transgenic alfalfa (Medicago sativa) vaccine containing Eg95-EgA31 fusion gene of Echinococcus granulosus dynamically,the leaf protein was extracted from the transgenic alfalfa by heat-coagulation method and prepared for a solution with a concentration of 20 g/ L.The 132 BALB/c mice were divided into 3 groups randomly and immunized intranasally or orally with the leaf protein solution once per 3 days for 2 months.At the same time,the group of intranasal immunization with leaf protein from the non-transgenic alfalfa was set as control group.4 mice randomized from each group were killed to get the spleens at Week 0,2,4,6,8,10,12,14,16,18 and 20 after last immunization,and the splenocytes were separated to measure CD_4~+and CD_8~+T cell subsets by FCM.In the oral group,CD_4~+subset increased significantly from Week 6 to Week 10 after the last immunization and reached the peak at Week 6.While CD+ 8 subset increased obviously from Week 4 to Week 12 and reached the highest level at Week 8.In the intranasal group,the significant increase of the CD_4~+subset was observed from Week 4 to Week 6 and also reached the peak at Week 6.The similar trend of CD_8~+ subset was observed from Week 4 to Week 10 and reached the highest level at Week 8.It was suggested that CD+ 4and CD+ 8subsets played an important role in the protection induced in mice immunized with the transgenic alfalfa vaccine.
2.Correlation between epidermal growth factor receptor gene mutations and CT signs in lung adenocarcinoma

Objective To explore the correlation between epidermal growth factor receptor (EGFR) gene mutations and computed tomography (CT) characteristics in lung adenocarcinoma.Methods Chest CT scan results of 200 postoperative lung adenocarcinoma patients were retrospectively studied,the EGFR gene mutations detection results were statistically analyzed,and the correlation between EGFR gene mutations status and CT characteristics was investigated.Results Single factor analysis results showed that non smoking,female patients,air bronchogram sign and small tumor diameter were significantly associated with EGFR gene mutations (P<0.05),and the incidence of GGO in the EGFR gene mutation group was higher than that in the wild group,but no statistical significance was found (P>0.05),while other clinical and CT signs,such as age,lobulation,spicule,calcification,cavity and pleural indentation were not associated with EGFR gene mutations (P>0.05).Logistics regression analysis results showed that non smoking and air bronchogram sign were significantly associated with EGFR gene mutations (P<0.05),but gender and tumor size were not associated with EGFR gene mutations(P>0.05).Conclusions Non smoking and air bronchogram sign may be used as predictive factors for EGFR gene mutations in lung adenocarcinoma.
3.Features of multislice spiral computed tomography in micropapil-lary-predominant lung adenocarcinomas

Objective:To examine the features of multislice spiral computed tomography (MSCT) in micropapillary-predominant lung adenocarcinomas to improve the understanding of this type of lung cancer. Methods:The MSCT features of 18 cases with micro-papillary-predominant lung adenocarcinoma (micropapillary component>50%) confirmed by histopathology were analyzed retrospec-tively. Results:Among the 18 cases of lung cancer, 1 was diffuse, 3 were central, and 14 were peripheral lung cancer (PLC). The size of the adenocarcinomas in the 14 PLC cases ranged from 1.3 cm to 8.5 cm, with an average of 3.56 cm, including the size of 8 cases greater than or equal to 3 cm. Among the 18 cases, 13 were lobulated, 9 showed spicule signs, 7 showed pleural indentation signs, 5 had pleural adhesions, 1 had bronchial truncation (i.e., cut-off sign), and 4 were surrounded by obstructive inflammation. In addition, calcifi-cation was observed in one case, uneven density in two large lesions, air bronchus sign in four, and solid and ground-glass mixed densi-ty in two. Among the total number of cases, a variety of the measurable enhanced CT values (ΔCT) of lesions were found in 16, ranging from 13 HU to 80 HU, with an average of 47.5 HU, of which 15 were cases ofΔCT≥15 HU and 15 were cases ofΔCT≥20 HU. Pleu-ral thickening was observed in two cases with pleural effusion, and pleural metastasis in one case was confirmed by histopathology. One case with pleural effusion suffering pleural metastasis was confirmed. Ground-glass density nodules in both lungs were observed in one case, with a few bilateral pleural and pericardial effusions. Eight cases had mediastinal or hilar enlarged lymph nodes with uneven density enhancement, and lymph node metastasis was pathologically confirmed in six cases. Lymph node metastasis was found in four cases, but no apparent enlargement of lymph nodes in MSCT was observed. Conclusion:Micropapillary-predominant lung adenocarci-nomas were common in non-smoking elderly female patients, whose lung cancer cases were mostly PLC. The typical features of PLC include lobulation, spicule, and pleural indentation signs. Solid density ranked first in the PLC cases, with evident enhancement and high rate of lymph node metastasis.
4.Design of Multipoint Temperature and Humidity Monitoring Software Based on VC.NET and MySQL

Objective To develop a monitoring software running on the host computer for the multipoint temperature and humidity monitoring system.Methods The software was developed in VC.NET.Its database was built based on MySQL.The software contains real-time part and history part.Real-time part completes the functions of communication,sample,store and display.History part supplies query and exportation of the historical data.Results The monitoring software stores data completely,and displays the real-time and historical data by curve and number.Besides,it supports 24 points monitoring.Conclusion The software has a lot of virtues such as friendly interface,easy operation,stable running without fault and high cost performance.

Objective To investigate the role of interleukin-1? (IL-1?) in regulating the gene expression of matrix metalloproteinase-13 (MMP-13) in the human hyaline chondrocyte. Objective The image segmentation of low-density gene chips based on genetic algorithm, which can implement the function of region identification, is achieved. Methods After image denoising by wavelet analysis, image segmentation is accomplished by genetic algorithm. Results The method can detect the region of sampling point more precisely. It can also effectively separate the valuable weak signal points and background or noise. 7.The effects of mesenchymal stem cells on the aging kidneys in rats

Objectives To study the effect of mesenchymal stem cells on the aging rat kidney and to explore the underlying mechanism. Methods Rat models of senile kidney were built with hypodermic injection of D-galactose daily. Injections of MSCs of 3 × 10<'6>were given to each rat through vena caudalis and CFSE was used as a tracing label to detect the distribution of MSCs in vivo. After 24 h, rats were dissected and their kidneys were frozen for section. MSCs were observed with Fluophot and quantitative analysis of the various parameters of kidney was performed under a light microscope with B12000 image analysis system.The contents of superoxide dismutase (SOD) and malondialdehyde (MDA) in serum and kidney were measured wtih hydroxylamine and chromatometry. The expression of VEGF and P16 mRNA in kidney tissue was detected with real-time PCR and Western blotting. Results MSCs was found homing in the rat kidney,and the glomerular size, sklerosis-rate and the average cell count of glomerulus in the treated group were different from those of the model group (P<0.05). In the treated group, the activity of SOD was significantly higher and the content of MDA was lower in serum and kidney than that in the model control group (P<0.05). The expression of VEGF mRNA and protein in the kidneys of MSCs group increased significantly as compared with the model group (P<0.05). The expression of P16 mRNA and protein in the kidney of MSCs group decreased significantly compared with the model group (P<0.05). Conclusion MSCs can increase the expression of VEGF while decrease the expression of P16, so as to play a key role in the anti-aging on rat kidney.
8.Relationship of serum C reactive protein level with serum leptin concentration in type 2 diabetic patients

Objective To explore the association of serum high-sensitivity C reactive protein (hs-CRP) level with leptin in patients with type 2 diabetes mellitns (T2DM). Methods 35 cases of T2DM patients were involved in the study and 16 normal subjects were taken as controls. Fasting serum leptin and CRP were measured .The relationship between them were also analyzed. Results Fasting serum hs-CRP concentration in T2DM group was higher than that in healthy control group, but serum ieptin levels is the same (P > 0.05). Partial correlation analysis demonstrated that fasting hs-CRP level was positively correlated with leptin (r =0.512 ,P =0. 018). Conclusion Fasting serum hs-CRP level is significantly raised in T2DM ,which is associated with leptin. There is chronic inflammatory reaction in T2DM patients,and leptin is possibly involved in inflammatory pathogenesis of T2DM.

10.Study on fingerprint and in vitro release of Xiaozheng-Zhitong cataplasm

Objective:To establish a method of evaluating the reproducibility of the preparation process of Xiaozheng-Zhitong cataplasm. Methods:Used fingerprint spectrum technology and in vitro release test to evaluate the reproducibility preparation process of Xiaozheng-Zhitong cataplasm. By using HPLC to establish the fingerprint of Xiaozheng-Zhitong cataplasm. The chromatographic column was an Agilent HC-C18 column (4.6 mm×250 mm, 5 μm); the detection wavelength was 280 nm; the flow rate was 1 ml/min; the column temperature was 25 ℃. The mobile phase was acetonitrile-0.2% phosphoric acid aqueous solution, used gradient elution method. An HPLC method for the determination of tetrahydropalmatine content was established, and the vertical Franz diffusion cell method was used to investigate its in vitro release. Results:The results showed that the similarity between the 10 batches of Xiaozheng-Zhitong cataplasm and the control map was above 0.95, indicating that the process of Xiaozheng-Zhitong cataplasm was stable and feasible; the cumulative release amount of tetrahydropalmatine was 0.23 mg in 24 hours, and the cumulative release rate in 8 hours was 91.54%, and the RSD of 6 samples at each time point was <3%. Conclusion:The method of evaluating the reproducibility of the preparation process of Xiaozheng-Zhitong cataplasm was established by using fingerprint technology and in vitro release; it can provide a reference and scientific basis for the evaluation of the reproducibility of the preparation process of the plaster.
