2.Mediating effect of inferiority feelings on relationship between childhood trauma and fear of intimacy in college students

Objective:To explore the mediating effect of inferiority feelings on the relationship between child-hood trauma and fear of intimacy in college students.Methods:Totally 420 college students (72 males and 324 fe-males,aged 18 to 24)from Langfang Normal School were assessed with the Feelings of Inadequacy Scale(FIS), Childhood Trauma Questionnaire(CTQ)and Fear-of-Intimacy Scale(FIS).The regression analysis and Bootstrap were used to test the mediating effect of inferiority feelings on the relationship between childhood trauma and fear of intimacy.Results:The total scores of CTQ and the scores of 4 childhood trauma factors were positively correla-ted with fear of intimacy score and feeling of inferiority score(r =0.18 -0.55,Ps <0.05).Feeling of inferiority scores were positively correlated with fear of intimacy scores (r =0.24 -0.44,Ps <0.05).The childhood trauma was positively associated with fear of intimacy (β=0.31,P <0.01).In addition,feeling of inferiority fully media-ted the relationship between childhood trauma and fear of intimacy,the value of mediating effect was 34%.Conclu-sion:Feeling of inferiority may play a mediating role between childhood trauma and fear of intimacy in college students.
3.Neural mechanism of salient target processing in college students

ObjectiveTo investigate the neural mechanism of processing the salient target during the visual search task with spatiotemporal patterns of event-related potential(ERP).MethodsSixteen subjects were asked to judge the existence of a low or high salient target from 4 items,in which one distractor might be also salient or not.A two-way repeated-measure ANOVA was carried out for behavioral performance and ERP data( distractor saliency:YES,NO)*( target:none,low salient,high salient).ResultsSignificant interaction was found between target and distractor for the reaction time (F( 1.99,29.79) =21.56,P=0.00).For the low salient target,significant difference of reaction time was found between the YES (552.57 ± 82.32 ms) and NO (540.47 ± 75.68 ms)of distractor saliency( t (15) =- 2.489,P =0.025 ).However,such difference of reaction time was not significant for the high salient target.The statistical parametric mapping of F-value of ERP suggested the effect of the distractor saliency revealed in the right fronto-parietal-temporal areas related to stimulus novelty and the effect of the target saliency appeared in the parietal-occipital regions related to visual processing during the same stage ( 350 ~ 450ms ).The significant interaction occurred in the left temporal regions (450 ~ 600 ms ) related to stimulus familiarity and its classification.ConclusionDue to both mechanisms of the parallel processing for the target and the distractor during the perceptual stage and the effective classification of the familiar network within left hemisphere during the stage of pattern classification,the salient target can be processed quickly in spite of the interference from the salient distractor.
4.Molecular cloning and alternative splicing analysis of hepatoma associated gene HTA

Objective:To obtain the full length cDNA sequences of hepatoma associated gene HTA, analyze its alternative splicing, detect the expression pattern of 2 HTA gene transcripts in different hepatic cell lines, and to establish a base for further study of HTA gene function in hepatocellular carcinoma (HCC) occurence and development. Methods:The full length cDNA of HTA gene was cloned by rapid amplification of cDNA 3' ends (3'-RACE), rapid ampliifcation of cDNA 5' ends (5'-RACE) and DNA sequencing. The gene structure and alternative splicing were analysed. Northern blot assay was performed to detect the expression pattern of 2 HTA gene transcripts in different hepatic cell lines. Results:The full length of HTA gene was 1414 bp, composed of 3 exons and 2 introns, and the second intron could be retained in mRNA. Northern blot assay showed that 2 transcripts of HTA mRNA(1.4 kb and 1.7 kb) could express in the HCC cell lines HepG2 and QGY-7703, but not in the non-malignant cell line L-02 and HUVEC. The expression level of 1.4 kb transcript was much higher than 1.7 kb one. Conclusion:This study successfully has obtained the full length cDNA of HTA gene, and analysed the gene sequence and alternative splicing, 2 transcripts of HTA mRNA specifically expressed in HCC cell lines. As a hepatoma associated gene, HTA deserves further investigation.
5.The Plasma Vitamin A and Subsets of T Lymphocyte in Lung Cancer Patients

Objective: Our aims were to evaluate the plasma vitamin A status and the function of T-cell dependent immunity in lung cancer patients, so as to explore the correlation between them. Methods:We measured the plasma vitamin A status with HPLC and checked the subsets of T lymphocyte with indirect immunofluorescence technique. Results: The levels of plasma vitamin A in lung cancer patients and in controls were(0.406±0.111)mg/L and(0.548±0.149)mg/L respectively . There was a significant difference between them(P<0.001) . The levels of CD3, CD4,and CD4/CD8 were lower in lung cancer patients than those in controls, while the level of CD8 was higher. The plasma vitamin A had positive correlation with CD3 , CD4,and negative with CD8. Conclusion: There is defect of the plasma vitamin A and low level of the function of T cell dependent immunity in lung cancer patients,which shows the obvious correlation between them. This research supplies the basis of clinical therapy to lung cancer with vitamin A.
6.Clinical research on the relationship between breast cancer and thyroid disorder

Objective To investigate the incidence of autoimmune and nonautoimmune thyroid diseases in patients with breast cancer.Methods Clinical and ultrasound evaluation of thyroid gland,detection of serum thyroid hormone and related antibodies,and fine-needle aspiration of thyroid gland were performed in 100 breast cancer patients and 100 control individuals during the period from 2004 to 2008.Results The mean values of anti-thyroid peroxidase antibodies were significantly higher in breast cancer patients than that in control individuals [(104.56±21.54) U/ml vs (22.16±4.65) U/ml,(P=0.030)].The incidence rates of autoimmune and nonautoimmune thyroid diseases were higher in breast cancer patients than that in control individuals[38 % (38/100) vs 17 % (17/100),P=0.0009,26 % (26/100) vs 9 % (9/100),P =0.0016,respectively].Conclusion The results indicate an increased incidence of autoimmune and nonautoimmune thyroid diseases in breast cancer patients.
7.Clinical study of ocular damnification after intensity-modulated radiotherapy in patients with nasopharyngeal carcinoma

Objective To study the effect on normal ocular tissues after intensity-modulated radiotherapy (IMRT) in the patients with nasopharyngeal carcinoma.Methods Nineteen nasopharyngeal carcinoma patients confirmed by pathology were enrolled (38 eyes).All Patients tnderwent visual acuity,slit lamp,fundus,visual evoked potential (VEP) and eleetroretinograms (ERG) examination before IMRT,at the end of IMRT,6 months and 12 months after IMRT.Results ① There was no statistically difference in uncorrected and corrected visual acuity at the various examination time points before and after radiotherapy ( P > 0.05 ).② The average latency of VEP P100 before IMRT,at the end of IMRT,at 6 months after IMRT and 12 months after IMRT was ( 99.684 ± 2.484) μV,( 99.947 ± 2.277 ) μV,( 104.000 ± 3.952 ) μV and ( 101.316 ±2.462) μV respectively.The average latency of VEP P100 was significantly prolonged at 6 months after IMRT compared with the time points of before IMRT,the end of IMRT and 12 months after IMRT ( P < 0.05 ).③ The average latency of ERG b wave before IMRT,at the end of IMRT,at 6 months after IMRT and 12months after IMRT was (44.974 ±3.774) ms,(44.816 ±3.368) ms,(43.184 ±2.837) ms and (44.000 ±3.154) ms respectively. The average amplitude of ERG b wave before IMRT,at the end of IMRT,at 6 months after IMRT and 12 months after IMRT was (421.237 ±27.353) μV,(414.763 ±26.188) μY,(419.026 ±24.876) μV,and (419.974 ± 25.894) μV respectively.No statistically difference was found in average latency and amplitude of ERG b wave at the various examination time points before and after radiotherapy ( P >0.(05).④ The average amplitude of Op2 wave before IMRT,at the end of IMRT,at 6 months after IMRT and 12 months after IMRT was (63.184 ± 6.028 ) μV,(48.605 ± 6.872 ) μV,( 50.421 ± 6.769 ) μV and (53.026 ±6.074) μV respectively.At the various time points after IMRT,the average amplitude of Op2 wave was significandy lower than before IMRT (P < 0.05 ).Conclusion IMRT can significantly reduce the incidence of ocular complications.There is a good clinical application value of ERG and VEP for evaluating retina and optical nerve functional changes induced by IMRT.

9.Systemic lupus erythmatosus and panniculitis presenting as multiple ulcers:one case report

SUMMARY A 36-year-old woman had an 8-year history of systemic lupus erythematosus (SLE) and was being treated with 10 mg/d of prednisone.She presented with a 6-month history of intermittent fever and multiple painful multi skin erythematous macules in her button, hips and extremities that had slowly en-larged to 8 cm ×4 cm in diameter.The lesions started as painful erythematous macules, which eventually ulcerated and scared.Laboratory tests showed leukopenia, protenuria, positive anti-double strand DNA and hypocomplementemia.Cultures of the bottom ulcer were E.Coli, fugus and Tuberculous mycobacteria were both negative.Biopsy was performed and revealed necrosis of epidermis, thrombus and cellulose de-generation in epidermis with neutrophils karyorrhexis and vasculitis.Her SLE was active, so she was pre-scribed antibiotics for 2 weeks and prednisone was added to 60 mg/d for a month.However her skin ul-cers did not relieve.When prednisone was added to 120 mg/d with combination therapy of cyclophospha-mide and hydroxychloroquine, her skin ulcer cicatrized gradually.
10.Multi-step purifications of botulinum neurotoxin A light chain and identification of its metalloproteases activity

Objective To obtain highly purified botulinum neurotoxin A light chain(BoNT-ALC) protein in E.coli by genetic engineering and multi-step purifications, and identify its metalloproteases activity.Methods The full-length of BoNT-ALC was cloned from BoNT A by PCR and inserted into plasmid pET-22b.Then pET-22b-ALC was transformed into E.coli BL21( DE3) strains and induced by IPTG.The protein was purified by Ni-NTA sepharose,anion exchange column and gel filtration.The enzymatic activity of the protein was identified by SNAP-25.Results and Conclusion A highly purified and homogeneous protein is obtained, which shows good enzymatic activity.
